RESUMO

The knowledge of how temperature influences elasmobranchs reproductive physiology allows a better understanding of their reproductive patterns. This study describes the relationship between temperature fluctuations and the plasmatic changes of the sex steroids related to reproduction: testosterone (T), estradiol (E2) and progesterone (P4), throughout the female reproductive cycle of the shark Mustelus schmitti. A total of 123 adult females were bi-monthly sampled in Buenos Aires, Argentina, coastal waters. Bottom temperatures were recorded at each sampling point and blood samples were taken from each female for plasma sex steroids measurement. Sex steroid plasma levels were analyzed in relation with maximum follicular diameter (MFD), uterosomatic index (USI, as indicator of pregnancy) and temperature using Generalized Additive Models. Plasmatic E2 and T increased during follicular growth until MFD reached 1.34 and 1.46 cm, respectively. Peak of T occurred at the follicular stage associated with parturition (MFD, 1.4-1.6 cm), just prior to final maturation and ovulation (MFD, 1.6-2.0 cm). Progesterone significantly increased at this last ovarian phase, while T and E2 decreased. The increase of USI with pregnancy was associated to a decrease in T and mainly E2 levels, while P4 remained unaffected. Prior to ovulation, T plasma levels decreased with temperature below to 13 °C and then increased progressively with a pronounced elevation above 17 °C, while E2 presented an opposite pattern. Progesterone plasma levels changed with temperature showing a similar pattern to that observed for T. Using M. schmitti shark as model species, this study shows a clear picture of how seawater temperature variations can affect the reproductive physiology in elasmobranch females. A hypothetical mechanism (based on T elevation driven by temperature increase and its connection by feedback with a P4 rise and parturition/ovulation induction) is proposed as evidence to support that the increase in temperature can trigger reproductive events in elasmobranchs. In addition to its ecological scope, this work contributes to reinforce the relatively scarce general knowledge of elasmobranchs reproductive physiology.

RESUMO

In the present study we histochemically and lectinhistochemically characterized the growing oocytes of the pink cuskeel (Genypterus blacodes). We used histochemical methods for the localization and characterization of glycoconjugates (GCs) and lectin histochemical techniques for the identification of specific sugar residues. We analyzed presence and distribution of GCs in the different structures of the growing follicles (cortical alveoli, globules, yolk granules and zona radiata). During the initial stage of vitellogenesis, the oocytes presented small yolk granules composed of GCs that gradually increased during exogenous vitellogenesis. These GCs contained moderate quantities of α-D-mannose, D-glucose, N-acetylglucosamine and N-acetyl-neuraminic acid. The cortical alveoli contained both neutral and carboxylated GCs, and lectin techniques detected N-acetylgalactosamine, galactose and L-fucose. The zona radiata showed a strong positive reaction to PAS and it reacted weakly with more specific techniques, such as KOH/PA*S and PA/Bh/KOH/PAS. This structure showed GCs with oxidizable vicinal diols, O-acyl sugars and sialic acid residues with different substitution types and presented N-acetylgalactosamine and L-fucose specific residues. The oocytes follicular envelope evidenced neutral and acidic non-sulfated GCs and high concentrations of α-D-mannose, D-glucose, galactose and N-acetylgalactosamine. The intergranular cytoplasmic GCs were mainly rich in α-D-mannose, D-glucose, N-acetylgalactosamine, N-acetylglucosamine and N-acetyl-neuraminic acid. These results enhance the comprehension of the structure and functionality of the pink cuskeel ovarian follicles, and provide a useful tool for the study of this tissue in other teleost species.

RESUMO

Pinguipes brasilianus is a conspicuous and abundant rocky-reef fish inhabiting the Northern Patagonian gulfs of Argentina, Southwest Atlantic. This study describes its reproductive biology in this region. We made macroscopic and histological descriptions of the testis and ovary development, analyzed the evolution of the gonadosomatic index (GSI) in both sexes throughout the year, and estimated fecundity and length at 50% maturity for females. Pinguipes brasilianus is a multiple spawner with indeterminate annual fecundity and an extended reproductive season, encompassing austral spring and summer. Low GSI values in males (monthly means ranged between 0.11% and 0.75%) and low relative fecundity in female fish (between 2 and 44 oocytes/g) are consistent with a need of close proximity between male and female during oocyte fertilization, sexual courtship and with spawning in pairs and/or within reef crevices. The estimated length at 50% maturity (± SD) for females was 15.22 ± 0.43 cm total length. Although P. brasilianus shares the main reproductive features of its congeneric species Pinguipes chilensis , male GSI and female length at 50% maturity in P. brasilianus were markedly lower than those reported for P. chilensis , while relative and batch fecundity were greater in fish of the same size.

Percophis brasiliensis is a demersal species that constitutes an important resource of Argentine coastal fisheries. Nevertheless, information about bioenergetic dynamic of reproduction has not been reported. Therefore, seasonal variations of condition factors, biochemical composition and energy density of different tissues were analyzed in order to determine the strategy of energy allocation during the reproductive cycle of this species. Condition indices (hepatosomatic and K) showed a seasonal pattern opposite to that observed for gonadosomatic index, which was characterized by higher values during the reproductive period (spring-summer), decreasing at the end of spawning. Biochemical composition of different tissues also showed a clearly seasonality associated to reproductive cycle. Analysis of energy density variation of liver indicates that P. brasiliensis accumulate reserves in winter before reproduction, which later decrease during the spawning season. In contrast, the energy density in muscle did not show significant differences among seasons, indicating that individuals could be also using an external source of energy during spawning. Therefore, it is possible that P. brasiliensis respond to an intermediate strategy of energy allocation, combining characteristics of both capital breeders (stores energy previous to the onset of reproductive activity) and income breeders (acquire energy by active feeding during spawning period).

The composition and distribution of the glycoconjugates (GCs) secreted by the epithelium of ovarian lamellae with reference to the reproductive biology of Genypterus blacodes (Schneider, 1801) through lectin hi stochemistry is here discussed. In this species, the epithelial cells that line the ovarian cavity presented sharp morphological variations along the reproductive cycle related to the mucus secretion that accompanies oocyte ma turation. During sp awning season, residues of mannose and N-acetylglucosamine were detected in the glycocalyx of those cells using lectinhistochemistry. N- acetylgalactosamine and fucose were also observed in the same zone. The greatest variations in the lectinhistochemical pattern were found in the apical cytoplasm composition in comparison to the basal zone of the cells. The results of the present study were discussed by comparing their possible functional implications.
