RESUMO

Although rare, Angiostrongylus costaricensis infection may be a more prevalent etiology of inflammatory bowel disease than ulcerative colitis and Chron's disease in endemic areas in Central and South America. The present study reviewed the occurrence of A. costaricensis in Brazil, its clinical presentation and pathology; and proposed diagnostic criteria and case definitions for abdominal angiostrongyliasis (AA). Southern and southeastern Brazilian regions are the main endemic areas, and AA affects both genders and all age groups. A review of all 23 published reports of 51 Brazilian patients highlighted the following features that were subsequently classified as minor diagnostic criteria: abdominal pain, palpable mass in the right lower abdominal quadrant, history of exposure, ileocecal tumor, and intestinal perforation with wall thickening. Proposed major criteria include right lower quadrant abdominal pain, blood eosinophilia, positive serology (antibody detection), intense eosinophilic infiltration that involves all strata of the intestinal wall, eosinophilic granulomatous reaction, and eosinophilic vasculitis. In addition to the definitions of suspected and possible cases according to increasing strength of evidence of this infection, demonstration of worms/eggs/larvae in tissues or Angiostrongylus DNA in tissues or serum are required for a confirmed diagnosis. The application of the proposed criteria and definitions may improve patient management, epidemiologic surveillance, and identification of new endemic areas.

RESUMO

Alzheimer's disease (AD) is the leading cause of dementia in older adults, having a significant global burden and increasing prevalence. Current treatments for AD only provide symptomatic relief and do not cure the disease. Physical activity has been extensively studied as a potential preventive measure against cognitive decline and AD. Recent research has identified a hormone called irisin, which is produced during exercise, that has shown promising effects on cognitive function. Irisin acts on the brain by promoting neuroprotection by enhancing the growth and survival of neurons. It also plays a role in metabolism, energy regulation, and glucose homeostasis. Furthermore, irisin has been found to modulate autophagy, which is a cellular process involved in the clearance of protein aggregates, which are a hallmark of AD. Additionally, irisin has been shown to protect against cell death, apoptosis, oxidative stress, and neuroinflammation, all of which are implicated in AD pathogenesis. However, further research is needed to fully understand the mechanisms and therapeutic potential of irisin in AD. Despite the current gaps in knowledge, irisin holds promise as a potential therapeutic target for slowing cognitive decline and improving quality of life in AD patients.

RESUMO

Eight outbreaks of acute and subacute fasciolosis are reported in sheep in the municipality of Santa Vitória do Palmar, southern Brazil, in areas used for irrigated rice cultivation, which are subject to frequent flooding. Two outbreaks occurred mid-winter during July and August, and six occurred during spring and early summer. Morbidity ranged from 3 to 66.7 %, and mortality ranged from 3 to 50 %. Clinical signs included weight loss, mucosal pallor, apathy, depression, muscle tremors, and drooling; these were followed by death within approximately 24 h. Some sheep were found dead. Gross lesions were characterized by an irregular liver capsule with fibrin deposition and hemorrhages. The liver parenchyma contained hemorrhagic tracts or irregular clear areas alternated with dark hemorrhagic areas. Histologically, hemorrhagic dark red tracts of necrotic liver parenchyma and an inflammatory infiltrate were noted; these lesions coincided with the presence of immature flukes. Langhans-type giant cells, fibrous tissue, and bile duct cell proliferation were observed in subacute cases. Chronic fasciolosis is a well-known disease to farmers in southern Brazil; nevertheless, acute and subacute fasciolosis, which are more difficult to diagnose and treat, may cause important economic losses. Efficient control of fasciolosis requires integration of measures to treat infections in the definitive host, to reduce the population of snails of the genus Lymnaea, and to avoid contact between the parasite and host by appropriate pasture management.

RESUMO

Alzheimer's disease (AD) is the most common type of dementia in the elderly. Several hypotheses emerged from AD pathophysiological mechanisms. However, no neuronal protective or regenerative drug is available nowadays. Researchers still work in drug development and are finding new molecular targets to treat AD. Therefore, this study aimed to summarize main advances in AD pharmacological therapy. Clinical trials registered in the National Library of Medicine database were selected and analyzed accordingly to molecular targets, therapeutic effects, and safety profile. The most common outcome was the lack of efficacy. Only seven trials concluded that tested drugs were safe and induced any kind of therapeutic improvement. Three works showed therapeutic effects followed by toxicity. In addition to aducanumab recent FDA approval, antibodies against amyloid-ß (Aß) showed no noteworthy results. 5-HT6 antagonists, tau inhibitors and nicotinic agonists' data were discouraging. However, anti-Aß vaccine, BACE inhibitor and anti-neuroinflammation drugs showed promising results.

