RESUMEN

Aggregation-induced emission (AIE) and aggregation-caused quenching (ACQ) are two classes of opposite luminescence phenomena. It is almost impossible to show both AIE and ACQ effect simultaneously by the same molecule. However, here we report that a simple TPE derivative TAP-TPE grows into both AIE crystals and ACQ ones. It is found that equatorial, contact distance-longer and weak ArH-π interactions exist in AIE crystals while vertical, contact distance-shorter and strong ArH-π interactions appear in ACQ crystals. Theoretical calculation of electron density on the interaction atoms unveils that ACQ crystals have much larger change in electron density than AIE ones, suggesting that the intermolecular electron transfer aroused by the strong ArH-π hydrogen bonds leads to ACQ phenomenon. This result provides a new insight into the emission mechanism in aggregation state. Interestingly, due to the ArH-pi interactions difference, only one of five kinds of crystals shows rapid photochromism, and can act as multimode anti-counterfeiting materials. Very exceptionally, for the first time we find that the photochromic micrometric rod-like crystal even makes forward rolling movement as it repeatedly bends and straightens by responding to on and off of the ultraviolet light irradiation, displaying potential for photo-actuator and light-driven micro-vehicle.

Artificial systems for sequential chirality transmission/amplification and energy relay are perpetual topics that entail learning from nature. However, engineering chiral light-harvesting supramolecular systems remains a challenge. Here, we developed new chiral light-harvesting systems with a sequential Förster resonance energy transfer process where a designed blue-violet-emitting BINOL (1,1'-Bi-2-naphthol) compound, BINOL-di-octadecylamide (BDA), functions as an initiator of chirality and light absorbance, a new green-emitting hexagonal tetraphenylethene-based macrocycle (TPEM) with aggregation-induced emission serves as a conveyor, and Nile red (NiR) or/and a near-infrared dye, tetraphenylethene (TPE)-based benzoselenodiazole (TPESe), are the terminal acceptors. Benefiting from the close contact and large optical overlap between donors and acceptors at each level, triad and tetrad relaying systems sequentially and efficiently furnish chirality transmission/amplification and energy transfer along the cascaded line BDA-TPEM-NiR (or/and TPESe), leading to bright customized-color circularly polarized luminescence (CPL) and bright white-light-emitting CPL (CIE coordinates: 0.33, 0.34) with an amplified dissymmetry factor (glum) of 3.5 × 10-2 over a wide wavelength range. This work provides a new direction for the construction of chiral light-harvesting systems for a broad range of applications in chiroptical physics and chemistry.

BACKGROUND: Studies on average-risk individuals undergoing gastroscopy screening in China are scarce. OBJECTIVE: To determine and compare the prevalence of lesions found by gastroscopy and the association between sex, age, Helicobacter pylori infection and gastric premalignant lesions. METHODS: Gastroscopy results were analysed for 60,519 individuals enrolled from January 2013 to December 2019. RESULTS: The median age was 49.84 years (SD, 9.47 years) for women and 48.90 years (SD, 8.82 years) for men, and the ratio of females to males was 35.10% (n = 21,240) to 64.90% (n = 39,279). The most common lesions detected by endoscopy were chronic gastritis, reflux oesophagitis, duodenitis and gastric polyps, detected in 24.48%, 10.28%, 3.96% and 3.61%, respectively. Oesophageal cancer and gastric cancer were detected in 0.33% and 0.47% of patients, respectively. The prevalence of chronic gastritis increased with age and was higher in males than in females (26.47% [n = 10396] versus 20.80% [n = 4417], p < .001). The prevalence of gastric ulcers was highest in the elderly group, and the H. pylori infection rate of gastric ulcer patients was 47.28%. The prevalence of gastric polyps was higher in females than in males (5.47% [n = 1161] versus 2.61% [n = 1024], p < .001), and the H. pylori infection rate in inflammatory polyp patients was higher than that in fundic gland polyp patients (28.32% [n = 442] versus 7.29% [n = 29], p < .001). CONCLUSION: The prevalence of upper gastrointestinal endoscopic lesions is high in the asymptomatic population undergoing physical examination and is associated with sex, age, and H. pylori infection.

