BACKGROUND: Establishing the existence of health inequalities remains a high research and policy agenda item in the United Kingdom. We describe ethnic and socio-economic differences in paediatric cancer survival, focusing specifically on the extent to which disparities have changed over a 20-year period. METHODS: Cancer registration data for 2674 children (0-14 years) in Yorkshire were analysed. Five-year survival estimates by ethnic group (south Asian/non-south Asian) and Townsend deprivation fifths (I-V) were compared over time (1997-2016) for leukaemia, lymphoma, central nervous system (CNS) and other solid tumours. Hazard ratios (HR: 95% CI) from adjusted Cox models quantified the joint effect of ethnicity and deprivation on mortality risk over time, framed through causal interpretation of the deprivation coefficient. RESULTS: Increasing deprivation was associated with significantly higher risk of death for children with leukaemia (1.11 (1.03-1.20)) and all cancers between 1997 and 2001. While we observed a trend towards reducing differences in survival over time in this group, a contrasting trend was observed for CNS tumours whereby sizeable variation in outcome remained for cases diagnosed until 2012. South Asian children with lymphoma had a 15% reduced chance of surviving at least 5 years compared to non-south Asian, across the study period. DISCUSSION: Even in the United Kingdom, with a universally accessible healthcare system, socio-economic and ethnic disparities in childhood cancer survival exist. Findings should inform where resources should be directed to provide all children with an equitable survival outcome following a cancer diagnosis.

RESUMO

A diode laser module emitting 1.4 kW optical in-pulse power near 780 nm optimized for high (≥ 10%) duty-cycle operation in a micro-channel free design is presented. With full collimation, a beam quality with a nearly symmetric M2 of 205 × 295 (vertical × horizontal direction) for a wide range of pulse widths is found.

The monoamine serotonin has been shown to regulate peripartal calcium homeostasis in multiparous cows and be a possible mitigation tool for hypocalcemia. Increasing circulating serotonin concentrations via prepartum intravenous (IV) administration of the serotonin precursor 5-hydroxy-L-tryptophan (5-HTP) increases postpartum calcium concentrations. However, the ability of 5-HTP to be used orally or ruminally to alter circulating serotonin concentrations has not been established. Hence, our objective was to determine if ruminal administration of 5-HTP altered circulating serotonin concentrations. Four ruminally cannulated, nonlactating, nonpregnant multiparous Holstein dairy cows were randomly assigned to 1 of 4 treatments in a 4 × 4 replicated Latin square with 4-d periods separated by a 7-d washout. On d 1 and 2 of each period, cows were dosed with 1 of 4 experimental treatments as follows: (1) 0 mg/kg of body weight (BW) of 5-HTP, (2) 1 mg/kg of BW of intraruminal 5-HTP, (3) 2 mg/kg of BW of intraruminal 5-HTP, or (4) 1 mg/kg of BW of IV 5-HTP. Infusions were administered over a 1-h period, and all groups not receiving 5-HTP IV were infused with an equal volume of IV saline to that of IV 1 mg/kg of BW of 5-HTP treatment. Continuous serial blood samples were collected beginning after d 2 of treatment administration. Whole blood serotonin concentrations were higher in cows dosed with 2 mg/kg of BW of intraruminal 5-HTP immediately after dosing when compared with cows dosed with 0 mg/kg of BW of 5-HTP on d 2, but were similar on d 3 and 4 of the experimental period. Cows receiving IV 5-HTP had the highest circulating serotonin concentrations relative to all other treatments. These findings demonstrated that 2 intraruminal dosings of 5-HTP at 2 mg/kg of BW resulted in elevated circulating serotonin concentrations relative to the control immediately after dosing. This supports the potential for 5-HTP to be used orally to manipulate circulating serotonin concentrations.

RESUMO

We demonstrate coherent beam combining of four high brightness tapered amplifiers in pulsed, quasi continuous wave (QCW) operation, seeded by a 976 nm laser diode. The maximum power of 22.7 W was achieved with > 64% combining efficiency in a close to diffraction limited beam. We discuss turn-on dynamics of tapered amplifiers operated in pulsed mode in detail.

