Two new vanadyl complexes of N-confused porphyrins (NCPs), [VONCTPP] (V-1) and [VONCP(OMe)8] (V-2), have been synthesized for the first time and investigated as a catalyst for the oxidative bromination reaction of phenol and its derivatives. This article further delineates crystal structures, photophysical, and redox properties of both the vanadyl complexes. Complexes V-1 and V-2 exhibited a significant red shift in their absorption spectra compared with their respective free bases. The single-crystal structure of V-1 revealed that the complex is in the 2H tautomeric form, while EPR studies revealed the +4 oxidation state of vanadium metal having an axial compression with dxy1 configuration. Catalytic potential for bromoperoxidases-like activity has been explored for both complexes V-1 and V-2 for the first time in NCP chemistry with excellent TOF values (4.7-6.3 s-1 for V-1 and 7.3-8.7 s-1 for V-2) using KBr as a source of bromine and H2O2 as a green oxidant in aqueous acidic medium at 298 K. Notably, both catalysts show excellent recyclability over five cycles. The vanadyl-metalated NCPs exhibit excellent stability in the air.

4-Methylbenzylidene camphor (4-MBC) is a widely used organic UV filter in personal care products. Extensive use of 4-MBC and its frequent detection in aquatic ecosystems defile the biota with muscular and neuronal impairments. This study investigates the neurobehavioral toxicity of 4-MBC using Danio rerio as a model organism. Embryos were exposed semi-statically to 4-MBC at 5, 50, and 500 µg/L concentrations for 10-day post fertilization (dpf). Embryos exhibited a significant thigmotaxis and decreased startle touch response with altered expression of nervous system mRNA transcripts on 5 & 10 dpf. Compared to the sham-exposed group, 4-MBC treated larvae exhibited changes in the expression of shha, ngn1, mbp, elavl3, α1-tubulin, syn2a, and gap43 genes. Since ngn1 induction is mediated by shh signaling during sensory neuron specification, the elevated protein expression of NGN1 indicates 4-MBC interference in the sonic hedgehog signaling pathway. This leads to sensory neuron impairment and function such as 'sense' as evident from reduced touch response. In addition, larval brain histology with a reduced number of cells in the Purkinje layer emblazing the defunct motor coordination. Predictive toxicity study also showed a higher affinity of 4-MBC to modeled Shh protein. Thus, the findings of the present work highlighted that 4-MBC is potential to induce developmental neurotoxicity at both behavioral and molecular functional perspectives, and developing D. rerio larvae could be considered as a suitable alternate animal model to assess the neurological dysfunction of organic UV filters.

Our study aimed to explore the genetic variation in the Toll-like receptor 4 (TLR4) gene and establish its association with somatic cell score (SCS) and milk production traits in four Indian camel breeds namely Bikaneri, Kachchhi, Jaisalmeri and Mewari. TLR4 gene fragment of 573 bp spanning 5' UTR, exon-1 and partial intron-1 region was amplified and genotyped using the PCR-sequence based typing method. Only one SNP located at position C472T was identified. Genotyping revealed two alleles (C and T) and three genotypes: CC, CT and TT. The genotype frequencies for CC, CT and TT were 0.116, 0.326 and 0.558 and allele frequencies for C and T alleles were 0.279 and 0.721, respectively. Association study inferred that the effect of genotype on SCS, lactation yield (LY) and peak yield (PY) was non-significant however heterozygote (CT) genotypes recorded lower SCS and higher LY and PY. It can be concluded that the TLR4 gene possesses limited genetic variation, depicting polymorphism at a single locus in Indian camel breeds with a predominance of the TT genotype. The association study indicated that heterozygote animals possess better udder health and production performance, the statistical significance of which needs to be established using a large data set.