BACKGROUND: The relationship between subclinical hypothyroidism (SCH) and blood lipid metabolism is controversial. This study is intended to evaluate the relationship between SCH and blood lipid profiles using well defined diagnostic criteria. METHODS: Data from 11,512 physical examinees in our hospital who had finished two tests with an interval of at least 3 months were analyzed, including 685 cases of SCH as stipulated in this study. In addition to common physical examination indicators, other parameters such as thyroid function indices and blood lipids were measured twice with an interval of at least 3 months. Subjects were diagnosed with SCH only when their levels of TT3, TT4, FT3 and FT4 were normal and TSH levels were increased on both tests, which meant these subjects were in a state of SCH for at least 3 months. The results of blood lipids for the second time were analyzed. RESULTS: Statistically significant differences were found in age, sex, BMI, hemoglobin, total cholesterol (TC), low density lipoprotein (LDL-C), high density lipoprotein (HDL-C), and BFP between the SCH and control groups (P < 0.001). However, there were no statistically significant differences in age, sex, blood pressure, blood lipids, blood glucose or BMI between patients with mild SCH and those with severe SCH (P > 0.05). After balancing the age and sex ratio, no factors were confirmed to be statistically significant independent factors of SCH. None of the parameters showed statistically significant differences between patients with mild SCH and those with severe SCH (P > 0.05). CONCLUSION: After defining rigorous criteria for the diagnosis of SCH, no definite association between SCH and TC, LDL-C or HDL-C was confirmed in this study. SCH may have no relationship to the most concerning blood lipid profile.

New hindered tetraphenylethylene (TPE) helicates with substitution at 2,6-position of phenyl rings were designed and synthesized. Due to the increased hindrance, the TPE helicates emit strong deep-blue to violet fluorescence both in the solid state and in solution, and could be resolved into enantiomers that emit strong and multicolor circularly polarized luminescence (CPL), and exhibit a high enantioselective recognition of chiral tartaric acid and its derivatives. Surprisingly, the derived helicate tetramines possess amino groups with an unpredented planar structure and sp2 -hybridized nitrogen, arousing the change between AIE effect and ACQ phenomenon through photoinduced electron transfer (PET). With advantages of short synthetic route, many modification positions, deep-blue to violet emission, wide CPL tuning, and high chiral recognition ability, the hindered TPE helicates show broad prospects as chiral materials.

BACKGROUND: ß-lapachone (ß-lap), the NQO1 bioactivatable drug, is thought to be a promising anticancer agent. However, the toxic side effects of ß-lap limit the drug use, highlighting the need for a thorough understanding of ß-lap's mechanism of action. ß-lap undergoes NQO1-dependent futile redox cycling, generating massive ROS and oxidative DNA lesions, leading to cell death. Thus, base excision repair (BER) pathway is an important resistance factor. XRCC1, a scaffolding component, plays a critical role in BER. METHODS: We knocked down XRCC1 expression by using pLVX-shXRCC1 in the MiaPaCa2 cells and BxPC3 cells and evaluated ß-lap-induced DNA lesions by γH2AX foci formation and alkaline comet assay. The cell death induced by XRCC1 knockdown + ß-lap treatment was analysed by relative survival, flow cytometry and Western blotting analysis. RESULTS: We found that knockdown of XRCC1 significantly increased ß-lap-induced DNA double-strand breaks, comet tail lengths and cell death in PDA cells. Furthermore, we observed combining XRCC1 knockdown with ß-lap treatment switched programmed necrosis with ß-lap monotherapy to caspase-dependent apoptosis. CONCLUSIONS: These results indicate that XRCC1 is involved in the repair of ß-lap-induced DNA damage, and XRCC1 loss amplifies sensitivity to ß-lap, suggesting targeting key components in BER pathways may have the potential to expand use and efficacy of ß-lap for gene-based therapy.

