BACKGROUND: Saliva is being increasingly used as a sample for measuring biomarkers in several species and shows a high potential of use to detect and monitor stress. The weaning and grouping in dairy calves are a particularly stressful time. Therefore, the objectives of this study were to evaluate a panel of antioxidant and oxidant biomarkers in the saliva of calves on the day of weaning (W0), 2 days after weaning or milk withdrawal (W + 2), and 4 days after grouping (G + 4). In addition, to verify if cortisol and oxytocin concentrations are related to the biomarkers measured. RESULTS: Salivary cupric reducing antioxidant capacity (CUPRAC), ferric reducing ability of saliva (FRAS), Trolox equivalent antioxidant capacity (TEAC), advanced oxidation protein products (AOPP), and ferrous oxidation-xylenol orange (FOX) were significantly higher (P < 0.02) 4 days after grouping than the day of weaning and 2 days after. The increases were 50 and 54% for CUPRAC, 93 and 116% for FRAS, 117 and 135% for TEAC, 22 and 49% for AOPP and 10 and 5% for FOX in comparison with weaning and 2 days after, respectively. In addition, oxytocin and cortisol showed significant negative and positive correlations (P < 0.05) respectively with the biomarkers of oxidative status. CONCLUSIONS: Our results showed that calves after grouping show increases in antioxidants and oxidants concentrations, indicating that a balance between these molecules has been tried to maintain during this stressful situation. The dynamic changes of biomarkers of oxidative status should be explored and characterised in other stressful conditions.

BACKGROUND: The possible use of oxytocin in saliva as an indicator of positive emotions in bovine species has been poorly investigated. In the present study, two new assays (one using a monoclonal antibody and the other using a polyclonal antibody) for the measurement of oxytocin in bovine saliva were developed and validated. Also, the changes in oxytocin in saliva were explored in two different situations. One was around parturition, and for this purpose, saliva samples from 13 cows were collected at three different times: 7 days before the parturition, the day of parturition and 7 days after the parturition. The second situation was weaning and grouping of calves, and for this purpose, saliva from 25 calves was collected at three different times: 1 day before weaning, 2 days after weaning or milk withdrawal and 4 days after grouping calves. RESULTS: In cows, oxytocin concentrations showed an increase on the day of parturition with both assays, while in calves, oxytocin concentrations showed a decrease 4 days after the grouping. CONCLUSIONS: The assays validated in this report could be used for the measurement of oxytocin in bovine saliva and detect changes in this analyte that can occur in different physiological or productive situations such as parturition and weaning.

BACKGROUND: Biomarkers of oxidative stress in pigs have been measured in serum/plasma samples. However, blood collection in pigs can be highly stressful to the animals. Saliva is a biological fluid with several advantages in pigs over blood, since it can be easily collected without stress to the animals, being therefore an ideal sample in this species. The objective of this study was the validation of assays for the evaluation of oxidative stress status in saliva of pigs. For this purpose, three assays commonly used to evaluate the total antioxidant capacity (TAC): trolox equivalent antioxidant capacity (TEAC), cupric reducing antioxidant capacity (CUPRAC), and ferric reducing ability of plasma (FRAP)), one individual antioxidant (uric acid) and two assays to evaluate oxidant concentrations (advanced oxidation protein products (AOPP) and hydrogen peroxide (H2O2)) were measured and validated in porcine saliva. In addition, the possible changes of these assays in sows' saliva during lactation were be studied. RESULTS: The methods had intra- and inter-assays coefficient of variation lower than 15%. They also showed an adequate linearity and recovery, and their detection limits were low enough to detect the analytes in saliva of pigs. Overall the analytical validation tests showed that the assays used in our study are valid and reliable for the evaluation of oxidative stress in porcine saliva. In addition, it was observed that these salivary biomarkers can change in a situation of oxidative stress such as lactation in sows. CONCLUSIONS: All assays for salivary biomarkers of oxidative stress evaluated in this study have demonstrated a high analytical accuracy and low imprecision. In addition, it has been observed that these biomarkers showed significant changes in a situation of oxidative stress such as lactation in sows. Therefore, this study opens a new possibility of using saliva as a non-invasive sample to evaluate oxidative stress in pigs.

