RESUMO

Under the present climatic emergency, the environmental quality of freshwater reservoirs is an increasingly urgent topic as its deterioration threatens humans and ecosystems. It is evident that pollution by natural and anthropogenic contaminants must be avoided or reduced. The Lake of Cavazzo (NE Italy) is a natural perialpine basin which, from the mid-20th century, has sustained several anthropogenic impacts that added to the effects of the intense regional seismicity. Starting from 2015, in response to concerns raised by local authorities, a multidisciplinary investigation of the lake floor and sub-floor was conducted, including a geophysical survey and the collection of sediment cores. Two of them were studied to detect contamination by Polycyclic Aromatic Hydrocarbons (PAHs) and specific Persistent Organic Pollutants (POPs; i.e. PolyChlorinated Biphenyls - PCBs, PolyBrominated Diphenyl Ethers - PBDEs, and OrganoChlorine Pesticides - OCPs), and to verify the link with known anthropogenic stressors. Results were interpreted in light of previous studies suggesting modified conditions after the '50s, and recognized the effects of the 1976-1977 MW 6.5 seismic sequence in resuspending sediments within the basin. Analyzed pollutants defined a potential critical situation only for few OCPs, above all 2,4'- and 4,4'-DDT isomers. In addition, PBDEs were found at concentrations exceeding those of other heavily polluted alpine lakes. Mass movements (either seismic or human induced) have likely resuspended and transferred pollutants from shallower locations to the lake depocenter, showing the potential of re-exposing contaminated layers to biomagnification processes along the lacustrine food chain. Local inputs of pollutants prevail over distributed sources, suggesting a link with local agricultural or industrial activities. Indeed, works connected to the construction of the hydroelectric power plant in the '50s might have reworked local sediments perturbing their natural accretion. Results of this work might inspire similar studies in other problematic lacustrine areas that sustain both natural and anthropogenic stressors.

RESUMO

Coastal areas are under continuous and increasing pressure from different human activities. A mixture of contaminants (e.g. hydrocarbons, pesticides, persistent organic pollutants (POPs), emerging contaminants, and others), originating mainly from populated, industrialised and agricultural areas, can reach the marine environment through different means such as wastewater discharge, soil runoffs, leaching from agriculture, and volatilisation/deposition. In this context, marine sediments have increasingly been considered repositories for a variety of pollutants that can accumulate and be stored for long periods, acting as a secondary source of contaminants during subsequent dredging operation or vessel manoeuvring. Chemical and ecotoxicological analyses of sediments are routinely conducted to evaluate the potential hazard/risk to the environment, either on bulk sediment or elutriate. In general, sediment elutriates are commonly prepared according to ASTM Guide even if alternative protocols are proposed by USACE for the various condition that they have to represent. The goal of the present study was to determine if the toxicological properties of ASTMprepared elutriates are comparable to those obtained from the USACE protocol. Sediment coming from 3 harbours (Olbia, Cagliari, and Toulon), as part of the "Se.D.Ri.Port" Interreg Project, were processed to obtain elutriates according to ASTM Guide and USACE Dredging Elutriate protocol and tested with the sea urchin Paracentrotus lividus embryo development test. Moreover, the significance of different stirring times of water/sediment mixture (1 h, 3 h, and 24 h) was tested with both the ASTM and USACE protocol. In addition to the biological analysis, for each sediment sample, heavy metals concentration, granulometry, and organic matter were determined. Even if for the ports of Toulon and Cagliari, the ASTM and USACE elutriates showed comparable results with P. lividus bioassay, for the port of Olbia the two protocols showed different criticalities. Preliminary results show that for the site Olbia elutriates prepared with the USACE protocol resulted in higher toxicity than elutriates obtained with ASTM (p < 0.001). In conclusion, differences in preparation protocols appear to be significant and can lead to different results in biological testing. To overcome this problem and to obtain more reliable evaluations of risk to the environment, standardisation and regulation must be the next goals in sediment management procedure.

