INTRODUCTION: Technological advancements enabled the analyses of limited sample volumes on 1H NMR. Manual spectral profiling of the data is, however, complex, and timely. OBJECTIVE: To evaluate the performance of BAYESIL for automated identification and quantification of 1H NMR spectra of limited volume samples. METHOD: Aliquots of a pooled African elephant serum sample were analyzed using standard and reduced volumes. Performance was evaluated on confidence scores, non-detects and laboratory CV. RESULTS: Of the 47 compounds detected, 28 had favorable performances. The approach could differentiate samples based on biological variation. CONCLUSIONS: BAYESIL is valuable for limited sample 1H NMR data analyses.

Natural food items and the additional benefits they provide have received considerable attention in recent years. Betalains are nutritious pigments which have valuable biological properties, e.g., antioxidant and free radical scavenging activity. In this study, aqueous betalain extracts were obtained from different coloured cactus pears (purple, red/pink, and orange), amaranth, and beetroot, with and without the addition of ascorbic acid, microwave-heated, and freeze-dried and subsequently analysed by thin layer chromatography (TLC). Beetroot samples without the addition of ascorbic acid (AA) had lower phenols, flavonoids, and ascorbic acid content than beetroot samples extracted with the addition of AA. Amaranth had significantly higher contents of antioxidants than all the other plants. Results for phenolic compounds showed that there were no significant differences between cactus pear cultivars, however, significant differences were seen between the two beetroot samples (microwave-heated with and without AA) as well as amaranth. For flavonoid compounds, amaranth had significantly higher values than all other samples. The lowest flavonoid content was found in beetroot without AA (0.49 mgCE/g). For ascorbic acid, significant differences were noticed between amaranth (71.71 mg/100 g) and samples from cactus pear and beetroot. TLC results showed that purple and red cactus pear samples had the most vivid colours, a reflection of the high betacyanin and betaxanthin contents in the cultivars. Moreover, extracts from cactus pear, beetroot, and amaranth were classified according to a decision tree which was designed by the Code of Federal Regulations/Food Additives Regulation of the EU. The classification of betalain pigment extracts as colouring foods was achieved through enrichment factor calculations and the colourant decision tree. The results showed that the betalain pigment extraction method used is inexpensive, time-saving, energy-saving, non-toxic, and chemical solvent free and yields high concentrations of betalains.

The proximate composition of milk from fifteen free-ranging white rhinoceroses at different lactation stages is reported with detailed analysis of fatty acid composition and minerals. Lactose is the main component at 7.93 ± 0.53%, followed by 0.93 ± 0.19% protein, 1.76 ± 0.39% fat, 0.40 ± 0.18% ash, 0.05 ± 0.01% glucose, and 0.04 ± 0.02% non protein nitrogen The interindividual variation of all the components is high, showing no trend of change over lactation. The K and P content decreased over lactation. The fatty acid composition of rhinoceros milk is characteristic with a high saturated fatty acids content of 62%-84%, of which the medium chain fatty acids form the major portion. The C10:0, C12:0, C16:0, and C18:1c9 are the major fatty acids in the milk fat. The results are compared with the two other Perissodactylae families, the Equidae and Tapiridae. Differences in gross composition are small, but the milk of the Rhinocerotidae have the lowest gross energy, while the milk fats consist of the highest amounts of saturated fatty acids, while the low levels of C16:1c9 and C18:1c9 indicate the lowest mammary Δ9-desaturase activity.