RESUMEN

The NUDT family is thought to play an important role in cancer growth and progression. However, the clinicopathologic significance and potential role of nucleotide diphosphate-linked X-component motif 21, NM_007006 (NUDT21) in pancreatic ductal adenocarcinoma (PDAC) remains largely unknown. In this study, we observed that NUDT21 was frequently up-expressed in PDAC. Clinical data revealed that its level positively correlated with poor survival of patients with PDAC. We found that knockdown of NUDT21 significantly inhibited cell proliferation and promoted apoptosis both in vitro and in vivo. Screening by microarray analysis and verifying by Western blot, we found that the EIF2 signaling pathway represented the main molecular mechanism underlying the effects of NUDT21 knockdown in PANC-1 cells, and PKR, HSPA5, EIF4E and DDIT3 may be its target genes. Thus, our results revealed for the first time that NUDT21, a valuable marker of PDAC prognosis, promotes tumor proliferation, inhibits cells apoptosis and might represent a potential target for gene-based therapy.

BACKGROUND: colon adenocarcinoma (COAD) is the most common malignant tumor of gastrointestinal tract. Our study attempts to explore the effect of miR-1307-3p on biological function of COAD cells and its connection with isthmin 1 (ISM1). METHODS: The miRNA dataset and clinical information of patients with COAD were downloaded from The Cancer Genome Atlas (TCGA) database. The survival prognosis was analyzed by GGSURV package from R. MicroRNA (miR)-1307-3p was identified by identifying overlapping miRNAs that target ISM1, across two databases (miRDB and Targetscan). Dual luciferase reporter assay was employed to scrutinize the relationship between miR-1307-3p and ISM1. RT-PCR was used to quantify miR-1307-3p and ISM1 expression of colon cancer tissues and cell lines. Western blot was performed to quantify related protein expression. Flow Cytometry, CCK8 and colony formation assays were performed to evaluate the apoptosis, cell cycle, cell viability and proliferation of COAD cells. RESULTS: miR-1307-3p mRNA level decreased in both COAD tissues and cell lines. Overexpression of miR-1307-3p suppressed the proliferation, promoted apoptosis and arrested cell cycle at G1 phase, meanwhile, downregulation of ISM1 accelerated the proliferation, inhibited apoptosis and promote cell cycleprogression. The result of dual luciferase reporter assay indicated that miR-1307-3p targeted ISM1 directly and inhibited its expression. The functions of miR-1307-3p regulating cleaved caspase-3, cyclinD1, Ki67 protein levels and activation of Wnt3a/ß-catenin signaling pathway were reversed by ISM1. CONCLUSIONS: miR-1307-3p inhibited activation of Wnt3a/ß-catenin signaling through targeting downregulation of ISM1, thereby inhibited proliferation and promote apoptosis of COAD cells.

Organic molecules with an aggregation-induced emission (AIE) effect have recently been attracting more and more attention due to their colossal potential in solid emitters and chemo/biosensors. The number and variety of AIEgen compounds are expanding very rapidly to obtain better application performance and a wider area of application. Among AIEgen systems, tetraphenylethylene (TPE) and its derivatives are the class that have received the most extensive study and the most rapid development because of their facile synthesis. Due to its C2 symmetry and at least tetratopic reaction positions, the TPE unit is also an ideal building block for constructing macrocycles and cages. The resultant cyclic TPE compounds have exhibited many exceptional performances that are difficult to access in their open chain counterparts, such as AIE enhancement, improvement in selectivity and sensitivity as sensors, emission tuning by guests, supramolecular catalysis, further disclosure of the AIE mechanism, molecular adsorption, storage and release, the propeller-like conformation exploitation of the TPE unit in chiral materials and so on. Recently, therefore, a large variety of studies about the synthesis, properties and application research of TPE macrocycles and cages have been reported. These TPE macrocycles and cages significantly expand the research area for the AIE phenomenon and its applications, and represent a development of the AIE area. However, up to now, no review of TPE macrocycles and cages has been available. Thus, this review serves as a summary of the designs, synthesis, photophysical properties, self-assembly, applications and prospects of TPE macrocycles and cages.

