RESUMO

Sry-box9 (SOX9) maintains stem cell properties and plays crucial roles in many cancers. However, whether SOX9 is correlated with cervical cancer cell stemness and its detailed mechanism remains obscure. We studied the relationship between SOX9 and prognosis of cervical cancer through public database, and SOX9 was related to poor prognosis of cervical cancer. Elevated SOX9 expression enhanced the self-renewal properties and promotes tumorigenicity in cervical cancer. Overexpression of SOX9 could promote the expression of stem cell-related factors in cervical cancer cells and xenografts. Meanwhile, overexpression of SOX9 could also enhance the expressions of FZD10, ß-catenin, and c-Myc in cervical cancer cells and xenografts, while inhibiting the expression of DDK1. The activation of Wnt pathway by chir-99 021 raised the tumor spheroid ability of SOX9 knockdown HeLa cells. In addition, SOX9 could transcriptional inhibit DKK1 and activate FZD10 and MYC by binding to their promoters to affect the Wnt/ß-catenin pathway. These results demonstrated SOX9 regulated the self-renewal and tumorigenicity of cervical cancer through Wnt/ß-catenin pathway by directly transcriptional activation of FZD10, MYC and transcriptional inhibition of DKK1.

BACKGROUND: Despite available clinical management strategies, chronic kidney disease (CKD) is associated with severe morbidity and mortality worldwide, which beckons new solutions. Host-microbial interactions with a depletion of Faecalibacterium prausnitzii in CKD are reported. However, the mechanisms about if and how F prausnitzii can be used as a probiotic to treat CKD remains unknown. METHODS: We evaluated the microbial compositions in 2 independent CKD populations for any potential probiotic. Next, we investigated if supplementation of such probiotic in a mouse CKD model can restore gut-renal homeostasis as monitored by its effects on suppression on renal inflammation, improvement in gut permeability and renal function. Last, we investigated the molecular mechanisms underlying the probiotic-induced beneficial outcomes. RESULTS: We observed significant depletion of Faecalibacterium in the patients with CKD in both Western (n=283) and Eastern populations (n=75). Supplementation of F prausnitzii to CKD mice reduced renal dysfunction, renal inflammation, and lowered the serum levels of various uremic toxins. These are coupled with improved gut microbial ecology and intestinal integrity. Moreover, we demonstrated that the beneficial effects in kidney induced by F prausnitzii-derived butyrate were through the GPR (G protein-coupled receptor)-43. CONCLUSIONS: Using a mouse CKD model, we uncovered a novel beneficial role of F prausnitzii in the restoration of renal function in CKD, which is, at least in part, attributed to the butyrate-mediated GPR-43 signaling in the kidney. Our study provides the necessary foundation to harness the therapeutic potential of F prausnitzii for ameliorating CKD.

Most soil ammonia (NH3) emissions originate from soil nitrogen (N) that has been in the form of exchangeable ammonium. Emitted NH3 not only induces nutrient loss but also has adverse effects on the cycling of N and accelerates global warming. There is evidence that arbuscular mycorrhizal (AM) fungi can alleviate N loss by reducing N2O emissions in N-limited ecosystems, however, some studies have also found that global changes, such as warming and N deposition, can affect the growth and development of AM fungi and alter their functionality. Up to now, the impact of AM fungi on NH3 emissions, and whether global changes reduce the AM fungi's contribution to NH3 emissions reduction, has remained unclear. In this study, we examined how warming, N addition, and AM fungi alter NH3 emissions from high pH saline soils typical of a temperate meadow through a controlled microscopic experiment. The results showed that warming significantly increased soil NH3 emissions, but N addition and combined warming plus N addition had no impact. Inoculations with AM fungi strongly reduced NH3 emissions both under warming and N addition, but AM fungi effects were more pronounced under warming than following N addition. Inoculation with AM fungi reduced soil NH4+-N content and soil pH, and increased plant N content and soil net N mineralization rate while increasing the abundance of ammonia-oxidizing bacterial (AOB) gene. Structural equation modeling (SEM) shows that the regulation of NH3 emissions by AM fungi may be related to soil NH4+-N content and soil pH. These findings highlight that AM fungi can reduce N loss in the form of NH3 by increasing N turnover and uptake under global changes; thus, AM fungi play a vital role in alleviating the aggravation of N loss caused by global changes and in mitigating environmental pollution in the future.

