RESUMO

The incidence and mortality rates of colorectal cancer (CRC) have significantly increased in recent years. It has been shown that early diagnosis of CRC improves the five-year survival of patients compared to late diagnosis, as patients with stage I disease have a five-year survival rate as high as 90 %. Through bioinformatics analysis, we identified Kallikrein 10 (KLK10), a member of the Kallikrein family, as a reliable predictor of CRC progression, particularly in patients with early-stage CRC. Furthermore, single-cell analysis revealed that KLK10 was highly expressed in tumor and partial immune cells. Analysis of the biological functions of KLK10 using the Kyoto encyclopedia of genes and genomes and gene ontology indicated that KLK10 plays a role in the proliferation and differentiation of cancer cells, along with the maintenance of tumor function and immune regulation, explicitly by T cells and macrophages. EdU cell proliferation staining, plate clone formation assay, and cell scratch assay demonstrated that KLK10 inhibition by siRNA affected the proliferation and migration of CRC cells. Cell cycle detection by flow cytometry demonstrated that KLK10 inhibition led to cell cycle arrest in the G1 phase. In addition, the proportion of M1 and M2 macrophages in 45 tumor specimens was analyzed by immunohistochemistry, the proportion of CD4+ T cells and CD8+ T cells in plasma was identified by flow cytometry, and their correlation with KLK10 was analyzed. The effects of KLK10 on T cells and macrophages were verified in independent cell experiments. The results revealed that KLK10 also activates CD4+ T cells, mediating M2-type macrophage polarization.

The effect of heat treatment on the allergenicity and microstructure of ovomucoid was studied by enzyme-linked immunosorbent assay (ELISA), CD spectra, ANS fluorescence probe emission spectra and UV absorption spectra. The results showed that the allergenicity of ovomucoid decreased with increasing temperatures and time. CD spectra were used to examine the changes in the secondary structure of the ovomucoid treated by different temperatures. When treated by different temperatures, the mutual transformation between a-helix, b-sheet, b-turn and the random coil was observed, and the disorderliness of the secondary structure was increased with alpha-helix and beta-sheet decreasing, but the random coil increased correspondingly. The ANS fluorescence probe emission spectra analysis demonstrated that heat treatment induced reducing in surface hydrophobicity with the increase in heat temperatures. With the increase in heat temperatures, the amino acid residues with the UV-absorption were exposed gradually, and the UV-absorption maximum value increased. The results indicated that when treated by heating, the changes in the microstructure of ovomucoid can lead to the changes in the allergenicity of ovomucoid.

The objective of the present paper was to study the effect of monomethoxypolyethlene glycol (mPEG) modification on the stability of chicken IgY and compare the stability of the modification products by Fourier transform infrared spectroscopy (FTIR), CD spectrooscopy and fluorescence spectroscopy. NHS-mPEG was used to modify IgY after mPEG was activated with N-hydroxysuccinimide (NHS). The optimal reaction condition for modification was 1:10 molar rate of IgY to mPEG at pH 7, reaction for 1 h, and the product was obtained with modification rate of 20.56% and activity reservation of 87. 62%. In addition, the thermal and pH stability of IgY and mPEG-IgY was compared by spectroscopic methods. The results showed that the alpha-helix, beta-sheet, beta-turn, and random content of IgY changed from 14.5%, 42.1%, 6.2% and 37.2% to 1.6%, 55.25%, 5.8% and 37.5%, while mPEG changed from 12.9%, 42.7%, 6.3% and 38. 1% to 3.1%, 50.5%, 7.2% and 39.2%, respectively, after incubating for 120 min at 70 degrees C. For the treatment with acid-base, similarly, the structure changes of mPEG-IgY were smaller than IgY. Thus, it is indicated that IgY modified by mPEG had greater stable properties.

