The aim of the present study was to validate methods of stool sample conservation for the egg hatch test (EHT). This study involved the use of a bovine naturally infected predominantly by Cooperia spp. and one equine naturally infected predominantly by cyathostomins characterized as susceptible to benzimidazoles in the EHT. Fecal samples were submitted to three treatments: aerobic methods (anaerobic storage in plastic bottles, anaerobic storage in vacuum-sealed bags or aerobic storage in plastic bags), under two temperature conditions (room temperature and refrigeration) analyzed at four different assessment times (48, 72, 96 and 120 h). As the standard test, an assay was also performed within 3 h. The tests were performed in triplicate for each drug concentration and with three experimental repetitions at one-week intervals. Two criteria were used for the storage methods: hatchability in the negative control group and sensitivity of the eggs to thiabendazole, comparing the EC50 and 95% confidence interval for each treatment to those of the standard test and the other repetitions. Bovine samples can be stored for up to 96 h and refrigerated vacuum storage can be used, ensuring hatchability of the negative control and sensitivity of the eggs to thiabendazole. For equine samples, no forms of storage were indicated due to the variation among the repetitions and the reduction in the sensitivity of the eggs to thiabendazole, which could result in a false positive detection of resistance.

Biomarkers are specific molecular, histological, or physiological characteristics of normal or pathogenic biological processes and are promising in the diagnosis of gastrointestinal nematodes (GINs). Although some biomarkers have been validated for infection by Ostertagia sp. in cattle raised in temperate regions, there is a lack of information for tropical regions. The aim of this project was to assess potential biomarkers and validate the most promising. In the first study, 36 bovines (Nelore breed) naturally infected by GINs were distributed into two groups: infected (not treated with anthelmintic) and treated (treated with fenbendazole on days 0, 7, 14, 21, 28, 42, and 56). The variables of interest were live weight, fecal egg count, hemogram, serum biochemical markers, phosphorus, gastrin, and pepsinogen. In the second step, pepsinogen was assessed in cattle of the Nelore breed distributed among three groups: infected (not treated with anthelmintic), MOX (treated with moxidectin), and IVM + BZD (treated with ivermectin + albendazole). In the first study, no difference between groups was found for weight, albumin, hematocrit (corpuscular volume [CV]), erythrocytes, or hemoglobin. Negative correlations were found between pepsinogen and both CV and albumin, and albumin was negatively correlated with the percentage of Haemonchus sp. in the fecal culture. Among the biomarkers, only pepsinogen differentiated treated and infected (beginning with the 28th day of the study). In the second study, a reduction in pepsinogen was found after anthelmintic treatment. Therefore, pepsinogen is a promising biomarker of worms in cattle naturally infected by the genera Haemonchus and Cooperia in tropical areas.

A possible synergistic effect of macrocyclic lactones' (MLs) combination has been previously described against resistant gastrointestinal nematodes of cattle. In addition to synergism, drug-drug interactions between MLs can also result in additive or antagonistic effect, considering the different MLs pharmacokinetics, pharmacodynamics, and interactions with molecular mechanisms of resistance. Therefore, the aim of the current work was evaluated the effect of different MLs combinations against Haemonchus contortus. Infecting larvae of two isolates (one susceptible and one resistant to ivermectin) were used in the larval migration inhibition test. After estimating the half maximal effective concentration of abamectin (ABA), eprinomectin, (EPR), ivermectin (IVM), and moxidectin (MOX) for both isolates, combinations were delineated by a simplex-centroid mixture experiment, and the mixture regression analysis was applied to the special cubic model. A synergistic effect was found for the EPR + MOX against the susceptible isolate as well as the EPR + MOX, IVM + MOX, and ABA + EPR + IVM against the resistant isolate. An antagonistic effect of ABA + IVM + MOX was found against the susceptible isolate. For the susceptible isolate, a higher inhibition was found with greater proportions of EPR and lower proportions of the other drugs compared to the reference mixture. For the resistant isolate, inhibition greater than that of the reference mixture was found with higher proportions of IVM as well as lower proportions of the other drugs. The synergistic and antagonistic effects were dependent on the following: (a) parasite drug resistance profile, (b) the composition of the combination, and (c) the proportions used, with EPR and IVM exerting a greater impact on these effects.