Perovskite-based photodetectors exhibit potential applications in communication, neuromorphic chips, and biomedical imaging due to their outstanding photoelectric properties and facile manufacturability. However, few of perovskite-based photodetectors focus on ultraviolet-visible-short-wavelength infrared (UV-Vis-SWIR) broadband photodetection because of the relatively large bandgap. Moreover, such broadband photodetectors with individual nanocrystal channel featuring monolithic integration with functional electronic/optical components have hardly been explored. Herein, an individual monocrystalline MAPbBr3 nanoplate-based photodetector is demonstrated that simultaneously achieves efficient UV-Vis-SWIR detection and fast-response. Nanoplate photodetectors (NPDs) are prepared by assembling single nanoplate on adjacent gold electrodes. NPDs exhibit high external quantum efficiency (EQE) and detectivity of 1200% and 5.37 × 1012 Jones, as well as fast response with rise time of 80 µs. Notably, NPDs simultaneously achieve high EQE and fast response, exceeding most perovskite devices with multi-nanocrystal channel. Benefiting from the high specific surface area of nanoplate with surface-trap-assisted absorption, NPDs achieve high performance in the near-infrared and SWIR spectral region of 850-1450 nm. Unencapsulated devices show outstanding UV-laser-irradiation endurance and decent periodicity and repeatability after 29-day-storage in atmospheric environment. Finally, imaging applications are demonstrated. This work verifies the potential of perovskite-based broadband photodetection, and stimulates the monolithic integration of various perovskite-based devices.

AIMS: To present the results of an exploratory analysis of the BEYOND V study in which Chinese individuals with uncontrolled type 2 diabetes (T2D) received short-term intensive insulin therapy (SIIT) during study run-in (prior to randomization) using a basal-first insulin titration method. MATERIALS AND METHODS: This was exclusively an exploratory analysis of the 7- to 10-day run-in period of BEYOND V. Participants were hospitalized and had oral therapies withdrawn (except metformin). They received SIIT with once-daily insulin glargine and three-times-daily premeal insulin glulisine, titrated daily from a total starting dose of 0.4 to 0.5 units/kg/d, first adjusting insulin glargine to achieve fasting blood glucose (FBG) of 4.4 to 6.1 mmol/L (79 to 119 mg/dL), then insulin glulisine to achieve pre-meal blood glucose of 4.4 to 6.1 mmol/L. Key outcomes were the proportions of participants achieving FBG and 2-hour postprandial blood glucose (PBG) targets. RESULTS: Overall, 397 entered the run-in (mean 54.2 years, 235 males [59.2%]). At the end of SIIT, 374/396 participants (94.4%) had both FBG <7.0 mmol/L (<126 mg/dL) and 2-hour PBG <10 mmol/L (<180 mg/dL) and 282/396 (71.2%) had both FBG <6.1 mmol/L (<100 mg/dL) and 2-hour PBG <10 mmol/L. The mean first time taken to achieve FBG <7 mmol/L, 2-hour PBG <10 mmol/L, and both, was 4.35, 3.88, and 5.04 days, respectively. Hypoglycaemia occurred in 99 participants (24.9%). There was no severe hypoglycaemia. CONCLUSIONS: Titrating basal insulin first is an effective and safe method of SIIT in individuals with T2D, rapidly achieving target glucose levels with a relatively low rate of hypoglycaemia.

NK-lysins are one of the most abundant antimicrobial peptides produced by cytotoxic T lymphocytes (CTLs) and natural killer cells (NKs), and identified as a new class of intrinsically disordered proteins, playing critical roles in the cell-mediated cytotoxicity response, as well as immunomodulatory and antimicrobial activities upon a significant range of pathogens. In the present study, an NK-lysin was identified from Obscure puffer Takifugu obscurus (ToNK-lysin). The open reading frame of ToNK-lysin sequence spans 423 bp, encoding a peptide with 140 amino acids which shares a moderate residue identity (18%-60%) with NK-lysin of mammals and other teleost species. Phylogenetic analysis revealed that ToNK-lysin was most closely related to NK-lysins from the Pleuronectiformes (Bastard halibut Paralichthys olivaceus and Pacific halibut Hippoglossus stenolepis). Comprehensive computational analysis revealed that ToNK-lysin have substantial level of intrinsic disorder, which might be contribute to its multifunction. The transcripts of the ToNK-lysin were detected in multiple examined tissues and most abundant in gills. After bacterial and Poly I:C challenge, the transcriptional levels of ToNK-lysin were significantly up-regulated in the head kidney, liver and spleen at different time points. The recombinant ToNK-lysin showed significant antibacterial activity against Vibrio harveyi and Escherichia coli, and the ToNK-lysin treatment not only reduced the bacterial loads in liver and head kidney, but also alleviated the pathogen-mediated upregulation of immune-related genes. In addition, the co-incubation with rToNK-lysin protein remarkably degraded bacterial genomic DNA, suggesting the potential mechanism of ToNK-lysin against microbes. These results suggest that ToNK-lysin possess antibacterial and immunoregulatory function both in vivo and in vitro, which may allow it a potential applicability to the aquaculture industry.