We report on a coherent beam combination of three high-brightness tapered amplifiers, which are seeded by a single-frequency laser at λ = 976 nm in a simple architecture with efficiently cooled emitters. The maximal combined power of 12.9 W is achieved at a combining efficiency of > 65%, which is limited by the amplifiers' intrinsic beam quality. The coherent combination cleans up the spatial profile, as the central lobe's power content increases by up to 86%. This high-brightness infrared beam is converted into the visible by second harmonic generation. This results in a high non-linear conversion efficiency of 4.5%/W and a maximum power over 2 W at 488 nm, which is limited by thermal effects in the periodically poled lithium niobate (PPLN).

The primary aim of this research was to describe the association between milking interval (MI) and milk production rate (MPR) at the quarter level in a large commercial farm using an automatic milking system. A secondary aim was to determine whether a 2-h decrease in MI would increase MPR at the cow level in midlactation multiparous cows. Six months of data from 1,280 cows were used to assess the association between MI (h) and quarter MPR (kg/h). Increasing MI was associated with decreased MPR for early, mid, and late lactation, both primiparous and multiparous cows, and all 4 quarter positions and across time. The decrease in MPR is approximately 2%/h of increasing MI for multiparous cows and 1.5%/h for primiparous cows. Regardless of quarter, multiparous cows had a greater MPR than primiparous cows, and rear quarters had greater MPR than front quarters. An experiment to test the causal relationship between changing MI and cow-level MPR was conducted using 26 animal pairs matched on MI, days in milk, and milk yield. During the 21-d treatment period, the average MI of treatment cows was decreased by 2.4 h compared with control cows. In both the 21-d treatment and 42-d posttreatment periods, no significant difference was found in cow-level MPR between the treatment and control groups. Despite the negative association between increasing MI and MPR being consistent across all assessed days in milk windows and all quarters, results from this experiment suggest that intervention to decrease MI might require an MI change greater than 2 h or be applied in early lactation to significantly increase MPR.

The primary aim of this observational study, in a single herd milked using multiple automatic milking system units, was to describe associations of quarter milk yield variability and quarter peak milk flow rate with cow-level factors. Information from the current lactation of 1,549 primiparous and multiparous cows was collected from January to December 2015. Data from each individual milking used in the analysis included quarter milk yield (QMY), udder milk yield, quarter peak milk flow rate (QPMF), quarter average milk flow rate (QAMF), quarter milking time, and milking interval. Milking interval and milk yield were used to calculate milk production rate (kg/h) at the quarter and udder levels. We investigated associations between QPMF and milking interval, QPMF and days in milk, and QMY and QAMF. A strong association between QPMF and both QAMF and milking interval was observed. A moderate association was found between QPMF and stage of lactation. However, QMY was not a useful indicator of QPMF because of the weak association observed between these variables. In this study, rear quarter QPMF was significantly increased by 3% compared with front quarter QPMF (1.45 vs 1.41 kg/min). Quarter milk yield was calculated as a percentage contribution of total udder milk yield per 10-d in milk window and ranked from lowest to highest contribution. Quarter contribution to udder milk yield showed a high level of variability, with 39% of animals having all 4 quarters change contribution rank at least once during part of or the whole lactation. Only 14% of cows were observed to have no change in quarter rank. When quarter contribution was assessed, irrespective of physical position of quarter within the udder, the percent of highest to lowest contribution across the lactation was relatively stable. The standard deviation of quarter milk production rate for each cow was regressed against the same cow's peak udder milk production rate, within a lactation, to ascertain whether quarter milk production rate variance could be used to predict peak udder milk production rate. Knowledge of the intra-udder quarter milk production rate standard deviation for an individual cow is not useful in predicting peak udder milk production rate. Quarter milking time appears to be a useful indicator to predict the optimal order of teatcup attachment. Analysis from this large, single-herd population indicates that QPMF is associated with the cow-level factors milking interval and days in milk, and that intra-udder QMY is highly variable.