Tetraphenylethylene (TPE) foldamers with double hairpin-turn linkers showing an aggregation-induced emission (AIE) effect have been synthesized for the first time. A crystal structure of a foldamer-TNT complex has been obtained, enabling unprecedented direct observation of the interactions between TNT molecules and the chromophores of the foldamer. Instead of π-π stacking interactions, which have often been considered to be the key mechanism in the binding of TNT by chromophoric receptors, strong n-π interactions between the nitro groups of TNT and the aromatic rings of the foldamer have been found. Exceptionally, by addition of 1 % NaF to a suspension of the foldamer in H2 O/THF (95:5), the fluorescence quenching efficiency by TNT vapor significantly increased from about 20 % to more than 90 %. Even after diluting TNT-saturated air at 25 °C by a factor of 2×104 , an obvious quenching response was observed, indicating that ultratrace TNT vapor (down to 3.4 fg per mL of air) could be detected.

BACKGROUND: Smoking and drinking are two predisposing factors for dyslipidemia. Exercise has been proposed as a strategy to improve the blood lipids. However, it remains unclear how smoking and drinking jointly affect blood lipids and whether exercise influences their effects. OBJECTIVES: To evaluate the effects of smoking and drinking, either alone or in combination, on lipid-related indices in both exercise and non-exercise groups among Chinese men. METHODS: This study was conducted in a health examination center between 2015 and 2016. A sample of 6,179 male subjects was divided into exercise and non-exercise groups. Logistic and linear regression analyses were used to calculate the odds ratios for abnormal lipid-related indices and correlation coefficients between smoking/drinking and lipid-related indices. RESULTS: In the study population, the percentage of stable smokers and stable drinkers was 46.3% (2,860/6,179) and 77.6% (4,795/6,179), respectively. An increased smoking amount was significantly associated with an elevated triglyceride (TG) level and a decreased high-density lipoprotein cholesterol (HDL-C) level. Heavier smokers had higher odds ratios for high TG and low HDL-C. Heavier drinkers had higher levels of total cholesterol (TC), TG, and HDL-C and higher odds ratios for high TC and high TG but lower odds ratio for low HDL-C. The exercise group had lower TG levels and higher HDL-C levels than did the non-exercise group. CONCLUSIONS: Both heavier smoking and heavier drinking were associated with poorer TG levels, and the results suggest that drinking may be helpful for HDL-C. Exercise may relieve the negative effects of smoking and drinking.

Tetraphenylethylene (TPE) and its derivatives are the most typical and most widely studied organic compounds showing aggregation-induced emission (AIE). Due to their propeller-like structures, V-like clefts exist between the aryl rings, which make them promising host compounds. However, such a possibility is seldom explored. Herein, it is reported that TPE derivatives bearing two or four pyridine rings at the para positions of the phenyl rings (TPE-Pys) can selectively include triangular (Δ-like) m-phthalic acid from a mixture of o-, m-, and p-phthalic acids due to their shape complementary to form host-guest co-crystals, which showed redder emission than the TPE-Pys themselves. The emission of co-crystals 1-5 could be reversibly switched between yellow and red by alternating exposure to HCl and ammonia vapor. The host-guest co-crystals not only exhibited great potential for selectively recognizing and separating m-phthalic acid and as multicolor emission materials, but are also suitable for use as secret ink due to their reversible color change on varying the host-guest interactions.

