RESUMO

The toxicity of a range of inorganic (Ag, Cu, Ni, Al(2)O(3), SiO(2), TiO(2) and ZrO(2)) nanoparticles (NP) and their corresponding metal salt or bulk metal oxide were screened for toxicity toward the earthworm Eisenia fetida using the limit-test design (1000 mg/kg). This study provides the first ecotoxicological life history trait data on earthworms for each these NPs, as well as for AgNO(3), Al(2)O(3), SiO(2), TiO(2) and ZrO(2). Significant effects were observed on survival for AgNO(3) (2.5% of controls), CuCl(2) (17.5% of controls) and NiCl(2) (32.5% of controls) and on reproduction (AgNO(3), CuCl(2), NiCl(2), Ag-NP, Cu-NP, TiO(2)-NP); with total reproductive failure in both silver treatments. Ag-NP, Cu-NP and TiO(2)-NP were the only NPs that caused toxic effects to E. fetida. The toxicity could not be singularly related to particle size or zeta potential or to the inherent element constituting the NPs (e.g. Ag).

RESUMO

To generate rough surfaces in Monte Carlo simulations, we use the 2 + 1 solid-on-solid model of deposition with rapid transient diffusion of newly arrived atoms supplied at glancing angle. The surfaces generated are employed to scrutinize the effect of surface roughness on adsorption of globular and anisotropic rodlike proteins. The obtained results are compared with the available experimental data for Ta deposition at glancing angle and for the bovine serum albumin and fibrinogen uptake on the corresponding Ta films.

It is well established in the field of biomaterials that hydroxyapatite (HA) may provide interesting osteoconductive properties. In this study, we investigated the osseointegrational effect of a 50/50 vol % composite of HA nanoparticles and poly-D,L-lactic acid (PDLLA) coated on model titanium bone implants in an in vivo animal model. The aim is to evaluate how the addition of HA to PDLLA may improve the bone formation and initial fixation of the implant. Two titanium implants coated with the PDLLA/HA composite and pure PDLLA, respectively, were implanted bilaterally in proximal part of humeri with a 2-mm peri-implant gap in 10 sheep. After 12 weeks, the remains of the coatings were present on 20.3 and 19.8% of PDLLA/HA composite- and PDLLA-coated implants, respectively. It was observed that newly formed bone (39.3%) and fibrous tissue (58.3%) had replaced the PDLLA/HA composite, whereas pure PDLLA was replaced almost completely by fibrous tissue (96.2%). Consequently, the PDLLA/HA composite-coated implants were better fixated as confirmed by push-out tests. Using quantification of peri-implant tissue and implant fixation as parameters, the present findings, therefore, clearly reveal that the addition of nanoparticulate HA to a PDLLA coating on titanium implants increases osseointegration.

This work introduces a novel chitosan-based siRNA nanoparticle delivery system for RNA interference in vitro and in vivo. The formation of interpolyelectrolyte complexes between siRNA duplexes (21-mers) and chitosan polymer into nanoparticles, ranging from 40 to 600 nm, was shown using atomic force microscopy and photon correlation spectroscopy. Rapid uptake (1 h) of Cy5-labeled nanoparticles into NIH 3T3 cells, followed by accumulation over a 24 h period, was visualized using fluorescence microscopy. Nanoparticle-mediated knockdown of endogenous enhanced green fluorescent protein (EGFP) was demonstrated in both H1299 human lung carcinoma cells and murine peritoneal macrophages (77.9% and 89.3% reduction in EGFP fluorescence, respectively). In addition, Western analysis showed approximately 90% reduced expression of BCR/ABL-1 leukemia fusion protein while BCR expression was unaffected in K562 (Ph(+)) cells after transfection using nanoparticles containing siRNA specific to the BCR/ABL-1 junction sequence. Effective in vivo RNA interference was achieved in bronchiole epithelial cells of transgenic EGFP mice after nasal administration of chitosan/siRNA formulations (37% and 43% reduction compared to mismatch and untreated control, respectively). These findings highlight the potential application of this novel chitosan-based system in RNA-mediated therapy of systemic and mucosal disease.