Hypocalcemia affects almost 50% of all dairy cows. Our laboratory has previously demonstrated that infusions of the serotonin precursor 5-hydroxy-l-tryptophan (5-HTP) increase circulating calcium concentrations in the Holstein transition cow. It is unknown whether feeding a negative dietary cation-anion difference (DCAD) diet alters the relationship between 5-HTP and hypocalcemia. The main objective of this study was to determine whether feeding a negative DCAD (-DCAD) diet before calving in conjunction with 5-HTP treatment could further diminish the magnitude of hypocalcemia at the time of calving. We used a randomized complete block design with a 2 × 2 factorial arrangement. Thirty-one multiparous Holstein cows were fed either a positive (+13 mEq/100 g) or negative (-13 mEq/100 g) DCAD diet 21 d before parturition and were intravenously infused daily with saline or 5-HTP (1 mg/kg) starting 7 d before the estimated date of parturition. Cows were blocked by parity and were randomly assigned to 1 of 4 treatment groups: positive DCAD plus saline, positive DCAD plus 5-HTP, negative DCAD plus saline, and negative DCAD plus 5-HTP, resulting in n = 8 per group. Total calcium (tCa), ionized calcium (iCa), and feed intake were recorded. The iCa was elevated prepartum in the -DCAD/5-HTP group compared with the other treatment groups as well as on d 0 and 1 postpartum. Although differences in tCa were not significant across the pre- or postpartum periods, tCa was numerically higher on d 0 and significantly higher on d 1 in -DCAD/5-HTP cows compared with all other groups. Prepartum the -DCAD/5-HTP treatment group ate less than the other treatment groups; however, postpartum dry matter intake differences were not significant. These findings demonstrate that feeding a -DCAD diet in conjunction with 5-HTP prepartum can increase postpartum circulating iCa concentrations and therefore diminish the magnitude of hypocalcemia at the time of parturition.

The aim of this study was to assess associations of cow-, udder-, and quarter-level factors with the risk of clinical mastitis (CM) in cows managed using an automatic milking system. The primary hypothesis was that quarter peak milk flow rate (QPMF) is associated with increased risk of CM. A retrospective, case-control study was undertaken using data from a 1,549 cow farm using 20 automatic milking system units. All data from cows milked during March to December 2015 was available for analysis. Cases (n = 82) were defined as cows diagnosed with their first case of CM between 24 and 300 d in milk in the current lactation. Healthy control cows (n = 6/case) were randomly matched based on identical parity, existence of milk records during the day in milk period corresponding to the 15-d window before case diagnosis, average conductivity of <5.5 mS/cm in that window, and no history of CM in the current lactation. Logistic regression was used to estimate effects of parity, quarter position, day in milk at diagnosis of CM, average of QPMF 15 d before CM diagnosis, udder milk yield, and milking interval on the probability of CM. Of the 6 predictor variables included in the model, only milking interval was significantly associated with the increased risk of quarter CM. We concluded that in a high-production, freestall-housed North American herd using automatic milking system, milking interval, but not QPMF, was associated with risk of CM.