The propeller-like conformation of tetraphenylethylene (TPE) with aggregation-induced emission (AIE) effect was partially and completely fixed by intramolecular cyclization for the first time. The immobilization of propeller-like conformation was found to show great advantages in determining the enantiomer purity, identifying the chiral amines. The completely fixed conformers are resolved into M- and P-enantiomer, which showed mirror imaged CD and almost quantitative fluorescence quantum yield. Furthermore, it also showed a mirror and large circularly polarized luminescence dissymmetric factor, depending on the helicity of the enantiomer. The result provides the most direct and persuasive evidence for AIE via the restriction of intramolecular rotation and finds the new insight of the compounds in chiroptical property.

A neutral chiral receptor based on TPE cyclohexylbisurea was synthesized and could discriminate the enantiomers of many different kinds of chiral reagents, including chiral acidic compounds, basic compounds, amino acids, and even neutral alcohols. The (1)H NMR spectra disclosed that the ability of chiral recognition could be ascribed to the multiple hydrogen bonds and CH-π interactions between the TPE urea receptor and the enantiomer of the chiral guest, which led to the selective aggregation of the receptor with one of the two enantiomers. This result exhibited a great potential in enantiomer discernment and high-throughput analysis of enantiomer composition of these chiral analytes by one chiral AIE molecule.

BACKGROUND: Ischemic stroke is the main cause of mortality and disability in older people worldwide. Recently epidemiological studies indicate that ischemic stroke is a complex disorder with a strong genetic component. Genome-wide association studies (GWAS) identified several single nucleotide polymorphisms (SNPs) associated with coronary artery disease (CAD) and myocardial infarction (MI) locus in ABO gene. Our study examined the association between four variants in the ABO gene and the risk of ischemic stroke and its subtypes, large-artery atherosclerosis (LAA) and small-vessel diseases (SVD) in the Chinese population. METHODS: In this case-control study, we recruited 1897 subjects, including 979 healthy controls and 918 ischemic stroke patients (465 with LAA and 453 with SVD). We selected four single nucleotide polymorphisms (rs579459, rs651007, rs514659 and rs529565) of the ABO gene and performed genotyping assays to assess the association with ischemic stroke and its subtypes. RESULTS: We found three polymorphisms, rs579459 and rs651007 were significantly associated with LAA using additive model and rs529565 was significantly associated with LAA using additive and dominant models. And we failed to find any significant association between these SNPs and ischemic stroke and SVD in the Chinese population. However, after the Bonferroni correction for multiple comparisons, the P-values of these SNPs failed to exceed significant threshold under any models. CONCLUSION: Our findings indicated that genetic variations of ABO gene may contribute to susceptibility of LAA but not ischemic stroke and SVD in the Chinese population. Our preliminary results should be further validated in prospective independent studies with expanded sample size.

AIM: To investigate the association of H-type hypertension with carotid atherosclerotic plaques in Chinese. METHODS: This hospital-based large population study included 13192 patients and all the patients were sequentially recruited between May 2010 and May 2013 at the Health Medical Center of the Chinese PLA General Hospital. The subjects were divided into four groups: the H-type of hypertension group, isolated systolic hypertension, simple homocysteine (Hcy) group, and the control group without hypertension and Hcy. Univariate logistic regression analysis was performed to investigate the association of H-type hypertension with the odds of carotid atherosclerotic plaques. RESULTS: Among the 13192 subjects, there were 9007 (68.28% of all subjects) patients with hyperhomocysteinemia and 3543 patients with H-type hypertension, which was 26.86% of all subjects and 74.45% of hypertension patients. 34.55% of all subjects (4558) had carotid atherosclerotic plaques. The carotid atherosclerotic plaques positive rate among four groups was significantly different (χ(2) = 647.8988, P = 0.000). The carotid atherosclerotic plaques' positive rate of patients with H-type hypertension was 49.31%, which was significantly higher than the other three groups. Univariate logistic regression analysis results indicated that significant correlations exist between high-Hcy hypertension and carotid atherosclerotic plaques. Incidence of carotid atherosclerotic plaques in H-type hypertension patients was 1.63 times that in patients with isolated systolic hypertension. CONCLUSION: H-type hypertension is independently associated with higher risk of carotid atherosclerotic plaques. H-type hypertension and Hcy should be the major intervention measures to decrease the incidence of carotid atherosclerotic plaques as well as the stroke in Health Practice Management.