Equalization-enhanced phase noise (EEPN) has emerged as one of the major impairments that cannot be ignored for a high baud rate Stokes vector direct detection (SVDD) system. When EEPN interacts with the rotation of state-of-polarization (RSOP) and chromatic dispersion (CD), the joint impairment effects become even more complicated. To achieve the joint equalization of EEPN, RSOP, and CD impairments of a high baud rate SVDD system, this paper first derives a joint impairment model of these three kinds of impairments, and then proposes a joint equalization scheme of EEPN, RSOP, and CD with a sliding window assisted extended Kalman filter (SWA-EKF). The SWA-EKF scheme first tracks RSOP in the time domain, subsequently compensates CD in the frequency domain, and finally performs EEPN mitigation in the time domain again. The effectiveness of the proposed scheme has been verified by a 60 GBaud SVDD-16QAM simulation system. The results show that when these three impairments are jointly equalized, the SWA-EKF scheme can track RSOP as fast as 3 Mrad/s, cumulative dispersion up to 1600 ps/nm, and EEPN caused by laser linewidth up to 3 MHz. In addition, with an optical signal-to-noise ratio penalty of 0.3 dB, it could increase 35 G baud rate under 3 MHz laser linewidth for the SVDD system. More importantly, its total complexity can be reduced to an order of O(N log N).

SNAI2 (Snai2) is a zinc-finger transcriptional repressor that belongs to the Snail family. The accumulated evidence suggests that SNAI2 exhibits biphasic effects on regulating a stem-like phenotype in various types of cells, both normal and malignant. In this study, by exogenously expressing SNAI2 in SiHa cells, SNAI2 exhibited the capacity to inhibit a stem-like phenotype in cervical cancer cells. The SNAI2-overexpressing cells inhibited cell growth, tumorsphere formation, tumor growth, enhanced sensitivity to cisplatin, reduced stem cell-related factors' expression, and lowered tumor initiating frequency. In addition, the EPCAMhigh cells sorted from SiHa cells exhibited an enhanced capacity to maintain a stem-like phenotype. Further study demonstrated that the trans-suppression of EPCAM expression by SNAI2 led to blockage of the nuclear translocation of ß-catenin, as well as reduction in SOX2 and c-Myc expression in SiHa and HeLa cells, but induction in SNAI2 knockdown cells (CaSki), which would be responsible for the attenuation of the stem-like phenotype in cervical cancer cells mediated by SNAI2. All of these results demonstrated that SNAI2 could attenuate the stem-like phenotype in cervical cancer cells through the EPCAM/ß-catenin axis.

BACKGROUND: Siraitia grosvenorii (Swingle) C. Jeffrey, also known as Luohanguo or monk fruit, is a famous traditional Chinese medicine ingredient with important medicinal value and broad development prospects. Diploid S. grosvenorii has too many seeds, which will increase the utilization cost of active ingredients. Thus, studying the molecular mechanism of seed abortion in triploid S. grosvenorii, identifying the abortion-related genes, and regulating their expression will be a new direction to obtain seedless S. grosvenorii. Herein, we examined the submicroscopic structure of triploid S. grosvenorii seeds during abortion. RESULTS: Upon measuring the endogenous hormone content, we found that abscisic acid (ABA) and trans-zeatin (ZR) levels were significantly downregulated after days 15 and 20 of flowering. RNA sequencing of triploid seeds at different developmental stages was performed to identify key genes regulating abortion in triploid S. grosvenorii seeds. Multiple genes with differential expression between adjacent stages were identified; seven genes were differentially expressed across all stages. Weight gene co-expression network analysis revealed that the enhancement of monoterpene and terpene metabolic processes might lead to seed abortion by reducing the substrate flow to ABA and ZR. CONCLUSIONS: These findings provide insights into the gene-regulatory network of seed abortion in triploid S. grosvenorii from different perspectives, thereby facilitating the innovation of the breeding technology of S. grosvenorii.