BACKGROUND: In recent decades there has been a growing concern about animal stress on intensive pig farms due to the undesirable consequences that stress produces in the normal physiology of pigs and its effects on their welfare and general productive performance. This review analyses the most important types of stress (social, environmental, metabolic, immunological and due to human handling), and their biological consequences for pigs. The physio-pathological changes associated with stress are described, as well as the negative effects of stress on pig production. In addition an update of the different biomarkers used for the evaluation of stress is provided. These biomarkers can be classified into four groups according to the physiological system or axis evaluated: sympathetic nervous system, hypothalamic-pituitary-adrenal axis, hypothalamic-pituitary-gonadal axis and immune system. CONCLUSIONS: Stress it is a process with multifactorial causes and produces an organic response that generates negative effects on animal health and production. Ideally, a panel of various biomarkers should be used to assess and evaluate the stress resulting from diverse causes and the different physiological systems involved in the stress response. We hope that this review will increase the understanding of the stress process, contribute to a better control and reduction of potential stressful stimuli in pigs and, finally, encourage future studies and developments to better monitor, detect and manage stress on pig farms.

Understanding the behavior of endangered species is crucial to improve the management tools to breed animals in captivity and, thus, to increase the success of ex situ conservation programs. In this study, we monitored suckling behavior of 26 cubs born between 2008 and 2012 at "El Acebuche" Iberian Lynx Breeding Centre. The cubs devoted 251 ± 19.7 min (mean ± SE) to lactation on the day of birth, while mothers spent 426 ± 27 min (mean ± SE) nursing their offspring. The time cubs spent suckling decreased exponentially as they grown, until they were fully weaned at 65 ± 2.6 days. The onset of weaning (first intake of solid food) occurred at 54 ± 1.35 days (mean ± SE). Thus, the strict lactation period occupied most of the overall lactation period. Both suckling and maternal behavior were affected by litter size. In twins and triplets, the competition between siblings caused a decrease in the time spent suckling, in spite of the mothers spending more time nursing their young. Finally, no significant differences were found in time spent suckling between littermates or depending on the sex of the cub. Lactation appeared to play a key role in the nutrition of the Iberian lynx and should therefore be conveniently managed in captive breeding programs of this threatened species. Ensuring welfare in captive wild animal populations is important not only for ethical and legal reasons, but also to maintain healthy individuals and populations. An increased level of social behaviors such as aggression can reduce welfare by causing physical damage and chronic stress to animals. Recently, cortisol in hair has been advanced as a non-invasive indicator to quantify long-lasting stress in many species. The sensitivity of social behavior and hair cortisol concentration was evaluated in several groups of dorcas gazelles (Gazella dorcas). Four different groups of gazelles from three different zoos were observed and the expression of intra-specific affiliative and negative social behaviors was assessed across the different groups. Hair samples were taken from sub-groups of animals and analyzed for cortisol concentrations. Significant differences between groups of dorcas gazelles were found in frequency of negative social behavior and hair cortisol concentration. Despite the low sample size, these two parameters had a positive Spearman correlation coefficient (rs = +0.80, P = 0.20). These results suggest that hair cortisol levels are sensitive to differences in the social structure of dorcas gazelles. Zoo Biol. 35:467-473, 2016. © 2016 Wiley Periodicals, Inc.

Knowledge on the growth and early development patterns of endangered species can become a useful conservation tool because it may allow detecting anomalous growth in newborns, both in captivity breeding and in the wild. We studied the growth and early development of 40 Iberian lynx (Lynx pardinus) cubs belonging to 21 litters born in captivity between 2005 and 2012 at "El Acebuche" Iberian Lynx Breeding Centre. This is the first study on growth in this critically endangered species. The Iberian lynx cubs were not fully developed at birth. During the first 3 weeks of life, cubs underwent many of the physical changes that allowed them to improve their interaction with the environment, such as the opening of eye and auditory channels, teeth eruptions, and the ability to walk. When the cubs were 1 month old, they were ready to leave the den and develop new behaviors such as the exploration of their environment, play, or hunt. Three different models had been fitted to the body mass growth of the Iberian lynx. The von Bertalanffy curve provided the best fit. The asymptotic adult mass was the only parameter that differed between males and females (males being 8% larger), due to the higher growth rate of males. The adult weight of hand-reared cubs (i.e., those abandoned at born) did not differ from that of cubs reared by their mothers. Both growth and development showed differences from other lynx species.