This research paper addresses the hypothesis that comparative genomics can give a new insight into the functionality of casein genes with respect to the casein micelle. Comparative genomics is a rapidly emerging field in computational biology whereby two or more genomes are compared in order to obtain a global view on genomes as well as assigning previously unknown functions for genes. Casein genes are among the most rapidly evolving mammalian genes, with the gene products mainly grouped into four types (αs1-, αs2-, ß- and κ-casein). Functionally, casein genes are central to the casein micelle, the exact structure of which is still a subject of intense debate. Moreover, and adding to this complexity, some mammals lack some of the casein genes, although casein micelles have been observed in their milk. This observation has prompted an investigation into the distribution of casein genes across a host of mammalian species. It was apparent from this study that casein gene sequences are very diverse from each other and we confirmed that many mammalian species lack one or more of the casein genes. The genes encoding ß- and κ-caseins are present in most mammals whereas α-casein encoding genes are less represented. This suggests different mechanisms for casein micelle formation in different species as well as the functions that are assigned to each individual casein.

Water soluble autolysate of yeast, usually utilised for microbial growth support, was used as additive in yoghurt fermentation. The yeast extract (YE) resulted in a decrease of fermentation time by 21% to reach a pH of 4·6. However, the YE resulted in unacceptable flavour and taste. By size exclusion chromatography, a fraction of the YE was obtained that could account for the observed 21% decrease in fermentation time. The fraction contained molecules of low molecular weight, consisting of minerals, free amino acids and peptides. The acceleration of the yoghurt fermentation was ascribed to the short peptides in the fraction. It is proposed that the application of this extract in industrial yoghurt manufacture would result in savings for both the industry and the consumer.

Elephant milk composition is unique, as are its changes over lactation. Presented here is the milk non-dedicated metabolite composition of three African elephants. Their lactation times are overlapping and span day one to thirty months. Metabolites were identified and quantified by 1H nuclear magnetic resonance spectroscopy. Lactose and short oligosaccharides are a large component of the metabolites, with lacto-N-difucohexaose I as the major oligosaccharide. These were followed by metabolites of lipids, amino acids, and the citric acid cycle. The content of lactose, lacto-N-difucohexaose I, 2'-fucosyllactose, and some unidentified oligosaccharides decrease over lactation, while that of difucosyllactose and other unidentified ones increase. The high content of glutamate, as a glucogenic amino acid, supported the uprated synthesis of saccharides by the milk gland cells. The content of succinate and choline increase over lactation, indicating higher energy expenditure and phospholipid synthesis during later lactation.

Maize is the staple food crop for millions of people in sub-Saharan Africa. Iron (Fe) and zinc (Zn) deficiency is a significant health risk that mainly affects low-income populations who rely solely on maize-based diets. This problem can be alleviated by developing micronutrient-rich maize grain. The aim of this study was to determine the adaptation and performance of hybrids for Fe and Zn concentration and grain yield under low soil nitrogen (N) and optimal conditions. Eighteen hybrids derived from lines and testers with low, medium, and high Fe and Zn concentration were grown during the summer rainy seasons of 2017 and 2018 at three locations under low and optimal N conditions. There were significant genotype and environment effects for grain yield, and Fe and Zn concentration, but the genotype by environment interaction effects were the largest, accounting for between 36% and 56% of variation under low N conditions. Low N levels significantly reduced grain yield, and Fe and Zn concentration. Hybrids G1, G2, G4, G7, G10, G11, and G16 were relatively stable, with relatively high mean Fe and Zn concentrations, and low additive main effects and multiplicative interaction (AMMI) stability values and iron stability index (FSI) and zinc stability index (ZSI) under low N conditions. These genotypes can be considered for production under low N stress conditions. Two environments (E4 and E3) were identified for good discriminatory power for genotype performance in terms of Fe and Zn content, respectively. Stable and high-yielding genotypes with high Fe and Zn concentration can be used as biofortified hybrids, which can contribute to a sustainable solution to malnutrition in the region, especially under low N conditions.