RESUMO

Stress is an important factor in the development of several human pathologies. The response of rodents and humans to stress depends on many factors; some people and rodents develop stress-related mood disorders, such as depression and anxiety in humans, depression-like and anxiety-like behavior in mice and rats, while others report no new psychological symptoms in response to chronic or acute stress, and are considered susceptible and resilient to stress, respectively. Resilience is defined as the ability to thrive in the face of adversity and is a learned process that can help protect against occupational stressors and mental illnesses. There is growing interest in the underlying mechanisms involved in resilience and vulnerability to depression caused by stress, and some studies have demonstrated that individual variability in the way animals and humans respond to stress depends on several mechanisms, such as oxidative stress, neuronal plasticity, immunology and genetic factors, among others not discussed in this review, this review provides a general overview about this mechanism.

RESUMO

Five years after the identification of Zika virus as a human teratogen, we reviewed the early clinical manifestations, collectively called congenital Zika syndrome (CZS). Children with CZS have a very poor prognosis with extremely low performance in motor, cognitive, and language development domains, and practically all feature severe forms of cerebral palsy. However, these manifestations are the tip of the iceberg, with some children presenting milder forms of deficits. Additionally, neurodevelopment can be in the normal range in the majority of the non-microcephalic children born without brain or eye abnormalities. Vertical transmission and the resulting disruption in development of the brain are much less frequent when maternal infection occurs in the second half of the pregnancy. Experimental studies have alerted to the possibility of other behavioral outcomes both in prenatally infected children and in postnatal and adult infections. Cofactors play a vital role in the development of CZS and involve genetic, environmental, nutritional, and social determinants leading to the asymmetric distribution of cases. Some of these social variables also limit access to multidisciplinary professional treatment.

RESUMO

ABSTRACT: Myelolipoma is a benign neoplasm composed of mature adipocytes and hematopoietic elements. This tumor is rare in dogs and cats and can develop in several organs and tissues. This report describes a case of splenic myelolipoma in a dog characterizing its clinical-pathological and ultrasonography aspects. A 9-year-old, female, mixed-breed, castrated dog was referred to the veterinary hospital with increased abdominal circumference. Ultrasonography revealed splenomegaly with masses of not measurable dimensions along the entire length of the parenchyma. The spleen was removed, and a fragment was sent for histopathological analysis. Macroscopically, the spleen was enlarged, with irregularly nodular growths that project above the surface of the organ. Nodules were red with small yellow areas. Microscopically, the spleen was effaced by a well-demarcated and not encapsulated mass composed of histologically well-differentiated neoplastic adipose tissue with islands and nests of varying proportions of hematopoietic elements. Histological examination confirmed the diagnosis of myelolipoma. Myelolipomas are observed on ultrasound images as hyperechoic masses or nodules with homogeneous echo texture. Here, ultrasound appearance involved mixed echogenicity and heterogeneous echotexture, due to the mixture of fat and non-fatty material components of the tumor.

Some plant species within the Convolvulaceae (morning glory family) from South America, Africa, and Australia cause a neurologic disease in grazing livestock caused by swainsonine. These convolvulaceous species including Ipomoea carnea contain the indolizidine alkaloid swainsonine, an inhibitor of α-mannosidase and mannosidase II, and polyhydroxy nortropane alkaloids, the calystegines which are glycosidase inhibitors. Swainsonine has been shown to be produced by a fungal endosymbiont in legumes of the Astragalus and Oxytropis genera, where it causes a similar neurologic disease in grazing livestock called locoism. Here we demonstrate that I. carnea plants are infected with a fungal endosymbiont that was cultured from its seeds and which produced swainsonine in pure culture but not the calystegines. The same fungal endosymbiont was detected by PCR and by culturing in I. carnea plants containing swainsonine. The fungal endosymbiont belongs to the Ascomycete order Chaetothyriales. Plants derived from fungicide-treated seeds lacked swainsonine, but calystegine concentrations were unaltered.