The objectives of this study were to measure performance responses and to evaluate indictors of N utilization in late-lactation cows fed diets with incremental reductions in crude protein (CP) concentration. Holstein cows (n = 128; 224 ± 54 d in milk) were stratified by parity and days pregnant (86 ± 25 d) and randomly assigned to 1 of 16 pens in a randomized complete block design. For 3 wk, all cows received a covariate diet containing 16.9% CP [dry matter (DM) basis]. For the subsequent 12 wk, pens were randomly assigned to 1 of 4 treatments that contained 16.2, 14.4, 13.1, or 11.8% CP (DM basis). Diets were offered once daily and contained 32.5% corn silage, 32.5% alfalfa silage, 13.5% high-moisture corn, and 21.5% concentrate mix. A reduction in dietary CP was achieved by replacing soybean meal with soy hulls in the concentrate mix (DM basis). Dry matter intake, milk urea N (MUN; mg/dL), and the yield of milk urea N (g/d) decreased linearly with dietary CP. Compared with a 16.2% CP diet, a 14.4% CP diet did not alter milk yield throughout the study, but the 13.1 and 11.8% CP diets reduced milk yield after 4 and 1 wk, respectively. Furthermore, milk protein percentage was reduced for all dietary CP less than 16.2%, but this negative effect was temporary and disappeared after 7 wk for the 14.4% CP diet. In contrast, MUN adjusted to a new steady state within 1 wk for all dietary treatments. Modeling quadratic responses with a plateau led to predictions of no reduction in fat- and protein-corrected milk (32.6 kg/d) and yields of fat (1.31 kg/d), lactose (1.49 kg/d), and true protein (1.12 kg/d) until dietary CP decreased below 15.5, 15.3, 15.9, and 16.2%, respectively. In this study, MUN and the yield of MUN were highly correlated with N intake, milk protein yield, and fat- and protein-corrected milk. Surprisingly, N use efficiency (milk protein N/intake N) was not correlated with any variables related to N utilization and reached an apparent upper limit of approximately 30%. Although this observation may be associated with feeding diets deficient in metabolizable protein, late-lactation cows in this study adjusted to low dietary CP concentration better than anticipated as milk production was 2.6, 3.6, 6.4, and 8.0 kg/d higher than National Research Council (2001)-predicted metabolizable protein-allowable milk for dietary CP of 16.2, 14.4, 13.1, and 11.8%, respectively.

The primary aim of this study was to assess the effects of incomplete milking on milk secretion and milk composition at the quarter level. Twelve cows were enrolled beginning at 5 d in milk and remained on study through 47 d in milk. Half of each contralateral udder was incompletely milked (treatment), detaching the teat cup early to leave approximately 30% of the total milk yield behind. This target milk remaining in the gland was based on weekly calibration milking measurements of quarter total milk yield. Control quarters were milked completely until milk flow had decreased to 0 kg/min based on visual assessment. Harvested milk yield was measured twice daily at each milking, and milk components (fat, protein, lactose, solids nonfat, milk urea nitrogen) and somatic cell count, were measured twice weekly at the quarter level. The experimental unit in this design was the half-udder, and a mixed-model approach was used to assess the main and interactive effects of experiment week and treatment on milk production rate, milk remaining in the gland, and milk composition. The effect of treatment on milk production rate was significant, with the average control half-udder producing 0.97 kg/h and the treatment half-udder 0.73 kg/h. The effect of week on milk production rate and the interaction of week × treatment were also significant. The effect of treatment on milk remaining in the gland was significant, illustrating that an increase in milk remaining in the cisternal compartment had been achieved. We detected a significant decrease in milk lactose percentage in treatment half-udders, and a significant increase in somatic cell count (log10). The increase was relatively small, from a geometric mean of 26,300 cells/mL in control quarters to 48,300 cells/mL in treatment quarters. The decrease in milk production rate in treatment half-udders supports current knowledge about how mammary epithelial cell secretion, proliferation, and apoptosis are modulated by autocrine-paracrine factors.

RESUMO

We describe a new coherent beam combining architecture based on passive phase locking of emitters in an extended cavity on the rear facet and their coherent combination on the front facet. This rear-side technique provides strong optical feedback for phase locking while maintaining a high electrical-to-optical efficiency. Two high-brightness high-power tapered laser diodes are coherently combined using a Michelson-based cavity. The combining efficiency is above 82% and results in an output power of 6.7 W in a nearly diffraction-limited beam with an M(4σ)(2)≤1.2. A semi-active automatic adjustment of the current enhances the long-term stability of the combination, while the short-term stability is passively ensured by the extended cavity. This new laser configuration exhibits the simplicity of passive self-organizing architectures while providing a power conversion efficiency of 27% that is comparable to master oscillator power amplifier architectures.