RESUMO

Despite the key role that areas close to the rivers Cau and Ngu Huyen Khê (Bac Giang Province, Northern Vietnam) play in the socio-economic development of Vietnam, poor information is available on the level of contaminants and their natural backgrounds in local soils and sediments. To partially fill this gap and to take into account for pressures and impacts on different zones and environment types (river sediments, crop fields, family wells, industrial and urban soils), the samples were collected and analyzed for fifteen trace elements at thirty sites distributed over the province. To overcome the lack of information on natural background and to identify the extent of the deviation from natural conditions, we coupled statistical analysis to contamination indices. The multivariate analysis was used to relate sediment chemical composition with a possible alteration from secondary inputs and to highlight those samples that most deviate from the distribution by category and are potentially more problematic. Geoaccumulation indexes and enrichment factors were calculated to discriminate between natural backgrounds and anthropic sources, SQGs were used for a screening evaluation of environmental risk in the study area. Results showed relatively high concentrations, sometimes exceeding international and national guidelines, and local sources could prevail over geogenic origins. Despite its significant natural contribution all over the study area, As evidenced some cases of anthropogenic contamination, similarly to that observed for Cd, Hg, and Zn. Their high concentrations may be a problem for public health, especially when found in family wells.

RESUMO

BACKGROUND: Developed countries have a high prevalence of vitamin D deficiency. In previous studies, 25(OH)D was predominantly measured by immunoassays. The present study assessed serum 25(OH)D in a very large Southern European outpatient cohort by liquid chromatography tandem mass spectrometry (LC-MS/MS). MATERIALS AND METHODS: 74,235 serum 25(OH)D results generated under routine conditions between 2015 and 2016 were extracted from the laboratory information system of the Department of Clinical Pathology at Bolzano Hospital (Italy). In 3801 cases, parathyroid hormone (PTH) was requested in parallel. Serum 25(OH)D was measured by a NIST-972 aligned commercial LC-MS/MS method. The distribution of serum 25(OH)D concentrations in males and females of different age groups, the prevalence of 25(OH)D2 and seasonal variability were studied. RESULTS: The average 25(OH)D concentration in the entire cohort was 68.6 nmol/L (7.5-1880 nmol/L). Females had a 7 nmol/L higher average 25(OH)D concentration than males, which increased significantly with age. 37.9 and 28.3% of males and females, respectively, had a deficient 25(OH)D concentration of < 50 nmol/L. 620 samples (0.84%) had measureable amounts of 25(OH)D2. In samples with a normal PTH, 25(OH)D was 11 nmol/L higher than in the entire cohort. Seasonal variation ranged between 20 and 30% and was most pronounced in young individuals. 25(OH)D2 remained constant throughout the year. CONCLUSION: Average serum 25(OH)D in South Tyrol is higher than in other parts of Europe. 25(OH)D and PTH show a continuous inverse relationship. Seasonal variation of serum 25(OH)D is an important aspect in young and middle-aged adults, but becomes less relevant in elderly subjects. 25(OH)D2 is of minor practical importance in South Tyrol.

RESUMO

The genetic dissection of root architecture and functions allows for a more effective and informed design of novel root ideotypes and paves the way to evaluate their effects on crop resilience to a number of abiotic stresses. In maize, limited attention has been devoted to the genetic analysis of root architecture diversity at the early stage. The difference in embryonic (including seminal and primary) root architecture between the maize reference line B73 (which mostly develops three seminal roots) and the landrace Gaspé Flint (with virtually no seminal roots) was genetically dissected using a collection of introgression lines grown in paper rolls and pots. Quantitative trait locus (QTL) analysis identified three QTLs controlling seminal root number (SRN) on chromosome bins 1.02, 3.07, and 8.04-8.05, which collectively explained 66% of the phenotypic variation. In all three cases, Gaspé Flint contributed the allele for lower SRN. Primary root dry weight was negatively correlated with SRN (r= -0.52), and QTLs for primary root size co-mapped with SRN QTLs, suggesting a pleiotropic effect of SRN QTLs on the primary root, most probably caused by competition for seed resources. Interestingly, two out of three SRN QTLs co-mapped with the only two known maize genes (rtcs and rum1) affecting the number of seminal roots. The strong additive effect of the three QTLs and the development of near isogenic lines for each QTL in the elite B73 background provide unique opportunities to characterize functionally the genes involved in root development and to evaluate how root architecture affects seedling establishment, early development, and eventually yield in maize.

