RESUMO

Duchenne Muscular Dystrophy (DMD) is the most common X-linked muscular disease affecting humans. The Golden Retriever Muscular Dystrophy model (GRMD) is considerthe most suitable for several studies. This assay aims to quantify lymphocyte subpopulations CD4, CD5, and CD8, and standardize, the serum electrophoretic profile, to understand their contribution to the pathologic process in normal Golden Retriever dogs (GR group) and dystrophic´s (GRMD group), through the umbilical cord blood, in dogs aged from 2 to 3 months (GR II and GRMD II), and in dogs over 1 year of age (GR III and GRMD III). No significant differences were observed between the CD8+ lymphocyte subpopulations of the groups studied. The CD4+ and CD5+ lymphocyte subpopulations were significantly higher in the GRMD III group compared to the GR III group. Twenty-two different proteins in the gel were identified. The serum concentrations of the proteins belonging to the GR I and GRMD I groups were significantly lower than those of the other groups. We show that expression of acute phase proteins are worst during the aging of the dogs. We hope to expand knowledge to better understand the GRMD model and the translational data.

INTRODUCTION: Ischemia-reperfusion (I/R) injury of the liver is a common area of interest to transplant and hepatic surgery. Nevertheless, most of the current knowledge of I/R of the liver derives from the hepatocyte and little is known of what happens to the cholangiocytes. Herein, we assess the sequence of early events involved in the I/R injury of the cholangiocytes. METHODS: Sixty Wistar rats were randomized in a SHAM group and I/R group. Serum biochemistry, histopathology, immunohistochemistry, transmission electron microscopy (TEM) and laser capture microdissection (LCM) were used for group comparison. RESULTS: There was peak of alkaline phosphatase 24 h after IR injury, and an increase of aspartate aminotransferase and alanine aminotransferase after 6 h of reperfusion, followed by a return to normal levels 24 h after injury. The I/R group presented the liver parenchyma with hepatocellular degeneration up to 6 h, followed by hepatocellular necrosis at 24 h. TEM showed cholangiocyte injury, including a progressive nuclear degeneration and cell membrane rupture, beginning at 6 h and peaking at 24 h after reperfusion. Cytokeratin-18 and caspase-3-positive areas were observed in the I/R group, peaking at 24-h reperfusion. Anti-apoptotic genes Bcl-2 and Bcl-xl activity were expressed from 6 through 24 h after reperfusion. BAX expression showed an increase for 24 h. CONCLUSIONS: I/R injury to the cholangiocyte occurs from 6 through 24 h after reperfusion and a combination of TEM, immunohistochemistry and LCM allows a better isolation of the cholangiocyte and a proper investigation of the events related to the I/R injury. Apoptosis is certainly involved in the I/R process, particularly mediated by BAX.

This study was designed to compare cutaneous mycoflora isolation and CD4+:CD8+ ratio in feline immunodeficiency virus (FIV)-infected cats with that in FIV-uninfected cats. Sixty cats were examined. Twenty-five were FIV-infected cats and 35 were FIV-uninfected cats. All 60 cats were FeLV-negative. Fungi were speciated and immunophenotyping of peripheral CD4+ and CD8+ T lymphocytes was performed. At least one fungal colony was isolated from 22/25 (88%) FIV-infected cats. Among the FIV-uninfected cats fungal colonies were recovered from 13/35 (37%) specimens. Dermatophytes were recovered from 2/25 (8%) FIV-infected cats (one Microsporum gypseum, one Microsporum canis) and 3/35 (8.5%) FIV-uninfected cats (M gypseum). Malassezia species was the most commonly isolated organism from both groups of cats (51.6%). Malassezia species was more commonly isolated from FIV-infected cats than FIV-uninfected cats (84% vs 28.6%). The CD4+ to CD8+ lymphocyte ratio for FIV-infected cats was significantly lower than the CD4+ to CD8+ ratio in the FIV-uninfected cats. The CD4+ to CD8+ lymphocyte ratio for FIV-infected cats with cutaneous overall fungal isolation was significantly lower than the CD4:CD8 lymphocyte ratio in the FIV-infected cats but without cutaneous fungal isolation. We can conclude that immunologic depletion due to retroviral infection might represent a risk factor to cutaneous fungal colonization in cats.

The domestic cat was named Felis catus by Carolus Linnaeus in his book Systema Na turae, in 1798. The family Felidae has many morphological similarities with wild felines. The study of the embryology of the domestic cat is of great value considering its importance as an experimental model for the wild cats endangered from extinction, especially in the research related to reproductive biology. The objective of this study is the descriptive embryology of the domestic cat at different stages of pregnancy, through macroscopic description of photographic records, radiographic and alizarin technique, and microscopic description of photographic records by light microscopy. In embryos with an estimated gestational age of 17 days we observed macroscopically an expansion corresponding to the rostral forebrain, the placoid site of lens, cervical flexure, the four pharyngeal arches with grooves dividing the cardiac prominence, a sign of the limb bud, and the presence of somites. In the caudal region of the embryo, we saw the cranio-caudal bend, allowing the same position in format of a "C". In embryos with an estimated age of 22 days, we noticed macroscopically the forebrain, optic vesicle pigmentation of the retina, the optic vesicle, fourth ventricle, liver, fore and hind limbs with a slight distinction between the digits and superficial vascularization. In embryos with an estimated age of 25 days we noticed presence of the forebrain and midbrain, the pronounced cervical curvature of the optic vesicle with strong pigmentation of the retina, the optic vesicle, limbs and chest well developed, distinguishing the digits and pronounced the liver Fetuses with estimated age of 52 days have internal and external structures easily identified in adult animals. With respect to the bone structure we noted that they did not have any radial bone formed, only bone shafts. Microscopically, the embryo of the domestic cat with CR of 0.9cm and estimated age of 19 days revealed the presence of beak, oral cavity with upper and lower nasal cavity, eye and opening of the 4th ventricle of the brain, esophagus, heart with atrium and ventricle, lung, liver, mesonephric ridge, primitive gonad, stomach, forelimb bud, spine and spinal cord in development. This paper is of great importance for study of the internal and external morphology of domestic cats for better understanding of the embryonic development of the species.


