Different starch-to-protein ratios were compared among neutered and spayed domiciled cats. Male and female obese and non-obese cats were fed kibble diets ad libitum for 4 months high in starch (HS (38 % crude protein (CP)): starch 32 %, protein 38 %; DM basis) or high in protein (HP (55 % CP): starch 19 %, protein 55 %) but similar in energy and fat in a crossover design. Physical activity was evaluated using an accelerometer, and body composition (BC), energy expenditure (EE) and water turnover (WT) using the doubly labelled water method. Results were compared in a 2 diet × 2 sex × 2 body condition factorial arrangement. Cats fed the HS (38 % CP) diet maintained a constant body weight, but lean mass (LM) tended to be reduced in female obese but to be increased in male non-obese (P < 0·08) and increased in female non-obese cats (P = 0·01). The HP (55 % CP) diet induced an increase in cat body weight and LM (P < 0·05) without altering BC proportion. EE tended to be higher in males (351 (se 8) kJ/kg0·67/d) than females (330 (se 8) kJ/kg0·67/d; P = 0·06), was unaffected by diet or BC, decreased as age increased (R 2 0·44; P < 0·01) and increased as physical activity increased (R 2 0·58; P < 0·01). WT was higher for the HP (55 % CP) diet (P < 0·01) and increased with EE (R 2 0·65; P < 0·01). The HS (38 % CP) diet favoured body weight control during 4 months of ad libitum feeding. Caution is necessary to balance protein in diets of female obese cats over 5 years, as they may have low energy and food intake, with LM loss.

Hydrolysed proteins have been shown to be potential ingredients in cat diets due to their high digestibility, presence of bioactive peptides, and relatively low antigenicity. The effects of the substitution of conventional low ash poultry byproduct meal (PBM) with hydrolysed poultry byproduct meal (HPM) as a protein source were evaluated in extruded cat diets. Five diets with similar nutrient contents were formulated: a control (CO) diet based on PBM and 4 diets with different inclusions of HPM (5%, 10%, 20%, and 30%, on an as-fed basis) replacing PBM as the protein source. The total tract apparent digestibility (CTTAD) of nutrients, faecal characteristics and microbial fermentation products, urine production and pH, nitrogen balance and urea renal excretion were evaluated using 30 healthy cats (15 males and 15 females; 4.18 ± 0.86 kg; 4.17 ± 1.38 years old), with 6 cats per diet in a complete randomised block design. When significant differences were found with the F test, the effects were evaluated by polynomial contrasts according to HPM inclusion (p < 0.05). The CTTADs of DM (89 ± 0.41%), CP (90 ± 0.36%), fat (93 ± 0.41%) and gross energy (90 ± 0.33%) were similar among treatments (p > 0.05). The faecal production, score, short-chain fatty acids and ammonia concentration were similar among treatments (p > 0.05). Isobutyric, isovaleric, valeric, and total branched-chain fatty acid contents increased quadratically (p < 0.05), with the highest level in the faeces of cats fed the diet with 20% HPM. Lactate concentration in faeces increased linearly with the inclusion of HPM (p < 0.05). Urine characteristics and urea renal excretion did not differ among treatments (p > 0.05). At 10% inclusion, HPM tended to increase the nitrogen retention of cats (p = 0.083), which may reflect the higher tryptophan, methionine, lysine, and available lysine contents of HPM in comparison to PBM. The inclusion of up to 30% HPM can be considered in cat formulations without affecting nutrient digestibility or faecal and urine characteristics. HPM tended to increase nitrogen retention and increased branched-chain fatty acids in faeces, aspects which deserves further studies.