RESUMO

The distinction between malignant mesothelioma and reactive mesothelial proliferation can be challenging both on histology and cytology. Recently, variants of the BRCA1-associated protein 1 (BAP1) gene resulting in nuclear protein loss were reported in hereditary and sporadic mesothelioma. Using immunohistochemistry, we evaluated the utility of BAP1 expression in the differential diagnosis between mesothelioma and other mesothelial proliferations on a large series of biopsies that included 212 mesotheliomas, 12 benign mesothelial tumors, and 42 reactive mesothelial proliferations. BAP1 stain was also performed in 70 cytological samples (45 mesotheliomas and 25 reactive mesothelial proliferations). BAP1 was expressed in all benign mesothelial tumors, whereas 139/212 (66%) mesotheliomas were BAP1 negative, especially in epithelioid/biphasic compared with sarcomatoid/desmoplastic subtypes (69% vs 15%). BAP1 loss was homogeneous in neoplastic cells except for two epithelioid mesotheliomas showing tumor heterogeneity. By fluorescence in situ hybridization, BAP1 protein loss was paralleled by homozygous deletion of the BAP1 locus in the vast majority of BAP1-negative tumors (31/41, 76%), whereas 9/10 BAP1-positive mesotheliomas were normal. In biopsies interpreted as reactive mesothelial proliferation BAP1 loss was 100% predictive of malignancy, as all 6 cases subsequently developed BAP1-negative mesothelioma, whereas only 3/36 (8%) BAP1-positive cases progressed to mesothelioma. On cytology/cell blocks, benign mesothelial cells were invariably positive for BAP1, whereas 64% of mesotheliomas showed loss of protein; all 6 cases showing BAP1 negativity were associated with histological diagnosis of BAP1-negative mesothelioma. BAP1 stain also showed utility in the differential of mesothelioma from most common pleural and peritoneal mimickers, such as lung and ovary carcinomas, with specificity and sensitivity of 99/70% and 100/70%, respectively. Our results show that BAP1 protein is frequently lost in mesothelioma, especially of epithelioid/biphasic subtype and is commonly associated with homozygous BAP1 deletion. BAP1 immunostain represents an excellent biomarker with an unprecedented specificity (100%) in the distinction between benign and malignant mesothelial proliferations. Finding BAP1 loss in mesothelial cells should prompt to immediately reevaluate the patient; moreover, it might be useful in mapping tumor extent and planning surgical resection.

RESUMO

Contamination levels by plastic debris, trace elements and persistent organic pollutants were assessed and related to macrobenthic diversity within soft bottoms of Grand Harbour (Malta, Central Mediterranean). Sediment toxicity was evaluated by ecotoxicological method, deploying Bacteria (Vibrio fischeri), Echinodermata (Paracentrotus lividus) and Crustacea (Corophium orientale). Univariate analysis (Pearson's test) was used to test relationships between biodiversity indices, pollutants and grain size. A multivariate approach (PERMANOVA) was applied to investigate for any significant differences among sampling stations concerning plastic abundances and to test the relationship between infaunal abundances and pollutant concentrations (the BIOENV test). Significant differences in the plastic abundances were found between sampling stations. The lowest value for Shannon-Wiener biodiversity index was associated to the highest sediment pollution level. Multivariate analyses suggest that MBT and TBT were factors that most influenced macrozoobenthic abundance and biodiversity. The bivalve Corbula gibba and the introduced polychaete Monticellina dorsobranchialis were the most abundant found species.

