RESUMO

Transmembrane domains (TMDs) contain information targeting membrane proteins to various compartments of the secretory pathway. In previous studies, short or hydrophilic TMDs have been shown to target membrane proteins either to the endoplasmic reticulum (ER), or to the Golgi apparatus. The basis for differential sorting to the ER and to the Golgi apparatus remained however unclear. To clarify this point, we analyzed quantitatively the intracellular targeting of a collection of proteins exhibiting a single TMD. Our results reveal that membrane topology is a major targeting element in the early secretory pathway: type I proteins with a short transmembrane domain are targeted to the ER, and type II proteins to the Golgi apparatus. A combination of three features accounts for the sorting of simple membrane proteins in the secretory pathway: membrane topology, length and hydrophilicity of the TMD, and size of the cytosolic domain. By clarifying the rules governing sorting to the ER and to the Golgi apparatus, our study may revive the search for sorting mechanisms in the early secretory pathway.

Nile tilapia (Oreochromis niloticus) is a species of worldwide importance for aquaculture. A crossbred lineage was developed through introgressive backcross breeding techniques and combines the high growth performance of the Chitralada (CHIT) lwith attractive reddish color of the Red Stirling (REDS) strains. Since the crossbreed has an unknown genetically improved background, the objective of this work was to characterize expression signatures that portray the advantageous phenotype of the crossbreeds. We characterized the microRNA transcriptome by high throughput sequencing (RNA-seq) and the proteome through mass spectrometry (ESI-Q-TOF-MS) and applied bioinformatics for the comparative analysis of such molecular data on the three strains. Crossbreed expressed a distinct set of miRNAs and proteins compared to the parents. They comprised several microRNAs regulate traits of economic interest. Proteomic profiles revealed differences between parental and crossbreed in expression of proteins associated with glycolisis. Distinctive miRNA and protein signatures contribute to the phenotype of crossbreed.

Phagocytic cells take up, kill and digest microbes by a process called phagocytosis. To this end, these cells bind the particle, rearrange their actin cytoskeleton, and orchestrate transport of digestive factors to the particle-containing phagosome. The mammalian lysosomal membrane protein LIMP-2 (also known as SCARB2) and CD36, members of the class B of scavenger receptors, play a crucial role in lysosomal enzyme trafficking and uptake of mycobacteria, respectively, and generally in host cell defences against intracellular pathogens. Here, we show that the Dictyostelium discoideum LIMP-2 homologue LmpA regulates phagocytosis and phagolysosome biogenesis. The lmpA knockdown mutant is highly affected in actin-dependent processes, such as particle uptake, cellular spreading and motility. Additionally, the cells are severely impaired in phagosomal acidification and proteolysis, likely explaining the higher susceptibility to infection with the pathogenic bacterium Mycobacterium marinum, a close cousin of the human pathogen Mycobacterium tuberculosis Furthermore, we bring evidence that LmpB is a functional homologue of CD36 and specifically mediates uptake of mycobacteria. Altogether, these data indicate a role for LmpA and LmpB, ancestors of the family of which LIMP-2 and CD36 are members, in lysosome biogenesis and host cell defence.

TM9 family proteins (also named Phg1 proteins) have been previously shown to control cell adhesion by determining the cell surface localization of adhesion proteins such as the Dictyostelium SibA protein. Here, we show that the glycine-rich transmembrane domain (TMD) of SibA is sufficient to confer Phg1A-dependent surface targeting to a reporter protein. Accordingly, in Dictyostelium phg1A-knockout (KO) cells, proteins with glycine-rich TMDs were less efficiently transported out of the endoplasmic reticulum (ER) and to the cell surface. Phg1A, as well as its human ortholog TM9SF4 specifically associated with glycine-rich TMDs. In human cells, genetic inactivation of TM9SF4 resulted in an increased retention of glycine-rich TMDs in the endoplasmic reticulum, whereas TM9SF4 overexpression enhanced their surface localization. The bulk of the TM9SF4 protein was localized in the Golgi complex and a proximity-ligation assay suggested that it might interact with glycine-rich TMDs. Taken together, these results suggest that one of the main roles of TM9 proteins is to serve as intramembrane cargo receptors controlling exocytosis and surface localization of a subset of membrane proteins.