When we implement the equalizations of polarization effects using a Kalman filter (KF) in a coherent optical fiber communication system, we will require to multiply many matrices. If the state vector describing the system has a dimension of n, the state error covariance matrix P will have the dimension of n × n, and other matrices used in the Kalman filter will also have the dimension of n × l (l is the dimension of the measurement vector). If n is very large, the KF-based algorithm will suffer from significant complexity, which results in an impractical KF-based polarization demultiplexing algorithm. In this paper, we propose a new structured KF-based polarization demultiplexing algorithm in which the state error covariance matrix P is diagonalized, which we call the diagonalized Kalman filter (DKF). We theoretically analyze the rationality of the DKF, and the validity of the DKF was verified in both 64 Gbaud polarization-division multiplexed (PDM) QPSK and 16QAM Nyquist coherent optical simulation systems. Compared with the conventional KF, simulation results proved that under a rotation of state of polarization from 1 to 10 Mrad/s for QPSK and 1 to 5 Mrad/s for 16QAM, a differential group delay from 15 to 75 ps, and a residual chromatic dispersion of 100 ps/nm, the OSNR penalties for the DKF are only within 0.5 dB for QPSK at the threshold BER = 3.8 × 10-3, and within 2 dB for 16-QAM at the threshold BER = 2 × 10-2, respectively, compare to the case of no impairment. In the meantime, for the proposed DKF, a computational complexity reduction of over 30% is achieved, compared with conventional KF, at the expense of about no more than 50 symbols convergence delay.

Moderate or strong shaping conditions reduce the occurrence probability of the outermost ring constellation points of probabilistically shaped (PS)-M quadrature amplitude modulation (QAM) signals, which easily causes the peaks in the 4th power periodogram of received signals be submerged, accordingly the classical frequency offset estimation (FOE) scheme using 4th power fast Fourier transform (FFT) cannot be applied in PS-MQAM system. To solve this issue, we have proposed an optimal decision threshold assisted quadrature phase shift keying (QPSK)-partition blind FOE scheme. Firstly, the proposed scheme utilizes an optimal decision threshold assisted method for the symbol decision of received symbols, then chooses the symbols on multiple specific QPSK-shape rings. Secondly, the amplitude of each symbol selected above is normalized and uniformly augmented to 18. Finally, it carries out FOE using an improved time-domain 4th power feedforward method that eliminates the time interval. The effectiveness of the proposed scheme has been verified by 28 GBaud polarization division multiplexing (PDM) PS-16/64QAM simulations and 28/8 GBaud PS-16/64QAM experiments. The results obtained by this scheme present that under moderate or strong shaping conditions, the generalized mutual information (GMI) increases with optical signal-to-noise ratio (OSNR) and eventually exceeds the corresponding GMI threshold. Besides that, the FOE range can reach [-Rs/8, Rs/8], where Rs denotes the baud rate. When OSNRs are higher than 16 dB and 19.5 dB, the NMSEs of PS-16QAM-3/3.6 are lower than 1e-7, respectively. For PS-64QAM-4.4/5, the NMSEs achieve lower than 1e-6 after OSNR increases to 20.3 dB and 23.4 dB, respectively. More importantly, the overall complexity can be reduced to O(N), which is at most as 26.5% as that of FFT FOE scheme.