Ferns and lycophytes are pioneer plants that can be useful for revegetation. Their natural distribution and interaction with soil fungal endophytes can increase plant fitness but have received little attention. This study aimed to identify these plant species in mine wastes, and determine colonization by arbuscular mycorrhizal fungi (AMF) and dark septate endophytes (DSE). The pseudo-total and diethylenetriamine pentaacetic acid (DTPA)-extractable rhizosphere concentrations of As, Cu, Cd, Pb, and Zn, bioavailability index (BI), and bioconcentration factor (BCF) were analyzed. Six ferns and one lycophyte were identified. Arsenic and metal concentrations were high, which were plant and site-dependent. All species showed hyperaccumulation of As in fronds, especially Argyrochosma formosa (2,883) and Notholaena affinis (2,160) had the highest concentrations (mg kg-1). All plants were colonized by AMF (3%-24%) and DSE (2%-33%). Astrolepis sinuata and Myriopteris notholaenoides had the maximum colonization by AMF and A. formosa by DSE. This study identifies for the first time five ferns and one lycophyte species on mine wastes, their As hyperaccumulation capacity and the simultaneous fungal colonization by AMF and DSE. These are relevant plant traits for phytoremediation. However, fungal identification and the role colonization by AMF and DSE requires full analysis.


Arsenic accumulator ferns and a lycophytes species naturally established on three polluted sites were found. Astrolepis integerrima, A. sinuata, Myriopteris notholaenoides, Notholaena affinis, N. sulphurea, and the lycophyte Selaginella lepidophylla are for the first time reported in these sites. This is the first evidence of DSE colonization in these plants growing on mine wastes in interaction with AMF-colonization. These plants may be useful in developing phytoremediation alternatives.

This retrospective study evaluated energy and nutrient intake of dogs in a weight loss programme. Ninety-four obese dogs were divided into three groups: G5-15: from 5% to 15% body weight (BW) loss (n = 55); G155-25: from 15.1% to 25% BW loss (n = 29); and G > 25: more than 25.1% BW loss (n = 10). Five brands of kibble diets designed for weight loss were analysed for crude protein, amino acids, fat, dietary fibre, and minerals. The food metabolizable energy (ME) was estimated (NRC, 2006). Data were compared inside each group using the paired t test and between groups with analysis of variance and Tukey tests (p < 0.05). The BW loss (weeks in regimen) was: G5-15, 9.8% ± 2.7% (13.5 ± 5.7 weeks); G15-25, 17.5% ± 2.7% (22.6 ± 11.9 weeks); G > 25, 30.0% ± 2.1% (50.4 ± 17.4% weeks; p < 0.01). The mean weekly BW loss rate was similar between groups (0.8% ± 0.3%; p > 0.05) but was higher during the first (0.96.6% ± 0.5%) than in the second (0.64% ± 0.4%) half of the regimen (p < 0.01). At beginning ME intake for BW loss did not differ (251.6 ± 32.2 kJ/kg0.75 /day), but it was lower for G > 25 in the second half of the regimen (230.3 ± 44.3; p = 0.02). Considering the observed ME intake of each dog, depending on the commercial product, intake below recommended for maintenance was verified for crude protein in 1%-20% of the dogs, methionine in 4%-38% of dogs, methionine plus cystine in 4%-22% of dogs, tryptophan in 7%-93% of dogs, potassium in 2%-85% of dogs and magnesium in 1% up to 95% of dogs. Although the diets presented elevated nutrient concentrations per MJ, due to the reduced energy allowance, the estimated intake of several nutrients was lower than the recommendations, highlighting the importance of changing the formulation perspective, which must prioritize the actual nutrient intake per kg of BW during the energy deficit.