Virus infection can increase drought tolerance of infected plants compared with noninfected plants; however, the mechanisms mediating virus-induced drought tolerance remain unclear. In this study, we demonstrate turnip mosaic virus (TuMV) infection increases Arabidopsis thaliana survival under drought compared with uninfected plants. To determine if specific TuMV proteins mediate drought tolerance, we cloned the coding sequence for each of the major viral proteins and generated transgenic A. thaliana that constitutively express each protein. Three TuMV proteins, 6K1, 6K2, and NIa-Pro, enhanced drought tolerance of A. thaliana when expressed constitutively in plants compared with controls. While in the control plant, transcripts related to abscisic acid (ABA) biosynthesis and ABA levels were induced under drought, there were no changes in ABA or related transcripts in plants expressing 6K2 under drought compared with well-watered conditions. Expression of 6K2 also conveyed drought tolerance in another host plant, Nicotiana benthamiana, when expressed using a virus overexpression construct. In contrast to ABA, 6K2 expression enhanced salicylic acid (SA) accumulation in both Arabidopsis and N. benthamiana. These results suggest 6K2-induced drought tolerance is mediated through increased SA levels and SA-dependent induction of plant secondary metabolites, osmolytes, and antioxidants that convey drought tolerance. Biotechnologies that use plant viruses as plant enhancement tools have shown great potential to flexibly engineer crop traits, but field applications of these technologies are still limited by efficient dissemination methods. Potyviruses can be rapidly inoculated into plants by aphid vectors due to the presence of the potyviral helper component proteinase (HC-Pro), which binds to the DAG motif of the coat protein (CP) of the virion. Previously it was determined that a naturally occurring DAG motif in the non-aphid-transmissible potexvirus, potato aucuba mosaic virus (PAMV), is functional when a potyviral HC-Pro is provided to aphids in plants. The DAG motif of PAMV was successfully transferred to the CP of another non-aphid-transmissible potexvirus, potato virus X, to convey aphid transmission capabilities in the presence of HC-Pro. Here, we demonstrate that DAG-containing segments of the CP from two different potyviruses (sugarcane mosaic virus and turnip mosaic virus), and from the previously used potexvirus, PAMV, can make the potexvirus, foxtail mosaic virus (FoMV), aphid-transmissible when fused with the FoMV CP. We show that DAG-containing FoMVs are transmissible by aphids that have prior access to HC-Pro through potyvirus-infected plants or ectopic expression of HC-Pro. The transmission efficiency of the DAG-containing FoMVs varied from less than 10 % to over 70 % depending on the length and composition of the surrounding amino acid sequences of the DAG-containing segment, as well as due to the recipient plant species. Finally, we show that the engineered aphid-transmissible FoMV is still functional as a plant enhancement resource, as endogenous host target genes were silenced in FoMV-infected plants after aphid transmission. These results suggest that aphid transmission could be engineered into non-aphid-transmissible plant enhancement viral resources to facilitate their field applications.

MAIN CONCLUSION: This review focuses on different factors involved in promoting symptom recovery in plants post-virus infection such as epigenetics, transcriptional reprogramming, phytohormones with an emphasis on RNA silencing as well as role of abiotic factors such as temperature on symptom recovery. Plants utilize several different strategies to defend themselves in the battle against invading viruses. Most of the viral proteins interact with plant proteins and interfere with molecular dynamics in a cell which eventually results in symptom development. This initial symptom development is countered by the plant utilizing various factors including the plant's adaptive immunity to develop a virus tolerant state. Infected plants can specifically target and impede the transcription of viral genes as well as degrade the viral transcripts to restrict their proliferation by the production of small-interfering RNA (siRNA) generated from the viral nucleic acid, known as virus-derived siRNA (vsiRNA). To further escalate the degradation of viral nucleic acid, secondary siRNAs are generated. The production of virus-activated siRNA (vasiRNA) from the host genome causes differential regulation of the host transcriptome which plays a major role in establishing a virus tolerant state within the infected plant. The systemic action of vsiRNAs, vasiRNA, and secondary siRNAs with the help of defense hormones like salicylic acid can curb viral proliferation, and thus the newly emerged leaves develop fewer symptoms, maintaining a state of tolerance.

