OBJECTIVE: In order to grasp the quality status of the first-aid ventilator in pre-hospital and field environment in time, the quality control and detection items of invasive ventilator were optimized to form a rapid detection operation process of first-aid ventilator and ensure the safety of patient treatment. METHODS: Based on the quality control detection data of invasive ventilator in hospital in recent years, methods such as narrowing the range of qualified criteria and analyzing confidence interval were adopted to extract indicators prone to deviation and verify their correlation with similar indicators, so as to form an optimized rapid detection portfolio. RESULTS: Compared with the original quality control detection procedures, the detection indicators of the rapid detection procedures were reduced from 5 categories of 24 indicators to 3 categories of 7 indicators. The detection time was shortened by 56.1% and the coincidence rate of the detection results was 100% after the actual measurement and verification. CONCLUSIONS: Under the premise of ensuring the testing quality, the operating procedure for rapid detection of emergency ventilator can greatly reduce the detection time, and realize the rapid and high frequency quality control detection, so as to ensure the quality and safety of the equipment.

RESUMO

In vivo imaging of cerebral ischemia/reperfusion injury remains an important challenge. We injected porous Ag/Au@SiO2 bimetallic hollow nanoshells carrying anti-tropomyosin 4 as a molecular probe into mice with cerebral ischemia/reperfusion injury and observed microvascular changes in the brain using photoacoustic imaging with ultrasonography. At each measured time point, the total photoacoustic signal was significantly higher on the affected side than on the healthy side. Twelve hours after reperfusion, cerebral perfusion on the affected side increased, cerebrovascular injury worsened, and anti-tropomyosin 4 expression increased. Twenty-four hours after reperfusion and later, perfusion on the affected side declined slowly and stabilized after 1 week; brain injury was also alleviated. Histopathological and immunohistochemical examinations confirmed the brain injury tissue changes. The nanoshell molecular probe carrying anti-tropomyosin 4 has potential for use in early diagnosis of cerebral ischemia/reperfusion injury and evaluating its progression.

Glycosylation is a common post-translational modification in eukaryotic cells. It is involved in the production of many biologically active glycoproteins and the regulation of protein structure and function. Core fucosylation plays a vital role in the immune response. Most immune system molecules are core fucosylated glycoproteins such as complements, cluster differentiation antigens, immunoglobulins, cytokines, major histocompatibility complex molecules, adhesion molecules, and immune molecule synthesis-related transcription factors. These core fucosylated glycoproteins play important roles in antigen recognition and clearance, cell adhesion, lymphocyte activation, apoptosis, signal transduction, and endocytosis. Core fucosylation is dominated by fucosyltransferase 8 (Fut8), which catalyzes the addition of α-1,6-fucose to the innermost GlcNAc residue of N-glycans. Fut8 is involved in humoral, cellular, and mucosal immunity. Tumor immunology is associated with aberrant core fucosylation. Here, we summarize the roles and potential modulatory mechanisms of Fut8 in various immune processes of the gastrointestinal system.

RESUMO

The inhibitory effect of three degraded sesquiterpene lactones, iso-seco-tanapartholide, arteludooicinolide A and millifolide A isolated from Achillea millefolium L., on anti-human lung cancer cells was examined using MTT and reporter gene assays. Millifolide A has significant inhibitory effects on the proliferation of human lung cancer cells probably through inducing cell apoptosis.

This study was conducted to investigate the effect of sulfate loading on methane production and organic matter degradation during the mesophilic anaerobic co-digestion of corn stover and bio-kerosene production wastewater (BKPW). The highest methane production of 192.04 mL/gVS was obtained at a sulfate concentration of 86 mg/L. This was 46.80% higher than that achieved by a sulfate concentration of 113 mg/L. Additional degradation of organic matter was obtained at a sulfate concentration of 113 mg/L because organic matter in the corn stover and BKPW was oxidized by sulfate-reducing bacteria (SRB). The concentration of sulfate declined by approximately 23% after 29 days of anaerobic co-digestion, and this reduction in sulfate was enhanced when the soluble chemical oxygen demand (SCOD)/sulfate ratio was less than 15. The results of a mass balance analysis showed that 34.87% of C element and 10.04% of S element in substrate, respectively, were converted to biogas during anaerobic co-digestion of corn stover and BKPW at a sulfate concentration of 86 mg/L. The microbial community was analysed using 16S rDNA sequencing technology, and the results showed that the relative abundance of Synergistes (related to methane production with acetic acid) at a sulfate concentration of 86 mg/L had obviously increased and was approximately 287% higher than the abundance achieved at a sulfate concentration of 113 mg/L.