RESUMO

ABSTRACT: The aim of the present study was to describe an outbreak of atypical interstitial pneumonia affecting 15 Angus cattle aged 7-30 months in the south of Rio Grande do Sul. The clinical course of the disease was approximately 1-3 days. The cattle were kept in the native field and had received feed containing the remains of wafers from a food factory before the outbreak. Clinical signs included dyspnea and abdominal breathing, with a clinical evolution period of up to 3 days. Nine animals presented apathy and respiratory difficulty and died within 24h, while three animals exhibited a clinical evolution period of 36-48h. Three other animals died without any clinical signs. The main macroscopic lesions were observed in the lungs, which did not collapse at the opening of the thoracic cavity and showed a rubbery consistency. On the cut surface, the organs were firm and dark red, with marked distension of the interlobular septa due to edema. Histopathological analysis revealed thickening of the interlobular pulmonary septa; interstitial edema and emphysema; mild mononuclear cell, eosinophil, and neutrophil infiltration; areas of type II pneumocyte proliferation, and thickening of the alveolar walls. Fibrin was observed in the alveolar lumen with the formation of hyaline membranes. On the basis of macroscopic and histological examinations and epidemiological data, a final diagnosis of atypical interstitial pneumonia was made. Although, the cause of the disease has not been identified, it is believed that the bran wafer meal contained ingredients that were not suitable for animals, consequently triggering the disease.

A survey of cases with history of sudden death in cattle was conducted in all necropsy protocols of the Regional Diagnostic Laboratory of the Veterinary School of the Federal University of Pelotas (LRD/UFPel) from 2000 to 2014. We considered animals that had died unexpectedly without prior illness or clinical signs 24 hours before death (Category 1), or before they were moved or suffered some type of management and died after falls or trembling (Category 2). We identified 72 cases (3.5%) from those two categories of a total of 2,031 materials of cattle received in LRD/UFPel. The cases occurred in all seasons of the year. In 34 cases (47.2%), affected cattle had more than four years of age, in 23 cases (31.9%) cattle had between two and three years, and in 11 cases (15.3%) they were up to one year of age. In four protocols (5.6%) age was not reported. In 62 cases (86.1%) the cattle were in an extensive grazing system, in seven (9.7%), they were in a semi-intensive grazing system, and in three cases (4.2%), they were maintained in an intensive farming system. Out of the 72 cases observed, 52 (72.2%) were classified in Category 1 and 20 (27.8%) in Category 2. The diseases that caused more often sudden death, were cerebral babesiosis (10/72), organophosphates poisoning (10/72), anthrax (7/72), bacillary hemoglobinuria (5/72), and fulguration (3/72). Out of the 18 cases considered inconclusive, just five full necropsiey were performed, and of the other 15, the material submitted was considered negative for Bacillus anthracis, and the material did not allow investigation for other diseases. The results showed that sudden deaths in southern Rio Grande do Sul are caused by diseases, mostly well-known and endemic to the region, which can be controlled or prevented by vaccination and appropriate management...

RESUMO

The oomycete Pythium insidiosum is the pathogenic cause of pythiosis, a life-threatening disease that affects several animal species. Canines are the second most affected species, and the disease is characterized by the development of cutaneous and gastrointestinal lesions. While concomitant cutaneous and gastrointestinal lesions are rarely found in the same animal, this report documents a case of concurrent cutaneous and gastrointestinal pythiosis in an 18-month-old female Labrador. This dog had an ulcerative cutaneous lesion on the right thoracic region for 12 months that was unresponsive to itraconazole and terbinafine therapy. Two months prior to death and concurrent with the cutaneous lesion, the dog became anorexic with frequent vomiting and bloody stools. At necropsy, a cutaneous lesion that extended subcutaneously into the intercostal muscles was observed. Additionally, the large intestine contained two lesions that caused luminal narrowing. Organs were collected, routinely processed and stained using hematoxylin and eosin and Gomori methenamine silver. Histological examination of the lesions in the large intestine and on the skin revealed areas of necrosis surrounded by a pyogranulomatous infiltrate. Occasionally, black, septate, branching hyphae were detected following staining with Gomori methenamine silver. The diagnosis of pythiosis was confirmed using immunohistochemical methods. This report describes the occurrence of concomitant gastrointestinal and cutaneous lesions in a dog and highlights the therapeutic difficulties encountered with this disease.