AIM: To compare the efficacy and safety of basal insulin glargine 100 units/ml (Gla) + 2-3 oral antihyperglycaemic drugs (OADs) with twice-daily premixed insulin aspart 70/30 (Asp30) + metformin (MET) after short-term intensive insulin therapy in adults with type 2 diabetes in China. MATERIALS AND METHODS: This open-label trial enrolled insulin-naïve adults with type 2 diabetes and an HbA1c of 7.5%-11.0% (58-97 mmol/mol) despite treatment with 2-3 OADs. All participants stopped previous OADs except MET, then received short-term intensive insulin therapy during the run-in period, when those with a fasting plasma glucose of less than 7.0 mmol/L and 2-hour postprandial glucose of less than 10.0 mmol/L were randomized to Gla + MET + a dipeptidyl peptidase-4 inhibitor or twice-daily Asp30 + MET. If HbA1c was more than 7.0% (>53 mmol/mol) at week 12, participants in the Gla group were added repaglinide or acarbose, at the physician's discretion, and participants in the Asp30 group continued to titrate insulin dose. The change in HbA1c from baseline to week 24 was assessed in the per protocol (PP) population (primary endpoint). RESULTS: There were 384 enrollees (192 each to Gla and Asp30); 367 were included in the PP analysis. The threshold for non-inferiority of Gla + OADs versus Asp30 + MET was met, with a least squares mean change from baseline in HbA1c of -1.72% and -1.70% (-42.2 and -42.1 mmol/mol), respectively (estimated difference -0.01%; 95% CI -0.20%, 0.17% [-0.1 mmol/mol; 95% CI -2.2, 1.9]). Achievement of HbA1c less than 7.0% (<53 mmol/mol) was comparable between the groups (60% vs. 57%). The proportion of participants with any (24% vs. 38%; P = .003), symptomatic (19% vs. 31%; P = .007) or confirmed hypoglycaemia (18% vs. 33%; P < .001) was lower in the Gla + OADs group. CONCLUSIONS: Compared with Asp30 + MET, Gla + 2-3 OADs showed similar efficacy but a lower hypoglycaemia risk in Chinese individuals with type 2 diabetes who had undergone short-term intensive insulin therapy.

PURPOSE: This study aimed to investigate the clinical usefulness of the enhanced-T1WI-based deep learning radiomics model (DLRM) in differentiating low- and high-grade meningiomas. METHODS: A total of 132 patients with pathologically confirmed meningiomas were consecutively enrolled (105 in the training cohort and 27 in the test cohort). Radiomics features and deep learning features were extracted from T1 weighted images (T1WI) (both axial and sagittal) and the maximum slice of the axial tumor lesion, respectively. Then, the synthetic minority oversampling technique (SMOTE) was utilized to balance the sample numbers. The optimal discriminative features were selected for model building. LightGBM algorithm was used to develop DLRM by a combination of radiomics features and deep learning features. For comparison, a radiomics model (RM) and a deep learning model (DLM) were constructed using a similar method as well. Differentiating efficacy was determined by using the receiver operating characteristic (ROC) analysis. RESULTS: A total of 15 features were selected to construct the DLRM with SMOTE, which showed good discrimination performance in both the training and test cohorts. The DLRM outperformed RM and DLM for differentiating low- and high-grade meningiomas (training AUC: 0.988 vs. 0.980 vs. 0.892; test AUC: 0.935 vs. 0.918 vs. 0.718). The accuracy, sensitivity, and specificity of the DLRM with SMOTE were 0.926, 0.900, and 0.924 in the test cohort, respectively. CONCLUSION: The DLRM with SMOTE based on enhanced T1WI images has favorable performance for noninvasively individualized prediction of meningioma grades, which exhibited favorable clinical usefulness superior over the radiomics features.

Cryptorchidism, as a common congenital disease of canine testes, is mainly caused by factors leading to endocrine abnormalities in testes and infertility in a heat stress and hypoxia microenvironment. Moreover, heat stress and hypoxia, as critical microenvironmental factors, promote epithelial-mesenchymal transition (EMT), which occurs during adult tissue remodelling responses including carcinogenesis and fibrosis and is the main cause of testicular tumours. In this study, we found by haematoxylin-eosin staining that the canine cryptorchid tissue produced a lot of collagen fibres. Also, the quantitative PCR and Western blot results showed that the mRNA and protein levels of the heat stress makers HSP70 and HO-1 and the hypoxia maker HIF-1α are significant higher compared with normal testes. Moreover, we found the expression levels of TGF-ßs and its two receptors TGF-ßRI and TGF-ßRII increased in case of cryptorchidism. From the study in vitro, we found both heat stress and COCl2 mimic hypoxia inhibited the secretion of testosterone (T) and androstenedione (A4) and promoted the expression of the EMT maker α-SMA and vimentin in Leydig cells, and also that heat stress and COCl2 stimulated with the TGF-ß signalling promoted the expression of TGF-ßs and its two type receptors and also the active phosphorylation of Smad2 and Smad3. The use of LY2109761, a receptor inhibitor of TGF-ßs/Smad signalling pathway, was associated with heat stress and COCl2 suppression of androgens' secretion and stimulated EMT in Leydig cells. These findings characterized a novel pathogenesis of cryptorchidism and provided a new idea for therapeutics.

