RESUMO

1. Hexamethylenediisocyanate (HDI) is a widely used aliphatic diisocyanate and a well-recognized cause of occupational asthma. 2. "Self" molecules (peptides/proteins) in the lower airways, susceptible to chemical reactivity with HDI, have been hypothesized to play a role in asthma pathogenesis and/or chemical metabolism, but remain poorly characterized. 3. This study employed unique approaches to identify and characterize "self" targets of HDI reactivity in the lower airways. Anesthetized rabbits free breathed through a tracheostomy tube connected to chambers containing either, O2, or O2 plus ∼200 ppb HDI vapors. Following 60 minutes of exposure, the airways were lavaged and the fluid was analyzed by LC-MS and LC-MS/MS. 4. The low-molecular weight (<3 kDa) fraction of HDI exposed, but not control rabbit bronchoalveolar lavage (BAL) fluid identified 783.26 and 476.18 m/z [M+H]+ ions with high energy collision-induced dissociation (HCD) fragmentation patterns consistent with bis glutathione (GSH)-HDI and mono(GSH)-HDI. Proteomic analyses of the high molecular weight (>3 kDa) fraction of exposed rabbit BAL fluid identified HDI modification of specific lysines in uteroglobin (aka clara cell protein) and albumin. 5. In summary, this study utilized a unique approach to chemical vapor exposure in rabbits, to identify HDI reaction products with "self" molecules in the lower airways.

Amyotrophic lateral sclerosis (ALS) attacks the corticomotor system, with motor cortex function affected early in disease. Younger females have a lower relative risk of succumbing to ALS than males and older females, implicating a role for female sex hormones in disease progression. However, the mechanisms driving this dimorphic incidence are still largely unknown. We endeavoured to determine if estrogen mitigates disease progression and pathogenesis, focussing upon the dendritic spine as a site of action. Using two-photon live imaging we identify, in the prpTDP-43A315T mouse model of ALS, that dendritic spines in the male motor cortex have a reduced capacity for remodelling than their wild-type controls. In contrast, females show higher capacity for remodelling, with peak plasticity corresponding to highest estrogen levels during the estrous cycle. Estrogen manipulation through ovariectomies and estrogen replacement with 17ß estradiol in vivo was found to significantly alter spine density and mitigate disease severity. Collectively, these findings reveal that synpatic plasticity is reduced in ALS, which can be amelioriated with estrogen, in conjuction with improved disease outcomes.

This report describes the clinical, radiographic, and pathologic findings in a female rhesus macaque that presented with acute abdominal distension and tympany. The macaque was euthanized after evidence of severe colonic distension on radiography and observation of widespread peritoneal adhesions on exploratory laparotomy. Gross and histopathologic evaluation revealed extensive entrapment of gastrointestinal and reproductive tracts by serosal fibrovascular proliferative tissue containing foci of endometriosis. The diagnosis of endometrial stromal sarcoma was supported by expression of CD10, Wilm tumor 1, estrogen receptor, and progesterone receptor and failure to express immunohistochemical markers characteristic of a range of differential diagnoses. In humans, this relatively uncommon neoplasm can arise from sites of endometriosis and often presents clinically as intestinal obstruction, similar to the presentation in this macaque.

Gray short-tailed opossums are used in a wide variety of research in the areas of developmental biology, oncology, immunology, and comparative biology. Despite many frequent experimental manipulations of these animals under anesthesia, few studies to date have characterized the effects of anesthesia in this species. Our aim was to identify safe and effective injectable anesthetic combinations using ketamine and xylazine or ketamine and dexmedetomidine at doses of 40 mg/kg to 100 mg/kg for ketamine, 5 mg/kg to 10 mg/kg for xylazine, and 0.05 mg/kg to 0.1 mg/kg for dexmedetomidine. Effects of the proposed regimens ranged from light sedation to surgical anesthesia, but only 100 mg/kg ketamine + 0.1 mg/kg dexmedetomidine induced surgical anesthesia in all opossums, with a mean duration of 25.4 min. The 2 lowest doses of ketamine and xylazine (40 mg/kg ketamine + 5 mg/kg xylazine and 40 mg/kg ketamine + 10 mg/kg xylazine) achieved sedation to light anesthesia in all animals but did not produce a surgical plane of anesthesia in any animal. All regimens that induced a surgical plane of anesthesia caused bradycardia and bradypnea, and 75 mg/kg ketamine + 10 mg/kg xylazine and 100 mg/kg ketamine + 0.1 mg/kg dexmedetomidine caused the greatest decreases in SpO2. Except for one opossum that died of unknown causes, all animals remained healthy and apparently free of anesthetic complications. Among all treatments, isoflurane delivered by a precision vaporizer provided the most consistent and reliable anesthesia; therefore, we recommend inhalant anesthesia over the injectable combinations used in this study.