BACKGROUND: Elevating dietary calcium (Ca) intake can reduce metal(loid)oral bioavailability. However, the ability of a range of Ca minerals to reduce oral bioavailability of lead (Pb), cadmium (Cd), and arsenic (As) from indoor dust remains unclear. OBJECTIVES: This study evaluated the ability of Ca minerals to reduce Pb, Cd, and As oral bioavailability from indoor dust and associated mechanisms. METHODS: A mouse bioassay was conducted to assess Pb, Cd, and As relative bioavailability (RBA) in three indoor dust samples, which were amended into mouse chow without and with addition of CaHPO4, CaCO3, Ca gluconate, Ca lactate, Ca aspartate, and Ca citrate at 200-5,000µg/g Ca. The mRNA expression of Ca and phosphate (P) transporters involved in transcellular Pb, Cd and As transport in the duodenum of mice was quantified using real-time polymerase chain reaction. Serum 1,25-Dihydroxyvitamin D3 [1,25(OH)2D3], parathyroid hormone (PTH), and renal CYP27B1 activity controlling 1,25(OH)2D3 synthesis were measured using ELISA kits. Metal(loid) speciation in the feces of mice was characterized using X-ray absorption near-edge structure (XANES) spectroscopy. RESULTS: In general, mice exposed to each of the Ca minerals exhibited lower Pb-, Cd-, and As-RBA for three dusts. However, RBAs with the different Ca minerals varied. Among minerals, mice fed dietary CaHPO4 did not exhibit lower duodenal mRNA expression of Ca transporters but did have the lowest Pb and Cd oral bioavailability at the highest Ca concentration (5,000µg/g Ca; 51%-95% and 52%-74% lower in comparison with the control). Lead phosphate precipitates (e.g., chloropyromorphite) were observed in feces of mice fed dietary CaHPO4. In comparison, mice fed organic Ca minerals (Ca gluconate, Ca lactate, Ca aspartate, and Ca citrate) had lower duodenal mRNA expression of Ca transporters, but Pb and Cd oral bioavailability was higher than in mice fed CaHPO4. In terms of As, mice fed Ca aspartate exhibited the lowest As oral bioavailability at the highest Ca concentration (5,000µg/g Ca; 41%-72% lower) and the lowest duodenal expression of P transporter (88% lower). The presence of aspartate was not associated with higher As solubility in the intestine. DISCUSSION: Our study used a mouse model of exposure to household dust with various concentrations and species of Ca to determine whether different Ca minerals can reduce bioavailability of Pb, Cd, and As in mice and elucidate the mechanism(s) involved. This study can contribute to the practical application of optimal Ca minerals to protect humans from Pb, Cd, and As coexposure in the environment. https://doi.org/10.1289/EHP11730.

Arsenic (As) hyperaccumulator Pteris vittata is efficient in As uptake, translocation and accumulation, but the impacts of soil As concentrations on As accumulation and distribution in P. vittata are still unclear. The impacts of soil As (7.3, 63 and 228 mg kg-1) on plant growth and As accumulation in P. vittata after 6 months of growth were evaluated. Arsenic concentrations in the roots, midribs and pinna margin of P. vittata fronds of different maturity were determined by inductively coupled plasma mass spectrometry (ICP-MS) and scanning electron microscopy coupled with an energy dispersive spectrometer (SEM-EDS). While moderate As level at As63 didn't impact P. vittata growth, higher As at As228 decreased plant biomass by 38%. Under As stress, more As was accumulated in the senescing fronds (47%) and mature fronds (11%) than the young fronds. In senescing fronds, As concentrations in pinna margin were 2.3 times that of the midribs, consistent with As-induced necrotic symptom. Arsenic distribution based on SEM-EDS analysis revealed good correlation between Si and As in the pinnae (r = 0.49). Our data showed that As accumulation in pinna margin caused necrosis and Si may have a potential role in As detoxification in P. vittata.

OBJECTIVE: To provide scientific basis for the utilization and development of Mucuna pruriens var. utilis by establishing its quality control standard. METHODS: The bioactive constituents were analyzed by TLC and HPLC. Moisture, ash and the extracts of Mucuna pruriens var. utilis were all determined. RESULTS: The TLC spots of levodopa had similar color with the control group at the same position. The results of HPLC quantitative analysis showed that the linear range of levodopa was 26.45 to approximately 132.25 microg/mL, r = 0.9992, and the average recovery rate was 103.8%, RSD = 1.85%. CONCLUSIONS: This method is convenient, accurate, reliable with good reproducibility, so it can be used to establish quality standard for the medicinal material.

