RESUMO

Fungal laccases play important roles in the degradation of lignocellulose. In this study, the laccase producing cotton straw medium for Pleurotus ostreatus was optimized by single-factor and orthogonal experiments, and to investigate the role of Lacc1 gene, one of the laccase-encoding genes, in the degradation of cotton straw lignin, an overexpression strain of Lacc1 gene was constructed, which was analyzed for the characteristics of lignin degradation. The results demonstrated that the culture conditions with the highest lignin degradation efficiency of the P. ostreatus were the cotton straw particle size of 0.75 mm, a solid-liquid ratio of 1:3 and containing 0.25 g/L of Tween in the medium, as well as an incubation temperature of 26 °C. Two overexpression strains (OE L1-1 and OE L1-4) of Lacc1 gene were obtained, and the gene expression increased 12.08- and 33.04-fold, respectively. The results of 1H-NMR and FTIR analyses of significant changes in lignin structure revealed that Lacc1 gene accelerated the degradation of lignin G-units and involved in the cleavage of ß-O-4 linkages and the demethylation of lignin units. These findings will help to improve the efficiency of biodelignification and expand our understanding of its mechanism.

BACKGROUND: Novel ß-lactams have in vitro activity against Pseudomonas aeruginosa (PA), but their clinical performances and the selection criteria for practical use are still not clear. We aimed to evaluate the efficacy of novel ß-lactams for PA infection in various sites and to compare the efficacy of each agent. METHODS: We searched PubMed, Embase, Cochrane Library, and Web of Science for randomized controlled trials that used novel ß-lactams to treat PA infection. The primary outcomes were clinical cure and favorable microbiological response. Subgroup analyses were performed based on drug type, drug resistance of pathogens, and site of infection. Network meta-analysis was carried out within a Bayesian framework. RESULTS: In all studies combined (16 randomized controlled trials), novel ß-lactams indicated comparable performance to other treatment regimens in both outcome measures (relative risk = 1.04; 95% confidence interval 0.94-1.15; P = .43) (relative risk = 0.97; 95% confidence interval 0.81-1.17; P = .76). Subgroup analyses showed that the efficacy of ceftolozane-tazobactam (TOL-TAZ), ceftazidime-avibactam (CAZ-AVI), imipenem-cilastatin-relebactam, and cefiderocol had no apparent differences compared to control groups among different infection sites, drug types and drug resistance of PA. In network meta-analysis, the results showed no statistically significant differences between TOL-TAZ, CAZ-AVI, and cefiderocol. CONCLUSIONS: TOL-TAZ, CAZ-AVI, imipenem-cilastatin-relebactam, and cefiderocol are not inferior to other agents in the treatment of PA infection. Their efficacy is also comparable between TOL-TAZ, CAZ-AVI, and cefiderocol.

With lifestyle changes, chronic diseases have become a public health problem worldwide, causing a huge burden on the global economy. Risk factors associated with chronic diseases mainly include abdominal obesity, insulin resistance, hypertension, dyslipidemia, elevated triglycerides, cancer, and other characteristics. Plant-sourced proteins have received more and more attention in the treatment and prevention of chronic diseases in recent years. Soybean is a low-cost, high-quality protein resource that contains 40% protein. Soybean peptides have been widely studied in the regulation of chronic diseases. In this review, the structure, function, absorption, and metabolism of soybean peptides are introduced briefly. The regulatory effects of soybean peptides on a few main chronic diseases were also reviewed, including obesity, diabetes mellitus, cardiovascular diseases (CVD), and cancer. We also addressed the shortcomings of functional research on soybean proteins and peptides in chronic diseases and the possible directions in the future.

Metabolomics, as a new omics technology, has been widely accepted by researchers and has shown great potential in the field of nutrition and health in recent years. This review briefly introduces the process of metabolomics analysis, including sample preparation and extraction, derivatization, separation and detection, and data processing. This paper focuses on the application of metabolomics in food-derived bioactive ingredients. For example, metabolomics techniques are used to analyze metabolites in food to find bioactive substances or new metabolites in food materials. Moreover, bioactive substances have been tested in vitro and in vivo, as well as in humans, to investigate the changes of metabolites and the underlying metabolic pathways, among which metabolomics is used to find potential biomarkers and targets. Metabolomics provides a new approach for the prevention and regulation of chronic diseases and the study of the underlying mechanisms. It also provides strong support for the development of functional food or drugs. Although metabolomics has some limitations such as low sensitivity, poor repeatability, and limited detection range, it is developing rapidly in general, and also in the field of nutrition and health. At the end of this paper, we put forward our own insights on the development prospects of metabolomics in the application of bioactive ingredients in food.