RESUMO

OBJECTIVE: The aim of this study was to assess the cultural adaptation and validation in Italian of the Seated Postural Control Measure for Adults 2.0 (SPCMA 2.0). METHODS: The original scale was translated and culturally adapted from French to Italian using the "Translation and Cultural Adaptation of Patient Reported Outcomes Measures-Principles of Good Practice" guidelines. Its internal consistency and test-retest reliability were examined. Its concurrent validity was evaluated using Pearson correlation coefficients with the Italian version of the Wheelchair use Confidence Scale and Wheelchair Skills Test 4.2. RESULTS: Fifty-nine people were evaluated and re-evaluated after 48 h. Most of the items and subscale totals were stable in the 2 evaluations as they reported an intraclass correlation coefficient value of >0.77. The test-retest analysis of the dynamic evaluation was performed on the same patients 48 h apart. The analysis for construct validity showed statistically significant correlations with Wheelchair use Confidence Scale and Wheelchair Skills Test 4.2. CONCLUSIONS: Seated Postural Control Measure for Adults 2.0 is one of the few tools that allow researchers to perform a quantitative and standardized posture assessment in a cost-effective and time-saving way. Furthermore, it has been demonstrated that it is an easy-to-administer scale and requires readily available tools. The limitations of this study highlighted above and the need to use quantitative and qualitative tools in clinical practice imply the need to conduct future studies.

RESUMO

Assessing pollution levels and trends in heavily impacted environments is important but hardly achievable due to the difficulty of recovering suitable undisturbed sediment records. An integrated approach is here presented to solve this kind of problem. It was adopted in the Augusta Bay (Italy) for the study of Hg historical inputs and present trends. Archive information on dredging and mud disposal, together with bathymetry and high-resolution seismic profiles, were used to identify suitable sampling sites. Undisturbed sediment cores were collected in the port and bay. Sediments were analyzed for depth distributions of radiotracers ((210)Pb and (137)Cs), Hg, and main sediment parameters (magnetic susceptibility, grain size, dry bulk density, mineralogy, and organic carbon and nitrogen contents). Hexachlorobenzene (HCB) was also analyzed as an additional time tracer, since its production history in the area was well-known. Results show that peak Hg contamination (up to 575 mg·kg(-1)) was reached in the 1970s. Technological improvements and waste treatment in the following years determined a consistent decrease, but high concentrations still affect surficial sediments (0.25-92 mg·kg(-1)). Hg-HCB correlation suggests that this situation is likely the effect of resuspension and redistribution of deep sediments by dredging and naval traffic.

RESUMO

Maize ear fasciation originates from excessive or abnormal proliferation of the ear meristem and usually manifests as flattened multiple-tipped ear and/or disordered kernel arrangement. Ear prolificacy expresses as multiple ears per plant or per node. Both ear fasciation and prolificacy can affect grain yield. The genetic control of the two traits was studied using two recombinant inbred line populations (B73 × Lo1016 and Lo964 × Lo1016) with Lo1016 and Lo964 as donors of ear fasciation and prolificacy, respectively. Ear fasciation-related traits, number of kernel rows (KRN), ear prolificacy and number of tillers were phenotyped in multi-year field experiments. Ear fasciation traits and KRN showed relatively high heritability (h 2 > 0.5) except ratio of ear diameters. For all ear fasciation-related traits, fasciation level positively correlated with KRN (0.30 ≤ r ≤ 0.68). Prolificacy and tillering were not correlated and their h 2 ranged from 0.41 to 0.78. QTL mapping identified four QTLs for ear fasciation, on chromosomes 1 (two QTLs), 5 and 7, the latter two overlapping with QTLs for number of kernel rows. Notably, at these QTLs, the Lo1016 alleles increased both ear fasciation and KRN across populations, thus showing potential breeding applicability. Four and five non-overlapping QTLs were mapped for ear prolificacy and tillering, respectively. Two ear fasciation QTLs, qFas1.2 and qFas7, overlapped with fasciation QTLs mapped in other studies and spanned compact plant2 and ramosa1 candidate genes. Our study identified novel ear fasciation loci and alleles positively affecting grain yield components, and ear prolificacy and tillering loci which are unexpectedly still segregating in elite maize materials, contributing useful information for genomics-assisted breeding programs.