Knowledge of energy requirements is necessary to optimise the nutritional management of animals. For tortoises, very little is known about their nutrient and energy requirements. Data on substrate oxidation and the implications of starch or fat intake on the energy metabolism are lacking. The present study compared the daily energy expenditures (DEE) of red-footed tortoises (Chelonoidis carbonaria) that were fed three extruded diets: a control diet high in fibre and two test diets, one with high starch content and another with high fat content. A total of 18 tortoises (5.5 ± 1.18 kg) were used in a completely randomised design, with 6 animals per diet. After 14 months of experimental diet intake and 48 h of preliminary fasting, the animals were kept for 12 h in 70-l respiratory chambers. An open "push mode" system was used to determine the carbon dioxide production and oxygen consumption levels for the subsequent calculations of DEE. The data were analysed with ANOVA, and the means were compared by using Tukey's test (p < 0.05). The body weights, chamber temperatures and food intakes of the tortoises were similar among the treatments (p > 0.05). There were no significant differences in oxygen consumption (21.7 ± 3.16 ml · kg-1 · h-1), carbon dioxide production (18.1 ± 1.96 ml · kg-1 · h-1), or DEE (9.7 ± 1.04 kJ · kg-1 d-1) between diets or sex (p > 0.05). The respiratory quotients (0.84 ± 0.11) were also similar among the diets (p > 0.05). The DEE of red footed tortoises did not differ after a long-term starch or fat intake.

The aim of the present study was to compare orange fibre, beet pulp and inulin as fibre sources for cats. A control diet (CON) was formulated without inclusion of a fibre source. Further experimental diets were also based on diet CON but were supplemented with 1% orange fibre (diet 1%OF); 3% orange fibre (diet 3%OF); 3% beet pulp (diet 3%BP) or 1% inulin (diet 1%IN). Forty cats were used in a randomised block design (4 blocks of 10 cats, 2 animals per food in each block, totalling 8 animals per treatment). Each block lasted 20 d, with 10 d of adaptation, 10 d of faecal collection for digestibility as well as evaluation of pH-value and fermentation products. The digestibility of dry matter, organic matter, crude protein, ether extract, starch and gross energy did not vary between diets. In diet 1%OF, dietary fibre revealed a greater digestibility than in diet CON (p < 0.05). Faecal production was higher in cats fed diets 3%OF and 3%BP than in those fed diet 1%IN (p < 0.05). Faecal concentrations of acetate and total short-chain fatty acids were higher for cats fed diet 3%BP than for those fed diets 1%IN and CON (p < 0.05), while diets 1%OF and 3%OF showed intermediate results. Faecal propionate concentration was higher for cats fed diet 3%BP, intermediate for diets 1%OF, 3%OF and 1%IN and lower for animals fed diet CON (p < 0.05). Compared with diets CON and 1%IN, the faecal concentration of tyramine was higher for cats fed diet 3%OF (p < 0.05). Orange fibre was fermentable; up to 3% inclusion it did not interfere with the digestibility of nutrients, faecal score and faecal moisture content, and promoted the formation of short-chain fatty acids and tyramine by the intestinal microbiota, with possible effects on intestinal function.

Most amino acid requirement trials appear for whole-body responses, but there is little information concerning amino acid incorporation in individual tissues, which may vary according to the age. L-[15 N] threonine was used to evaluate its incorporation rate and distribution among broiler tissues in different ages. Seventy-two male broiler chickens were distributed into three different phases: starter (4 to 9 days old), grower (18 to 23 days old) and finisher phase (32 to 37 days old). L-[15 N] threonine was added on balanced diets, and birds were fed for five days in each phase. Enriched samples of breast muscle, feathers, liver, jejunum and plasma were collected at 0, 6, 12, 24, 48, 72, 96 and 120 hr after fed birds with the tracer in each phase. In the tissues were analysed dry matter, nitrogen and stable nitrogen. The 15 N isotope abundance according to the time was fitted into exponential or linear equations using a same intercept. The ratio of the steepness or slope coefficients was determined to compare the L-[15 N] threonine incorporation according to the age. In addition, L-[15 N] threonine mass balances were performed to assess the L-[15 N] threonine distribution among the evaluated tissues. Except for feathers, the L-[15 N] threonine incorporation rate decreased with ageing. Taking into account the L-[15 N] threonine distribution in the tissues, only in the jejunum was not observed an increase as the broiler grew. The L-[15 N] incorporation varied in each tissue and according to the age of the broiler chickens. These outcomes could be useful to comprehend changes in amino acid requirements tissue-specific according to age.