Two new meso-substituted oxido-molybdenum corroles were synthesized and characterized by various spectroscopic techniques. In the thermogram, MoO[TTC] (1) exhibited excellent thermal stability up to 491 °C while MoO[TNPC] (2) exhibited good stability up to 318 °C. The oxidation states of the molybdenum(V) were verified by electron paramagnetic resonance (EPR) spectroscopy and exhibited an axial compression with dxy1 configuration. Oxido-molybdenum(V) complexes were utilized for the selective epoxidation of various olefins with high TOF values (2066-3287 h-1) in good yields in a CH3CN/H2O (3:2, v/v) mixture in the presence of hydrogen peroxide as a green oxidant and NaHCO3 as a promoter. The oxidative bromination catalytic activity of oxido-molybdenum(V) complexes in an aqueous medium has been reported for the first time. Surprisingly, MoO[TNPC] (2) biomimics of the vanadium bromoperoxidase (VBPO) enzyme activity exhibited remarkably high TOF values (36 988-61 646 h-1) for the selective oxidative bromination of p-cresol and other phenol derivatives. Catalyst MoO[TNPC] (2) exhibited higher TOF values and better catalytic activity than catalyst MoO[TTC] (1) due to the presence of electron-withdrawing nitro groups evident from cyclic voltammetric studies.

The economic viability of the camel in the current scenario can be ensured by improving its dairy potential. The study on the udder and teat characteristics of dromedary camel and understanding its relationship with milk yield and milkability can be of great value in establishing camel as a dairy animal. The present study was conducted on 45 lactating she-camels of four Indian camel breeds, viz., Bikaneri, Jaisalmeri, Kachchhi, and Mewari, stationed at ICAR-NRCC Bikaner, Rajasthan, India. The udder, teat, and milk vein measurements traits, factors affecting these traits and their relationship with milkability traits, were studied in hand-milked Indian dromedary camel. The means ± S.E. of teat lengths (TL), namely, left fore (LF), left rear (LR), right fore (RF), and right rear (RR), were observed as 52.21 ± 1.66, 58.52 ± 2.11, 50.13 ± 1.74, and 54.37 ± 1.82 mm, respectively. The means ± S.E. of teat diameter (TD), namely, left fore, left rear, right fore, and right rear teat diameters, were observed as 42.44 ± 1.60, 46.01 ± 1.68, 39.29 ± 1.31, and 45.20 ± 1.56 mm, respectively. The means ± S.E for udder depth, udder length, udder height from the ground, milk vein diameter, and milk vein length were observed as 25.44 ± 0.42, 37.29 ± 0.80, 114.80 ± 0.80, 2.02 ± 0.08, and 88.70 ± 0.96 cm, respectively. Udder and milk vein measurements did not differ significantly between breeds. Kachchhi breed has largest teat length and diameter. The breed differences were significant (p ≤ 0.05) for TL-LF, TL-RF, and TD-RR only. The effect of parity was non-significant on udder, teat, and milk vein measurement traits except TD-RR (p ≤ 0.05); however, second parity animals had higher values for all the studied traits except udder height from ground. Positive and highly significant (p ≤ 0.01) correlation of milk yield was observed with the majority of udder, teat, and milk vein measurements, milking time, and milk flow rate, while a negative correlation was found with udder height from ground and milk let-down time. It can be concluded that udder characteristics are influenced by various genetic and non-genetic factors and its relationship with milk yield and milkability can be used for selection and dairy management purposes.

Human oral squamous cell carcinoma is the sixth most frequent malignant cancer, with an unacceptably high death rate that affects people's health. Albeit, there are several clinical approaches for diagnosing and treating oral cancer they are still far from ideal. We previously synthesised and characterised the docetaxel nanoformulation (PLGA-Dtx) and discovered that docetaxel nanoencapsulation may suppress oral cancer cells. The goal of this study was to figure out the mechanism involved in the suppression of oral cancer cell proliferation. We discovered that PLGA-Dtx inhibited SCC-9 cell growth considerably as compared to free docetaxel (Dtx), and that the viability of SCC-9 cells treated with PLGA-Dtx was decreased dose-dependently. MTT assay showed that PLGA-Dtx selectively inhibited the growth of PBMCs from oral cancer patients while sparing PBMCs from normal healthy controls. Further, flow cytometry analysis showed that PLGA-Dtx induced apoptosis and necroptosis in SCC-9 cells. G2/M cell cycle arrest has been confirmed on exposure of PLGA-Dtx for 24 h in SCC-9 cells. Interestingly, western blot investigation found that PLGA-Dtx increased the amounts of necroptic proteins and apoptosis-related proteins more efficiently than Dtx. Furthermore, PLGA-Dtx was more effective in terms of ROS generation, and mitochondrial membrane potential depletion. Pretreatment with necroptosis inhibitor Nec-1 efficiently reversed the ROS production and further recover MMP caused by PLGA-Dtx. Overall, this study revealed a mechanistic model of therapeutic response for PLGA-Dtx in SCC-9 cells and proposed its potency by inducing cell death via activation of concurrent apoptosis and necroptosis in SCC-9 cells via TNF-α/RIP1/RIP3 and caspase-dependent pathway.