OBJECTIVE: To investigate the psychological features of relief crew in critical period after Wenchuan earthquake and to improve their mental health. METHODS: On May 12 of 2008, severe earthquake of a magnitude of 8.0 on Richter scale occurred in Wenchuan county, Sichuan province of China. Within 20 days after the earthquake, a research group conducted a study of psychological features of 600 relief crew with symptom checklist 90 (SCL-90), and the general symptom index and factorial scores were compared with Chinese norm (results from 1 388 personnel, published in 1986) and army norm (results from 19,662 servicemen, published in 1999). The positive items were measured and the relative factors were evaluated. RESULTS: (1) In the critical period after the earthquake, the relief workers had lower general symptomatic index than Chinese normal personnel or normal servicemen (both P<0.01). (2) Obsessive-compulsive symptoms (factorial score: 1.29+/-0.36) and somatization symptoms (factorial score: 1.22+/-0.29) were the main psychologic problems in this period. (3) Psychology of relief workers was correlated positively with working zones and education (r1=0.29, r2=0.15, both P<0.01), while negatively with working period (r=-0.28 , P<0.01). CONCLUSION: In immediate post-tremor period, the mental health of relief crew is predominantly fine, with obsessive-compulsive symptoms and somatization symptoms as outstanding mental disorders. Working zone, working period and education are main factors which affect their mental health.

OBJECTIVE: To compare patient-reported outcomes after implantation of the ZA9003 intraocular lens (IOLs), or the MCX11 ASP IOLs or the spherical IOLs (HQ-201HEP). METHODS: Prospective nonrandomized controlled trial was used. A total of 105 patients (210 eyes) were divided into three groups according to the type of IOLs: ZA9003 (35 patients, 70 eyes), MCX11 ASP (35 patients, 70 eyes) or HQ-201HEP (35 patients, 70 eyes). The main outcome was scores of Catquest nine-item short-form questionnaire. Additional outcome was best corrected visual acuities, spherical aberration (SA) and total higher-order aberrations (HOAs). RESULTS: The global score was significantly lower in the spherical IOL group than the aspherical IOL group of -020 µm SA (P < 0.05) and the aspherical IOL group of -027 µm SA (P < 0.05), and no significant difference was found in the global score between the aspherical IOL group of -020 µm SA than the aspherical IOL group of -027 µm SA (P > 0.05). Significant differences were also found in question 2, question 5, question 6 and question 8 between the spherical IOLs and the aspherical IOLs. CONCLUSION: Implantation of an aspherical IOL could improve vision-related quality of life compared with a spherical IOL. However, there were no statistically significant differences in vision-related quality of life between aspheric IOLs with different negative spherical aberrations.

OBJECTIVE: To study the effects of water and alcohol extracts of several Chinese herbal medicines and other medicines on alcohol dehydrogenase activity in order to provide enzymology basis on new medicine. METHODS: Water or alcohol extracts of Chinese herbal medicine and other medicine were tested on the effects of alcohol dehydrogenase activity by Valle and Hoch method. RESULTS: Among them, 8 were found to have the effect of activation on alcohol dehydrogenase. They were water extracts of Amomum kravanh and Pueraria flowers, the alcohol extracts of Pueraria flowers, compound hepatcare Chinese medicine and compound Pueraria medicine, L-cysteine, notoginseng saponin. Others had inhibiting action. CONCLUSION: To decrease alcohol concentration in the body through activating the activity of ADH may be one of the mechanisms for some traditional Chinese herbal medicine in neutralizing the effect of alcohol drink.

The reactions of *OH and e(aq)-, with AO7 in aqueous solutions were studied by nano-second pulse radiolysis techniques. The transient absorption spectra were attributed and the second order reaction rate constants were determined by following the buildup kinetics of the transient absorptions. In combination with the analysis of the UV-Vis absorption changes of AO7 after gamma-radiolysis, the underlying reaction mechanisms were elucidated.