The trial was conducted to evaluate the supplementation of E. coli phytase on performance, weight and ash of bones, as well as to determine the bioavailability of P and cost/benefit of its use in diets. A total 1,890 Cobb male day old chicks were assigned to six treatments and seven replicates with 45 birds each, distributed in a completely randomized design. The treatments were: Positive Control; Negative Control (NC1) - reduction of 0.06% avP; Negative Control 2 (NC2) - reduction of 0.12% avP; NC2 + Phytase (120 OTU); NC2 + Phytase (180 OTU); NC2 + Phytase (240 OTU), being 1 OTU equivalent to approximately 2 FTU. With different phytase inclusions, it was possible to verify a gradual increase on body weight gain, feed intake, feed conversion ratio, viability and even the bone characteristics of broilers fed diets containing reduction of P. The closest levels to the highest studied (240 OTU) showed the best results. The replacement of dicalcium phosphate by phytase supplementation is economically viable when the cost per OTU does not exceed US$ 1.4 × 10-5, US$1.2 × 10-5 and US$ 1.0 × 10-5 for the concentrations of 120, 180 and 240 OTU, respectively.

The aim of this study was to develop a method to determine the efficiency of utilization of Met and Cys using stable isotopes in order to reduce the number of sacrificed animals relative to the comparative slaughter technique. Met and Cys efficiencies were obtained separately and as total SAA values. Twenty-one 14- to 28-day-old broiler chickens were fed experimental diets containing different Met:Cys ratios (44:56, 50:50 and 56:44). Birds were given diets with daily supplements of L-(15 N) Met (60 mmol/kg) or L-(15 N2 ) Cys (35 mmol/kg) throughout the entire experimental period. Excreta were collected daily, and birds were euthanized at the end of the trial to collect feather-free bodies and feathers. Samples were analysed for 15 N and 15 N-Met content. The utilization efficiency for Met, Cys and Met + Cys for feather-free bodies was 55%, 75%, and 60%, while the efficiencies for feathers were estimated at 96%, 77% and 84% respectively.

BACKGROUND: Assuming that part of Methionine (Met) is converted into Cystine (Cys), but ignoring the rates with which such phenomenon occurs may lead to an excessive supply of Met in poultry diets. Such inconvenient could be easily avoided with the knowledge of the ideal Met:Cys/Total sulfur amino acids (TSAA) ratio and the rates of Met conversion into Cys. RESULTS: Met sources did not affect performance. Met:Cys/TSAA ideal ratio was determined using curvilinear-plateau regression model. Both optimum body weight gain and feed conversion ratio were estimated in 1007 g/day and 1.49, respectively, at 52% Met/TSAA ratio. Feed intake was not affected by Met:Cys/TSAA ratios. In the labelled amino acid assay, the rates with which Met was converted into Cys ranged from 27 to 43% in response to changes in Met:Cys/TSAA ratios, being higher at 56:44. CONCLUSION: Based on performance outcomes, the minimum concentration of Met relative to Cys in diets for broilers from 14 to 28 d of age based on a TSAA basis, is 52% (52:48 Met:Cys/TSAA). The outcomes from labelled amino acid assay indicate that highest the Met supply in diets, the highest is its conversion into Cys.