RESUMO

This data article presents the dataset collected for selected organic pollutants in the framework of a larger research project aimed at assessing the effects of different environmental stressors (natural and anthropogenic) in sediments of the Lake of Cavazzo, a basin of glacial origin located in a seismically active region of the Italian Eastern Alps. Information relative to sampling strategy and operations, location of sampling sites, sedimentary chronological benchmarks, and profiles of RGB (Red-Green-Blue) color code determined from high resolution photos taken at cores CAV-04 and CAV-06 are reported, together with analytical data for 15 polycyclic aromatic hydrocarbons, 21 polychlorinated biphenyls' congeners (including the non-Aroclor CB-11), 14 polybrominated diphenyl ethers' congeners, and 22 organochlorine pesticides, whose concentrations were determined by Gas Chromatography coupled both to Low-Resolution and High-Resolution Mass Spectrometry. Interpretation of this dataset is fully discussed in the companion article by Pizzini et al. (2022) and relys on the multi-proxy analysis of sediment samples presented in Polonia et al. (2021) that highlighted lake stratigraphy and major changes occurring at a decadal scale since the 1950s.

RESUMO

Kidney transplant recipients are a vulnerable population at risk of a life-threatening COVID-19 infection with an incidence of death four-times higher than in the general population. The availability of mRNA COVID-19 vaccines has dramatically changed the fate of this infection also within this fragile population. Transplanted patients have an impaired immunological response also to mRNA vaccines. In March 2021, however, we started a vaccination campaign. These preliminary results show that both the incidence of death and of hospitalization dropped from 13% to 2.4% and from 45% to 12.5% compared to the previous outbreaks reported by our group. In univariate analysis, two variables were associated with an increased risk of hospitalization: older age and dyspnea (p = 0.023, p < 0.0001, respectively). In multivariate analysis, dyspnea (p < 0.0001) and mycophenolate therapy (p = 0.003) were independently associated with the risk of hospitalization. The association was even stronger when the two variables were combined (p < 0.0001). Vaccinations did not reduce the incidence of COVID-19 infections among our transplanted patients, but provided certain protection that was associated with a significantly better outcome for this infection.

RESUMO

We describe the application of complexity reduction of polymorphic sequences (CRoPS(®)) technology for the discovery of SNP markers in tetraploid durum wheat (Triticum durum Desf.). A next-generation sequencing experiment was carried out on reduced representation libraries obtained from four durum cultivars. SNP validation and minor allele frequency (MAF) estimate were carried out on a panel of 12 cultivars, and the feasibility of genotyping these SNPs in segregating populations was tested using the Illumina Golden Gate (GG) technology. A total of 2,659 SNPs were identified on 1,206 consensus sequences. Among the 768 SNPs that were chosen irrespective of their genomic repetitiveness level and assayed on the Illumina BeadExpress genotyping system, 275 (35.8%) SNPs matched the expected genotypes observed in the SNP discovery phase. MAF data indicated that the overall SNP informativeness was high: a total of 196 (71.3%) SNPs had MAF >0.2, of which 76 (27.6%) showed MAF >0.4. Of these SNPs, 157 were mapped in one of two mapping populations (Meridiano × Claudio and Colosseo × Lloyd) and integrated into a common genetic map. Despite the relatively low genotyping efficiency of the GG assay, the validated CRoPS-derived SNPs showed valuable features for genomics and breeding applications such as a uniform distribution across the wheat genome, a prevailing single-locus codominant nature and a high polymorphism. Here, we report a new set of 275 highly robust genome-wide Triticum SNPs that are readily available for breeding purposes.