Cellulose is the most abundant and widely used biopolymer on earth and can be produced by both plants and micro-organisms. Among bacterial cellulose (BC)-producing bacteria, the strains in genus Komagataeibacter have attracted wide attention due to their particular ability in furthering BC production. Our previous study reported a new strain of genus Komagataeibacter from a vinegar factory. To evaluate its capacity for BC production from different carbon sources, the present study subjected the strain to media spiked with 2% acetate, ethanol, fructose, glucose, lactose, mannitol or sucrose. Then the BC productivity, BC characteristics and biochemical transformation pathways of various carbon sources were fully investigated. After 14 days of incubation, strain W1 produced 0.040⁻1.529 g L-1 BC, the highest yield being observed in fructose. Unlike BC yields, the morphology and microfibrils of BCs from different carbon sources were similar, with an average diameter of 35⁻50 nm. X-ray diffraction analysis showed that all membranes produced from various carbon sources had 1⁻3 typical diffraction peaks, and the highest crystallinity (i.e., 90%) was found for BC produced from mannitol. Similarly, several typical spectra bands obtained by Fourier transform infrared spectroscopy were similar for the BCs produced from different carbon sources, as was the Iα fraction. The genome annotation and Kyoto Encyclopedia of Genes and Genomes analysis revealed that the biochemical transformation pathways associated with the utilization of and BC production from fructose, glucose, glycerol, and mannitol were found in strain W1, but this was not the case for other carbon sources. Our data provides suggestions for further investigations of strain W1 to produce BC by using low molecular weight sugars and gives clues to understand how this strain produces BC based on metabolic pathway analysis.

North China Plain is an important region of grain production.Soil fertility and grain production in this region are significantly influenced by the levels of soil carbon and nitrogen.In order to explore the effects of agricultural practices on the levels of soil carbon and nitrogen,a long-term field experiment was started in 1999 in Quzhou County,Hebei Province.Four treatments,including following nature (F),tillage without straw (N),no tillage with crushed straw incorporation (S),and tillage with crushed straw incorporation (TS),were chosen to collect soil samples at the layers of 0-20 cm and 20-40 cm in 2013.Soil organic carbon (SOC),soil inorganic carbon (SIC),total carbon (TC),total nitrogen (TN),δ13CSOC,δ13CSIC and δ15 N were analyzed.The results indicated that compared with F,SOC stocks of N,S and TS decreased by 21.6%,12.3% and 3.4% in the 0-20 cm soil layer,but the changes of SIC stocks were not significant.In the 20-40 cm soil layer,the changes of the SOC stocks were not significant,but the SIC stocks increased by 4.1%(N),7.3%(S) and 5.0%(TS) compared to F,respectively.Major contribution of SIC increase was the pedogenic inorganic carbonate (PIC),which increased by 97%-261% in the farmland soil.In the soil layer of 0-20 cm,the values of δ15N,δ13CSIC and δ13CSOC in the farmland treatments were higher than those of F,meanwhile,the values of δ13 CSOC were significantly higher than that of F.In the soil layer of 20-40 cm,the values of δ15N and δ13CSIC were lower than those of F,but the value of δ13CSOC showed the opposite trend.In North China Plain,lithogenic inorganic carbonate (LIC) of farmland soil decomposed and PIC increased by the soil-crop system,which provided CO2 for the formation of PIC,and straw returning was an effective agricultural practice to restore the soil carbon decreased by tillage.There should be more long-term monitoring and studies for the impacts of crop straw incorporation and tillage on SOC and SIC,especially for soil in deeper layers.