Ticks and fleas are arthropods widely distributed around the world involved in the transmission of various vector-borne diseases (VBDs), including Brazilian Spotted Fever (BSF), Baggio-Yoshinari Syndrome and the plague, with outstanding consequences for the public health. The aim of this study was to investigate the presence of Rickettsia spp., Borrelia spp. and Yersinia pestis in arthropods collected from dogs, cats and horses living in the state of Pernambuco, Northeastern Brazil. From January 2017 to April 2019, ectoparasites were collected, morphologically identified and molecularly analysed through PCR and sequencing. In total 401 specimens were collected from 86 animals, being 68% (n = 273) and 32% (n = 128) from rural and urban areas, respectively. The most commonly detected species were the ticks Dermacentor nitens, Amblyomma sculptum, Rhipicephalus sanguineus sensu lato (s.l.), Rhipicephalus microplus, and Amblyomma ovale, and the fleas Ctenocephalides felis and Ctenocephalides canis. DNA of Rickettsia felis was detected in D. nitens collected from horses, and C. felis, and R. sanguineus s.l. collected from dogs. All samples scored negative for Borrelia spp. and Y. pestis DNA. This study provides valuable data on ectoparasite fauna from domestic animals and identifies the circulation of a zoonotic pathogen (i.e., R. felis) in the population of the arthropods assessed. Therefore, preventive measures should be adopted in order to reduce the risk of occurrence of neglected VBD caused by this pathogen in animal and human hosts.

BACKGROUND: The occurrence of higher winter temperatures in Brazilian areas with tropical and highland climates may result in a fifth peak of tick populations during winter in addition to the four generations previously described. Therefore, a strategic control protocol was developed with treatments in two seasons with the objective of controlling the generations of ticks that occur in spring/summer and those that occur in autumn/winter. METHODS: The study was conducted in Mato Grosso do Sul, Brazil, from the beginning of the rainy season, November 2020, to October 2021. In a randomized block design, 36 calves were distributed into three groups: (i) negative control; (ii) traditional strategic control in one season (SC1S), at the beginning of the rainy season; and (iii) strategic control in two seasons (SC2S), at the beginning and end of the rainy season. The SC1S strategic control group was treated on day 0, November 2020, and twice more with intervals of 42 days. The SC2S group received three more treatments beginning on day 182, May 2021, with intervals of 42 days. All treatments consisted of 5% fluralaner (Exzolt® 5%) delivered via a pour-on dose of 1 mL/20 kg body weight. Counts of semi-engorged female ticks were performed on day 3 and every 14 days thereafter, and the animals were weighed at the same time. RESULTS: Fluralaner showed a mean efficacy of more than 95% up to day 294. The two treated groups showed a decrease (P < 0.05) in the average number of ticks on day 3. In the SC2S group, the means were close or equal to zero throughout the study, while in the SC1S group, the means did not differ (P > 0.05) from those of the control group from day 231 onward. The final mean weight gain of each group was 76.40 kg, 98.63 kg, and 115.38 kg for the control, SC1S, and SC2S groups, respectively, differing (P < 0.05) from each other. CONCLUSIONS: Therefore, three applications of fluralaner, with one application every 42 days from the beginning of the rainy season in the middle spring, resulted in effective tick control for 224 days. When three additional treatments were given in autumn/winter with intervals of 42 days between applications, tick counts were reduced throughout the year. This strategic control approach may be indicated in years with climatic conditions that allow that population peaks are expected to occur in the autumn/winter period.