Society is showing a growing concern about the welfare of cetaceans in captivity as well as cetaceans in the wild threatened by anthropogenic disturbances. The study of the physiological stress response is increasingly being used to address cetacean conservation and welfare issues. Within it, a newly described technique of extracting cortisol from epidermal desquamation may serve as a non-invasive, more integrated measure of a cetacean's stress response and welfare. However, confounding factors are common when measuring glucocorticoid hormones. In this study, we validated a steroid hormone extraction protocol and the use of a commercial enzyme immunoassay (EIA) test to measure cortisol concentrations in common bottlenose dolphin (Tursiops truncatus) and beluga (Delphinapterus leucas) epidermal samples. Moreover, we examined the effect of sample mass and body location on cortisol concentrations. Validation tests (i.e., assay specificity, accuracy, precision, and sensitivity) suggested that the method was suitable for the quantification of cortisol concentrations. Cortisol was extracted from small samples (0.01 g), but the amount of cortisol detected and the variability between duplicate extractions increased as the sample mass decreased. In common bottlenose dolphins, epidermal skin cortisol concentrations did not vary significantly across body locations while there was a significant effect of the individual. Overall, we present a contribution towards advancing and standardizing epidermis hormone assessments in cetaceans.

Alternative farrowing systems that have been developed in recent years could have a positive effect on the welfare of sows during farrowing and lactation. Oxytocin measurements in saliva may provide information about positive animal welfare status. The purpose of this study was to evaluate the changes in salivary oxytocin concentrations in sows during the lactation period in three different farrowing systems and in two different seasons. Crossbred Duroc sows (n = 34, average parity = 3.6 ± 1.80) were housed in conventional farrowing crates (FC) (n = 10) or in farrowing pens with temporary crating (TC), including SWAP (n = 12) and JFL15 (n = 12) in two different seasons: summer and winter. Saliva samples were collected for six days during lactation: days 2, 4, 12, 23, 25 (i.e., 1-day post-weaning) and 26 (i.e., 2-day post-weaning) after farrowing. Moreover, behavioral data from sows was recorded on days 2, 4, 12 and 23 after farrowing, using a 30-s scan sampling method for 3 min per pen to record the behaviors which were assessed by the same observer. The results showed that the salivary oxytocin concentrations were 472.5 pg/mL and 399.4 pg/mL higher in both TC (SWAP and JLF15, respectively) than in the FC in early-lactation period, and these differences were more pronounced in summer and at the end of lactation in winter. In terms of behavior, higher number of mother-young interactions were observed in TC than FC in early- and mid-lactation period. In conclusion, TC is associated to a higher salivary oxytocin concentration that could indicated an increased mother-young interaction, although oxytocin concentration can be influenced by other factors, such as season or day of lactation.

Ensuring high standards of animal welfare is not only an ethical duty for zoos and aquariums, but it is also essential to achieve their conservation, education, and research goals. While for some species, animal welfare assessment frameworks are already in place, little has been done for marine animals under human care. Responding to this demand, the welfare committee of the European Association for Aquatic Mammals (EAAM) set up a group of experts on welfare science, cetacean biology, and zoo animal medicine across Europe. Their objective was to develop a comprehensive tool to evaluate the welfare of bottlenose dolphins (Tursiops truncatus), named Dolphin-WET. The tool encompasses 49 indicators that were either validated through peer review or management-based expertise. The first of its kind, the Dolphin-WET is a species-specific welfare assessment tool that provides a holistic approach to evaluating dolphin welfare. Inspired by Mellor's Five Domains Model and the Welfare Quality®, its hierarchical structure allows for detailed assessments from overall welfare down to specific indicators. Through combining 37 animal-based and 12 resource-based indicators that are evaluated based on a two- or three-level scoring, the protocol offers a detailed evaluation of individual dolphins. This approach allows for regular internal monitoring and targeted welfare management, enabling caretakers to address specific welfare concerns effectively.