BACKGROUND AND OBJECTIVES: To identify the main dietary patterns of adults and investigate the cross-sectional associations of these dietary patterns with prediabetes and undiagnosed or diagnosed diabetes mellitus (DM) in Qingdao, China. METHODS AND STUDY DESIGN: This study included 4,457 participants who were administered the semi-quantitative food frequency questionnaire (FFQ). Dietary patterns were identified through principal component analysis (PCA). Logistic regression analysis was performed to determine the associations of each pattern with the risks of prediabetes and undiagnosed or diagnosed DM. RESULTS: PCA revealed two major dietary patterns. The Fruits-Vegetables and Poultry-Seafood patterns were not significantly associated with the risk of prediabetes in either crude or adjusted models (all p>0.05). The highest quartile of the Fruits-Vegetables pattern was significantly associated with decreased risks of undiagnosed DM (crude: OR=0.55, 95% CI: 0.41-0.72; Model 1: OR=0.61, 95% CI: 0.46-0.81; Model 2: OR=0.57, 95% CI: 0.42-0.77; Model 3: OR=0.56, 95% CI: 0.41-0.76) and diagnosed DM (crude: OR=0.51, 95% Cl: 0.34-0.75; Model 1: OR=0.59, 95% CI: 0.39-0.88; Model 2: OR=0.60, 95% CI: 0.39-0.93; Model 3: OR=0.59, 95% CI: 0.38-0.91) compared with the lowest quartile in crude and adjusted models. The Poultry-Seafood pattern was not significantly associated with the risk of undiagnosed or diagnosed DM in crude or adjusted models (all p>0.05). CONCLUSIONS: The Fruits-Vegetables pattern was associated with a decreased risk of undiagnosed or diagnosed DM.

Having access to and visiting urban green space (UGS) improves liveability and provides considerable benefits to residents. However, traditional methods of investigating UGS visitation, such as questionnaires and social surveys, are usually time- and resource-intensive, and frequently provide less transferable, site-specific outcomes. This study uses social media data (Twitter) to examine spatio-temporal changes in UGS use in London associated with COVID-19 related lockdowns. It compares georeferenced Tweets posted in a 3 month period from 23 March to 23 June for 3 years covering the first lockdown in the UK in 2020, with Tweets for the same period in 2019 and 2021. The results show that (1) the land-use type of Public Park and Garden was the most frequently visited type of UGS, which may be correlated with these UGS areas remaining opening during the lockdown period; (2) the usage of UGS decreased in central London and increased in other areas during lockdown, which may correlated with working from home restrictions; (3) activities were positively associated with Physical activities maybe as a result of allowing people to take a single daily exercise, and (4) people spent more time in UGS areas on weekdays than weekends compared to pre-lockdown. This is the first study to examine social media data over consistent time period before, during and after the lockdown in relation to UGS. The results show that the findings and method can inform policy makers in their management and planning of UGS, especially in a period of social crisis like the COVID-19 pandemic.

In optical fiber communication systems, the tendency to prefer increasingly higher baud rates, up to 100 or 200 Gbaud, compels us to deal with wideband fiber channels in which polarization effects must be treated in a more complicated manner. In contrast to the conventional 50 GHz bandwidth wavelength division multiplexing channel, polarization mode dispersion (PMD) is observed in an all-order manner in a wideband fiber channel beyond 100 GHz, rather than only first- and second-order. In this study, certain principles that should be followed in establishing a PMD emulation model in a wideband fiber channel were put forward. Then, a temporal and spectral evolution PMD model that can reflect and emulate the evolution features of the polarization effects in a real fiber channel was proposed and demonstrated. The analyses show the PMD model satisfies all the principles for the wideband channels. Finally, we suggested some approaches to define a narrowband or wideband channel only considering the polarization effects to deal with PMD in a high baud rate optical fiber communication system.