Iron (Fe) and zinc (Zn) nutrient enrichment of staple crops through biofortification can contribute to alleviating micronutrient deficiency in sub-Saharan Africa. A line × tester mating design was used to determine the general combining ability (GCA), specific combining ability (SCA) and heterosis for grain yield, iron, Zn and phytic concentration of six lines crossed with three testers. Lines and testers were selected for high, intermediate and low mineral content. The F1 hybrids and parental lines were evaluated under low nitrogen (N) and optimum conditions across four environments over two seasons. Under low N conditions, Fe and Zn concentration in grain, and grain yield of genotypes were reduced by 9%, 9%, and 59%, respectively. However, phytic acid concentration in grain was increased by 10% under low N conditions. Both additive and non-additive gene effects were important in controlling Fe, Zn and phytic acid concentration in grain and grain yield of maize under both N conditions. The preponderance of GCA effects indicates the importance of additive gene effects in the inheritance of grain yield. Line GCA effects were more sensitive to N conditions across the environments than the tester GCA. High and significant positive SCA effects for grain yield, Fe and Zn content under low N conditions, would be a good indicator of possible heterosis in these traits. Hybrid CBY101 LM-1600 × CBY358 LM-1857 had high and significant positive SCA for grain yield under low N conditions and is a promising candidate for production in low N environments. CBY358 LM-1857 (tester) and CBY102 LM-1601 (line) are a good general combiners for Fe, Zn and GY can be used as parents in future maize hybrid breeding programs to develop high-yielding maize genotypes with high Fe and Zn content.

Pectin and alginate satisfy multiple functional requirements in the food industry, especially relating to natural packaging formulation. The continuous need for economic and environmental benefits has promoted sourcing and investigating alternative biomaterials, such as cactus pear mucilage from the cladodes of Opuntia spp., as natural packaging alternatives. The structural and mechanical properties of mucilage, pectin and alginate films developed at a 5% (w/w) concentration were modified by treating the films with calcium (Ca) in the calcium chloride (CaCl2) form. Scanning electron microscopy (SEM) showed the 5% (w/w) 'Algerian' and 'Morado' films to display considerable microstructure variation compared to the 5% (w/w) pectin and alginate films, with calcium treatment of the films influencing homogeneity and film orientation. Treating the alginate films with a 10% (w/w) stock CaCl2 solution significantly increased (p < 0.05) the alginate films' tensile strength (TS) and puncture force (PF) values. Consequently, the alginate films reported significantly higher (p < 0.05) film strength (TS and PF) than the pectin + Ca and mucilage + Ca films. The mucilage film's elasticity was negatively influenced by CaCl2, while the pectin and alginate films' elasticity was positively influenced by calcium treatment. These results suggest that the overall decreased calcium sensitivity and poor mechanical strength displayed by 'the Algerian' and 'Morado' films would not make them viable replacements for the commercial pectin and alginate films unless alternative applications were found.

Pectin and alginate are well-established biopolymers used in natural film development. Single-polymer mucilage films were developed from freeze-dried native mucilage powder of two cultivars, 'Algerian' and 'Morado', and the films' mechanical properties were compared to single-polymer pectin and alginate films developed from commercially available pectin and alginate powders. The casting method prepared films forming solutions at 2.5%, 5%, and 7.5% (w/w) for each polymer. Considerable variations were observed in the films' strength and elasticity between the various films at different polymer concentrations. Although mucilage films could be produced at 5% (w/w), both cultivars could not produce films with a tensile strength (TS) greater than 1 MPa. Mucilage films, however, displayed > 20% elongation at break (%E) values, being noticeably more elastic than the pectin and alginate films. The mechanical properties of the various films were further modified by varying the pH of the film-forming solution. The various films showed increased TS and puncture force (PF) values, although these increases were more noticeable for pectin and alginate than mucilage films. Although single-polymer mucilage films exhibit the potential to be used in developing natural packaging, pectin and alginate films possess more suitable mechanical attributes.