Medical records of 120 Iberian lynx (Lynx pardinus) from the captive breeding population (CBP), 96 of which were older than 1 wk old, were studied from January 2004 to June 2010. From a total of 413 clinical signs recorded, it was possible to obtain a diagnosis in 258 (62.5%). Inappetence, skin wound, and vomiting had the highest incidence. Adult (2 to 6 yr old) and juvenile (1 wk to 1 yr old) animals accounted for most of the clinical signs. Vitamin D toxicosis and intraspecific trauma accounted for 55.4% and 15.1% of the clinical signs, respectively. Renal toxicosis due to the administration of supplements with an excess of vitamin D occurred in 2009 and affected a total of 39 individuals. Intraspecific trauma cases were predominantly observed from sibling aggression. Diet-related conditions consisted of sporadic cases of fatal salmonellosis, dermatophytosis, and gastrointestinal episodes. Suspected idiopathic epilepsy and femoral neck metaphyseal osteopathy were also observed. A total of 15 animals older than 1 wk old died including five vitamin D toxicosis cases and three juveniles due to intraspecific trauma. Mycobacterium bovis was found as a secondary infection in two animals that died from vitamin D toxicosis. Abortions, premature births, and stillbirths accounted for 12 mortalities, and 13 neonatal deaths due to maternal neglect or bacterial sepsis were observed. Data show that improvement of diet-related conditions is a key factor in preserving the health of animals in the CBP. Thus, the control of food and supplement composition, rabbit farm suppliers, and hygiene should be standardized and improved. Furthermore, data recording and diagnostic protocols should be standardized.

RESUMO

Perphenazine enanthate was used to allow adaptation to captivity in 11 Pyrenean chamois (Rupicapra pyrenaica). At the time of capture, all animals received 0.10 mg/kg of acepromazine maleate and 2.5 mg/kg of perphenazine enanthate intramuscularly. The effect was evaluated by means of three behaviors: alertness, defecation, and flight distance. The tranquilization and lack of fear of humans of all animals were determined and the usefulness of this long-acting tranquilizer for chamois adaptation to captivity was confirmed.

Animal-welfare-assessment protocols are important for identifying welfare problems in conservation programs. This study aimed to develop a baseline welfare protocol for the assessment of the welfare of Patagonian huemuls (Hippocamelus bisulcus) in conservation centers. This protocol is based on the Welfare Quality® (WQ) framework for cattle and was developed with the consideration of the available research on the behaviors, ecology, conservation, and captive management of this species, as well as welfare-assessment protocols for other ungulate species. As a result, the protocol was specifically developed for Patagonian huemuls and included four principles, 12 criteria, and 23 animal- and resource-based indicators. The twelve criteria of the WQ protocol were reduced to nine, and three new criteria were added because they were both feasible and essential for welfare assessment in captive Patagonian huemuls. This protocol is mainly intended to identify welfare problems in endangered species in the context of conservation centers (reproduction, rescue, rehabilitation, or treatment centers). Thus, the aggregation of different measures to obtain a global score was not proposed. However, a scoring system that assigns a value on a 0-2 scale (0 = no welfare concern; 1 = welfare concern; 2 = urgent welfare concern) was proposed for each category. Although further research is still needed to fully validate the protocol, this is the first development of a protocol to assess Patagonian huemul welfare, and it can be used as a basis for the development of welfare-assessment protocols for other deer species in captivity.

Zoological institutions, such as zoos and aquariums, have made animal welfare a top priority, as it is not only a moral obligation but also crucial for fulfilling their roles in education and conservation. There is a need for science-based tools to assess and monitor animal welfare in these settings. However, assessing the welfare of zoo animals is challenging due to the diversity of species and lack of knowledge on their specific needs. This review aims to discuss the advantages and disadvantages of existing methodologies for assessing zoo animal welfare through: (1) A critical analysis of the main approaches to zoo animal welfare assessment; (2) A description of the most relevant animal-based welfare indicators for zoo animals with a particular focus on behavioural and physiological indicators; and (3) An identification of areas that require further research.