RESUMO

Trace element concentrations have been measured in soil and sediment samples taken from the Thua Thien Hue (TT-H) Province and the Tam Giang-Cau Hai (TG-CH) Lagoon in 2002 and 2004. Results show that the lagoon is only slightly contaminated by elements such as Ag, Cd, Cr, Cu, Ni, Pb and Zn, whereas As, probably naturally enriched in this Asian region, reaches values above the lowest internationally accepted sediment quality guidelines. Concentrations in soils are ascribable to natural sources and distributions in the TG-CH Lagoon are mainly influenced by river inputs, with Ag and Cd undergoing estuarine desorption processes. However, concentration-depth profiles of most trace elements in sediments show a recent increasing trend that might be linked to the economic development of the area. The comparison of porosity and (210)Pb depth profiles (confirmed also by Cd and As) from repeated samplings of the same core locations in the TG-CH Lagoon seems to account for a loss of 5-10 cm of surficial sediment that took place during the time interval between the two samplings. It is hypothesized that extreme meteorological events (e.g. a major typhoon that hit the area in November 2003) could be responsible for such sediment displacement.

RESUMO

Polychlorinated biphenyls (PCBs) were measured in two dated salt marsh cores of the Venice Lagoon to assess their input chronology and to evaluate the importance of atmospheric deposition as a source. Sampling sites were chosen in order to evidence the differences between areas located leeward and windward with respect to inputs originating in both the city of Venice and the industrial area. Concentrations of PCB indicators (0.13-15.6 ng g⁻¹) increased gradually from the 1930s, reached maxima from the 1950s to the late 1970s, and then decreased. PCB loadings to marshes are driven by both the atmospheric deposition and the resuspension of subtidal sediments, this latter being more important for heavier congeners. The downwind marsh recorded higher fluxes (0.06-9.72 ng cm⁻² year⁻¹) than the upwind one (0.01-0.53 ng cm⁻² year⁻¹). Recent fluxes are rather consistent with bulk deposition measurements. A higher contribution of CB-101 and CB-118 was detected in the intermediate layers of the downwind site, suggesting a different PCB source for the corresponding time interval. In the other marsh, PCBs showed a rather constant composition at all levels (mostly CB-153, CB-138 and CB-180), accounting for a regional influence. Deep layers showed an enrichment of higher chlorinated congeners at both sites, whereas recent samples conserve the patterns typical of surficial and subsurficial subtidal sediments. The scientific approach adopted in this research can be considered as a sort of methodological procedure for the determination of fluxes and pathways of PCBs through the study of marsh cores.

RESUMO

The present dataset was collected to evaluate the environmental stressors on a lacustrine basin in the Eastern Alps of glacial origin that has been affected in recent years by natural and anthropogenic events such as the construction of a hydroelectric power plant and a series of strong earthquakes during 1976-1977. We collected sediment cores in different sites from the lake margins to the depocenter and performed a multiproxy analysis of sediment sample to highlight lake stratigraphy and major changes occurring at a decadal scale (Polonia et al., [1]). The integrated analyses of sedimentological, geochemical, isotopic, mineralogical and micropaleontological analyses aimed at reconstructing changes in sediment composition and define the triggering mechanisms of altered environmental conditions. The dataset demonstrates that evaluating ex post the effects of artificial modification in a natural environment during relatively long time spans (decades) can provide important insights for managing and protection strategies in similar environments worldwide.