Our objective was to evaluate the effect of a second PGF2α treatment and duration of an Ovsynch protocol that included a progesterone-releasing intravaginal device (PRID) on progesterone (P4) concentrations and pregnancies per artificial insemination (P/AI) after resynchronization of ovulation and timed artificial insemination (TAI). Lactating Holstein cows (n=821) were assigned randomly at a nonpregnancy diagnosis (d 0) to 3 resynchronization protocols: (1) GnRH, d 0; PGF2α, d 7; GnRH, d 9.5 (7D1PGF); (2) GnRH, d 0; PGF2α, d 7; PGF2α, d 8; GnRH, d 9.5); (7D2PGF); or (3) GnRH, d 2; PGF2α, d 7; PGF2α, d 8; GnRH, d 9.5 (5D2PGF). All cows received a PRID at the first GnRH treatment of the resynchronization protocol, which was removed at the first PGF2α treatment, and all cows received TAI approximately 16h after the second GnRH treatment. Blood samples were collected from a subgroup of cows at each treatment of the resynchronization protocols. At 32 d after TAI, cows receiving a second PGF2α treatment (7D2PGF + 5D2PGF cows) had more P/AI (42.6 vs. 35.7%) than cows receiving a single PGF2α treatment (7D1PGF cows). For cows treated with a second PGF2α treatment, decreasing the duration of the protocol did not increase P/AI (41.4 vs. 43.8% for 7D2PGF vs. 5D2PGF cows). At 60 d after TAI, P/AI did not differ between cows treated with the 1 PGF2α (7D1PGF cows) or 2 PGF2α (7D2PGF + 5D1PGF cows) treatments (32.5 vs. 37.9%, respectively). In addition, reducing the duration of the protocol did not increase P/AI at 60 d after TAI (37.8 vs. 38.5% for 7D2PGF vs. 5D2PGF cows). Pregnancy loss from 32 to 60 d after TAI was not affected by the number of PGF2α treatments (8.5 vs. 10.6%, for 7D1PGF vs. 7D2PGF + 5D2PGF cows) or the duration of the protocol (9.1 vs. 12.1%, for 7D2PGF vs. 5D2PGF cows). The percentage of cows with incomplete luteal regression at the second GnRH treatment tended to differ among treatments and was lowest for 7D2PGF cows, intermediate for 5D2PGF cows, and greatest for 7D1PGF cows (1.9 vs. 6.9 vs. 11.0%, respectively). In conclusion, addition of a second PGF2α treatment tended to decrease the percentage of cows with incomplete luteal regression and increased P/AI 32 d after AI, whereas decreasing the duration of the Ovsynch protocol did not increase P/AI.

The objective of this study was to evaluate the effects of ensiling time and exogenous protease addition on soluble CP (% of CP), ammonia-N (% of N), and ruminal in vitro starch digestibility (ivSD) of whole-plant corn silage (WPCS) from 3 hybrids, 2 maturities, and 2 chop lengths. Samples from 3 nonisogenic hybrids [brown midrib containing the bm3 gene mutation (BM3), dual-purpose (DP), or floury-leafy (LFY)] at 2 harvest maturities [2/3 kernel milk line (early) or 7d later (late)] with 2 theoretical lengths of cut settings (0.64 or 1.95cm) on a forage harvester were collected at harvest, treated with or without exogenous protease, and ensiled in triplicate in vacuum heat-sealed plastic bags for 0, 30, 60, 120, and 240d. Thus, the experiment consisted of 120 treatments (3 hybrids × 2 maturities × 2 chop lengths × 2 protease treatments × 5 time points) and 360 mini-silos (3 replications per treatment). Vitreousness, measured by dissection on unfermented kernels on the day of harvest, averaged 66.8, 65.0, and 59.0% for BM3, DP, and LFY, respectively. A protease × maturity interaction was observed with protease increasing ivSD in late but not early maturity. Ensiling time × hybrid interactions were observed for ammonia-N and soluble CP concentrations with greater values for FLY than other hybrids only after 120d of ensiling. Ensiling time × hybrid or protease × hybrid interactions were not observed for ivSD. Measurements of ivSD were greatest for FLY and lowest for BM3. Length of the ensiling period did not attenuate negative effects of kernel vitreousness or maturity on ivSD in WPCS. Results suggest that the dosage of exogenous protease addition used in the present study may reduce but not overcome the negative effects of maturity on ivSD in WPCS. No interactions between chop length and ensiling time or exogenous protease addition were observed for ivSD.