The gut microbiome is critical for maintaining host health. In healthy humans, the aging process is one of the main factors modulating the changes in the intestinal microbiota. However, little is known about the relationship between gut health, microbiota, and the aging process in dogs. The present study aims to explore the differences in the intestinal microbiota and intestinal health based on fecal biomarkers in a population of dogs of different ages. The study involved 106 dogs of different breeds aged between 0.2 and 15 years categorized as senior (>7 years; n = 40), adult (2-7 years; n = 50), and junior (< 2 years; n = 16). Fecal samples were collected during the same period at the same facilities. The analysis included the following gut health indicators: 16S rRNA gene sequencing to investigate the differences in the fecal microbiota; qPCR to determine the dysbiosis index; fecal short-chain fatty acid concentrations; fecal calprotectin; and immunoglobulin A. Beta diversity analysis revealed a significant difference with a small effect size (p = 0.003; R = 0.087) among age categories based on the unweighted UniFrac metric, but no significance was observed based on the weighted UniFrac metric or Bray-Curtis distances. There were no significant differences in the alpha diversity measures or the fecal dysbiosis index among age categories. Senior dogs had significantly higher relative abundance proportions in phyla Bacteroidota and Pseudomonadota and the genus Faecalibacterium, but not on qPCR analysis. At the family level, Ruminococcaceae, Uncl. Clostridiales.1, Veillonellaceae, Prevotellaceae, Succinivibrionaceae, and Bacteroidaceae abundances were higher in the senior category than in the adult and/or junior categories. Relative proportions, but not concentrations of fecal acetate, were higher in the senior category, while butyrate, isovaleric acid, and valeric acid were lower. The valeric acid concentration was significantly lower in the senior category than in the adult category. Calprotectin and immunoglobulin A levels did not differ significantly across groups. In conclusion, this study observed multiple minor changes in the fecal microbiota composition and the relative amount of short-chain fatty acids in dogs among different age groups, but studies in larger populations representative of all ages are warranted to refine the present results.

Cognitive dysfunction syndrome is the most common cause of cognitive decline in aged dogs. Early diagnosis is crucial because the sooner treatment is implemented, the greater the chance of slowing the progression of the disease. Assessment tools to assess cognitive decline may differ depending on the environment in which the dogs live. The aims of this study were threefold, first, to describe two feasible methods to evaluate cognitive impairment in aged dogs living in different environments: (i) a Canine Cognitive Assessment Scale (CCAS) for dogs living in a home environment and (ii) a practical cognitive test (PCT) potentially useful for dogs not living in a home environment (NHE); second, to assess the effect of age on the outcome of both tools and, finally, to compare the results of the CCAS with those of the PCT. Both methods were found to be practical to perform. Age was found to significantly predict the score obtained by the CCAS (p = 0.0011) and the outcome of the PCT (p = 0.009). However, the reversal phase from the PCT did not significantly predict the outcomes of the CCAS (p = 0.97). Taken together, these findings suggest that the CCAS is a practical method to evaluate age related cognitive changes in owned dogs. The fact that the PCT has not been proven to be related with the CCAS calls into question the use of the PCT as a sensitive tool to assess cognitive impairment. Further studies in this field are suggested.

The main conditions and diseases considered painful in dairy cows are mastitis, lameness, calving (including dystocia and caesarean section) and metritis. The cattle literature reports that deviation from normal daily activity patterns (both increased and/or reduced daily lying time) can be indicative of painful conditions and diseases in cows. This narrative review discusses on how pain due to several health conditions in dairy cows modifies its activity pattern and explores if non-steroidal anti-inflammatory drugs (NSAIDs) are capable of restoring it. Divergent outcomes may differ depending upon the painful cause, the severity and the moment, and consequently its interpretation should be properly explained. For instance, cows with clinical mastitis reduced their time lying and increased the number of lying bouts and stepping due to pain caused by the swollen udder when cows are lying. However, lame cows show longer lying times, with a lower number of lying bouts and longer and more variable lying bouts duration, as compared to non-lame cows. When the relationship between painful disorders and daily activity patterns is studied, factors such as parity, bedding type and severity of disease are important factors to take into consideration. The potential benefits of the NSAIDs treatment in painful health disorders depend upon the type of drug administered, its dosage and administration mode, and the time of administration relative to the painful health disorder. This narrative review can be used as a tool to properly interpret and grade pain in cows through behavioural activity patterns and proposes directions for future investigations.

