Maturing male and female Atlantic salmon (Salmo salar L.) were held under three temperature regimes for 10 weeks between September and December: warm (constant 14-16 °C), ambient (decreasing from 11 to 5 °C), and cold (decreasing from 7 to 3 °C). Blood samples were analyzed for plasma steroid levels, and the fish were inspected for the presence of expressible milt (total volume and spermatocrit) and ovulation weekly. Samples of eggs were dry-fertilized with milt stripped from three males held at the same temperatures and incubated until the eyed stage. In females, levels of plasma testosterone (T) and 17ß-oestradiol (E2) dropped as ovulation approached, concurrent with a rapid increase in levels of plasma 17α,20ß-dihydroxy-4-pregnen-3-one (17,20ß-P). In males, levels of T and 11-ketotestosterone (11-KT) peaked 2-3 weeks after the first appearance of expressible milt, while levels of 17,20ß-P increased steadily and did not exhibit a definite peak. Exposure of females to cold water amplified and advanced the profiles of all three steroids compared with the ambient group, and increased the survival rates to the eyed egg stage. Cold water had no immediate effect on the male steroid profiles, but later, higher levels of 17,20ß-P were evident compared with both the ambient controls and the warm water group, while the effects on 11-KT and T were more variable. Exposure to warm water completely inhibited both milt production and ovulation. Moreover, warm water modulated the steroid profiles of the males with lower 11-KT levels compared with ambient controls and lower 17,20ß-P level compared with cold-water-treated males. In females, warm water resulted in total inhibition of the peri-ovulatory peak in 17,20ß-P and prevented the normal decline of T and E2 levels associated with ovulation. The findings of the present study are highly relevant for broodstock management in aquaculture, as well in understanding the impact of climate change/temperature variability on wild salmon spawning.

Reproduction of many temperate fishes is seasonal and maturation and spawning of gametes are under photothermal control. Reproductive success of first generation (G1) common sole Solea solea in captivity has been low. In this study, the sexual maturation status has been assessed during the prespawning months in G1 sole that were housed (a) outdoor under the natural photoperiod and temperature, or (b) indoor under artificial photothermal induction. Maturation was assessed in male and female G1 broodstock in November as controls, after which the remaining population was divided over two outdoor flow-through tanks placed in a pond and two indoor recirculating aquaculture system (RAS) tanks. Subsequently, maturation status (gonadosomatic index GSI and plasma levels of testosterone T and 17ß-estradiol E2) was assessed in one tank for each condition in January, February and during spawning in early April, while fish in the other tank were not disturbed in achieving reproductive success. Quantitative real-time PCR was performed to determine species-specific gonadotropin mRNA expression in females. Successful G1 spawning and egg fertilisation occurred in all experimental tanks. Gonadal development was similar under both conditions. Higher E2 and T levels were found in indoor housed females. Gonadotropin expression revealed similar profiles between outdoor and indoor housed females. G1 sole could be reproduced in the outdoor tanks under the natural photoperiod and in the indoor tanks under artificial simulation of this regime that includes a potentially crucial chilling period of 2-3 months at 5-7 °C.

Circanual variations in plasma testosterone (T), 17-estradiol (E2), and 17,20-dihydroxy-4-pregnen-3-one (17,20-P) levels and ovarian steroid synthetic potential of Tenualosa ilisha of river Hooghly, West Bengal, India were examined. This fish exhibited bi-annual spawning; one during April-May and another during August-September. Coinciding with the GSI values, present study recorded a decline in plasma T and E2 levels from October, reaching their lowest values in January followed by a rapid rise in March when the ovary contained mostly vitellogenic follicles and remained high up to April (postvitellogenic stage). Plasma 17,20ß-P level was detected in March and reached peak value in April during oocyte maturation. After spawning, all the steroid levels declined to reach lowest values in June. From June onwards, T and E2 levels again increased for the next cycle and peaked at the end of vitellogenesis. Plasma 17,20ß-P was reappeared in August and reached maximum in September during oocyte maturation and spawning. Of the two gonadotropins tested, in vitro production of both T and E2 by the vitellogenic and postvitellogenic follicles was regulated by FSH and LH respectively. Production of 17,20-P by the post-vitellogenic follicles was regulated by LH only. Acquisition of in vitro oocyte maturational competence (OMC) was developed by the addition of HCG in culture medium. Treatment of a 3ß-HSD inhibitor blocked LH-induced steroid production, but not development of OMC. Both Cycloheximide and actinomycin D inhibited LH-induced development of OMC, indicating the requirement of de novo protein synthesis for this process.