ACAPs and ASAPs are Arf-GTPase-activating proteins with BAR, PH, GAP and ankyrin repeat domains and are known to regulate vesicular traffic and actin cytoskeleton dynamics in mammalian cells. The amoeba Dictyostelium has only two proteins with this domain organization, instead of the six in human, enabling a more precise functional analysis. Genetic invalidation of acapA resulted in multinucleated cells with cytokinesis defects. Mutant acapA(-) cells were hardly motile and their multicellular development was significantly delayed. In addition, formation of filopodial protrusions was deficient in these cells. Conversely, re-expression of ACAP-A-GFP resulted in numerous and long filopodia-like protrusions. Mutagenesis studies showed that the ACAP-A actin remodeling function was dependent on its ability to activate its substrate, the small GTPase ArfA. Likewise, the expression of a constitutively active ArfA•GTP mutant in wild-type cells led to a significant reduction in filopodia length. Together, our data support a role for ACAP-A in the control of the actin cytoskeleton organization and dynamics through an ArfA-dependent mechanism.

The objective of this study was to evaluate the salivary glands in diabetic mice, analyzing alterations in the secretory epithelium and interactions with the stromal compartment acquired during a prolonged period of treatment with Bauhinia forficata extract. Female mice were divided into two groups: Nonobese diabetic (NOD) mice treated with Bauhinia forficata (I), and NOD mice not treated with the hypoglycemic agent (II). After treatment, the salivary glands were collected for analysis by transmitted and polarized light microscopy, complemented by three-dimensional analysis of these tissues. The results showed weight loss in animals of group II and weight recovery in treated animals. Glucose levels were elevated in group II, but declined in group I. In the two groups, the salivary glands were characterized by involution of the secretory epithelium, presence of an inflammatory infiltrate and an increase of extracellular fibrillar components. It can be concluded that treatment with Bauhinia forficata reduced glucose levels and contributed to weight recovery in treated animals. However, the observation of tissue destructuring and compromised epithelial-stromal interactions, with consequent impairment of glandular function, demonstrates that Bauhinia forficata exerts an effect on the recovery of body metabolism but this improvement does not influence in the tissue recovery.

Objetivo: identificar em pesquisas, no âmbito da comunidade científica, os principais aspectos sobre a atuação de enfermeiros na informática em saúde no contexto hospitalar. Método: revisão integrativa da literatura com buscas realizadas nas bases de dados MEDLINE/PubMed, PubMed Central, Scopus, Cochrane Library e Web of Science utilizando os descritores: nurses; information technology; medical informatics; nursing informatics; hospitals; computer systems. Utilizou-se o método PRISMA e, após a seleção e análise, a amostra final foi constituída por 16 artigos. Resultados: foram identificadas duas dimensões: "Aspectos atitudinais de enfermeiros em relação à informática em saúde" e "Aspectos sobre o suporte organizacional". Nas questões atitudinais, percebeu-se que o nível de conhecimento, treinamento, habilidades e trabalho interdisciplinar influencia a atuação e o desempenho de funções assistenciais e gerenciais. Por sua vez, o suporte organizacional foi considerado importante na participação de enfermeiros para a construção e o aperfeiçoamento dos sistemas informatizados. Conclusão: as pesquisas evidenciam que o uso de sistemas informatizados em ambientes hospitalares e ambulatoriais tem relações com questões atitudinais dos profissionais da Enfermagem tanto quanto o suporte organizacional para o uso de recursos tecnológicos de forma eficiente, mostrando caminhos para uma atualização na formação profissional, bem como a inserção em novos ambientes de trabalho.(AU)


Objective: to identify, within the scientific community, the main aspects about nurses' performance in health informatics in hospital settings. Method: integrative literature review with searches conducted in the databases MEDLINE/PubMed, PubMed Central, Scopus, Cochrane Library, and Web of Science using the keywords: nurses; information technology; medical informatics; nursing informatics; hospitals; computer systems. The PRISMA method was used and, after selection and analysis, the final sample consisted of 16 articles. Results: two dimensions were identified: "Attitude aspects of nurses towards health informatics" and "Aspects of organizational support". In the questions regarding attitude, it was perceived that the level of knowledge, training, skills, and interdisciplinary work influence the performance of care and management functions. In turn, organizational support was considered important in the participation of nurses for the implementation and improvement of computerized systems. Conclusion: research shows that the use of computerized systems in hospital and outpatient settings is related to attitude issues of nursing professionals as well as organizational support for the efficient use of technological resources, showing paths for an update in professional training, as well as the insertion in new work environments.(AU)

A su vez, se consideró importante el apoyo organizacional en la participación de los enfermeros en la construcción y perfeccionamiento de los sistemas informatizados. Conclusión: la investigación muestra que el uso de sistemas informatizados en ambientes hospitalarios y ambulatorios está relacionado con cuestiones actitudinales de los profesionales de Enfermería, así como el apoyo organizacional para el uso eficiente de los recursos tecnológicos, mostrando formas de actualización en la formación profesional, así como la inserción en nuevos entornos de trabajo.(AU)