BACKGROUND: Slug (Snai2) is a pivotal player in initiating epithelial-mesenchymal transition (EMT) through its trans-suppression effect on E-cadherin in various normal and malignant cells. In this study, the positive effect of Slug on promoting cell motility and metastasis in cervical cancer was further confirmed in this study. METHODS: RNA-Seq was performed to explore the potential molecules that participate in Slug-mediated EMT in cervical cancer cells. The negative correlation between Slug and EpCAM expression in cervical cancer cells was detected in this study, and linked them with in vitro migration and invasion assay, in vivo metastasis experiments, luciferase reporter assay and Chromatin immunoprecipitation. RESULTS: Transcriptome sequencing analysis revealed that epithelial cell adhesion molecule (EpCAM) was significantly decreased in Slug-overexpressing SiHa cells. Simultaneously, an absence of EpCAM expression was observed in Slug-overexpressing cells. Further studies revealed the trans-suppression effect of Slug on EpCAM through its binding to the E-boxes in the proximal promoter region of EpCAM in cervical cancer cells. Restoring EpCAM in Slug-overexpressing cells by transiently transfecting an EpCAM recombinant plasmid attenuated cell motility and promoted cell growth. Moreover, the negative correlation between Slug and EpCAM expression in human squamous cervical carcinoma (SCC) samples was verified by using Pearson correlation analysis. CONCLUSIONS: These results demonstrated that the absence of EpCAM under Slug expression in cervical cancer cells probably participated in Slug-regulated EMT and further promoted tumor metastasis. Additionally, this study supports a potential way for Slug to initiate EMT progression in cervical cancer cells in addition to inhibiting E-cadherin.

BACKGROUND: Hexokinases 2 (HK2) is a member of the hexokinases, linking with malignant tumor growth and distant metastasis. However, evidence regarding the potential role of HK2 in regulating cell motility and tumor metastasis during the cervical cancer malignant progression remains limited. METHODS: In vitro migration and invasion assay, in vivo metastasis experiments were performed to detect the effective of HK2 on regulating cell motility and tumor metastasis in cervical cancer cells. RNA-Seq was performed to explore the potential molecules that participate in HK2-mediated cell motility and tumor metastasis in cervical cancer cells. The correlation between HK2 and Akt1, p-Akt1, FN1 expression in cervical cancer cells and human squamous cervical carcinoma (SCC) samples was verified in this study. RESULTS: In this study, cervical cancer cells with exogenous HK2 expression exhibited enhanced cell motility and distant metastasis. Transcriptome sequencing analysis revealed that fibronectin (FN1) was significantly increased in HK2-overexpressing HeLa cells, and the PI3K/Akt signaling pathway was identified by KEGG pathway enrichment analysis. Further studies demonstrated that this promotion of cell motility by HK2 was probably a result of it inducing FN1, MMP2 and MMP9 expression by activating Akt1 in cervical cancer cells. Additionally, HK2 expression was altered with the changing of Akt1/p-Akt1 expression, implying that HK2 expression is also modulated by Akt1/p-Akt1. Moreover, the positive correlation between HK2 and Akt1, p-Akt1, FN1 expression in human squamous cervical carcinoma (SCC) samples was verified by using Pearson correlation analysis. CONCLUSIONS: This study demonstrated that HK2 could activate Akt1 in cervical cancer cells, subsequently enhancing cell motility and tumor metastasis by inducing FN1, MMP2 and MMP9 expression. There likely exists an interactive regulatory mechanism between HK2 and Akt1 during the malignant process of cervical cancer.

BACKGROUND: Extranodal natural killer/T cell lymphoma (ENKL) is a rare subtype of non-Hodgkin lymphoma, and lung involvement is extremely rare. The patients with pulmonary ENKL always presented unspecific symptoms of the respiratory system, such as cough with sputum and varying degrees of fever, while developing into acute respiratory distress (ARDS) was seldomly reported, especially promoted by the surgical procedure. CASE PRESENTATION: Here we describe a patient with nasal ENKL and most likely lung dissemination that was regarded as an infection at first. After nonresponse to a period of anti-infective therapy, this patient received surgical debridement. While the histopathology did not show the evidence of infection, but consistent with ENKL. The patient got refractory hypoxemia rapidly after surgery, with the LDH surging to a much higher level than before surgery. The ARDS was diagnosed, and he died on the 5th day after surgery. We postulate that ARDS was due to aggressive lymphoma proliferation promoted by the surgical procedure. CONCLUSIONS: Pulmonary ENKL developing into ARDS was scarce, and was likely attributed to the aggressive tumor cell proliferation after surgery in this case.