The epidemiological, clinical and pathological aspects of paratuberculosis diagnosed in southern Rio Grande do Sul in a beef cattle property are described. Two cattle raised extensively and that presented progressive weight loss and chronic diarrhea were necropsied. The mesenteric lymph nodes were enlarged and edematous. The intestinal mucosa was thickened and wrinkled with cerebroid aspect, especially in the final portion of the ileum, ileocecal valve and cecum. Fragments of the organs were fixed in 10% formalin, embedded in paraffin, sectioned and stained with hematoxylin and eosin (HE) and Ziehl-Neelsen (ZN). Feces samples were referred to Departamento de Medicina Veterinária, Área de Medicina Veterinária Preventiva da Universidade Federal Rural de Pernambuco, for the cultivation of Mycobacterium avium subsp. paratuberculosis with Lõwenstein Jensen for mycobacterial and PCR analysis. Histologically, granulomatous enteritis was observed in the jejunum, ileum, cecum and rectum, and in multifocal areas the duodenum and colon. Lymphangitis and granulomatous adenitis was also observed. In ZN staining numerous resistant acid-fast bacilli (AFB) within macrophages, giant cells of Langhans and mesenteric lymph nodes in the jejunum, ileum cecum and rectum were observed. There was no bacterial growth in stool samples. Five samples amplified the gene sequence IS900 specific for Mycobacterium aviumsubsp. paratuberculosis. It can be concluded that paratuberculosis, despite the few reports, occurs in beef cattle raised extensively in southern Rio Grande do Sul, Paratuberculosis may have a higher prevalence in the region than it is assumed. There is need for diagnosis of this disease and to assume effective measures for its control, as for many it is still considered an exotic malady in Brazil.

RESUMO

A retrospective study of equine skin diseases diagnosed in the Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas was conducted between 1978 and 2013. The necropsy and biopsy protocols for horses received for diagnosis were reviewed to determine the prevalence of dermatopathies in southern Brazil. The most prevalent skin diseases in decreasing order were: sarcoid [234/710 (32.9%)], exuberant granulation tissue [81/710 (11.4%)], pythiosis [67/710 (9.4%)], squamous cell carcinoma [55/710 (7.7%)], papillomatosis [33/710 (4.6%)] and habronemiasis [30/710 (4.2%)]. Other skin lesions accounted for 25.3% of all cases studied. The Crioulo breed was the most prevalent [310/710 (43.6%)]. Horses aged between 2-5 years old [230/710 (32.3%)] were the most frequently affected. The data obtained in this study demonstrate the importance of skin diseases that affect horses in southern Brazil. The most of the dermatopathies observed in horses, although not resulting in death could cause aesthetic damage resulting in animal rejection, the inability to participate in collective sports activities and economic losses due to treatment and surgery costs.

A retrospective study of the diagnoses of injuries and deaths in equine in Southern Brazil was conducted between 1978 and 2012. All necropsy protocols and other materials of the Regional Diagnostic Laboratory of the Veterinary School of the Federal University of Pelotas were reviewed. The collected data consisted of 2,026 equine materials, including 514 necropsy protocols of and 1,512 other biologic materials, including the protocols of biopsies, organs, swabs, feces, blood, and skin scrapings. From the resulting 2,026 diagnoses, 467 (23.05%) corresponded to neoplasms and tumor like lesions; 168 (8.29%) to parasitic diseases; 135 (6.66%) to bacterial diseases; 31 (1.53%) to viral diseases; 86 (4.24%) to diseases caused by fungi and oomycetes; 50 (2.47%) to intoxication and poisoning by mycotoxins; 9 (0.44%) to metabolic diseases; 60 (2.96%) to other diseases; and 75 (3.70%) to non-transmittable diseases of the digestive tract. Three hundred twenty cases (15.79%) were classified as diseases of unknown etiology. Other diagnoses accounted for 489 of the total 2,026 cases (24.14%). In 44 (8.56%) of the 514 necropsies and 91 (9.47%) of the 961 biopsies of organs mailed into the laboratory, the diagnoses were inconclusive, totaling 135 (9.15%) of the 1,475 in the category. This study demonstrates the importance of skin lesions in horses, as 31.88% (642) of the biopsies received were lesions on the skin of the animals. The most significant tumors observed were equine sarcoid, at 33.18%, and squamous cell carcinoma, at 7.94%. The most common observed causes of death were leucoenceflomalacia (7.59%), rabies (3.70%) thromboembolism by Strongylus vulgaris (2.33%) and monocytic ehrlichiosis (1.75%).

