RESUMO

A novel self-assembled hybrid nanocompound consisting of bismuth sulfide nanocrystals (Bi2S3 NCs) and Ag@SiO2 nanoparticles (NPs) is used to study the enhancement of photoluminescence by localized surface plasmon resonance (LSPR). Ag@SiO2 core-shell NPs were prepared by deposition of silica onto the surface of Ag NPs through the sol-gel method and followed by surface modification via 3-aminopropyltriethoxysilane for the coming conjugation with Bi2S3 NCs. We propose the photoluminescence enhancement by the LSPR effect through adjusting the thickness of silica shell and the Ag@SiO2 NP concentration. By modulating the thickness of the silica shell and the concentration of Ag NPs, the maximum enhancement of a 5.7 fold can be reached with the thickness of an SiO2 shell at 22.5 nm. A clear red shift of the emission peaks in the Bi2S3 NCs-Ag@SiO2 NPs hybrid structures is observed. Such a metal-enhanced Bi2S3 quantum dot (QD) fluorescence system may have promising applications in optoelectronic device.

Nasopharyngeal carcinoma (NPC), a malignant cancer originating from the epithelial cells of the nasopharynx, presents diagnostic challenges with current methods such as plasma Epstein-Barr virus (EBV) DNA testing showing limited efficacy. This study focused on identifying small extracellular vesicle (sEV) proteins as potential noninvasive biomarkers to enhance NPC diagnostic accuracy. We isolated sEVs from plasma and utilized 4D label-free proteomics to identify differentially expressed proteins (DEPs) among healthy controls (NC = 10), early-stage NPC (E-NPC = 10), and late-stage NPC (L-NPC = 10). Eighteen sEV proteins were identified as potential biomarkers. Subsequently, parallel reaction monitoring (PRM) proteomic analysis preliminarily confirmed sEV carbonic anhydrase 1 (CA1) as a highly promising biomarker for NPC, particularly in early-stage diagnosis (NC = 15; E-NPC = 10; L-NPC = 15). To facilitate this, we developed an automated, high-throughput and highly sensitive CA1 immune-chemiluminescence chip technology characterized by a broad linear detection range and robust controls. Further validation in an independent retrospective cohort (NC = 89; E-NPC = 39; L-NPC = 172) using this technology confirmed sEV CA1 as a reliable diagnostic biomarker for NPC (AUC = 0.9809) and E-NPC (AUC = 0.9893), independent of EBV-DNA testing. Notably, sEV CA1 exhibited superior diagnostic performance compared to EBV-DNA, with a significant incremental net reclassification improvement of 27.61 % for NPC and 72.11 % for E-NPC detection. Thus, this study identifies sEV CA1 as an innovative diagnostic biomarker for NPC and E-NPC independent of EBV-DNA. Additionally, it establishes an immune-chemiluminescence chip technology for the detection of sEV CA1 protein, paving the way for further validation and clinical application.

The fluorescence characteristics of oxidation reaction between MDA and cooked ground meat were analyzed by front face three dimensional synchronous fluorescence spectroscopy, parallel factor and two dimensional correlation technique. The results showed that the reaction system has two synchronous fluorescence peaks, one is Ex 292 nm and deltalambda 50 nm, assigned to the fluorescence characteristics of tryptophan residues in proteins; the other is Ex 400 nm, delta 70 nm, corresponding with the fluorescence characteristics of MDA-protein adducts formed during oxidation; The synchronous fluorescence landscape was analyzed using PARAFAC. The loading profiles of 1st and 2nd components had an optimal lambda 50 and 70 nm, respectively. During oxidation reaction, the synchronous fluorescence intensity of tryptophan gradually decreased, while the synchronous fluorescence intensity of MDA-protein adducts gradually increased. Two dimensional correlation synchronous fluorescence spectroscopy technique showed that the variation ratio of fluorescence intensity of tryptophan preceded that of MDA-protein adducts.

This study aimed to develop a model using Epstein-Barr virus (EBV)-associated hub genes in order to predict the prognosis of nasopharyngeal carcinoma (NPC). Differential expression analysis, univariate regression analysis, and machine learning were performed in three microarray datasets (GSE2371, GSE12452, and GSE102349) collected from the GEO database. Three hundred and sixty-six EBV-DEGs were identified, 25 of which were found to be significantly associated with NPC prognosis. These 25 genes were used to classify NPC into two subtypes, and six genes (C16orf54, CD27, CD53, CRIP1, RARRES3, and TBC1D10C) were found to be hub genes in NPC related to immune infiltration and cell cycle regulation. It was shown that these genes could be used to predict the prognosis of NPC, with functions related to tumor proliferation and immune infiltration, making them potential therapeutic targets. The findings of this study could aid in the development of screening and prognostic methods for NPC based on EBV-related features.

Nasopharyngeal carcinoma (NPC) is an Epstein-Barr virus (EBV)-associated cancer characterized by a high degree of recurrence, angiogenesis, and metastasis. The importance of alternative pro-angiogenesis pathways including viral factors has emerged after decades of directly targeting various signaling components. Using NPC as a model, we identified an essential oncogenic pathway underlying angiogenesis regulation that involves the inhibition of a tumor suppressor, Spry3, and its downstream targets by EBV-miR-BART10-5p (BART10-5p) and hsa-miR-18a (miR-18a). Overexpression of EBV-miR-BART10-5p and hsa-miR-18a strongly promotes angiogenesis in vitro and in vivo by regulating the expression of VEGF and HIF1-α in a Spry3-dependent manner. In vitro or in vivo treatment with iRGD-tagged exosomes containing antagomiR-BART10-5p and antagomiR-18a preferentially suppressed the angiogenesis and growth of NPC. Our findings first highlight the role of EBV-miR-BART10-5p and oncogenic hsa-miR-18a in NPC angiogenesis and also shed new insights into the clinical intervention and therapeutic strategies for nasopharyngeal carcinoma and other virus-associated tumors.

The failure of children to acknowledge mixed, contradictory emotions is equally of developmental and clinical interest. Developmentally, children do not ordinarily acknowledge the existence of mixed emotions until late in middle childhood. Clinically, the failure to recognise mixed feelings toward others or self is a common presenting problem. The question addressed here is, how readily can such limitations be corrected in children of different ages. Two studies are reported showing that children as young as 6 and 7 years, who initially revealed little understanding of mixed feelings, showed more insight after a short training session. In Experiment 1. two groups of children were equated for their inability to diagnose the mixed feelings of a story character. Subsequently, both groups were presented with a second story containing a similar conflictual event, but only one group was prompted to consider the character's emotional reaction to each component of the conflict. Children in the prompted group were more accurate in diagnosing the character's emotional reaction at the end of the story than the control group, and they maintained their superiority on a post-test story where no prompts were given. Experiment 2 included a similar training procedure. but with a more stringent measure of post-test generalisation: Children were asked to describe or invent their own examples of emotionally charged conflictual situations. Four- and five-year-olds showed little benefit from the training session, but six- and seven-year-olds again showed considerable benefit. Taken together, the two experiments suggest that young school age children often fail to acknowledge mixed feelings because they engage in a cursory appraisal of the elements of an emotionally charged situation; highlighting the elements is sufficient to improve performance. Preschool children, however, appear to suffer from more basic limitations in their ability to integrate the relevant information.