Quercetin is a natural compound that has several biological activities including anticancer activity. However, the use of this drug has been limited mostly because of its poor water solubility and low bioavailability. Therefore, the development of quercetin-loaded nanocarrier systems may be considered a promising advance to exploit its therapeutic properties in clinical setting including cancer treatment. This study evaluates the effect of orally administered nanosized emulsion containing quercetin (QU-NE) on the cytotoxicity activity against B16-F10 cells in vitro, and on subcutaneous melanoma in mice inoculated with B16-F1O cells. In vivo experiments, also evaluate the co-administration of quercetin with cisplatin in order to predict synergic effects and the renal and hepatic toxicity. The nanocarriers were prepared through the hot solvent diffusion associated with the phase inversion temperature methods. In vitro study showed reduction of cell viability in a concentration-depend manner for free quercetin and QU-NE. In vivo study, quercetin either as a free drug or colloidal dispersion was administrated at a dose of 5 mg kg(-1) twice a week for 17 days via oral route. Cisplatin was administrated at dose of 1 mg kg(-1) once a week intraperitoneally. Free quercetin and QU-NE reduced tumor growth, however, the reduction observed for QU-NE (P < 0.001 vs. control) was significantly higher than free quercetin (P < 0.05 vs. control). The association of both drugs did not show synergic effect. Besides, no renal or hepatic toxicities were observed after administration of free quercetin and QU-NE. These results suggest that an improvement in the oral bioavailability of quercetin occurred when this compound was dissolved in the oily phase of a nanosized emulsion, indicating that it might have a potential application in the treatment of melanoma.

Biliary complications like cholelithiasis and choledocholithiasis are more common in bariatric surgery patients due to obesity and rapid weight loss. Patients with a body mass index > 40 face an eightfold risk of developing cholelithiasis. Post-bariatric surgery, especially after laparoscopic Roux-en-Y gastric bypass (LRYGB), 30% of patients develop biliary disease due to rapid weight loss. The aim of this review is to analyze the main biliary complications that occur after bariatric surgery and its management. A review of the literature was conducted mainly from 2010 up to 2023 with regard to biliary complications associated with bariatric patients in SciELO, PubMed, and MEDLINE. Patients undergoing LRYGB have a higher incidence (14.5%) of symptomatic calculi post-surgery compared to those undergoing laparoscopic sleeve gastrectomy at 4.1%. Key biliary complications within 6 to 12 months post-surgery include: Cholelithiasis: 36%; Biliary colic/dyskinesia: 3.86%; Acute cholecystitis: 0.98%-18.1%; Chronic cholecystitis: 70.2%; Choledocholithiasis: 0.2%-5.7% and Pancreatitis: 0.46%-9.4%. Surgeons need to be aware of these complications and consider surgical treatments based on patient symptoms to enhance their quality of life.

Stable isotope methods have been used to study protein metabolism in humans; however, there application in dogs has not been frequently explored. The present study compared the methods of precursor (13C-Leucine), end-products (15N-Glycine), and amino acid oxidation (13C-Phenylalanine) to determine the whole-body protein turnover rate in senior dogs. Six dogs (12.7 ± 2.6 years age, 13.6 ± 0.6 kg bodyweight) received a dry food diet for maintenance and were subjected to all the above-mentioned methods in succession. To establish 13C and 15N kinetics, according to different methodologies blood plasma, urine, and expired air were collected using a specifically designed mask. The volume of CO2 was determined using respirometry. The study included four methods viz. 13C-Leucine, 13C-Phenylalanine evaluated with expired air, 13C-Phenylalanine evaluated with urine, and 15N-Glycine, with six dogs (repetitions) per method. Data was subjected to variance analysis and means were compared using the Tukey test (P<0.05). In addition, the agreement between the methods was evaluated using Pearson correlation and Bland-Altman statistics. Protein synthesis (3.39 ± 0.33 g.kg-0,75. d-1), breakdown (3.26 ± 0.18 g.kg-0.75.d-1), and flux estimations were similar among the four methods of study (P>0.05). However, only 13C-Leucine and 13C-Phenylalanine (expired air) presented an elevated Pearson correlation and concordance. This suggested that caution should be applied while comparing the results with the other methodologies.