Dietary fatty acids can affect both milk fat yield and fatty acid (FA) composition. This relationship is well established when the dietary level of FA exceeds 3% of diet dry matter (DM). We could find no reports directly examining the effects of dietary FA profile on milk fat at levels below 3%. Twenty-four primiparous and 36 multiparous lactating cows were paired by production (1 high with 1 low, within parity) to form 30 experimental units. Pairs were fed 6 diets in five 6×6 balanced Latin squares with 21-d periods, and data were collected during the last 5d of each period. Two control diets were fed: a corn control diet (CC; 29% corn silage, 16% alfalfa silage, 19% corn grain, and 8% distillers grain on a DM basis) containing 1.8% FA; and a low-oil control diet (LOC; 9% corn silage, 35% alfalfa silage, 20% food-grade corn starch, and 8% corn gluten feed on a DM basis) containing 1.2% FA. A portion of the food-grade corn starch in LOC was replaced with 4 different FA supplements to create the 4 treatment diets. Treatments were 1.7% (DM basis) of a 50:50 blend of corn oil and high-linoleic safflower oil (LO), 1.7% high-oleic sunflower oil (OO), 1.7% palm oil (PO), or 1.8% calcium salts of palm fatty acids (PFA). The resultant diets were thus enriched in linoleic (LO), oleic (OO), or palmitic acid (PO and PFA). Dietary treatments did not affect dry matter intake. Addition of any of the fat sources to LOC resulted in increased milk yield, but milk fat yields and milk FA composition were variable for the different treatments. The LO treatment resulted in lower milk fat yield, fat concentration, and C16:0 yield but increased both trans-10 C18:1 and trans-10,cis-12 C18:2 yields compared with the other added FA treatments. Diets PO and PFA resulted in increased milk C16:0 yield and decreased total milk C18 yield compared with OO. Regression analysis revealed a negative coefficient for dietary linoleic acid content over basal (LOC) for both milk short-chain FA yield and C16:0 yield. Dietary linoleic acid content also had a positive coefficient for milk trans-10 C18:1 and trans-10,cis-12 conjugated linoleic acid yield. These results demonstrate that even when total dietary FA are below 3%, free oils rich in linoleic acid can reduce milk fat yield by reducing secretion of milk FA with fewer than 18 carbons. Fatty acid composition of fat supplements is important even at this low level of total dietary fat.

The objective was to study repeatability and sources of variation in feed conversion efficiency [FCE, milk kg/kg dry matter intake (DMI)] of lactating cows in mid to late lactation. Trials 1 and 2 used 16 cows (106 to 368 d in milk) grouped in 8 pairs of 1 high- and 1 low-FCE cow less than 16 d in milk apart. Trial 1 determined the repeatability of FCE during a 12-wk period. Trial 2 quantified the digestive and metabolic partitioning of energy and N with a 3-d total fecal and urine collection and measurement of CH4 and CO2 emission. Trial 3 studied selected ruminal methanogens in 2 pairs of cows fitted with rumen cannulas. Cows received a single diet including 28% corn silage, 27% alfalfa silage, 17% crude protein, and 28% neutral detergent fiber (dry matter basis). In trial 1, mean FCE remained repeatedly different and averaged 1.83 and 1.03 for high- and low-FCE cows, respectively. In trial 2, high-FCE cows consumed 21% more DMI, produced 98% more fat- and protein-corrected milk, excreted 42% less manure per kilogram of fat- and protein-corrected milk, but emitted the same daily amount of CH4 and CO2 compared with low-FCE cows. Percentage of gross energy intake lost in feces was higher (28.6 vs. 25.9%), but urinary (2.76 vs. 3.40%) and CH4 (5.23 vs. 6.99%) losses were lower in high- than low-FCE cows. Furthermore, high-FCE cows partitioned 15% more of gross energy intake toward net energy for maintenance, body gain, and lactation (37.5 vs. 32.6%) than low-FCE cows. Lower metabolic efficiency and greater heat loss in low-FCE cows might have been associated in part with greater energy demand for immune function related to subclinical mastitis, as somatic cell count was 3.8 fold greater in low- than high-FCE cows. As a percentage of N intake, high-FCE cows tended to have greater fecal N (32.4 vs. 30.3%) and had lower urinary N (32.2 vs. 41.7%) and greater milk N (30.3 vs. 19.1%) than low-FCE cows. In trial 3, Methanobrevibacter spp. strain AbM4 was less prevalent in ruminal content of high-FCE cows, which emitted less CH4 per unit of DMI and per unit of neutral detergent fiber digested than low-FCE cows. Thus lower digestive efficiency was more than compensated by greater metabolic efficiencies in high- compared with low-FCE cows. There was not a single factor, but rather a series of mechanisms involved in the observed differences in efficiency of energy utilization of the lactating cows in this study.