Arsenic (As) in soils is often adsorbed on Fe-(hydro)oxides surface, rendering them more resistant to dissolution, which is undesirable for phytoremediation of As-contaminated soils. Arsenic hyperaccumulator Pteris vittata prefers to grow in calcareous soils where available Fe and As are low. To elucidate its mechanisms of acquiring Fe and As from insoluble sources in soils, we investigated dissolution of goethite with pre-adsorbed arsenate (AsV; As-goethite) in presence of four organic ligands, including two root exudates (oxalate and phytate, dominant in P. vittata) and two microbial siderophores (PG12-siderophore and desferrioxamine B). Their presence increased As solubilization from As-goethite from 0.03 to 0.27-5.33 mg L-1 compared to the control. The siderophore/phytate bi-ligand treatment released 7.42 mg L-1 soluble Fe, which was 1.2-fold that of the sum of siderophore and phytate, showing a synergy in promoting As-goethite dissolution. In the ligand-mineral-plant system, siderophore/phytate was most effective in releasing As and Fe from As-goethite. Moreover, the continuous plant uptake induced more As-goethite dissolution. The continued release of As and Fe significantly enhanced their plant uptake (from 0.01 to 0.43 mg plant-1 As and 2.7-14.8 mg plant-1 Fe) and plant growth (from 1.2 to 3.1 g plant-1 fw) in P. vittata. Since microbial siderophores and root exudates often coexist in soil rhizosphere, their synergy in enhancing dissolution of insoluble As-Fe minerals may play an important role in efficient phytoremediation of As-contaminated soils.

Biochar application to agricultural soil is an appealing approach to mitigate nitrous oxide (N2O) and methane (CH4) emissions. However, the underlying microbial mechanisms are unclear. In this study, a paddy soil slurry was incubated anaerobically for 14d with biochar amendments produced from rice straw at 300, 500, or 700°C (B300, B500, and B700) to study their influences on greenhouse gas emissions. Illumina sequencing was used to characterize shift of soil bacterial and archaeal community composition. After peaking at day 1, N2O emission then sharply decreased to low levels while CH4 started to emit at day 3 then continually increased with incubation. Compared to control soil (57.9mgkg-1 soil), B300, B500, and B700 amendments decreased N2O peak emission to 17.9, 1.28, and 0.59mgkg-1, mainly due to increased soil pH. In contrast, the amendments enhanced CH4 production from 58.2 to 93.4, 62.6, and 63.4mgkg-1 at day 14 due to increased soil dissolved organic carbon. Abundance of denitrifying bacteria (e.g., Bacilli, 7.07-13.6 vs. 16.9%) was reduced with biochar amendments, especially with B500 and B700, contributing to the decreased N2O emissions. However, larger pore size of B500 and B700 (surface area of 68.1 and 161m2g-1) than B300 (4.40m2g-1) favored electron transfer between bacteria and iron minerals, leading to increased abundance of iron-reducing bacteria, (e.g., Clostridia, 48.2-50.6 vs. 33.3%), which competed with methanogens to produce CH4, thereby leading to lower increase in CH4 emission. Biochar amendments with high pH and surface area might be effective to mitigate emission of both N2O and CH4 from paddy soil.

In order to investigate the internal exposure levels of polycyclic aromatic hydrocarbons (PAHs) in primary school students of Guangzhou, the research collected urine of 78 and 86 primary school students from two primary schools in the summer of 2014, one school located in the ordinary residential area and the other in the industrial area. The contents of 10 kinds of OH-PAHs were tested by the rapid liquid chromatography coupled to triple quadruple tandem mass spectrometry. The results showed that the concentrations of total OH-PAHs in primary school students in the residential zone ranged from 0.83 µmol · mol⁻¹ to 80.63 µmol · mol⁻¹, while those in industrial area ranged from 1.06 µmol · mol⁻¹ to 72.47 µmol · mol⁻¹. The geometric average concentrations were 6.18 µmol · mol⁻¹ and 6.47 µmol · mol⁻¹, respectively, and there was no statistical significance between them (P > 0.05). Comparison of the exposure levels of different components of PAHs in the two areas found that all the OH-PAHs had no significant difference except for the levels of 1- OHP (P < 0.05). We should also pay attention to the higher exposure levels of PAHs in both areas when compared with other researches. In addition, the OH-PAHs in primary school students in the ordinary residential area had a good correlation between 0. 511 and 0.928 (P < 0.01), whereas there was no correlation between 1-OHP and 2-OHN, 1-OHN in the primary school students in the industrial area and other OH-PAHs had relatively weak correlation ranging from 0.338 to 0.855 (P < 0.01). This difference might indicate different pollution sources of PAHs in different functional areas, which was relatively single in the residential area, while the industrial area was polluted by multiple sources of industrial enterprises and logistics transportation emissions.
