RESUMO

A cryptic virus, named mulberry cryptic virus 1 (MuCV1), was identified in a mulberry (Morus alba) transcriptome dataset and confirmed using RACE methods. The genome of MuCV1 is composed of two double-stranded RNAs, 1605 bp and 1627 bp in size, encoding an RNA-dependent RNA polymerase (RdRp) and a coat protein (CP), respectively. The 5'-AGAAUU-3' sequence in the 5' untranslated region was conserved in the two dsRNAs. Sequence comparisons and phylogenetic analysis based on RdRp and CP sequences both indicated that MuCV1 is a deltapartitivirus (family Partitiviridae).

RESUMO

The silkworm Bombyx mori is an important economic insect for producing silk, the "queen of fabrics". The currently available genomes limit the understanding of its genetic diversity and the discovery of valuable alleles for breeding. Here, we deeply re-sequence 1,078 silkworms and assemble long-read genomes for 545 representatives. We construct a high-resolution pan-genome dataset representing almost the entire genomic content in the silkworm. We find that the silkworm population harbors a high density of genomic variants and identify 7308 new genes, 4260 (22%) core genes, and 3,432,266 non-redundant structure variations (SVs). We reveal hundreds of genes and SVs that may contribute to the artificial selection (domestication and breeding) of silkworm. Further, we focus on four genes responsible, respectively, for two economic (silk yield and silk fineness) and two ecologically adaptive traits (egg diapause and aposematic coloration). Taken together, our population-scale genomic resources will promote functional genomics studies and breeding improvement for silkworm.

Bombyx mandarina is generally thought to be the wild ancestor nearest to the domesticated silkworm, Bombyx mori. Here, we report the complete mitochondrial genome (mitogenome) of B. mandarina strain Shiquan. The mitogenome contains 13 protein-coding genes (PCGs), 22 tRNA genes, two rRNA genes, and one A + T-rich region. Phylogenetic analysis provides solid evidence that B. mandarina Shiquan belongs to Chinese B. mandarina, rather than Japanese B. mandarina. The new mitogenome provides useful information to further explore the origin and domestication of this species.