BACKGROUND: Global and country specific recommendations on Delayed umbilical cord clamping (DCC) are available, though guidance on their implementation in program settings is lacking. In India, DCC (clamping not earlier than 1 min after birth) is a component in the package of services delivered as part of the India Newborn Action Plan (INAP) supported by Nutrition International (NI) in two states. The objective of this case study was to document the learnings from implementation of DCC in these two states and to understand the health system factors that affected its operationalization. METHODS: Mixed methods were followed. Using the World Health Organization (WHO) Health Systems building blocks as a framework, 20 Key-Informant Interviews were conducted to explore facilitators and barriers to routine implementation of DCC in public health settings. Existing quantitative program data and secondary data from labour-room registers from eight NI- supported districts were analysed to assess the prevalence of DCC implementation in public health systems settings. RESULTS: A demonstrated commitment from the government to implement DCC at all delivery points in NI supported districts was observed. Funds were sufficient, trainings were optimal, knowledge of the health workforce was adequate and a recording mechanism was in place. According to record reviews, DCC was more likely to happen in facilities that provide Basic Emergency Obstetric services and among normal deliveries. It was less likely to be followed in babies delivered by Caesarean section (OR 0.03; 95%CI 0.02,0.05), birthweight < 2000 g (OR 0.22; 95%CI 0.12,0.47), multiple pregnancies (OR 0.17, 95%CI 0.05,0.63), birth asphyxia requiring resuscitation (0.37; 95%CI 0.26,0.52), and those delivered during day shift (OR 0.59, 95%CI 0.40, 0.83). CONCLUSIONS: Wide coverage of DCC in public health settings in the two states was observed. Good governance, adequate funding, commitment of health workforce has likely contributed to its success in these contexts. These are critical elements to guide DCC implementation in India and for consideration in other settings.

Acute kidney injury (AKI) in children with Transposition of Great arteries (TGA) undergoing Arterial Switch operation (ASO) is an important complication in the post-operative period associated with worse outcomes. AKI in children post open cardiac surgery has been well studied, with lesser data in literature pertaining to TGA and its sub-types specifically. This was a prospective, observational study enrolling infants with TGA undergoing ASO at a single center over a span of a decade from January 2010 to December 2020. The infants were followed during the duration of ICU and hospital stay, with documentation of baseline and intraoperative parameters as well as post-operative course. Out of 145 infants enrolled in the study, 83.1% developed AKI with majority (83.9%) having stage 1 AKI. Higher odds of AKI were seen in infants requiring Norepinephrine [odds ratio - 16.76 (95% CI 2.19-128.2), p < 0.001] and those who developed gram-negative infections [2.81 (1.04-7.56), p - 0.036]. Infants with AKI had significantly higher vasoactive-inotropic support at day 1 than those without AKI [16 (12.5-21.50 vs 13 (10.25-15.75), p - 0.014]. Seventeen infants in the AKI group (14%) died as opposed to none in the non-AKI group (p = 0.076). Median hours of ventilator support required were significantly higher in those with AKI than those who did not develop AKI (48 vs 45.5 p = 0.015). The infants with ASO + ASD + PDA (53% of neonates who died) were younger, had less weight at admission, more gram-negative sepsis and need for dopamine, as compared to ASO + VSD + ASD (23.5% of mortality) and ASO + ASD + VSD + aortic arch repair (23.5% of mortality). AKI in infants with TGA undergoing ASO is common and associated with poorer outcomes. In this subpopulation, AKI development is associated most commonly with hemodynamic instability and infections. This is the first study, looking at outcomes of TGA depending on the sub-types of ASO surgeries done in the infants [ASO with ASD + PDA or ASD + VSD or ASD + VSD + Arch Repair].

