RESUMO

The dissolution of soluble rocks (gypsum/anhydrite) beneath the Mosul Dam by water seepage has been observed upon the initial impoundment; consequently, several sinkholes have been manifested in the vicinity of the dam site. Traditional grouting has been envisaged as a potential remedy; however this measure has not eradicated the problem. The main purpose of this study is to overcome the solubility of the gypsum/anhydrite rocks using chemical grouts. Rock samples were acquired from the Fatha Formation outcrop and problematic layers of brecciated gypsum situated at varying depths beneath the Mosul Dam. Two commercially available liquid polymers, polyurethane (PU) and a mixture of acrylic and cement (ARC) were used to investigate their sealing performance in halting of the solubility of the rocks (gypsum/anhydrite). To simulate the dissolution phenomenon under the influence of artificial hydraulic pressure of the dam and the water flow in its abutments, two distinct laboratory models were devised. The outcomes from the experimental study on both untreated and treated samples revealed that the acrylic-cement composite (ARC) and polyurethane (PU) are influential polymers in halting the solubility of the gypsum rock samples under both factors of water pressure and high-velocity water flow.

BACKGROUND: Polycystic ovary syndrome (PCOS) is one of the most common endocrine disorders in reproductive-age women. The present study aimed to evaluate the effects of Rosa damascena (RD) extract in estradiol valerate (EV) induced polycystic ovary syndrome rats. METHODS: Adult female Wistar rats were divided into control (n = 12) and PCOS groups (n = 36). The PCOS model was induced using EV (4 mg/kg/day), which was confirmed in 6 rats in each control and PCOS group by observation of irregular estrous cycles in vaginal smears and ovarian multiple cystic. Then, the rest of the control group (n = 6) and PCOS rats (n = 30 in 5 divided groups) were treated orally for 28 days with metformin (MET) as a positive control (200 mg/kg/day) and RD extract (400, 800, and 1200 mg/kg/day, respectively). Body and ovary weights, biochemical and histological parameters, and expression of the IGF-1 gene were measured. RESULTS: Compared to the PCOS group, metformin and higher doses of RD extract (800 and 1200 mg/kg/day) significantly reduced BW, HOMA-IR, FBS, FINS, TG, LDL, TT, E2, LH, TC, and liver enzymes, and increased HDL and FSH levels. In addition, ovarian weight and CFs decreased, and the findings showed an increment in PFs, CLs, PAFs, AFs, and GFs. IGF-1 gene expression levels were significantly decreased (p < 0.001). CONCLUSION: RD extract seems to have the potential therapeutic effect of alleviating PCOS complications, and IGF-1 signaling may be involved in the beneficial effects of RD on PCOS.

Background: Male infertility is a growing health problem. It is proposed that infertility is associated with some metabolic abnormalities. Objectives: This study aimed to examine the prevalence of self-reported male infertility and related metabolic disturbances. Methods: This is a cross-sectional analysis of the Tehran Lipid and Glucose Study (TLGS). A total of 1526 males participated in the study. Logistic regression was used to examine metabolic factors associated with self-reported male infertility. Results: The total prevalence of self-reported male infertility was 6.42%. The mean (SD) body mass index (BMI) of participants among fertile and infertile males was 26.80 (3.93) and 26.92 (4.36), respectively. The majority of participants in both groups were in the age group of 40-50 years old. In the fully adjusted model, the odds of infertility were significantly increased by each unit increase in total cholesterol [TC; odds ratio (OR), 1.01; 95% CI, 1.01 - 1.01; P = 0.03] and hip circumference (HC; OR, 1.06; 95% CI, 1.00 - 1.12; P = 0.02), respectively. Conclusions: The prevalence of self-reported male infertility was 6.42%. Male infertility was positively associated with TC and HC, indicating that knowledge about these risks might assist health care professionals and governments in developing and executing measures to change the status quo.

Vitrification negatively affects the mitochondrial membrane potential (ΔΨm) in oocytes while also leading to increased reactive oxygen species (ROS), ATP depletion and induction of apoptosis in oocytes. Mitoquinone (MitoQ) is an antioxidant that protects mitochondrial membrane integrity from ROS. This study examined the effect of adding MitoQ to vitrification medium on mitochondrial function and embryo development in vitrified oocytes. Metaphase II (MII) stage oocytes were collected from NMRI mouse ovaries and preincubated for 20 min in a medium containing 0.02 µM of MitoQ. Next, oocytes were vitrified in medium supplemented with 0.02µM of MitoQ (treatment group). The control group was processed in the same way but without exposure to MitoQ. After warming, oocyte survival rate, ΔΨm, cytoplasmic ROS and glutathione (GSH) levels and gene expression levels (Bcl2, BAX, and caspase3) were measured. In addition, the vitrified oocytes were fertilized in-vitro to assess developmental competence. The results showed that MitoQ improved survival and ΔΨm in treated vitrified oocytes. Treated oocytes showed lower ROS levels and higher GSH levels than did the control group. Furthermore, mRNA expression of the Bax/ Bcl2 ratio and caspase3 were significantly lower in treated oocytes. These findings indicate that medium supplementation with 0.02 µM of MitoQ during vitrification can improve oocyte survival and developmental competency in mouse oocytes.
