The microscopic fluorescence spectroscopy has become a mature technology of fluid inclusions test and analysis system, which is used to distinguish different types of crude oil and oil inclusions. These would be the important basis to study the history of hydrocarbon accumulation of petroleum basins. The mixture of crude oil from different sources could occur in migration and accumulation process. In order to effectively identify the type of geological process, mixing ratio of crude oil experiment has been carried out. This study result shows that mixing of crude oil make fluorescence color and spectral parameters(λmax, QF535 and CIE-XY) change nonlinearly. Fluorescence spectral parameters of mixed oil is between end member oil A and B. The greater A or B ratio of mixed oil, the closer to A or B. Fluorescence color of mixed oil show nonlinear and gradual change in CIE-XY chromaticity diagram. Variation of spectral spectrum shape show that single peak is changed into double and three peaks. The relationship between QF535 and degree of mixing could calculate quantitatively relative contribution. Mixing different types of crude oil make spectral spectrum shape changes, which present characteristics of two peaks and three peaks but not unimodal peak. The main and subsidiary wavelength reserve wavelength information of end member oils. Based on variation characteristics of fluorescence spectrum, there are three different types of oil including blue, blue-green and yellow fluorescing oil filling in the bottom member of Pinghu formation in A gas field. At the same time, there also was a mixing process of blue-green fluorescing oil and yellow fluorescing oil. The degree of mixing is 47%~55%.

Based on oil samples and rock samples in several petroliferous basins, this paper studied that secondary alterations influence on fluorescence color and spectral parameters of crude oil and oil inclusions by using microscopic fluorescent spectral and geochemical parameters. The results demonstrated that secondary alterations influence on spectral parameters by changing the ratio of saturated hydrocarbon and aromatic hydrocarbon (saturation/aromatics). Biodegradation and water washing lead to redshift of fluorescence parameters while evaporative fractionation and oil cracking lead to blueshift. Beside, mixed oil lead to redshift and blueshift. The secondary alterations and main period of accumulation in research area were recognized by frequency histogram of crude oil and oil inclusion. The relationship between QF-535 and the parameters measuring the secondary alterations could the extent of secondary alterations from which crude oil suffered. In the microscopic fluorescence test, oil inclusions suffered from stretch or leakage, thermal cracking and photooxidation should be rejected.

RESUMO

A novel four-component one-pot approach for the synthesis of 2-amino-1,3,4-thiadiazoles from primary amines, carbon disulfide, hydrazine, and acyl chlorides has been developed. A series of 5-substituted-2-amino-1,3,4-thiadiazoles were synthesized in medium-to-good yields utilizing this newly developed method.

Mitochondrial transcription termination factor (mTERF) is a DNA-binding protein that is encoded by nuclear genes, ultimately functions in mitochondria and can affect gene expression. By combining with mitochondrial nucleic acids, mTERF regulates the replication, transcription and translation of mitochondrial genes and plays an important role in the response of plants to abiotic stress. However, there are few studies on mTERF genes in tomato, which limits the in-depth study and utilization of mTERF family genes in tomato stress resistance regulation. In this study, a total of 28 mTERF gene family members were obtained through genome-wide mining and identification of the tomato mTERF gene family. Bioinformatics analysis showed that all members of the family contained environmental stress or hormone response elements. Gene expression pattern analysis showed that the selected genes had different responses to drought, high salt and low temperature stress. Most of the genes played key roles under drought and salt stress, and the response patterns were more similar. The VIGS method was used to silence the SLmTERF13 gene, which was significantly upregulated under drought and salt stress, and it was found that the resistance ability of silenced plants was decreased under both kinds of stress, indicating that the SLmTERF13 gene was involved in the regulation of the tomato abiotic stress response. These results provide important insights for further evolutionary studies and contribute to a better understanding of the role of the mTERF genes in tomato growth and development and abiotic stress response, which will ultimately play a role in future studies of tomato gene function.

Coral-zooxanthellae holobionts are one of the most productive ecosystems in the ocean. With global warming and ocean acidification, coral ecosystems are facing unprecedented challenges. To save the coral ecosystems, we need to understand the symbiosis of coral-zooxanthellae. Although some Scleractinia (stony corals) transcriptomes have been sequenced, the reliable full-length transcriptome is still lacking due to the short-read length of second-generation sequencing and the uncertainty of the assembly results. Herein, PacBio Sequel II sequencing technology polished with the Illumina RNA-seq platform was used to obtain relatively complete scleractinian coral M. foliosa transcriptome data and to quantify M. foliosa gene expression. A total of 38,365 consensus sequences and 20,751 unique genes were identified. Seven databases were used for the gene function annotation, and 19,972 genes were annotated in at least one database. We found 131 zooxanthellae transcripts and 18,829 M. foliosa transcripts. A total of 6328 lncRNAs, 847 M. foliosa transcription factors (TFs), and 2 zooxanthellae TF were identified. In zooxanthellae we found pathways related to symbiosis, such as photosynthesis and nitrogen metabolism. Pathways related to symbiosis in M. foliosa include oxidative phosphorylation and nitrogen metabolism, etc. We summarized the isoforms and expression level of the symbiont recognition genes. Among the membrane proteins, we found three pathways of glycan biosynthesis, which may be involved in the organic matter storage and monosaccharide stabilization in M. foliosa. Our results provide better material for studying coral symbiosis.

The Chinese tree shrew (Tupaia belangeri yaoshanensis) has long been proposed to serve as an animal model for studying human diseases. However, its overall genetic diversity and population structure remain largely unknown. In the present study, we investigated the genetic diversity of population microsatellite DNA in wild Tupaia belangeri yaoshanensis. Sixteen microsatellite loci were assessed in 76 wild Tupaia belangeri yaoshanensis. The target microsatellite DNA fragments were amplified from the peripheral blood DNA of the animals by polymerase chain reaction (PCR), and the PCR-amplified products were verified by DNA sequencing and used for the analysis of allele, effective allele, genetic heterozygosity, polymorphism and population structure. Our results showed that of the 16 microsatellite loci examined, 5 microsatellite loci were monomorphic and 11 microsatellite loci were polymorphic. We detected 61 alleles in the polymorphic loci and found 2-10 (with an average of 5.5455) alleles per locus. Our data also showed that the observed and expected heterozygosities ranged from 0.087 to 0.8947 and 0.1368 to 0.7892 with an average of 0.3968 and 0.4796, respectively. Taken together, the results revealed a considerably high heterozygosity and high genetic diversity at the molecular level in the population of wild Tupaia belangeri yaoshanensis. The identified markers from the present study may be useful for individual identification and parentage testing, as well as for the quantification of population heterogeneity in the Chinese tree shrew.