Chronic obstructive pulmonary disease (COPD), a heterogeneous lung disorder that is characterized by airflow obstruction and the third leading cause of death, globally. COPD is influenced by environmental and genetic factors. Here, we measured the serum level of matrix metalloproteinase-9 (MMP-9), cyclooxygenase-2 (COX-2) and prostaglandin E-2 (PGE-2) and reveal the correlation between their levels in COPD subjects. In this study, we included a total of 79 COPD and 79 healthy controls. We assessed demographic profile, risk factors, respiratory symptoms, clinical history, COPD Assessment Test (CAT) score and spirometry. Further, we determined the serum levels of MMP-9, COX-2 and PGE-2 by enzyme-linked immunosorbent assay (ELISA). The correlation between their serum levels was also determined. Among the studied population age, gender, body mass index and socioeconomic status were comparable. Serum levels of MMP-9, COX-2 and PGE-2 were significantly increased in the COPD group than in healthy controls (P < 0.0001). Moreover, MMP-9, COX-2 and PGE-2 levels were increased with the GOLD grades and CAT score (> 10). Serum levels of MMP-9, COX-2 and PGE-2 was enhanced in patients with larger clinical history (> 20 years) than those with lower clinical history (< 10 years). Serum levels of MMP-9 and COX-2; MMP-9 and PGE-2; COX-2 and PGE-2 showed a positive correlation (P < 0.0001) with the COPD group. Our data demonstrate that serum levels of MMP-9, COX-2 and PGE-2 were correlated with the GOLD grade, CAT score and clinical history of the COPD group, pointing that they can be used as a indicators to understand the disease progression. The growing production of cosmetic products such as organic UV filters (OUVFs) in recent years has raised concern regarding their safety to human and environmental health. The inability of wastewater treatment plants in removing these chemical entities and their high octanol-water partition coefficient values tend to result in the persistence of OUVFs in several environmental matrices, leading these to be categorized as "emerging environmental contaminants" because of their unknown risk. Besides aquatic ecosystem contamination, the application of sludge disposal equally threatens terrestrial biota. Besides, the available reviews focusing on levels of OUVFs in aqueous systems (freshwater and marine), instrumental analysis from various samples, and specific toxicity effects, compiled information on the ecotoxicity of OUVFs is currently lacking. Hence, the present manuscript systematically reviews the ecotoxicity of OUVFs in freshwater and marine organisms occupying lower to higher trophic levels, including the underlying mechanisms of action and current knowledge gaps. The available scientific evidence suggests that OUVFs are a prime candidate for environmental concern due to their potential toxic effects. To the best of our knowledge, this is the first document detailing the toxicological effects of OUVFs in aquatic organisms.

The advent of the nanotechnology era offers a unique opportunity for sustainable agriculture provided that the exposure and toxicity are adequately assessed and properly controlled. The global production and application of cerium oxide nanoparticles (CeO2-NPs) in various industrial sectors have tremendously increased. Most of the nanoparticles end up in water and soil where they interact with soil microorganisms and plants. Investigating the uptake, translocation and accumulation of CeO2-NPs is critical for its safe application in agriculture. Plant uptake of CeO2-NPs may lead to their accumulation in different plant tissues and interference with key metabolic processes of plants. Soil microbes can also be affected by increasing CeO2-NPs in soil, leading to changes in the physiology and enzymatic activity of soil microorganisms. The interactions between CeO2-NPs, microbes and plants in the agricultural system need systemic research in ecologically relevant conditions. In the present review, The uptake pathways and in-planta translocation of CeO2-NPs,and their impact on plant morphology, nutritional values, antioxidant enzymes and molecular determinants are presented. The role of CeO2-NPs in modifying soil microbial community in plant rhizosphere is also discussed. Overall, the review aims to provide a comprehensive account on the behaviour of CeO2-NPs in soil-plant systems and their potential impacts on the soil microbial community and plant health.