This study describes the natural poisoning of goats by Rhododendron simsii in Southern Brazil. Five of eight goats died after ingesting branches of plants, which were pruned and placed near the goats. Clinical signs observed were restlessness, shrill bleating, profuse regurgitation, looking and kicking at the flank, a loss of balance, irregular gait, staggering, and opisthotonos. At necropsy or histopathology affected goats had not showed lesions. Two goats were fed with 21.5g kg-1 bw and 30g kg-1 bw of fresh leaves of R. simsii. Both goats developed signs similar to those naturally affected goats. These findings indicated that R. simsii is highly toxic and lethal to goats.

Two outbreaks of Senecio madagascariensis poisoning in cattle occurred in October 2013 in southern Brazil. Morbidity rate ranged were 3.2% and 6.1%, and lethality rate was 100%. A third case of poisoning occurred on a property in which out of 54 cattle one died with clinical signs of intoxication. In all cases, the cattle were in areas highly infested by S. madagascariensis that was in bloom. The clinical signs were diarrhea, tenesmus, opisthotonus, and progressive weight loss. Death occurred between 10 and 15 days after the onset of clinical signs. At necropsy, lesions were edema in the mesentery, in the wall of the rumen and abomasum, and in the walls of the gall bladder; the liver was firm with marbled aspect. Histologically, the liver had proliferation of fibrous connective tissue, especially in portal areas, hepatomegalocytosis, and bile duct hyperplasia. The observation of large number of S. madagascariensis in several farms in the counties of Arroio Grande, Pedro Osório and Capão do Leão reinforces that this plant is in the process of adaptation and dissemination in this region and that other outbreaks may occur in coming years. The cases reported here apparently resulted from the consumption of the plant during the fall/winter of 2013, when it was in bloom. The quantification of alkaloids in S. madagascariensis revealed the presence of 500µg/g and 4000µg/g of pyrrolizidine alkaloids in the dry plant from two properties. It is believed that the large amount of the plant in the areas where the animals were and the amount of pyrrolizidine alkaloids found were factors that contributed to the occurrence of outbreaks...

In order to determine the cause of the sudden increase in the number of outbreaks of organophosphate poisoning, nine outbreaks diagnosed in cattle were analyzed at the Laboratório Regional de Diagnóstico, Faculdade de Veterinária, Universidade Federal de Pelotas (LRD/UFPel) between November 2013 and February 2014. In all outbreaks the animals were treated with concentration from two to 151 times higher than the concentration recommended of diazinon for tick treatment. The incorrect route of application, and the intense heat recorded at the time of the outbreaks also contributed to the large number of poisoning.

The aim of this study was to describe and discuss the epidemiological aspects of spontaneous outbreaks of poisoning by Ramaria flavo-brunnescens in cattle maintained in silvipasture system in Southern Brazil. Three outbreaks of the disease between 2011 and 2013 were diagnosed. In two outbreaks morbidity was 35 percent and 37.4 percent, and the mortality rate was 36.12 percent and 16 percent, respectively. The fatality rate in these outbreaks was 45.71 percent and 96.55 percent. The diagnosis was confirmed by the epidemiology and the presence of the mushroom in the areas where the cattle grazed, in addition to the clinical signs, gross and histological lesions. The climatic data of temperature, humidity and rainfall were statistically analyzed and no significant differences were observed between the years 2007-2013. It was not possible to confirm that the presence of R. flavo-brunnescens in eucalyptus groves is associated with rainy autumns and warm dry summers. It is likely that other factors are associated with the presence of the mushroom in the woods and its toxicity. The soil and type of forest planted may also determine the presence of the mushroom in the area. It is possible that the species of eucalyptus is also a determining factor for the occurrence of the mushroom, since in many eucalyptus forests R. flavo-brunnescens does not occur. The recognition of the mushroom by workers and farmers and the time for its development in the eucalyptus woods is essential to minimize economic losses caused by the poisoning.