Pleopeltis crassinervata is a fern documented in ethnobotanical records for its use in Mexican traditional medicine to treat gastric disorders and mouth ulcers. Consequently, conducting biological and pharmacological assays is crucial to validate the therapeutic efficacy of this plant within the context of traditional medicine. In the present study, we investigated the biological activity of extracts and fractions obtained from P. crassinervata organs against bacteria (Salmonella typhimurium, Salmonella typhi, Staphylococcus aureus, Proteus mirabilis, Shigella flexneri, Bacillus subtilis, Escherichia coli) and Trichomonas vaginalis using in vitro models. The precipitate fraction obtained from the frond methanolic extract showed significant antibacterial activity (minimal inhibitory concentration [MIC] 120 µg/mL) against the Staphylococcus aureus strain and was effective against both Gram-positive and Gram-negative bacteria. The hexane fraction also obtained from frond methanolic extract, showed a trichomonacidal effect with an IC50 of 82.8 µg/mL and a low cytotoxic effect. Hsf6 exhibited the highest activity against T. vaginalis, and the GC-MS analysis revealed that the predominant compound was 16-pregnenolone. The remaining identified compounds were primarily terpene-type compounds.

The objective of this study was to evaluate the inclusion of the autolyzed yeast (AY) Saccharomyces cerevisiae with or without an immunomodulator (1,3/1,6 ß-glucans) as a total/partial substitute for blood plasma (BP) in the diet of post-weaning piglets; zootechnical performance, intestinal health and microbiota, immune responses and energy metabolism were assessed. A total of 240 castrated male and female piglets, with a mean age of 22 days and mean initial weight of 5.24 ± 0.82 kg, were randomly divided into blocks of four treatments with 12 replicates. The dietary inclusions were blood plasma (BP), autolyzed yeast (AY), autolyzed yeast + immunomodulator (AYI) and 50% BP and 50% AY (BPAY). In pre-initial phase II (29-35 days), piglets fed AY showed better feed conversion (FCR = 1.358) than the piglets in the BP (1.484), AYI (1.379) and BPAY (1.442) groups, i.e., 8.49% (0.126), 1.52% (0.021) and 4.50% (0.084), respectively (p = 0.0293). In the total period (21-42 days), better FCR was observed in the AYI (1.458) group, i.e., 4.64% (0.071), 1.15% (0.017) and 4.58% (0.070), than in the BP (1.529), AY (1.475) and BPAY (1.528) groups, respectively (p = 0.0150). In piglets fed AY (n = 3) and BPAY (n = 2), there was a reduction in the number of medications, i.e., 82.35% (-14n) and 88.23% (-15n), respectively (p = 0.0001), compared with that in the BP group (n = 17). In the AY group (73.83 mg/dL), AYI group (69.92 mg/dL), and BPAY group (69.58 mg/dL), piglets exhibited increases in triglyceride levels of 79.32%, 69.83%, and 69.00%, respectively, in comparison to those in the BP group, which had triglyceride levels of 41.17 mg/dL (p = 0.0400). The beta-hydroxybutyrate concentration in the AY group (79.96 ng/µL) was lower by 31.95%, 22.64%, and 5.89% compared to the BP group (117.50 ng/µL), AYI group (103.36 ng/µL), and BPAY group (84.67 ng/µL), respectively (p = 0.0072). In the AYI group, there was modulation of the microbiota, with an increase in the relative abundance of bacteria of the genera Lactobacillus, Collinsella and Bulleidia. AY, associated or not associated with an immunomodulator, is a potential substitute for BP in diets for piglets in the nursery phase, with positive effects on immune, metabolic, and intestinal microbial performance.