Dictyostelium discoideum is a widely used model to study molecular mechanisms controlling cell adhesion, cell spreading on a surface, and phagocytosis. In this study we isolated and characterize a new mutant created by insertion of a mutagenic vector in the heretofore uncharacterized spdA gene. SpdA-ins mutant cells produce an altered, slightly shortened version of the SpdA protein. They spread more efficiently than WT cells when allowed to adhere to a glass substrate, and phagocytose particles more efficiently. On the contrary, a functional spdA knockout mutant where a large segment of the gene was deleted phagocytosed less efficiently and spread less efficiently on a substrate. These phenotypes were highly dependent on the cellular density, and were most visible at high cell densities, where secreted quorum-sensing factors inhibiting cell motility, spreading and phagocytosis are most active. These results identify the involvement of SpdA in the control of cell spreading and phagocytosis. The underlying molecular mechanisms, as well as the exact link between SpdA and cell spreading, remain to be established.

Ovarian xenotransplantation is a promising alternative to preserve fertility of oncologic patients. However, several functional aspects of this procedure remained to be addressed. The aim of this study was evaluate the feasibility of xenotransplantation as a strategy to maintain bovine ovarian grafts and produce oocytes. Adult ovarian cortical pieces were xenotransplanted to the dorsal subcutaneous of female NOD-SCID mice (n = 62). Grafts were recovered ten days after xenotransplantation. Host and graft weights; folliculogenesis progression; blood perfusion, relative gene expression and number of macrophage and neutrophil of xenografts; in vitro developmental competence of graft-derived oocytes were evaluated. Folliculogenesis was supported in the grafts, as indicated by the presence of primordial, primary, secondary, antral, and atretic follicles. The xenografts showed a greater volumetric density of atretic follicles and higher hyperemia and number of host-derived macrophage and neutrophil (P<0.05), when compared to non-grafted fragments. There was a higher blood perfusion under the back skin in the transplantation sites of host animals than in control and non-grafted (P<0.01). BAX and PRDX1 genes were up-regulated, while BCL2, FSHR, IGF1R and IGF2R were down-regulated, when compared to the control (P<0.01). Twenty seven oocytes were successfully harvested from grafts, and some of these oocytes were able to give rise to blastocysts after in vitro fertilization. However, cleavage and blastocyst rates of xenograft derived oocytes were lower than in control (P<0.01). Despite showing some functional modifications, the ovarian xenografts were able to support folliculogenesis and produce functional oocytes.

Dictyostelium discoideum has largely been used to study phagocytosis and intracellular killing of bacteria. Previous studies have shown that Phg1A, Kil1 and Kil2 proteins are necessary for efficient intracellular killing of Klebsiella bacteria. Here we show that in phg1a KO cells, cellular levels of lysosomal glycosidases and lysozyme are decreased, and lysosomal pH is increased. Surprisingly, overexpression of Kil1 restores efficient killing in phg1a KO cells without correcting these lysosomal anomalies. Conversely, kil1 KO cells are defective for killing, but their enzymatic content and lysosomal pH are indistinguishable from WT cells. The killing defect of phg1a KO cells can be accounted for by the observation that in these cells the stability and the cellular amount of Kil1 are markedly reduced. Since Kil1 is the only sulfotransferase characterized in Dictyostelium, an (unidentified) sulfated factor, defective in both phg1a and kil1 KO cells, may play a key role in intracellular killing of Klebsiella bacteria. In addition, Phg1B plays a redundant role with Phg1A in controlling cellular amounts of Kil1 and intracellular killing. Finally, cellular levels of Kil1 are unaffected in kil2 KO cells, and Kil1 overexpression does not correct the killing defect of kil2 KO cells, suggesting that Kil2 plays a distinct role in intracellular killing.

OBJECTIVES: The incretin-based therapy might be effective in patients possessing certain levels of preserved pancreatic beta-cells. However, doubts still exist regarding the efficacy of this atment in the recovery of tissues damaged by type 1 diabetes. Thus, the objective of this study was to evaluate the treatment with MK0431 in salivary glands of spontaneously diabetic mice, focusing mainly on the possible therapeutic and hypoglycaemic effects of this dipeptidyl peptidase IV inhibitor in the recovery of these salivary tissues. METHODS AND RESULTS: Twenty mice were divided into two groups of 10 animals each: group I (NOD diabetic/untreated) and group II (NOD diabetic MK0431/treated). The group II was treated during 4 weeks with MK0431 mixed in the food. The group I was maintained in the same way without receiving, however, any treatment. Glucose levels were monitored during treatment and salivary glands samples were collected at the end of treatment for the histological examination under both transmitted and polarized light microscopy. High glucose levels were observed in untreated animals, while in animals with treatment, reduction of these levels was observed. Tissue restructuring was also observed in animals submitted to therapy with MK0431, mainly in relation to the attempt to extracellular matrix reorganization. CONCLUSIONS: According to results, the treatment with this dipeptidyl peptidase IV inhibitor contributed to the general homeostasis of the organism and to the reestablishment of both epithelial and stromal compartments which were damaged by the hyperglycaemic condition, demonstrating that the incretin-based therapy may be an important complementary treatment for the type 1 diabetic condition.