The objective was to report an outbreak of tick-borne disease (TBD) on riverside property in the Western Amazon. The death of 25 Nellore cattle was reported on a rural property on the banks of the Purus River, state of Acre. The producer observed animals with staggering walking, drop in productivity, weight loss and evolution to death in approximately 30 days. Fifteen animals from the same batch were selected for clinical evaluation and the ear tip was punctured for hemoparasite research, in addition to blood collection for hematological, biochemical and molecular evaluation. The main laboratory findings were leukocytosis, thrombocytopenia, hypoproteinemia, elevated creatine kinase and reduced urea, creatinine and albumin, as well as visualization of forms suggestive of Anaplasma spp. in 13.33% of the samples. Through PCR, 20% positivity was observed for Anaplasmamarginale and 53.33% for Babesia sp. Hematological and biochemical changes, although highly suggestive, may suffer changes from other factors not related to TBD. Therefore, the presumptive identification of the etiological agent in the blood or confirmatory by molecular methods is essential in the diagnosis. Depending on the stage of the disease, low parasitemia occurs, making it difficult to see hemoparasites in blood smears. The Babesia sp. was the main agent of the outbreak of TBD in the population evaluated, which, when associated with early clinical and laboratory diagnosis, results in adequate therapeutic direction and prophylactic measures, promoting a balance between host, agent and vector.
Objetivou-se relatar um surto de tristeza parasitária bovina (TPB) em propriedade ribeirinha na Amazônia Ocidental. Foi notificado o óbito de 25 bovinos da raça Nelore, em uma propriedade rural às margens do rio Purus, estado do Acre. O produtor observou animais com andar cambaleante, queda na produtividade, perda de peso e evolução ao óbito em aproximadamente 30 dias. Quinze animais do mesmo lote foram selecionados para avaliação clínica e foi procedida a punção a ponta de orelha para pesquisa de hemoparasitos, além da coleta de sangue para avaliação hematológica, bioquímica e molecular. Os principais achados laboratoriais foram anemia, leucocitose, trombocitopenia, hipoproteinemia, elevação da creatina quinase e redução de ureia, creatinina e albumina, além da visualização de formas sugestivas de Anaplasma spp. em 13,33% das amostras. Por meio da PCR, foi observado 20% de positividade para Anaplasma marginale e 53,33% para Babesia sp. As alterações hematológicas e bioquímicas, embora bastante sugestivas, podem sofrer alterações de outros fatores não relacionados à TPB. Por isso, a identificação presuntiva do agente etiológico no sangue ou confirmatória por métodos moleculares é essencial no diagnóstico. A depender da fase da doença, ocorre baixa parasitemia, dificultando a visualização de hemoparasitos em esfregaços sanguíneos. A Babesia sp. foi o principal agente do surto de TPB na população avaliada, que, quando associado ao diagnóstico clínico e laboratorial precoce, resulta no direcionamento terapêutico adequado e medidas profiláticas, promovendo uma relação de equilíbrio entre hospedeiro, agente e vetor.

Bovine brucellosis (BB) is a compulsory notifiable bacterial disease caused by Brucella abortus, which is responsible for severe reproductive disorders in cattle, considerable damages to trade and public health damage. The objectives of this study were to identify the frequency of BB and to analyze the risk factors in the dairy herds of the Rio Branco microregion, in the state of Acre, Brazil. Thus, 527-bovine-serological samples were submitted to the buffered acidified antigen test and to 2-mercaptoethanol. Risk factors were characterized by the administration of epidemiological questionnaires. The logistic regression test was used to evaluate the existence of a significant association between the variables. The frequency of BB in the Rio Branco microregion was 10.6% (56/527). The inappropriate destination of the aborted fetus and placental remnants (odds ratio OR = 13.6), the slaughter of cattle within the property (OR = 3.4) and the age range above 4 years old (OR = 2.9) were the most significant risk factors identified by the logistic regression model. According to the results of this study, it is recommended to intensify BB control and prophylaxis measures in the microregion of Rio Branco, especially those related to sanitary and reproductive management in the livestock breeding.(AU)
A brucelose bovina (BB) é uma bacteriose de notificação obrigatória, causada pela Brucella abortus, responsável por distúrbios reprodutivos graves em bovinos, prejuízos consideráveis ao comércio e agravos em saúde pública. Objetivaram-se com este estudo identificar a frequência de BB e analisar os fatores de risco nos rebanhos leiteiros da microrregião de Rio Branco, Acre. Assim, 527 amostras sorológicas de bovinos foram submetidas ao teste do antígeno acidificado tamponado e ao 2-mercaptoetanol. Os fatores de risco foram caracterizados por aplicação de questionários epidemiológicos. Utilizou-se o teste de regressão logística para avaliar a existência de associação significativa entre as variáveis. A frequência da BB na microrregião de Rio Branco foi de 10,6% (56/527). O destino inadequado do feto abortado e restos placentários (odds ratio OR = 13,6), o abate de bovinos na propriedade (OR = 3,4) e a faixa etária acima de 4 anos (OR = 2,9) foram os fatores de risco mais significativos apontados pelo modelo de regressão logística. Conforme os resultados deste estudo, recomenda-se a intensificação nas medidas de controle e profilaxia da BB na microrregião de Rio Branco, sobretudo relacionadas ao manejo sanitário e reprodutivo nas criações.(AU)