The objective of this study was to evaluate the effects of yeast products (YP) and essential oils (EO) in total or partial replacement to in-feed antibiotic protocols (growth promoter and prophylactic), both in recommended doses and in overdose of prophylactic antibiotics (PA), on growth performance, and diarrhea incidence in the growing-finishing pigs; and fecal microbiota in market hogs. Four hundred pigs (20.36 ±â€…2.64 kg) were assigned to five treatments in a randomized block design: diets with prophylactic and growth promoter antibiotics (ANT); ANT with 30% more PA (ANT+30); diets with less PA and YP (ANT+Y); diets with less PA, YP and EO (ANT+Y+EO); and antibiotics-free diets with YP and EO (Y+EO). The content of the active components of the YP was 60% purified ß-1,3/1,6-glucans extracted from Saccharomyces cerevisiae yeast (Macrogard), 20% functional water-soluble MOS (HyperGen), and 18% MOS, extracted from Saccharomyces cerevisiae yeast (ActiveMOS). From 0 to 14 d, pigs of the ANT+30, ANT+Y, and ANT+Y+EO treatments showed a greater body weight (BW) and average daily gain (ADG) compared to pigs from the Y+EO group. From 14 to 35 d, pigs of ANT+30 and ANT+Y+EO treatments were heavier than Y+EO group. At 105 d, ANT pigs had a higher BW than the Y+EO group. For the entire period, ADG of ANT pigs was greater, and feed conversion ratio better than Y+EO pigs. From 0 to 35 d, pigs of the Y+EO treatment showed a higher diarrhea incidence compared to pigs of the other groups. From 49 to 70 d, ANT+Y and ANT+Y+EO treatments showed a lower diarrhea incidence than Y+EO group, which remained the case during the overall period. At 105 d, the alpha diversity of fecal microbiota by Shannon Entropy was lower in ANT, ANT+30, and Y+EO groups than observed for ANT+Y+EO group. The abundance of Firmicutes phylum and Firmicutes/Bacteroidetes ratio was higher in ANT than in ANT+Y+EO pigs. Proteobacteria phylum abundance in ANT+Y+EO was higher than ANT, ANT+Y, and Y+EO. Peptostreptococcaceae family abundance was higher in ANT, ANT+30, and ANT+Y groups than in ANT+Y+EO and Y+EO groups. ANT+Y+EO and Y+EO groups show a lower abundance of SMB53 genus than ANT and ANT+30 groups. In conclusion, the use of YP and EO, in partial replacement to the in-feed antibiotic protocols, does not reduce the growth performance, can replace antibiotic growth promotors, and reduce the in-feed use of PA in growing-finishing pigs. The use of YP and EO, together with PA, increases the microbial diversity, despite having important genera for weight gain in less abundance. Overdose of PA does not improve growth performance and reduces microbial diversity, which does not characterize it as an efficient preventive protocol.

In this work we described the synthesis, the antileishmanial activity and the molecular modeling and structure-activity relationship (SAR) evaluations of a series of chalcone derivatives. Among these compounds, the methoxychalcones 2h, 2i, 2j, 2k and 2l showed significant antileishmanial activity (IC(50)<10 µM). Interestingly 2i (IC(50)=2.7 µM), 2j (IC(50)=3.9 µM) and 2k (IC(50)=4.6 µM) derivatives presented better antileishmanial activity than the control drug pentamidine (IC(50)=6.0 µM). Our SAR study showed the importance of methoxy di-ortho substitution at phenyl ring A and the relationship between the frontier orbital HOMO coefficients distribution of these molecules and their activity. The most active compounds 2h, 2i, 2j, 2k, and 2l fulfilled the Lipinski rule-of-five which theoretically is important for good drug absorption and permeation through biological membranes. The potential profile of 2j (IC(50)=3.9 µM and CC(50)=216 µM) pointed this chalcone derivative as a hit compound to be further explored in antileishmanial drug design.