The study aimed to investigate the incidence, risk factors and economic impact of clinical mastitis in an organized camel herd located at Bikaner, Rajasthan, India. Furthermore, bacterial pathogens were isolated and identified using MALDI-TOF mass spectrometry analysis. The overall annual incidence of clinical mastitis was 25%, with highest incidence in Bikaneri and Mewari breed followed by Kachchhi and Jaisalmeri breeds. Animals with high peak yield appeared at higher risk, though statistical significance could not be established for any risk factor included in the study. Highest incidence was recorded in camels falling in 4th or higher parity group. In 80% cases, only one udder quarter was involved, while in rest 20% cases, two udder quarters were involved. Quarter-wise incidence was maximum in right fore-quarter followed by left fore-quarter, right hind-quarter and left hind-quarter. Important pathogens identified included Staphylococcus aureus, E. coli, Klebsiella pneumoniae, Staphylococcus epidermidis, Streptococcus uberis, Bacillus clausii and Corynebacterium amylocolatum. Other isolates included Staphylococcus hominis, Streptococcus pluranimalium, Aerococcus viridians and Moraxella osloensis. The mean cost of clinical mastitis per animal per month was calculated as Indian Rs. 181.63 (~ US$ 2.45), while the mean cost per animal per annum was Indian Rs. 2179.29 (~ US$ 29.37). The maximum cost incurred in purchase of drugs (43.63%), followed by cost of discarded milk plus milk production loss (18.17%) and cost of veterinary services (17.03%). To our knowledge, this is the first report documenting economic impact of clinical mastitis in camel.

Asthma is a common chronic airways inflammatory disorder in which the expression of genes for the transcription factors FoxP3 and GATA3 plays crucial roles in activation of specific T cells population and pathogenesis of asthma. Recent data have shown that Hb, Eosinophils, total leucocytes count (TLC), absolute eosinophil count (AEC), and IgE, may be involved in adversely influencing the status of several chronic diseases including asthma. In this communication, we have carried out a case control study in order to evaluate the expression of FoxP3, GATA-3 genes in 80 bronchial asthmatic patients using real time polymerase chain reaction technique, and also to analyse and compare the values of Hb, TLC, AEC, and IgE in asthmatics with 80 control subjects. The numbers of eosinophils and total leucocytes and the level of serum IgE were higher in asthmatics compared to healthy subjects. The relative expressions of FoxP3 and GATA-3 genes in control versus asthmatics were 12.42 ± 1.413 versus 5.79 ± 0.260 (P value = < 0.0001) and 4.731 ± 0.350 versus 8.415 ± 0.359 (P value = 0.0043), respectively. The asthmatics displayed comparatively decreased level of FoxP3 expression and higher level of GATA-3 expression. There was a positive and significant correlation between the level of IgE and expression of GATA-3 in asthmatics. Relatively lower level of FoxP3 mRNA expression in bronchial asthmatics may be linked with the sustained inflammatory process and decreased immune tolerance by asthmatics. A positive correlation of GATA-3 expression with the increase in IgE level shows it to be a characteristic of asthma. However, extensive work is required to delineate the targets involved in the pathogenesis of asthma for adequate therapeutic interventions.