Biodelignification is widely regarded as a low-efficiency process because it is usually slow and difficult to control. To improve its efficiency and understand its mechanism, the present study analyzed the delignification characteristics of Pleurotus ostreatus grown on a cotton stalk medium. The results demonstrated that all strains of P. ostreatus can selectively degrade the cotton stalk lignin. When cultured in a cotton stalk medium for 60 days, P. ostreatus degraded lignin primarily during its mycelium growth with up to 54.04% lignin degradation and produced laccase and manganese dependent peroxidase with high activity levels at the peaks of 70.17 U/ml and 62.39 U/ml, respectively, but no detectable lignin peroxidase. The results of nuclear magnetic resonance spectroscopy and Fourier transform infrared spectroscopy analyses of significant changes in lignin structure revealed that syringyl (S) lignin units were more degraded than guaiacyl (G) lignin units, with a significantly elevated G/S ratio. The Gas Chromatography-Mass Spectrometer analysis of low-molecular-weight compounds revealed that the delignification resulted in the formation of alcohols, organic acids, benzodiazepines, and alkanes. Identified benzodiazepines implied the degradation of G and S units of lignin. These findings will help to improve the efficiency of biodelignification and expand our understanding of its mechanism.

In this study, a natural polymer-based organic flocculant (sodium alginate-methacrylatoethyl trimethyl ammonium chloride, SA-PDMC) was synthesized by graft copolymerization. The optimum preparation procedures were determined by single factor experiments. The flocculation behaviors of SA-PDMC were investigated in humic acid (HA) and kaolin suspension considering the effects of flocculant doses and initial pH. The results indicated that charge neutralization and bridging action played an important role in the removals of HA and kaolin. Also, SA-PDMC performed well in a wide pH range of 5.0-10.0. Besides, SA-PDMC, as polyaluminium (PAC) aid, was investigated in the lake water treatment. The ratio of PAC and SA-PDMC was optimized through response surface methodology based on a central composite design. Results showed that SA-PDMC and PAC have a strong synergy, under optimal conditions SA-PDMC can reduce the dose of PAC by 40 % while ensuring the water quality.

The charge densities (CD) and molecular weights (MW) of the flocculants are closely related to their application performances, but seldom researches focus on the effects of flocculant CD and MW on decolorization efficiencies. Herein, a series of flocculants with various CD and MW levels, named as PBF1-9, were designed and synthesized from papermaking sludge. The physicochemical characteristics of the PBF1-9 were measured by fourier transform infrared spectroscopy (FTIR), gel permeation chromatography (GPC), X-ray photoelectron spectroscopy (XPS) and particle charge density analyzer (PCD). The efficiencies of PBF1-9 were studied in the reactive blue (RB) dye removals by flocculation under different process conditions. The operation costs of the flocculants were evaluated at their optimal dosages. Also, the pH-independences and ion-tolerances of the aforementioned flocculants were studied in terms of the molecular levels. The experimental results exhibited that the flocculants CD or MW values were relevant to their flocculation behaviors and operation costs. CD values played a dominant role in color removal efficiencies and the costs, whereas MW values were critical to the floc structure. The pH or ion-independences of the flocculants were significantly dependent on the CD and MW values. However, some conclusions, conflicted with prior studies, were observed in this work. For instance, flocculant with the highest CD and MW levels was not the most effective one in enduring pH variation and the coexisting ions. The floc properties, including floc size, resistance and recovery ability, were relatively insensitive to flocculant intrinsic CD and MW levels when the flocculants were used at their optimal dosages. Furthermore, the possible relevance between CD or MW levels and the flocculation mechanisms have been proposed in this work. Exploring the effects of flocculants CD and MW levels could precisely control the flocculant characteristics to achieve satisfactory decontamination efficiencies with low costs.