A survey was conducted in Wisconsin (WI) and Michigan (MI) to quantify the proportion of farms that use a single diet for all lactating cows and to better understand the reasons for current grouping strategies and the limitations to grouping for better nutritional management. A questionnaire was mailed to all WI dairy farmers with ≥200 lactating cows (971 farms) and to a random sample of grade-A MI dairy farmers (800 farms) of varying herd sizes. The survey return rate was 20% in WI (196 farms) and 26% in MI (211 farms; 59 of them had ≥200 lactating cows). Feeding 2 or more different diets to lactating cows was predominant: 63% in WI (124 farms, all ≥200 lactating cows), 76% in MI farms with ≥200 lactating cows (45 farms), and 28% in MI farms with <200 lactating cows (43 farms). Farmers feeding more than 1 diet used 1 or more of the following criteria for grouping lactating cows: stage of lactation, milk production, or body condition score. Overall for both states, 52% of the farms (211 from 407 farms) feeding more than 1 diet grouped cows according to their nutritional needs. However, a notable population of farms fed the same diet to all lactating cows: 37% in WI (72 farms), 24% in MI (14 farms) for herds of ≥200 lactating cows, and 72% in MI for herds of <200 lactating cows (109 MI farms). "Desire to keep it simple" and "milk drops when cows are moved to a different group" were identified as main constraints to having more groups within a farm for nutritional purposes. Farm facilities and labor were also limiting factors to grouping in farms with herd sizes of <200 lactating cows.

The objective of this study was to evaluate the associations of variable intensity in rearing dairy heifers on 33 commercial dairy herds, including 23,008 cows and 18,139 heifers, with age at first calving (AFC), average daily weight gain (ADG), and milk yield (MY) level on reproduction traits and profitability. Milk yield during the production period was analyzed relative to reproduction and economic parameters. Data were collected during a 1-yr period (2011). The farms were located in 12 regions in the Czech Republic. The results show that those herds with more intensive rearing periods had lower conception rates among heifers at first and overall services. The differences in those conception rates between the group with the greatest ADG (≥0.800 kg/d) and the group with the least ADG (≤0.699 kg/d) were approximately 10 percentage points in favor of the least ADG. All the evaluated reproduction traits differed between AFC groups. Conception at first and overall services (cows) was greatest in herds with AFC ≥800 d. The shortest days open (105 d) and calving interval (396 d) were found in the middle AFC group (799 to 750 d). The highest number of completed lactations (2.67) was observed in the group with latest AFC (≥800 d). The earliest AFC group (≤749 d) was characterized by the highest depreciation costs per cow at 8,275 Czech crowns (US$414), and the highest culling rate for cows of 41%. The most profitable rearing approach was reflected in the middle AFC (799 to 750 d) and middle ADG (0.799 to 0.700 kg) groups. The highest MY (≥8,500 kg) occurred with the earliest AFC of 780 d. Higher MY led to lower conception rates in cows, but the highest MY group also had the shortest days open (106 d) and a calving interval of 386 d. The same MY group had the highest cow depreciation costs, net profit, and profitability without subsidies of 2.67%. We conclude that achieving low AFC will not always be the most profitable approach, which will depend upon farm-specific herd management. The MY is a very important factor for dairy farm profitability. The group of farms having the highest MY achieved the highest net profit despite having greater fertility problems.

