RESUMO

The synthesis of linear symmetric ethynyl- and acetylide-amidinates of the coinage metals is presented. Starting with the desilylation of the complexes [{Me3 SiC≡CC(NDipp)2 }2 M2 ] (Dipp=2,6-diisopropylphenyl) (M=Cu, Au) it is demonstrated that this compound class is suitable to serve as a versatile metalloligand. Deprotonation with n-butyllithium and subsequent salt metathesis reactions yield symmetric tetranuclear gold(I) acetylide complexes of the form [{(PPh3 )AuC≡CC(NDipp)2 }2 M2 ] (M=Cu, Au). The corresponding Ag complex [{(PPh3 )AuC≡CC(NDipp)2 }2 Ag2 ] was obtained by a different route via metal rearrangement. All compounds show bright blue or blue-green microsecond long phosphorescence in the solid state, hence their photophysical properties were thoroughly investigated in a temperature range of 20-295 K. Emission quantum yields of up to 41 % at room temperature were determined. Furthermore, similar emissions with quantum yields of 15 % were observed for the two most brightly luminescent complexes in thf solution.

Highly alkaline hydrogels that allow the restoration of alkaline buffer in cementitious materials can be obtained from diallyldimethylammonium hydroxide. The latter must be prepared in dilute solutions and polymerised at ambient temperatures in order to avoid decomposition. Using methacrylamide as a neutral co-monomer capable of forming hydrogen bonds, the rheological properties of the gels can be adjusted over a wide range; e.g. the viscosity increases a thousandfold from 0.35 Pa s to >350 Pa s by using 10 mol% methacrylamide. For the proof of principle experiments, gels with 9 mol% methacrylamide were used, which contain approx. 1.6 mol hydroxide ions per kg gel. Ion exchange between this and a neutral chloride containing gel provided an apparent diffusion coefficient of 4.12 × 10-7 m2 s-1 for the hydroxide ions and confirmed the transport of chloride ions into the alkaline gel. The re-alkalisation was tested on fully carbonated mortar prisms made from Portland cement. Re-alkalisation of the mortar was confirmed by the phenolphthalein test according to DIN EN 14630:2007-01 and by a control experiment with pure calcium carbonate using IR spectroscopy.

Binary diblock copolymers and corresponding ternary miktoarm stars are studied at oil-water interfaces. All polymers contain oil-soluble poly(propylene oxide) PPO, water-soluble poly(dimethylaminoethyl methacrylate) PDMAEMA and/or poly(ethylene oxide) PEO. The features of their Langmuir compression isotherms are well related to the ones of the corresponding homopolymers. Within the Langmuir-trough, PEO-b-PPO acts as the most effective amphiphile compared to the other PPO-containing copolymers. In contrast, the compression isotherms show a complexation of PPO and PDMAEMA for PPO-b-PDMAEMA and the star, reducing their overall amphiphilicity. Such complex formation between the blocks of PPO-b-PDMAEMA is prevented in bulk water but facilitated at the interface. The weakly-interacting blocks of PPO-b-PDMAEMA form a complex due to their enhanced proximity in such confined environments. Scanning force microscopy and Monte Carlo simulations with varying confinement support our results, which are regarded as compliant with the mathematical random walk theorem by Pólya. Finally, the results are expected to be of relevance for e.g. emulsion formulation and macromolecular engineering.

RESUMO

The versatile metalloligand [{HCCC(NDipp)2}2Au2] (dipp = 2,6-diisopropylphenyl) was converted into early-late heterotetrametallic complexes [{ClCp2MCCC(NDipp)2}2Au2] (M = Ti, Zr). These compounds show photoluminescence with either remarkably different (Ti) or similar (Zr) features as compared to related solely coinage metal containing acetylide amidinate complexes.

Superabsorbent polymers (SAP) are of major interest as materials to control the cement hydration process. The swelling behavior of the SAPs significantly influences the performance of the resulting concrete by slowly releasing polymer-bound water in order to maintain a consistent w/c value. A round-robin test conducted by the RILEM Technical Committee 260-RSC showed that the same batch of polymer can lead to large deviations in concrete performance and this was assumed to originate in different storage conditions of the SAP. In this contribution the change in the performance of two SAPs, a crosslinked poly(acrylate) and a crosslinked poly(acrylate-co-acrylamide), was assessed after ageing in standard climate, at 50 °C, and under UV irradiation. During storage in standard climate or 50 °C, ageing led to dehydration of the SAP, and this subsequently led to a higher water uptake during swelling. By contrast, UV irradiation reduced the water uptake, most likely as a result of photo-crosslinking. Dynamic water vapor sorption experiments indicated a strong dependence of the water uptake on both the ambient humidity and the temperature. As a result, cement mixtures containing SAP must be calculated on the dry mass of the SAP rather than the actual weight on site. A standard procedure of how to pack and handle SAP to be used in concrete is also provided.

Autologous peripheral blood stem/progenitor cell transplantation (APBSCT) has been investigated as a potential therapeutic option to improve outcome in patients with acute myelogenous leukemia (AML). However, its optimal role in treatment for adults in remission has not been clearly established. We performed a retrospective analysis on 45 patients aged 21 to 73 years (median 51 years) with de novo AML who underwent APBSCT stratified by age, complete remission status, and cytogenetic risk. The 5-year disease-free survival (DFS) for all patients was 33.9% (95% confidence interval [CI], 20.1%-53.7%) and overall survival (OS) was 43.6% (CI, 29.2%-62.8%). For patients under the age of 60 years, the 5-year DFS for intermediate and high cytogenetic risk was 53.3% (CI, 23.5%-85.6%) and 50.0% (CI, 16.1%-100.0%); the 5-year OS for patients under the age of 60 years with low, intermediate, and high cytogenetic risk was 80.0% (CI, 40.0%-100.0%), 60.0% (CI, 31.2%-90.7%), and 75.0% (CI, 39.0%-100.0%), respectively. For patients over the age of 60 years, the 5-year DFS and OS for intermediate cytogenetic risk was 21.4% (CI, 7.9%-58.4%) and 21.4% (CI, 7.9%-58.4%). The DFS and OS of these patients are comparable to the historic survival of those who underwent allogeneic stem cell transplantation when adjusted by age. In addition, there was no treatment-related mortality (TRM). We conclude that APBSCT is a reasonable and safe intensive consolidation for patients with AML who do not have a suitable HLA-matched donor.