Diverse reproductive strategies shown by ferns and lycophytes allow them to colonize a variety of habitats, particularly after the incidence of natural or anthropogenic disturbances. This study assessed the presence, abundance and reproductive strategies of ferns growing in soils of temperate forests with different levels of disturbance at the Sierra Nevada mountain range in the State of Mexico. Vegetation, soil and environmental variables were recorded in 200 m2 permanent plots located in four forest stands. One-Way ANOVA and Canonical Correspondence Analysis resulted in the recognition of three landscape types defined by the degree of environmental alteration: low, moderate and severe. Also, from five soil samples collected in each stand, germination of ferns and lycophytes was induced. A positive relationship was found between the alteration degree and Cheilanthes abundance. Under a low landscape alteration regime, species richness is restricted to Cheilanthes bonariensis, C. marginata and Pellaea ternifolia subsp. ternifolia. The soil is a reservoir of spores of Cheilanthes and Pellaea ternifolia subsp. ternifolia because their spores can remain viable for different time intervals. Apogamy is the usual reproductive strategy of Cheilanthes species in disturbed ecosystems, although these species also show sexual reproduction in natural ecosystems with adequate water availability. Apogamy may be related to a shorter generation time in comparison with a low disturbed ecosystem. On the other hand, Pellaea ternifolia subsp. ternifolia only has sexual reproduction. Apogamy might be related to a faster generation in comparison with a low disturbed ecosystem.

Sepsis activates the renin-angiotensin system and the production of angiotensin II, which has a key role in the regulation of blood pressure through AT1 receptors. However, excessive activation of AT1 receptor is associated with deleterious effects. We investigated the consequences of a differential blockade of AT1 receptor caused by two doses of losartan (0.25 mg/kg or 15 mg/kg, s.c), a selective AT1 receptor antagonist on sepsis outcome. These doses reduced the effect of angiotensin II in normal rats by 30% and >90% 8 h after administration, respectively, but only the higher dose maintained its inhibitory effect (~70%) 24 h after injection. Sepsis was induced by cecal ligation and puncture (CLP). Losartan was injected 2 h after CLP and parameters were evaluated 6 and 24 h after CLP. Septic rats developed hypotension and hyporesponsiveness to vasoconstrictors, an intense inflammatory process and increase in plasma markers of organ dysfunction. The lower dose of losartan improved the vasoconstrictive response to phenylephrine and angiotensin II, reduced lung myeloperoxidase and prevented leukopenia 24 h after CLP, but it did not reduce NOS-2 expression, plasma IL-6 levels or organ injury parameters of septic rats. On the other hand, the higher dose of losartan worsened the response to vasoconstrictors, potentiated the hypotension and increased further levels of creatine, urea and lactate in septic rats. Therefore, an early and partial blockade of AT1 receptor with a low dose of losartan may counteract sepsis-induced refractoriness to vasoconstrictors thus providing an opportunity to improve the outcome of this condition.

BACKGROUND: Chronic prostatitis has been a common disease reported with high frequency in ankylosing spondylitis (AS) even from decades ago. Infectious (Chlamydia trachomatis) or non-infectious (uric acid) prostatitis can hypothetically trigger vertebral inflammation in AS. This study aimed to assess the features of chronic prostatitis in patients with AS compared to healthy controls. METHODS: A cross-sectional study including male patients with AS and healthy controls who agreed to undergo a prostate examination was conducted. Structured clinical interviews, prostate physical examinations, and cytological, biochemical, and microbiological tests on urinary samples collected before and after standardized prostatic massage (pre- and post-massage test) were performed. RESULTS: Ninety participants (45 AS patients, mean age: 52.5 ± 10.0 years, with longstanding disease, 12.4 ± 6.9 years, and 45 controls, mean age: 52.8 ± 12.1 years) were included. National Institutes of Health - Chronic Prostatitis Symptom Index (NIH-CPSI) scores were similar in the AS and control groups (4.0 [1.0-12.0] vs. 5.0 [1.0-8.5], p = 0.994). The frequencies of symptoms of chronic prostatitis (NIH-CPSI Pain Domain ≥4) were also similar in both groups (23.3% vs. 22.7%, p = 0.953). Results of polymerase chain reaction tests for Chlamydia trachomatis were negative in all tested urinary samples, and uric acid concentrations and leukocyte counts were similar in all pre- and post-massage urinary samples. CONCLUSIONS: In this study, chronic prostatitis occurred in male patients with AS, but its frequency and characteristics did not differ from those found in the healthy male population of similar age.
