We analyzed the phylogenetic relationships of the genus Trisetacus using two genes [cytochrome c oxidase subunit I (COI) and D1-D2 region of 28S rDNA (D1-D2 28S)], a representive taxon sampling (nearly 40% of known diversity), and a large set of close and distant outgroups. Our analyses suggest the presence of a dichotomy between Trisetacus associated with Cupressaceae and Pinaceae. The following smaller molecular clades were found: Pin-1 (bud mites, twig sheath mites, bark gall mites, and endoparasitic mites from pinaceans), Pin-2 (needle sheath mites from pines), Pin-2a (putative Nearctic group of needle sheath mites), Pin-2b (putative Palearctic group of needle sheath mites), Cup-1 and 2 (bud, cone, seed mites and mites living under bark scales from cupressaceans). The monophyly of the recently proposed subgenus Brevithecus nested within clade Cup-2 was confirmed. Ancestral character reconstruction analyses recovered: (1) Pinaceae as the ancestral hosts of Nalepellidae and Trisetacus, (2) repetitive reductions of the spermathecal tube independently occurred in two lineages of Trisetacus from Cupressaceae, and (3) several mite habitats on host (galls, cones, twig sheaths, seeds, inside leaves, and under scales) are evolutionarily derived states, whereas living in buds or needle sheaths are ancestral states for Trisetacus clades Cup and Pin. Using confocal microscopy, we identified six basic types of the female internal genitalia of Trisetacus based on shapes of the spermatheca and spermathecal tube. These genitalic types are strongly correlated with lineages recovered by molecular phylogenetic analyses, suggesting that the female genital morphology is both evolutionarily conserved and is a factor influencing macroevolutionary patterns in this group of mites.

The genetic structure and diversity of species is determined by both current population dynamics and historical processes. Population genetic structure at the edge of the distribution is often expected to differ substantially from populations at the centre, as these edge populations are often small and fragmented. In addition, populations located in regions that have experienced repeated glaciations throughout the Pleistocene, may still carry imprints from the genetic consequences of frequent distribution shifts. Using chloroplast DNA sequences and nuclear microsatellite markers we studied the genetic structure of Epipactis atrorubens at the northern edge of its distribution. Contrary to populations in the centre of the distribution, populations at the northern range are regionally endangered as they are small and disjunct. Sequence data of 2 chloroplast loci and allelic data from 6 nuclear microsatellite markers were obtained from 297 samples from Finland, Estonia and Russia. We sought for genetic indicators of past population processes, such as post-glacial colonisation history of E. atrorubens. As expected, we observed low genetic variation, in terms of numbers of substitutions, haplotypes and alleles, and significant levels of differentiation, especially pronounced in the chloroplast DNA. These features suggest that the edge populations could be prone to extinction.

We present a review of Orchidaceae Juss. of the northern part of Kazakhstan, within the steppe, forest-steppe and semi-desert habitats of the country (Pavlodar, northern Kazakhstan, Kostanay, Akmola, Aktobe, West Kazakhstan, partially Karaganda and East Kazakhstan regions). The investigation is based on herbarium materials, literature data and field observations. We examined material from the following herbarium collections: LE, MW, TK, MHA, SVER, KUZ, ALTB, AA, NUR, KG, KSPI, NS, NSK, MOSP, ORIS, PPIU, totalling 288 herbarium specimens. The paper presents data in the form of revision, focusing on orchids of the northern part of Kazakhstan. It is accompanied by maps indicating localities, notes on habitat preferences, phenology and conservation status. A total of 25 species of 16 genera were recorded, of which eight are included in the Red Book of Kazakhstan (2014). According to our data, we propose to enlarge the number of protected orchids by adding the following nine species: Corallorhizatrifida, Epipactisatrorubens, Gymnadeniaconopsea, Hammarbyapaludosa, Herminiummonorchis, Liparisloeselii, Malaxismonophyllos, Neottiacamtschatea and Spiranthesaustralis. The most widespread species in the studied region are Dactylorhizaincarnata, D.umbrosa and Epipactispalustris. The rarest species (single locality only) are Epipactisatrorubens, E.helleborine, Epipogiumaphyllum, Hammarbyapaludosa and Herminiummonorchis.

In this article we present and discuss the main factors that threaten natural populations of Cypripedium calceolus (lady's slipper orchid) in Europe, and we propose conservation strategies and directions for protective actions of its population on a regional scale. European C. calceolus populations have decreased significantly in the last two decades, in both number and size. A key result of the present study is an evaluation of the effectiveness of the Natura 2000 network across the European Union (EU) countries. Northern and/or mountainous countries present higher percentages of potentially suitable areas within the Natura 2000 network. Finland and the United Kingdom are the exceptions to this rule. It is predicted that, due to global warming, the coverage of niches suitable for C. calceolus will decrease in countries in which now-healthy colonies exist. However, as plant species can occur in micro-sites with suitable environmental conditions (e.g., microclimate, vegetation, soil factors) which cannot be predicted as suitable at coarser spatial resolutions, conservation efforts should be focused on management of local healthy populations. For the effective protection of C. calceolus in Natura 2000 sites, the participation of experts in botany, including orchid biology, is necessary at several stages.