A time-saving phase-diagram screening is introduced for the self-assembly of miktoarm star polymers with different arm numbers for the insoluble part. Agreeing with theory, all conventional micellar morphologies (spherical star-like micelles, cylindrical micelles and vesicles) can be accessed by adjusting the average arm number when blending miktoarm stars with diblock copolymers (at constant arm/block lengths). Additionally, a rare clustered vesicle phase is detected. Hence, this approach permits an easy tuning of the equilibrium morphology and the size of the solvophobic domain. Such screening by scattering, ultracentrifugation, and electron microscopy techniques assists the targeted synthesis of miktoarm stars with a well-defined arm number, aimed at the morphology control of the nanostructures without blending. Specifically, we demonstrate a systematic variation of all classical micellar morphologies based on interpolyelectrolyte complexes (IPECs), consisting of a water-insoluble part formed by electrostatically coupled poly(styrenesulfonate) chains/quaternized poly(2-(dimethylamino)ethyl methacrylate) blocks, being stabilized by hydrophilic poly(ethylene oxide) blocks.

Near-equilibrium stimulus-responsive polymers have been used extensively to introduce morphological variations in dependence of adaptable conditions. Far-less-well studied are triggered transformations at constant conditions. These require the involvement of metastable states, which are either able to approach the equilibrium state after deviation from metastability or can be frozen on returning from nonequilibrium to equilibrium. Such functional nonequilibrium macromolecular systems hold great promise for on-demand transformations, which result in substantial changes in their material properties, as seen for triggered gelations. Herein, a diblock copolymer system consisting of a hydrophilic block and a block that is responsive to both pressure and temperature, is introduced. This species demonstrates various micellar transformations upon leaving equilibrium/nonequilibrium states, which are triggered by a temperature deflection or a temporary application of hydrostatic pressure.

PURPOSE: We evaluated the influence of RAS mutation status on the treatment effect of panitumumab in a prospective-retrospective analysis of a randomized, multicenter phase III study of panitumumab plus fluorouracil, leucovorin, and irinotecan (FOLFIRI) versus FOLFIRI alone as second-line therapy in patients with metastatic colorectal cancer (mCRC; ClinicalTrials.gov, NCT0039183). EXPERIMENTAL DESIGN: Outcomes were from the study's primary analysis. RAS mutations beyond KRAS exon 2 (KRAS exons 3, 4; NRAS exons 2, 3, 4; BRAF exon 15) were detected by bidirectional Sanger sequencing in wild-type KRAS exon 2 tumor specimens. Progression-free survival (PFS) and overall survival (OS) were coprimary endpoints. RESULTS: The RAS ascertainment rate was 85%; 18% of wild-type KRAS exon 2 tumors harbored other RAS mutations. For PFS and OS, the hazard ratio (HR) for panitumumab plus FOLFIRI versus FOLFIRI alone more strongly favored panitumumab in the wild-type RAS population than in the wild-type KRAS exon 2 population [PFS HR, 0.70 (95% confidence interval [CI], 0.54-0.91); P = 0.007 vs. 0.73 (95% CI, 0.59-0.90); P = 0.004; OS HR, 0.81 (95% CI, 0.63-1.03); P = 0.08 vs. 0.85 (95% CI, 0.70-1.04); P = 0.12]. Patients with RAS mutations were unlikely to benefit from panitumumab. Among RAS wild-type patients, the objective response rate was 41% in the panitumumab-FOLFIRI group versus 10% in the FOLFIRI group. CONCLUSIONS: Patients with RAS mutations were unlikely to benefit from panitumumab-FOLFIRI and the benefit-risk of panitumumab-FOLFIRI was improved in the wild-type RAS population compared with the wild-type KRAS exon 2 population. Thrombopoietin (TPO) and its receptor c-Mpl are essential in the regulation of the hematopoietic stem and progenitors cells as well as for the differentiation of megakaryocytes into mature platelets. Once TPO binds to its receptor, an intracellular signaling process is initiated through Janus kinase (JAK-2)-induced phosphorylation of the c-Mpl intracellular domain. Although some protein mediators that transmit the effects of TPO have been identified, many remain undiscovered. Using an unbiased approach with peptide microarrays that contained virtually every Src Homology (SH)2 and Phosphotyrosine Binding (PTB) domains in the human genome, we discovered a previously unreported interaction between c-Mpl at phospho-Tyrosine631 (pY 631) and Tensin2, a protein for which limited information is available. Confirming the findings of the microarrays, we discovered that Tensin2 co-precipitates with a pY 631 bearing peptide. Furthermore, we found that Tensin2 becomes phosphorylated in a TPO dependent manner. The functional consequence of Tensin2 was tested via knockdown of Tensin2, which dramatically decreased TPO-dependent cellular proliferation of UT7-TPO cell line as well as their activation of Akt signaling. These studies affirm the use of these arrays as an unbiased screening tool of protein-protein interactions. We conclude that Tensin2 is an important new mediator in TPO/c-Mpl pathway and has a positive affect on cellular growth, at least in part through its effect on the PI3K/Akt signaling.