OBJECTIVE: To determine the incidence of sleep disorder at a follow-up examination from 1 to 4 years, in demented patients diagnosed at first visit, besides analyzing associated demographic and comorbidities characteristics. METHODS: A total of 122 elderly patients aged 60 years or older and diagnosed with dementia (Alzheimer and other) were followed in a reference geriatric center for dementia. The clinical protocols included interviews with patient and caregiver, complete physical examination, laboratory and imaging tests. Criteria for the diagnosis of sleep disorder included complain of insomnia from the patient or caregiver using the Neuropsychiatric Inventory nighttime. RESULTS: The incidence density of sleep disorder among dements was 18.7/100 person/years. The risk of developing sleep disorder within the frst and fourth years of follow-up was 9.8% and 50.9%, respectively. Multivariate Coxregression analysis revealed that educational level less than 8 years and report of aggressiveness at baseline were an independent predictor of sleep disorder, increased risk in 3.1 (95%CI: 1.30-9.22) and 2.1 times (95%CI: 1.16-4.17), respectively. CONCLUSION: The incidence of sleep disorder in demented patients was elevated, and was particularly associated to low educational level and aggressiveness at admission.

Seleção para características do crescimento pode ser feita após ajustadas funções de crescimento para todos os animais sob seleção. Porém, a curva de crescimento de fêmeas é influenciada pelas gestações e lactações subsequentes. Neste trabalho, métodos são descritos para selecionar uma função que ajuste a curva de crescimento de fêmeas bovinas considerando-se os efeitos de variação de peso corporal devido a gestação e a lactação. Foram utilizados dados provenientes do Instituto Agronômico do Paraná, constituindo-se de 15.271 observações de pesos de 103 fêmeas, sendo 43 Nelore, 10 F1 Guzerá x Nelore, 29 F1 Red Angus x Nelore, 5 F1 Marchigiana x Nelore e 16 F1 Simental x Nelore. Comparou-se o ajuste das funções de Brody, Gompertz, Logística, Von Bertalanffy e Michaelis-Menten reparametrizada (MMr) aos efeitos do ganho de peso em função da idade, sendo adicionado a todas as funções o ganho de peso durante a gestação e a perda de peso na lactação de fêmeas da raça Nelore e Cruzadas. Ao ajustar as funções, observou-se um Coeficiente de Determinação Múltiplo Ajustado variando entre 0,7552 e 0,9267. Observou-se o menor Quadrado Médio do Resíduo e o Desvio Médio Absoluto para a função de MMr. Para descrição da curva de crescimento em função da idade das fêmeas, deve ser ajustada a função de MMr.


Selection for growth characteristics can be done after fitting growth functions for all breeding animals. However, in the female population the growth curve is influenced by gestation and subsequent lactations. In this work methods are described to select a function that fits a growth curve of bovine females considering the varying effects of body weight due to gestation and lactation. Data from the Paraná Agronomic Institute (IAPAR) were used consisting of 15.271 weight observations of 103 females distributed as 43 Nelore, 10 F1 Guzerá x Nelore, 29 F1 Red Angus x Nelore, 5 F1 Marchigiana x Nelore and 16 F1 Simental x Nelore. The function adjustment of Brody, Gompertz, Logística, Von Bertalanffy and Michaelis-Menten reparametrized MMr were compared to the effects of weight gain as a function of age, adding to all functions the weight gain during gestation and the weight loss in the lactation of Nelore and crossbreed females. It was observed an Adjusted Coefficient of Multiple Determination varying from 0.7552 to 0.9267. Lower Residue Mean Square and Absolute Mean Deviation were observed for the MMr function. The MMr function must be adjusted in order to describe the growth curve as a function of the female age.