RESUMO

Nicotine has been studied for the first time by pulse radiolysis techniques. It has been found that hydrated electron, hydrogen radical, and hydroxyl radical can react on nicotine to produce an anion radical and neutral radical, respectively. Different absorption spectra were obtained, and the related rate constants were determined. The results provide some advices for scientists to study the harmfulness of smoking nicotine to people, which can give some new ideas to make harmless tobacco.

Vitamin C, an antioxidant that reduces reactive oxygen species (ROS) in cells, is capable of significantly improving the developmental competence of porcine and mouse somatic cell nuclear transfer (SCNT) embryos, both in vitro and in vivo. In the present study, the effects of vitamin C on the developmental competence of bovine SCNT embryos were investigated. The results indicated that vitamin C (40 µg/mL) positively affected the scavenging of intracellular ROS, cleavage rate at 24 h (76.67 vs. 68.26%, p<0.05), compact morulae formation (60.83 vs. 51.30%, p<0.05), and the blastomere apoptosis index (3.70 ± 1.41 vs. 4.43% ± 1.65, p<0.05) of bovine SCNT embryos. However, vitamin C supplementation did not significantly affect the blastocyst formation rate and proportion of inner cell mass over total cells per blastocyst on day 7. Moreover, vitamin C supplementation obviously impaired the total cell numbers per blastocyst (97.20 ± 11.35 vs. 88.57 ± 10.43, p<0.05) on day 7 and the hatching blastocysts formation rate on day 9 (26.51 vs. 50.65%, p<0.05) compared with that of the untreated group. Vitamin C supplementation preferentially improved the viability of bovine SCNT embryos prior to the blastocyst stage, but did not enhance the formation and quality of blastocysts in vitro. In conclusion, the effect of vitamin C on the development of bovine SCNT embryos is complex, and vitamin C is not a suitable antioxidant chemical for the in vitro culture of bovine SCNT embryos.

The homeostasis of intracellular diadenosine 5',5″'-P(1),P(4)-tetraphosphate (Ap4A) in the yeast Saccharomyces cerevisiae is maintained by two 60% sequence-identical paralogs of Ap4A phosphorylases (Apa1 and Apa2). Enzymatic assays show that, compared to Apa1, Apa2 has a relatively higher phosphorylase activity towards Ap3A (5',5″'-P(1),P(3)-tetraphosphate), Ap4A, and Ap5A (5',5″'-P(1),P(5)-tetraphosphate), and Ap4A is the favorable substrate for both enzymes. To decipher the catalytic insights, we determined the crystal structures of Apa2 in the apo-, AMP-, and Ap4A-complexed forms at 2.30, 2.80, and 2.70Å resolution, respectively. Apa2 is an α/ß protein with a core domain of a twisted eight-stranded antiparallel ß-sheet flanked by several α-helices, similar to the galactose-1-phosphate uridylyltransferase (GalT) members of the histidine triad (HIT) superfamily. However, a unique auxiliary domain enables an individual Apa2 monomer to possess an intact substrate-binding cleft, which is distinct from previously reported dimeric GalT proteins. This cleft is perfectly complementary to the favorable substrate Ap4A, the AMP and ATP moieties of which are perpendicular to each other, leaving the α-phosphate group exposed at the sharp turn against the catalytic residue His161. Structural comparisons combined with site-directed mutagenesis and activity assays enable us to define the key residues for catalysis. Furthermore, multiple-sequence alignment reveals that Apa2 and homologs represent canonical Ap4A phosphorylases, which could be grouped as a unique branch in the GalT family.

AIM: Investigated the relationship between NF-kappaB activation and cell apoptosis in mouse macrophages treated with 7-ketochesterol (7-KC). METHODS: Cell apoptosis was detected by MTT assay, DNA fragmentation assay and flow cytometric analysis. NF-kappaB activation was detected by western blot and immunohistochemistry. Inhibitory assay was used to show the effect of the activation of NF-kappaB on the apoptosis induced by 7-KC. RESULTS: 7-KC inhibited macrophages proliferation, and then induced apoptosis, which is associated with NF-kappaB activation. Moreover, cell apoptosis with NF-kappaB activation was inhibited by pyrrolidine dithiocarbamate (PDTC), an inhibitor of NF-kappaB. CONCLUSION: 7-KC induced the activation of NF-kappaB and following cell apoptosis.