RESUMEN

New chiral tetraphenylethylene (TPE) macrocycles bearing optically pure amine groups were synthesized and found to have a discriminating ability between the two enantiomers of not only chiral acidic compounds but also α-amino acids by enantioselective aggregation and aggregation-induced emission (AIE) effects. NMR spectra, including 2D-NOESY, disclosed that the host-guest interaction of the macrocycle receptor played a key role in addition to the acid-base interactions.

OBJECTIVE: To explore the correlation of food intolerance and hyperlipidemia. METHODS: A cross-sectional survey was conducted from August 2008 to June 2009. It recruited a total of 11 434 adults from Health Sciences Center at our hospital. Non-conditional Logistic regression model was utilized for determining odd ratio (OR) and 95% confidencer interval (CI). And statistic analysis was performed with SPSS 13.0. RESULTS: Among them 6 897 participants were food intolerance positive and 4 537 food intolerance negative. The prevalence of food intolerance was 60.32%. Age, gender, smoking, body mass index, total cholesterol, triglycerides and high density lipoprotein (HDL) cholesterol were significantly different between positive and negative ones (all P < 0.05). Multiple regression analysis showed that food intolerance was significantly correlated with age, gender, body mass index, but negatively with hyperlipidemia. Compared with age group ≤ 40 years, the OR (95%CI) of age groups 41-50, 51-60 and >60 years were 1.125(1.021-1.238), 1.350(1.207-1.510) and 1.564(1.279-1.912) respectively. Compared with males, the OR (95%CI) of females was 1.528(1.382-1.689). Compared with normal weighters, the OR (95%CI) of low weighters was 1.720 (1.147-2.580). Compared with normal blood lipids, the OR (95%CI) of hyperlipidemia was 0.879 (0.810-0.955). CONCLUSION: Food intolerance may be negatively correlated with hyperlipidemia in healthcare population.

RESUMEN

A quadrangular prismatic tricyclooxacalixarene cage 1 based on tetraphenylethylene (TPE) was efficiently synthesized by a one-pot S(N)Ar condensation reaction. As a result of the porous internal structure in the solid state, cage 1 exhibited a good CO2 uptake capacity of 12.5 wt% and a high selectivity for CO2 over N2 adsorption of 80 (273 K, 1 bar) with a BET surface area of 432 m(2) g(-1). Formation of cage 1 led to the fluorescence of TPE being switched on in solution. The system was employed as a single-molecule platform to study the mechanism of aggregation-induced emission (AIE) by examining the restriction of intramolecular rotation (RIR).

RESUMEN

A tetraphenylethylene (TPE) Schiff-base macrocycle showing an aggregation-induced emission (AIE) effect has been synthesized, which could aggregate into nanospheres and emit yellow fluorescence in aqueous media. By virtue of its AIE effect, the macrocycle showed a sensitive and selective response to 2,4,6-trinitrophenol (TNP) and 2,4-dinitrophenol (DNP) among a number of nitroaromatic compounds, which could be used to detect TNP and DNP at nanomolar levels. Moreover, it exhibited a superamplified quenching effect with DNP but not with TNP, providing a possible means of discriminating these two compounds. In comparison with open-chain TPE Schiff-bases, the cavity of the macrocycle is essential for the selectivity for DNP over TNP. In addition, quantitative analyses of both DNP and TNP in real water samples and qualitative detection of these two analytes in the solid state by the macrocycle have been tested. The reliability of the quantitative analysis has been confirmed by HPLC. Our findings demonstrate that the TPE Schiff-base macrocycle has great potential as an excellent sensor for DNP and TNP.