The major nutrient and fatty acid composition of the milk of impala and tsessebe is reported and compared with other Bovidae and species. The proximate composition of impala milk was 5.56 ± 1.96% fat, 6.60 ± 0.51% protein, and 4.36 ± 0.94% lactose, and that of tsessebe milk was 8.44 ± 3.19%, 5.15 ± 0.49%, and 6.10 ± 3.85%, respectively. The high protein content of impala milk accounted for 42% of gross energy, which is typical for African Bovids that use a "hider" postnatal care system, compared to the 25% of the tsessebe, a "follower". Electrophoresis showed that the molecular size and surface charge of the tsessebe caseins resembled that of other Alcelaphinae members, while that of the impala resembled that of Hippotraginae. The milk composition of these two species was compared by statistical methods with 13 other species representing eight suborders, families, or subfamilies of African Artiodactyla. This showed that the tsessebe milk resembled that of four other species of the Alcelaphinae sub-family and that the milk of this sub-family differs from other Artiodactyla by its specific margins of nutrient contents and milk fat with a high content of medium-length fatty acids (C8-C12) above 17% of the total fatty acids.

The results presented describe and compare the fatty acid composition and melting properties of captive, healthy wild, and pansteatitis-affected wild crocodiles (Crocodylus niloticus). Differences in fatty acid composition between intramuscular and adipose fat is noted in captive crocodiles, and the latter differs from wild crocodiles as a result of different diets. Adipose fat of healthy wild crocodiles differs minimally from diseased ones, respectively with 37.3+/-2.6% vs. 43.2+/-2.3% monounsaturated fatty acids, and 43.2+/-2.9% in dead crocodiles, while polyunsaturated fatty acids decrease from 27.3+/-1.9% to as low as 21.9+/-3.6% respectively. Of the unsaturated fatty acids 18:2n-6 decreased from 6.5+/-2.6% in unaffected crocodiles to 3.5+/-0.6% in highly affected and 3.2+/-0.4% in dead crocodiles, and 22:5n-3 from 2.8+/-0.6% to 1.8+/-0.3% and 2.2+/-0.3% respectively. The melting properties as determined by differential scanning calorimetry show that extracted adipose fat is a small degree softer in pansteatitis-affected tissue, specifically in the temperature range 7-36 degrees C, and does not contribute to the hard texture noted for adipose fat tissue of pansteatitis-affected animals. A high moisture content of 51.0+/-19.7% of the fat tissue of pansteatitis-affected animals vs.17.1+/-8.0% of healthy ones, suggests that physiological changes due to interstitial inflammation may contribute to the hard texture.

Little is known about the performance of South African cactus pear varieties in different agro-ecological regions. Effects of locality on internal quality parameters of available cactus pear varieties were examined. With only one exception, no significant differences among the mean replication values for the different parameters between the different locations were observed. The differences between mean values for most individual parameters at the three localities were highly significant. Highly significant differences between the mean values for the measured characteristics were observed, not only among the locations (except for the pulp glucose values), but also for the influences of genotype and interaction between locality and genotype. Significant variations existed between mean values of the different characteristics between localities. Genotype x environmental interactions were noted. It was concluded that Meyers is the most appropriate cultivar for economical purposes in South Africa.

The combined data of milk composition of 14 African elephants over 25 months of lactation are presented. The milk density was constant during lactation. The total protein content increased with progressing lactation, with caseins as the predominant protein fraction. The total carbohydrates steadily decreased, with the oligosaccharides becoming the major fraction. Lactose and isoglobotriose reached equal levels at mid lactation. The milk fat content increased during lactation, as did the caprylic and capric acids, while the 12 carbon and longer fatty acids decreased. The fatty acid composition of the milk phospholipids fluctuated, and their total saturated fatty acid composition was low compared to the triacylglycerides. The milk ash and content of the major minerals, Na, K, Mg, P, and Ca, increased. Vitamin content was low, Vitamin E occurred in quantifiable amounts, with traces of vitamins A, D3, and K. The energy levels of African elephant milk did not change much in the first ten months of lactation, but they increased thereafter due to the increase in protein and fat content. The overall changes in milk composition appeared to be in two stages: (a) strong changes up to approximately 12 months of lactation and (b) little or no changes thereafter.