RESUMO

A previous study on maize F(2:3) families derived from Lo964xLo1016 highlighted one QTL in bin 1.06 (hereafter named root-yield-1.06) affecting root and agronomic traits of plants grown in well-watered (WW) and water-stressed (WS) conditions. Starting from different F(4) families, two pairs of near isogenic lines (NILs) were developed at root-yield-1.06. The objective of this study was to evaluate root-yield-1.06 effects across different water regimes, genetic backgrounds, and inbreeding levels. The NILs per se and their crosses with Lo1016 and Lo964 were tested in 2008 and 2009 near to Bologna, with the well-watered (WW) and water-stressed (WS) treatments providing, on average, 70 mm and 35 mm of water, respectively. For NILs per se, the interactions QTL x water regime and QTL x family were negligible in most cases; the QTL additive effects across families were significant for several traits, especially root clump weight. For NILs crosses, analogously to NILs per se, the interactions were generally negligible and the additive effects across water regimes and families were significant for most traits, especially grain yield. A meta-analysis carried out considering the QTLs described in this and previous studies inferred one single locus as responsible for the effects on roots and agronomic traits. Our results show that root-yield-1.06 has a major constitutive effect on root traits, plant vigour and productivity across water regimes, genetic backgrounds, and inbreeding levels. These features suggest that root-yield-1.06 is a valuable candidate for cloning the sequence underlying its effects and for marker-assisted selection to improve yield stability in maize.

RESUMO

This data article presents the datasets obtained during the years 2011-2012 in the Provinces of Bac Giang and Bac Ninh (Northern Vietnam) in the framework of an International Cooperation project aimed at the restoration and preservation of natural resources together with the improvement of life conditions of the local population. All information relative to sampling strategy, location and description of the studied sites are reported, together with analytical data (i.e. grain size, Total Organic Carbon, Organic Nitrogen, C/N ratio, δ13C, PolyChlorinated Biphenyls, Polycyclic Aromatic Hydrocarbons, PolyBrominated Diphenyls Ethers, PolyChlorinated Dibenzo-p-Dioxins and Furans, OrganoChlorine Pesticides, Total Petroleum Hydrocarbons). In addition, maximum admitted levels, quality standards and emission limits set by the Vietnamese environmental regulations are reported in comparison to Italian ones for the following parameters and environmental compartments: noise, soils, air, drinking and groundwaters, sediments. Interpretation of this dataset is fully discussed in Giuliani et al. (2019) [1].

RESUMO

In situ toxicity tests represent a technique rarely performed owing to the lack of standard methodologies or to technical or economic problems. Nevertheless, its application would allow for a more realistic interpretation of pollution effects than those obtained by standard laboratory tests. The goal of this study is to develop and validate a specific exposure chamber for in situ exposition of Paracentrotus lividus embryos to obtain a defined methodology to perform the sea urchin embryo toxicity test in field conditions. After a first part of the study to verify the feasibility of the test chamber and the methodology, this approach was used as a tool to investigate whether the cruise ship "Costa Concordia" shipwrecked on Giglio Island (Tuscany, Italy), could have acted as a source of pollution. The results obtained for in situ tests showed, on average, percentages of normal embryos lower than those obtained in laboratory conditions and a greater sensitivity than for those obtained in the laboratory owing to the time-integration of results. Thus the exposure chamber and the in situ methodology so far developed would appear to be suitable tools for future application in the environmental quality evaluation of marine waters.

RESUMO

Major and trace element, PAH, and PCB concentrations were measured in surface sediments and particles from sediment traps collected in the First and Second Basin of the Mar Piccolo (Gulf of Taranto) in two periods (June-July and August-September, 2013). The aim of the study was to evaluate pollution degree, sediment transport and particle redistribution dynamic within the area. Results confirm the higher contamination of sediments from the First Basin observed by previous researches, particularly for Cu, Hg, Pb, total PAHs, and total PCBs. Advective transport from the First to the Second Basin appears to be the leading transfer mechanism of particles and adsorbed contaminants, as evidenced by measured fluxes and statistical analyses of contaminant concentrations in surficial sediments and particles from sediment traps. Long-range selective transports of PAHs and microbial anaerobic degradation processes for PCBs have been also observed. These results are limited to a restricted time window but are consistent with the presence of transport fluxes at the bottom of the water column. This mechanism deserves further investigation and monitoring activities, potentially being the main responsible of pollutant delivering to the less contaminated sectors of the Mar Piccolo.
