RESUMO

The new species Pandora cacopsyllae Eilenberg, Keller & Humber (Entomophthorales) is described. The fungus was found on infected pear psyllids Cacopsylla pyri (Hemiptera: Psyllidae) in a pear orchard in Zealand, Denmark. Morphological structures (conidia, rhizoids, cystidia) were described on the designated type host C. pyri. In addition, conidia from an in vitro culture were described. Pandora cacopsyllae differs from other Pandora species by a) C. pyri is the natural host; b) conidia are different from other Pandora species infecting Psylloidea; c) ITS differs from other Pandora species infecting Hemiptera. The fungus has a high potential for future use in biological control of Cacopsylla pest species as well as other psyllids.

Pollen beetles Meligethes aeneus were collected in oilseed rape fields at different sites in Switzerland in spring 2004-2005 and 32 isolates of the fungal genus Beauveria occurring as latent infections in the beetles were obtained and molecularly characterized. Three major clades, Beauveria bassiana sensu stricto (Clade A: n=13), Beauveriabrongniartii (Clade B: n=1) and Beauveria Clade C (n=18) were identified among the isolates based on sequences of the ITS region and the 5' end of EF1-α. B. bassiana s.s. was further separated in the two clades, Eu_1 (n=10) and Eu_4 (n=3). The intergenic region Bloc provided best resolution of the individual clades B. bassiana s.s. Eu_1, Eu_4 and B. brongniartii. No specific clade of Beauveria appeared to be associated with adult M. aeneus populations. However, data suggested high relative abundance of Beauveria Clade C among the fungal entomopathogens infecting M. aeneus. Characterization of the isolates by simple sequence repeats (SSR) revealed further genotypic diversity within the clades except B. bassiana s.s. Eu_4 which appeared to be clonal. However, the individual SSR markers were differentially amplifiable from isolates of the different clades. It is therefore important to identify the underlying phylogenetic affinity of Beauveria isolates to interpret results based on SSR markers. The data suggest that not all available SSR markers are suitable for reliable characterization of diversity within Beauveria Clade C.

The effects of abiotic and biotic soil factors on occurrence of the entomopathogenic fungus Beauveria brongniartii after application at different times of the year were examined in Switzerland. Applications made from May to August generally resulted in an increase of 1-5 x 10(3) CFU g(-1) dry soil compared to untreated control plots. Conversely, soils treated in October and November yielded no increase. Soil temperatures between 20 and 25 degrees C, and high clay content of the soil had a positive effect on the occurrence and density of B. brongniartii whereas increased catalase activity and temperatures above 27 degrees C had a negative influence. Laboratory experiments revealed that a higher number of CFUs developed after one month of incubation at 22 degrees C than at 12 degrees C. Differences were not detected after three months of incubation, indicating that growth rate was simply slower at sub-optimal temperatures. The increase was different in three native soils, but was not correlated with different clay contents of the soil. In sterilized soil, though, the differences were not detected, suggesting that biotic factors have a greater influence rather than soil texture.
