RESUMO

The objective of the present paper is fast and nondestructive estimate of kalium content using ASD FieldSpec3 spectrometer determined hyperspectral data in apple florescence canopy. According to detection of hyperspectral data of the apple florescence canopy and kalium content data at laboratory in Qixia city of experimental orchards in 2008 and 2009, the correlation analysis of hyperspectral reflectance and its eleven transforms with kalium content was proceeded. The biggest correlation coefficient as independent variable and the estimation model of kalium content were established based on fuzzy recognition algorithms. The model was tested by sample inspection in 2008 and verified by data in 2009. The results showed that the correlation is less for the original spectral reflectance (R) and its reciprocal(1/R), logarithm (lgR), square root (R1/2) and the kalium content, but it is enhanced obviously for their first derivative and second derivative. The correlation coefficient(r) of kalium content estimating model y = 11.344 5h + 1.309 7 is 0.985 1, the total root mean square difference (RMSE) is 0.355 7 and F statistics is 3 085.6. The average relative error of measured values and estimated values for 24 inspection sample is 9.8%, estimation accuracy is 90.2% and verification accuracy is 83.3% utilizing test data in 2009. It was showed that this model is more stable by estimating apple florescence canopy of kalium content and the model precision is able to meet the needs of production.

The present study aims to systematically analyze the hyperspectral characteristics of apple florescence canopy and explore the sensitive spectra to provide the theoretical basis for large area apple information extracting and remote sensing retrieval for nutrition diagnosis. Based on the 120 hyperspectral data of apple florescence canopy acquired with ASD Field Spec 3 portable object spectrometer, the effects of different sample numbers on hyperspectral characteristics were analyzed. Using variance analysis method, the hyperspectral characteristics of apple florescence canopy and the sensitive wave bands were obtained. The results showed that with the increase in cumulative sample numbers, the hyperspectrum curves of apple florescence became stable and smooth. At the 550 nm green peak and the 760-1,300 nm reflection plateau, the reflection rate reduced with the increase in flowering amount, while in the red valley of 670 nm, the reflection rate increased with the increase in flowering amount; At the wave bands of 350-500, 600-680 and 760-1,300 nm, the variance analysis results showed very significant differences, indicating that they were sensitive wave bands of florescence canopy. With the increase in flowering amount, the red-edge position, the red-edge slope and red edge area tended to decrease gradually.

MSC transplantation has been explored as a new clinical approach to stem cell-based therapies for bone diseases in regenerative medicine due to their osteogenic capability. However, only a small population of implanted MSC could successfully reach the injured areas. Therefore, enhancing MSC migration could be a beneficial strategy to improve the therapeutic potential of cell transplantation. Catharmus tinctorius volatile oil (CTVO) was found to facilitate MSC migration. Further exploration of the underlying molecular mechanism participating in the pro-migratory ability may provide a novel strategy to improve MSC transplantation efficacy. This study indicated that CTVO promotes MSC migration through enhancing ROCK2 mRNA and protein expressions. MSC migration induced by CTVO was blunted by ROCK2 inhibitor, which also decreased myosin light chain (MLC) phosphorylation. Meanwhile, the siRNA for ROCK2 inhibited the effect of CTVO on MSC migration ability and attenuated MLC phosphorylation, suggesting that CTVO may promote BMSC migration via the ROCK2/MLC signaling. Taken together, this study indicates that C. tinctorius volatile oil could enhance MSC migration via ROCK2/MLC signaling in vitro. C. tinctorius volatile oil-targeted therapy could be a beneficial strategy to improve the therapeutic potential of cell transplantation for bone diseases in regenerative medicine.

OBJECTIVE: To observe the effect of extract components from Plastrum Testudinis and their combination component on the proliferaiton of mesenchymal stem cell (MSC) to screen out the effective components. METHODS: Mesenchymal stem cells were dissociated from bone marrow of rat and were marked by Brdu, and the expression of CD44, CD54 and double label of Brdu and CD44 extract components (A to approximately J) with different concentration (2 microl, 5 microl,10 microl,20 microl)added to cultured MSC. The cell viability was tested by MTT. RESULTS: Cell viability in component A, B, H, I with 20 microl, C, F with 10 microl and J with 5 microl concentration group increased compared with that in control group (P < 0.05) and cell viability in compponent E with 2 microl to approximately 20 microl concentration groups decreased compared with that in control group (P < 0.05). Cell viability in component D and G with 2 microl to approximately 20 microl concentration groups was similar to that in control group. Cell viability in combination component with J component increased, but cell viability in combination component with E component decrased. CONCLUSION: All compoents A, B,C,F, H, I and J can improve proliferation of MSC. Anong them, component J and its combination are the best. Component E and its combination can inhibit proliferation. But component D and G have no effect on proliferation of MSC.