O gato doméstico (Felis catus) foi nomeado por Carolus Linnaeus em seu livro Systema Naturae, em 1798. A família Felidea apresenta muita semelhança morfológica com os felinos selvagens. O estudo da embriologia do gato doméstico é de grande valia, uma vez que, é considerado um importante modelo animal quando comparado aos gatos selvagem em extinção, especialmente relacionado às pesquisas sobre biologia reprodutiva. Este trabalho objetivou análisar e comparar as fases embrionárias de quatro embriões e um feto de felinos domésticos. Nos embriões com idade gestacional estimada em 17 dias (0,5cm CR) podemos observar pela análise macroscópica a presença de dilatação rostral correspondente ao prosencéfalo, o local placóide do cristalino, a flexura cervical, os quatro arcos faríngeos com os sulcos que o dividem, a proeminência cardíaca, o indício do brotamento do membro pélvico, além da presença de somitos. Na região caudal do embrião, visualizamos a curvatura cranio-caudal, permitindo ao mesmo uma posição em formato de "C". Nos embriões com idade gestacional estimada em 22 dias (1,2cm CR), na análise macroscópica foi visualizado o prosencéfalo, vesícula óptica com pigmentação da retina, vesícula ótica, quarto ventrículo, fígado, membros torácicos e pélvicos com discreta distinção dos dígitos e vascularização superficial. Nos embriões com idade gestacional estimada em 25 dias (1,5cm CR) notamos a presença do prosencéfalo e mesencéfalo, a curvatura cervical pronunciada, vesícula óptica com forte pigmentação da retina, vesícula ótica, membros pélvicos e torácicos bem desenvolvidos, com distinção dos dígitos e fígado bem pronunciado. Os fetos com idade gestacional estimada em 52 dias (10cm CR) possuem estruturas internas e externas facilmente identificadas em animais adultos. Com relação às estruturas ósseas notamos que as mesmas não apresentam nenhuma epífise óssea formada, sendo visíveis somente as diáfises ósseas. Na análise microscópica, o embrião de idade gestacional de 19 dias (0,9cm CR) revelou a presença do rostro, cavidade oral com lábio superior e inferior, cavidade nasal, olho e a abertura do 4º ventrículo encefálico, esôfago, coração com átrio e ventrículo, pulmão, fígado, crista mesonéfrica, gônada primitiva, estômago, broto do membro torácico, coluna vertebral e a medula espinhal em formação. Esse trabalho é de grande importância para o estudo da morfologia externa e interna de gatos domésticos, principalmente no que diz respeito ao desenvolvimento ósseo e articular, considerando as alterações que podem ou não ser promovidas pelo uso de terapias medicamentosas ou celulares durante o desenvolvimento embrionário e fetal.

The umbilical cord blood (UCB) is an important source of pluripotent stem cells, which motivated researches on ontogeny and transplantation. The morphological characterization of umbilical cord cells is the first step to establish subsequent experiments on these areas. Although some information on humans can be found, no data on UCB is available for bovines. Therefore, this work is the first attempt to conduct an ultrastructural characterization of bovine umbilical cord blood. Blood was collected from the umbilical cord of twenty fetuses by punction of the umbilical vein. Samples were processed for whole leucocytes observation by centrifugation and the buffy coat was collected. Cells were washed and pelleted and prepared according to the standard protocol of the transmission electron microscopy. The presence of cells with morphologic characteristics compatible with the precursors from the erythrocytic, neutrophilic, eosinophilic, basophilic, and lymphocytic lineages was observed. Atypical cells with peculiar morphological features, strongly similar to apoptotic cells, were seen. Bovine neutrophils with three types of cytoplasmic granules were also found in the blood. The ultrastructural characteristics of observed bovine UCB cells where similar to those found in other species, suggesting that bovines could possibly constitute an experimental model for approaches on UCB cells research.


O sangue de cordão umbilical (SCU) é uma importante fonte de células progenitoras pluripotentes, que motiva pesquisas em ontogenia e transplantes. A caracterização morfológica das células de cordão umbilical é o primeiro passo para se estabelecer experimentos subsequentes nessas áreas. Embora algumas informações sobre SCU em humanos possam ser encontradas, não existe nenhuma informação disponível sobre elas em bovinos. Portanto, este trabalho é a primeira tentativa de se conduzir uma caracterização ultra-estrutural do sangue de cordão umbilical bovino. O sangue foi coletado do cordão umbilical de 20 fetos por punção da veia umbilical. As amostras foram processadas para observação dos leucócitos totais por centrifugação pela coleta do botão leucocitário. As células foram lavadas, peletizadas e preparadas de acordo com protocolo padrão para microscopia eletrônica de transmissão. A presença de células com características morfológicas compatíveis com precursores das linhagens eritrocítica, neutrofílica, eosinofílica, basofílica e linfocítica foram observadas. Células atípicas com características morfológicas peculiares muito semelhantes a células em apoptose foram observadas. No sangue do cordão umbilical também foi encontrado neutrófilos bovinos apresentando três tipos de grânulos citoplasmáticos. As características ultraestruturais do SCU bovino foram semelhantes às encontradas em outras espécies, sugerindo que esta espécie possa servir como modelo experimental para abordagens em pesquisa sobre sangue de cordão umbilical.