The study investigated the effect of farrowing environment on the weaning adaptability of sows and piglets. One farrowing crate (FC) and two farrowing pens with temporary crating (TC: SWAP and JLF15) were compared. Sixty-four sows and 663 piglets were followed until 5 days post-weaning. At weaning (D24), sows and piglets were moved to group pens and nursery pens, respectively. Sows and piglets' behaviors were observed on D24, D25, and D26. On D23, D25, and D26, piglets' skin lesions were counted, and sows and piglets' saliva samples were collected for stress biomarkers (cortisol and chromogranin A, CgA). Piglets were weighed on D23 and D29. All the piglets' skin lesions increased on D25 and decreased on D26 (p < 0.05). Compared to D23, cortisol of JLF15 and CgA of FC piglets increased, whereas those of SWAP piglets remained similar after weaning (p < 0.05). Post-weaning performance in piglets was similar across farrowing systems. SWAP sows vocalized more than FC and JLF15 on D24 and D25 (p < 0.001). Results suggested that SWAP piglets showed a lower weaning stress response. Frequent post-weaning vocalization in SWAP sows might be linked with a negative effect of the abrupt separation from the piglets.

The intense nature of pig production has increased the animals' exposure to stressful conditions, which may be detrimental to their welfare and productivity. Some of the most common sources of stress in pigs are extreme thermal conditions (thermal stress), density and mixing during housing (social stress), or exposure to pathogens and other microorganisms that may challenge their immune system (immune-related stress). The stress response can be monitored based on the animals' coping mechanisms, as a result of specific environmental, social, and health conditions. These animal-based indicators may support decision making to maintain animal welfare and productivity. The present study aimed to systematically review animal-based indicators of social, thermal, and immune-related stresses in farmed pigs, and the methods used to monitor them. Peer-reviewed scientific literature related to pig production was collected using three online search engines: ScienceDirect, Scopus, and PubMed. The manuscripts selected were grouped based on the indicators measured during the study. According to our results, body temperature measured with a rectal thermometer was the most commonly utilized method for the evaluation of thermal stress in pigs (87.62%), as described in 144 studies. Of the 197 studies that evaluated social stress, aggressive behavior was the most frequently-used indicator (81.81%). Of the 535 publications examined regarding immune-related stress, cytokine concentration in blood samples was the most widely used indicator (80.1%). Information about the methods used to measure animal-based indicators is discussed in terms of validity, reliability, and feasibility. Additionally, the introduction and wide spreading of alternative, less invasive methods with which to measure animal-based indicators, such as cortisol in saliva, skin temperature and respiratory rate via infrared thermography, and various animal welfare threats via vocalization analysis are highlighted. The information reviewed was used to discuss the feasible and most reliable methods with which to monitor the impact of relevant stressors commonly presented by intense production systems on the welfare of farmed pigs.

Tryptophan, a precursor of important molecules such as serotonin, melatonin and niacin, is an essential amino acid for dogs. In pigs, tryptophan supplementation has been shown to induce a significant increase in food intake. The aim of the present study was to assess whether long-term tryptophan supplementation increases voluntary food intake in dogs and to observe whether this was accompanied by a change in serum ghrelin. In the present study, sixteen adult Beagle dogs were used, with four male and four female dogs fed diets supplemented with tryptophan (1 g/dog per d) during 81 d (Trp) and four male and four female dogs that were not supplemented (control). A voluntary food intake test was performed during 5 d following the supplementation period. The Trp group tended to show a higher food intake during the voluntary food intake test (58.0 (SE 5.37) v. 77.5 (SE 3.65) g/kg metabolic weight per d; P = 0.074). No significant differences were found for serum ghrelin concentrations.