Taking the Qihe County in Shandong Province of East China as the study area, soil samples were collected from the field, and based on the hyperspectral reflectance measurement of the soil samples and the transformation with the first deviation, the spectra were denoised and compressed by discrete wavelet transform (DWT), the variables for the soil alkali hydrolysable nitrogen quantitative estimation models were selected by genetic algorithms (GA), and the estimation models for the soil alkali hydrolysable nitrogen content were built by using partial least squares (PLS) regression. The discrete wavelet transform and genetic algorithm in combining with partial least squares (DWT-GA-PLS) could not only compress the spectrum variables and reduce the model variables, but also improve the quantitative estimation accuracy of soil alkali hydrolysable nitrogen content. Based on the 1-2 levels low frequency coefficients of discrete wavelet transform, and under the condition of large scale decrement of spectrum variables, the calibration models could achieve the higher or the same prediction accuracy as the soil full spectra. The model based on the second level low frequency coefficients had the highest precision, with the model predicting R2 being 0.85, the RMSE being 8.11 mg x kg(-1), and RPD being 2.53, indicating the effectiveness of DWT-GA-PLS method in estimating soil alkali hydrolysable nitrogen content.

A total of 60 soil samples with approximate contents of N, P, and K and greatly different content of organic matter were selected by statistical analysis. Through hyper-spectral detection and analysis, the first derivative spectrum of the soil logarithmic reflectance was obtained, and was decomposed by the Bior 1.3 wavelet function. The approximative signal of the lowest frequency and the noise signal of the highest frequency were removed from the input spectrum so as to obtain the characteristic spectrum corresponding to soil physical and chemical parameters. The sensitive bands of soil organic matter were selected by correlation analysis, and the forecasting models were built by multiple regression analysis, based on the sensitive bands and the characteristic spectrum, respectively. Through comparison analysis, the optimal wavelet decomposing resolution for extracting the characteristic spectrum of soil organic matter was ascertained, and the best forecasting model was established. The best wavelet decomposing resolution was 9, followed by 8 and 10. Based on the characteristic spectrum of wavelet decomposing of 9 resolutions, the model R2 reached 0.89, which was increased by 0.31 as compared to the model based on sensitive bands, and increased by 0.10 as compared to the model based on the original spectrum.

Novel 2-vinyl-8-hydroxyquinoline derivatives as potential antioxidants and regulators of H(2)O(2)-induced oxidative stress in rat bone marrow mesenchymal stem cells (MSCs) are first reported. The antiradical properties and the reducing power of these compounds were assessed using 2, 2-diphenyl-1-picrylhydrazyl (DPPH) and auto-oxidation of pyrogallol method, respectively. The activity against lipid peroxidation was determined using ammonium thiocyanate method. The results revealed that introduction of electron-donating groups at 2nd position decreased the antioxidant activities of 8-hydroxyquinoline derivatives. In addition, compound 4, the structure of which is similar to melatonin, exhibited superior antioxidant activities in scavenging DPPH free radical, O(2) free radical, and anti-LPO activities. Except for compounds 7, 12, and 15, the other compounds exhibited a stimulatory effect on MSCs growth. Using hydrogen peroxide (H(2)O(2)), we also investigated the protective efficacy of 2-vinyl-8-hydroxyquinoline derivatives against oxidative stress-induced cell death of MSCs. Cell viability assayed by MTT method indicated that exposure of MSCs cultures to hydrogen peroxide resulted in a concentration-dependent decrease in cell viability, and compounds 4 and 5 at given concentration (2.62 x 10(-3) m) could protect MSCs against H(2)O(2)-induced oxidative stress in bone mesenchymal stem cell (BMSCs).