The objective of this study was to evaluate the effect of body condition score (BCS), body weight (BW), average daily weight gain (ADG), and age at first calving (AFC) of Holstein heifers on production and reproduction parameters in the 3 subsequent lactations. The data set consisted of 780 Holstein heifers calved at 2 dairy farms in the Czech Republic from 2007 to 2011. Their BW and BCS were measured at monthly intervals during the rearing period (5 to 18 mo of age), and the milk production and reproduction data of the first 3 lactations were collected over an 8-yr period (2005 to 2012). The highest milk yield in the first lactation was found in the group with medium ADG (5 to 14 mo of age; 0.949 to 0.850 kg of ADG). The highest average milk yield over lifetime performance was detected in heifers with the highest total ADG (≥ 0.950 kg/d). The difference in milk yield between the evaluated groups of highest ADG (in total and postpubertal growth ≥ 0.950 kg/d and in prepubertal growth ≥ 0.970 kg/d) and the lowest ADG (≤ 0.849 kg/d) was approximately 1,000 kg/305 d per cow. The highest milk yield in the first lactation was found in the group with the highest AFC ≥ 751 d, for which fat and protein content in the milk was not reduced. Postpubertal growth (11 to 14 mo of age) had the greatest effect on AFC. The group with lowest AFC ≤ 699 d showed a negative effect on milk yield but only in the first 100 d of the first parity. The highest ADG was detrimental to reproduction parameters in the first lactation. The highest BW at 14 mo (≥ 420 kg) led to lower AFC. Groups according to BCS at 14 mo showed no differences in AFC or milk yield in the first lactation or lifetime average production per lactation. We concluded that low AFC ≤ 699 d did not show a negative effect on subsequent production and reproduction parameters. Therefore, a shorter rearing period is recommended for dairy herds with suitable management.

Multiple metabolic and hormonal factors can affect the success of protocols for ovarian superstimulation. In this study, the effect of acute feed restriction and increased LH content in the superstimulatory FSH preparation on numbers of ovulations, fertilization, and embryo quality in lactating dairy cows was evaluated. Two experiments were performed using a Latin square design with treatments arranged as a 2 × 2 factorial: feed restriction (FR; 25% reduction in dry matter intake) compared with ad libitum (AL) feeding, combined with high (H) versus low (L) LH in the last 4 injections of the superstimulatory protocol. As expected, FR decreased circulating insulin concentrations (26.7 vs. 46.0 µU/mL). Two analyses were performed: one that evaluated the complete Latin square in experiment 2 and a second that evaluated only the first periods of experiments 1 and 2. For both analyses, follicle numbers, ovulation rates, and corpora lutea on d 7 were not different. In the first period analysis of experiments 1 and 2, we observed an interaction between feed allowance and amount of LH on fertilization rates, percentage of embryos or oocytes that were quality 1 and 2 embryos, and number of embryos or oocytes that were degenerate. Fertilization rates were greater for the AL-L (89.4%) and FR-H (80.1%) treatments compared with the AL-H (47.9%) and FR-L (59.9%) treatments. Similarly, the proportion of total embryos or oocytes designated as quality 1 and 2 embryos was greater for AL-L (76.7%) and FR-H (73.4%) treatments compared with AL-H (35.6%) and FR-L (47.3%) treatments. In addition, the number of degenerate embryos was decreased for AL-L (1.3) and FR-H (0.4) treatments compared with the AL-H (2.6) and FR-L (2.3) treatments. Thus, cows with either too low (FR-L) or too high (AL-H) insulin and LH stimulation had lesser embryo production after superstimulation because of reduced fertilization rate and increased percentage of degenerate embryos. Therefore, interaction of the gonadotropin content of the superstimulatory preparation with the nutritional program of the donor cow needs to be considered to optimize success of ovarian superstimulatory protocols.