MAIN CONCLUSION: RNA-dependent RNA polymerase 1 of Nicotiana tabacum modulates ToLCGV pathogenesis by influencing a number of defence-related genes in N. benthamiana plants. Key means of plants protecting themselves from the invading viruses is through RNA silencing. RNA-dependent RNA polymerase-1 (RDR1) is one of the crucial proteins of the RNA silencing pathway, which is induced after infection by viruses. RDR1 functions in the generation of small interfering RNAs (siRNAs) against the viral genome, thus it is antiviral in nature. Here, we used the transgenic Nicotiana benthamiana plant expressing N. tabacum NtRDR1 and observed reduced susceptibility towards Tomato leaf curl Gujarat virus (ToLCGV) infection compared to the wild-type N. benthamiana plants. To understand the reason for such reduced susceptibility, we prepared high-definition small RNA (sRNA) cDNA libraries from ToLCGV-infected wild-type N. benthamiana and NtRDR1 expressing N. benthamiana lines and carried out next-generation sequencing (NGS). We found that upon ToLCGV infection the majority of siRNAs generated from the host genome were of the 24 nucleotide (nt) class, while viral siRNAs (vsiRNAs) were of the 21-22-nt class, indicating that transcriptional gene silencing (TGS) is the major pathway for silencing of host genes while viral genes are silenced, predominantly, by post transcriptional gene silencing (PTGS) pathways. We estimated the changes in the expression of various defence-related genes, such as Constitutively Photomorphogenic-9 (COP9) signalosome (CSN) complex subunit-7, Pentatricopeptide repeat containing protein (PPRP), Laccase-3, Glutathione peroxidase-1 (GPX-1), Universal stress protein (USP) A-like protein, Heat shock transcription factor B4 (HSTF-B4), Auxin response factor-18 (ARF18), WRKY-6 and Short chain dehydrogenase reductase-3a. The differential expression of these genes might be linked with the enhanced tolerance of NtRDR1 N. benthamiana transgenic plants to ToLCGV. Our study suggests that reduced expression of subunit-7 of CSN complex and WRKY6, and increased expression of USPA-like protein might be linked with the reduced susceptibility of NtRDR1-transgenic N. benthamiana plants to ToLCGV.

Over the past decades the role of nitric oxide (NO) and reactive oxygen species (ROS) in signaling and cellular responses to stress has witnessed an exponential trend line. Despite advances in the subject, our knowledge of the role of NO and ROS as regulators of stress and plant growth and their implication in signaling pathways is still partial. The crosstalk between NO and ROS during root formation offers new domains to be explored, as it regulates several plant functions. Previous findings indicate that plants utilize these signaling molecules for regulating physiological responses and development. Depending upon cellular concentration, NO either can stimulate or impede root system architecture (RSA) by modulating enzymes through post-translational modifications. Similarly, the ROS signaling molecule network, in association with other hormonal signaling pathways, control the RSA. The spatial regulation of ROS controls cell growth and ROS determine primary root and act in concert with NO to promote lateral root primordia. NO and ROS are two central messenger molecules which act differentially to upregulate or downregulate the expression of genes pertaining to auxin synthesis and to the configuration of root architecture. The investigation concerning the contribution of donors and inhibitors of NO and ROS can further aid in deciphering their role in root development. With this background, this review provides comprehensive details about the effect and function of NO and ROS in the development of RSA.

IgG4-Related Disease(IgG4-RD) is a rare disease that can present with myriad clinical features. We report a tuberculosis contact case who presented with fever and constitutional complaints with imaging evidence of paravertebral and retroperitoneal soft tissue thickening. Further workup, including tissue biopsy ruled out tuberculosis and revealed diagnosis to be IgG4-related disease. Patient was started on oral steroids, which led to symptomatic improvement. In a TB endemic country such as ours, for a patient presenting with pleural and/or peritoneal fibrosis, a differential diagnosis of IgG4-RD must be kept.

RNA silencing phenomenon in plants provides resistance to various pathogens and also, it maintains genome integrity. The process of RNA silencing is regulated by diverse proteins, among which RNA dependent RNA polymerases (RDRs) are very crucial for the amplification of small RNAs (sRNAs). Out of various RDR proteins present in plants, role of RDR1, RDR2 and RDR6 for providing resistance against various biotic stresses have been well documented. In contrast, very few information is available regarding the role of RDR3, RDR4 and RDR5 proteins in plant biology and stress response. Furthermore, the regulation of RDRs is not yet known. Here, we have carried out in silico studies for identification of the transcription factor (TF) binding sites on the promoter of RDR1-6 genes of various plant species. Among the TFs predicted to bind on the promoter of RDRs, MYB44, AS1/AS2, WRKY1 are the major one. Furthermore, putative interacting protein partners of RDRs proteins of tomato and rice were also predicted by STRING database which suggests that DCL (Dicer-like) proteins are strong candidate proteins as the interacting partners of RDRs. The knowledge of regulation of RDRs and its interacting protein partners might help in developing resistant plants to biotic stresses.
