RESUMO

Background. Cancer-related fatigue (CRF) is a painful, persistent feeling of physical and cognitive subjective fatigue related to cancer or cancer remedy. The occurrence of CRF may be related to the hypothalamic-pituitary-adrenaline (HPA) axis, inflammatory mediator theory, and gut microbiota. Moreover, acupuncture could play a vital part in the therapy of CRF. The study will evaluate whether acupuncture can improve fatigue symptoms of CRF patients after breast cancer chemotherapy by regulating the gut-brain axis. Methods/design. Seventy patients with CRF will be enrolled in this study, with 35 patients randomly assigned to each group. Blood and feces will be collected at the beginning and end of treatment. Piper fatigue scale, KPS score scale, and quality-of-life scale will be used to observe the changes of fatigue symptoms and life quality of CRF patients and to evaluate the effect of acupuncture on CRF. Then, the method of ELISA will be used to explore the regulatory effect of acupuncture on the HPA axis and inflammatory cytokines. Moreover, based on 16SrDNA, the changes of gut microbiota structure and flora of CRF patients will be observed. Ultimately, the correlation analysis of gut microbiota can be related to inflammatory cytokines, HPA axis, and clinical efficacy evaluation. Discussion. This study will identify the effect and the mechanism of acupuncture therapy in CRF. And it will offer an alternative treatment modality for the treatment of CRF after chemotherapy for breast cancer. Furthermore, it will also provide the clinical and theoretical bases for the extensive application of acupuncture therapy in tumor rehabilitation. Trial Status. OBJECTIVE: To investigate the expression and clinical significance of microRNA-195 in patients with diffuse large B cell lymphoma(DLBCL). METHODS: Sixty patients with DLBCL were selected from nearly four years in our hospital, and at the same time 30 healthy people with physical examination of the same period and with the same age in our hospital were choosed as control group. Real-time fluorescent quantitative PCR (RT-qPCR) was used to detect the miR-195 expression of the patients and controls, the relationship between miR-195 expression and clinicopathological characteristics of DLBCL and survival time of patients was analyzed. RESULTS: The expression level of miR-195 in DLBCL patients was significantly lower than that in the controls (P<0.001). The expression level of miR-195 closely related with tumor diameter, IPI score and Ann Arbor stage of patients with DLBCL. Overall survival(OS) time of DLBCL patients with highly expressed miR-195 was significantly longer than that of patients with low expression (P<0.001). CONCLUSION: miR-195 expression decrease in DLBCL patients, and miR-195 closely relates with tumor characteristics of patients with DLBCL. DLBCL patients with higher expression of miR-195 show longer overall survival time.

Hypoxia­inducible­factor 1α (HIF­1α) is a marker for poor prognosis in the majority of the cancer types, and it has been revealed to be essential for maintaining cancer stem cells (CSCs). In the present study, it was determined that the expression of HIF­1α and CSC­related genes under hypoxic conditions was upregulated. Stable knockdown of HIF­1α significantly inhibited cell proliferation, migration and tumour growth in vivo in oesophageal squamous cell carcinoma (ESCC). A previous study revealed that the Wnt/ß­catenin pathway may play a key role in maintenance and progression of CSCs. Therefore, it was also revealed that stable knockdown of HIF­1α reduced the formation of spheroid body cells, the expression of CSC­related genes and Wnt/ß­catenin pathway­related target genes, as well as the activity of the Wnt/ß­catenin pathway. Collectively, the present results indicated that HIF­1α may regulate the stemness of ESCC by activating the Wnt/ß­catenin pathway.

With four-year-old potted Prunus persica L. cv. Qingfeng as test material, this paper studied the change pattern of its leaf betaine content under water stress, and its physiological responses under effects of foliage-spraying exogenous betaine. The results showed that under normal water supply, the betaine content in Qingfeng' s leaf was 75.9-80.5 microg x g(-1) FM, which was increased with increasing water stress, and up to 278.9 microg x g(-1) FM on the 16th day after cutting off the water supply. The leaf plasma membrane permeability was 8.06% - 8.61% under normal water supply, but increased to 28.62% under water stress. When 100 and 500 mg x L(-1) of betaine were applied exogenously, the plasma membrane permeability was 26.25% and 21.79% after 16 days, respectively. The hydrogen peroxide (H2O2) content increased from 27.2-32.5 micromol g(-1) FM to 76.4 micromol x g(-1) FM in the course of water stress, and decreased to 73.2 and 68.5 micromol x g(-1) FM after spraying 100 and 500 mg betaine x L(-1), respectively. During the period of intensified water stress, the peak value of ascorbate peroxidase (AsA-POD) activity was 0.435 mg x g(-1) FM, and up to 0.490 mg x g(-1) FM when treated with exogenous betaine. When the peach tree was subjected to water stress, the contents of free proline and soluble sugar accumulated dramatically, but produced on approximately decrease in 500 mg x L(-1) endogenous betaine application on the 16th day which was slightly less than that of control and 100 mg x L(-1) betaine application. There was a gradual decline in the content of soluble protein under water stress, and an increment of 20. 3% was observed when betaine was applied exogenously. These results strongly suggested that foliage-spraying exogenous betaine could increase the drought resistance of peach tree through decreasing its leaf plasma membrane permeability and H2O2, free proline and soluble sugar contents and increasing its leaf AsA-POD activity and soluble protein content.