O câncer colorretal é responsável por cerca de 8.000 óbitos/ano no Brasil. Acredita-se que haja subestadiamento pós-operatório. O objetivo deste trabalho é pesquisar sobre linfonodo-sentinela em pacientes com adenocarcinoma de cólon. A amostra foi composta de 18 pacientes, todos com diagnóstico de adenocarcinoma de cólon, submetidos à laparotomia com injeção dos marcadores de linfonodos na subserosa peritumoral. RESULTADOS: a identificação intra-operatória de linfonodo-sentinela com os marcadores ocorreu em 16 (88,8 por cento) pacientes. O azul patente identificou linfonodos-sentinela em 72,2 por cento e o fitato marcado com tecnécio em 88,8 por cento. Obtiveram-se linfocintilografias do espécime cirúrgico removido em 15 pacientes. A sensibilidade global do método foi de 66,7 por cento e o falso negativo de 33,3 por cento. Depois do exame histológico com multissecção e imunoistoquímica de 11 pacientes, foi diagnosticada metástase em uma (9 por cento) ocorrência, sendo considerada ultra-estadiamento. CONCLUSÕES: pode-se afirmar que o procedimento é viável; o radiofármaco é mais eficaz; a linfocintilografia da peça cirúrgica é capaz de certificar a presença de captação de radiofármaco pelo linfonodo; a incidência de metástases linfonodais é, proporcionalmente, a mesma nos linfonodos-sentinela e não-sentinela; as técnicas de multissecção e imunoistoquímica contribuem para melhorar a acuidade diagnóstica de metástase linfonodal.


The colorectal cancer is responsible for 8.000 deaths a year in Brazil. It is believed that there is post operative sub staged. The objective of this study is to research on the sentinel lymph node in patients with colon cancer. The sample was composed by 18 patients, all of them with diagnose of cancer, undertaken to laparotomy with injection of the markers of lymph nodes in the subserosa peritumoral. RESULTS: intraoperative identification sentinel lymph nodes with the markers occurred in 16 (88,8 percent) patients. The patent blue dye identified sentinel lymph nodes in 13 (72,2 percent) and the radioisotopic in 16 (88,8 percent). Lymphoscintigraphy of surgical specimen were obtained from 15 patients. The global sensitivity of this method was of 66,7 percent and the false-negative of 33,3 percent. After the histological examination with multilevel section and immunohistochemical in 11 patients, one (9 percent) case of micrometastase was diagnosed being considered ultrastaging. CONCLUSIONS: It can be said that the procedure is viable; the radioisotope is more effective; the lymphoscintigraphy of the surgical specimen is capable of certifying the presence of absorption of the radioisotope by the lymph node; the incidence of lymph node metastases is, proportionally, the same as the one of the sentinel and non sentinel nodes; the techniques of the multilevel section and immunohistochemical contribute to improve the activity of the lymph node metastases diagnose.

RESUMO

É apresentado o caso de paciente de 49 anos, sexo feminino, com ecocardiograma e cateterismo cardíaco mostrando massa intra-atrial esquerda, sugerindo mixoma, encaminhada para avaliacão cardiológica devido à dispnéia. A paciente era ainda portadora de hepatoesplenomegalia esquistossomótica. Após preparo pré-operatório, que incluiu assistência hematológica, a paciente foi submetida a tratamento cirúrgico, com auxílio de circulacão extracorpórea. Apresentou sangramento aumentado no pós-operatório imediato, melhorando após transfusão de plaquetas. O estudo anatomopatológico revelou tratar-se de hemangioma capilar benigno.

RESUMO

No período de junho de 1988 a junho de 1989 foram realizadas 46 punçöes com agulha fina guiada pela ultra-sonografia em 41 pacientes. As punçöes destinavam-se a esclarecer a natureza das massas abdominais ou o estadiamento de tumores conhecidos. Tentou-se confirmar a posiçäo da agulha na massa pela ultra-sonografia. Após aspiraçäo, foram confeccionadas lâminas fixadas em álcool 95%. O órgäo mais puncionado foi o fígado, seguido pelo pâncreas, massas abdominais, rim e retroperitônio. A exceçäo de um hematoma subcapsular esplênico, näo houve complicaçöes imediatas, bem como infecçöes. Foram diagnosticadas 21 neoplasias, a maioria adenocarcinomas. Houve 9 falsos negativos, a maioria por falta de material adequado na aspiraçäo; destes, 7 ocorreram nos seis primeiros meses de estudo. Em um dos casos, a repetiçäo do procedimento foi diagnóstica. Näo houve falso positivo. Em 10 casos, o resultado foi verdadeiro negativo. A sensibilidade geral do método foi de 70%. Concluimos ser a biópsia com agulha fina guiada por ultra-sonografia um procedimento seguro e valioso no diagnóstico e estadiamento de tumores abdominais
