RESUMO

The first flush occurs during urban runoff events. In this study, we aimed to assess the characteristics of different-sized particles in the first flush of roof runoff, and runoff was collected from an asphalt roof (AR), metal roof (MR), and cement roof (CR) for analysis. There were no clear patterns in the particle size distributions in the runoff from the three roofs and were affected by several factors. The strength of the first flushes differed significantly for particles in different size categories in AR, MR, and CR runoff and were very different from suspended solids (SS). The comparison showed that it would be possible to meet the SS control design expectation required by the Chinese national standard for runoff pollution control (VFF = 3 mm) for particles <45 µm but not for particles >45 µm. The methods presented provide an alternative for assessing the ability to control the transport of different-sized particles in runoff.

RESUMO

Bioretention is commonly used for runoff pollution control. The first flush strength of pollutants can affect bioretention performance. To examine the influence of the first flush strength on bioretention performance, bioretention columns filled with garden soil as the main media were established. Activated carbon and water treatment residuals (WTR) were added and compared for their ability to enhance phosphorus removal. Waste edible fungus culture medium (WEFCM) as a carbon source was also explored. When WEFCM was used as a carbon source instead of wood chips, total nitrogen (TN) removal increased from 60.83 ± 21.22 to 62.21 ± 16.43%, but chemical oxygen demand (COD) leaching was observed. WTR was better able than activated carbon to enhance phosphorus removal (87.97 ± 8.87 vs. 81.66 ± 9.27%) without impacting TN removal. NH4+-N removal increased with increasing first flush strength, but there was no trend for suspended solids (SS), COD, TN, or total phosphorus. First leaching phenomenon in bioretention outflow was proposed in this study. A low first leaching was observed in the outflow when the inflow had a uniform pollutant mass (i.e., no first flush) because of media leaching. A weak first leaching outflow was observed for SS and COD when they were present at strong first flush inflow.

RESUMO

First flush is a common phenomenon in urban runoff pollution. Typical cement roof and asphalt road runoff in Beijing, China were monitored for 2 years. Based on the M(v) curve, the suspended solids (SS), chemical oxygen demand (COD), total phosphorus (TP) and particulate phosphorus in cement roof runoff presented a stronger first flush than those in asphalt road runoff. The first flush volume (VFF) of SS, COD, total nitrogen (TN) and TP in asphalt road runoff differed slightly from the cement roof. There were also differences in the first flush assessment depending on which method was used. We proposed a new method based on the runoff depth versus pollutant cumulative mass curve. According to the national standards in China (VFF = 3 mm), various masses of different pollutants, such as 91.42 ± 9.80% (cement roof) and 78.49 ± 19.41% (asphalt road) of SS and 86.85 ± 13.54% (cement roof) and 72.80 ± 25.79% (asphalt road) of COD, can be effectively controlled, but our mass control efficiencies were 55.91%-66.65% when VFF = 1 mm. The new method proposed in this study provides an alternative approach for assessing runoff pollution control efficiency of different VFF.

RESUMO

Cardiac hypertrophy has become a major cardiovascular problem wordwide and is considered the early stage of heart failure. Treatment and prevention strategies are needed due to the suboptimal efficacy of current treatment methods. Recently, many studies have demonstrated the important role of histone acetylation in myocardium remodelling along with cardiac hypertrophy. A Chinese herbal extract containing anacardic acid (AA) is known to possess strong histone acetylation inhibitory effects. In previous studies, we demonstrated that AA could reverse alcohol-induced cardiac hypertrophy in an animal model at the foetal stage. Here, we investigated whether AA could attenuate cardiac hypertrophy through the modulation of histone acetylation and explored its potential mechanisms in the hearts of transverse aortic constriction (TAC) mice. This study showed that AA attenuated hyperacetylation of acetylated lysine 9 on histone H3 (H3K9ac) by inhibiting the expression of p300 and p300/CBP-associated factor (PCAF) in TAC mice. Moreover, AA normalized the transcriptional activity of the heart nuclear transcription factor MEF2A. The high expression of cardiac hypertrophy-linked genes (ANP, ß-MHC) was reversed through AA treatment in the hearts of TAC mice. Additionally, we found that AA improved cardiac function and survival rate in TAC mice. The current results further highlight the mechanism by which histone acetylation is controlled by AA treatment, which may help prevent and treat hypertrophic cardiomyopathy.

RESUMO

Bioretention is an effective technology for urban stormwater management, but the nutrient removal in conventional bioretention systems is highly variable. Thus, a pilot bioretention column experiment was performed to evaluate the nutrient control of systems with commercial activated carbon and river sediment-derived biochar. Significant chemical oxygen demand (COD) and total phosphorus (TP) leaching were found with the addition of activated carbon and biochar, but total nitrogen (TN) leaching was significantly improved when activated carbon was used as the medium. During a semi-synthetic runoff experiment, the bioretention systems containing two types of fluvial biochar showed relatively better COD and TN control (average mass removal efficiencies and cumulative removal efficiencies) than commercial activated carbon. However, the average TP mass removal efficiency with commercial activated carbon (95% ± 3%) was significantly higher than biochar (48% ± 20% and 56 ± 14%). The addition of biochar in the media increased the nitrogen removal efficiency, and the addition of activated carbon significantly increased the phosphorous removal efficiency. Therefore, both biochar and activated carbon are effective materials for bioretention, and fluvial biochar provides an alternative approach to comprehensively utilize river sediment.

RESUMO

In this study, laboratory-scale green (e.g. living) roof platforms were established to assess the potential use of polluted river sediment in their substrate mixture. The mean runoff retention of the green roof platforms, which contained peat and/or river sediment, after 11 artificial rainfall events was >72%, significantly higher than traditional roofs. However, green roof platforms that had been filled with peat soil showed chemical oxygen demand (COD), total nitrogen (TN) and total phosphorus (TP) leaching. Green roofs that had used river sediment showed good leaching control for COD, TN and TP. The cumulative leaching masses from the green roofs contained 30% (COD), 42% (TN) and 47% (TP) as much as the total leaching mass from traditional roofs, and the Cu, Zn, Cd and Pb leaching risk from green roofs when river sediments are used as part of a substrate mixture was relatively low. Despite some nutrient leaching in the initial phase of runoff from the green roofs, river sediment has the potential to be used as a substrate for extensive green roofs.

RESUMO

Diastolic cardiac dysfunction can be caused by abnormality in cTnI expression during cardiogenesis. In this study, we investigated the effects of estrogen on the abnormal expression of cTnI in the hearts of neonatal mice and its potential epigenetic mechanisms. We then evaluated suberoylanilide hydroxamic acid (SAHA), a HDAC inhibitor, as a new target treatment of diastolic cardiac dysfunction. Postnatal day 0.5 C57BL/6 mice were injected with estrogen for 1 week, then the hearts of 7-day-old neonatal mice were retrieved for examination. The activities of HDAC and HAT were assayed by colorimetry, and the interaction of cTnI with HDAC5 in mice hearts were examined using chromatin immunoprecipitation assays. The expression of cTnI was tested by quantitative real-time RT-PCR and Western blot. Estrogen treated groups displayed a significantly increased HDAC activity in the hearts of neonatal mice while HAT activity remained unchanged. Additionally, HDAC5 was higher at the cTnI promoter, as compared to the saline treated control groups. The acetylation of histone H3K9ac on cTnI promoter significantly decreased in the hearts of neonatal mice treated with estrogen, and the expression of cTnI at transcriptional and protein levels also decreased. SAHA was shown to increase the acetylation of histone H3K9ac and upregulate the expression of cTnI. The data demonstrated that SAHA can correct cTnI expression abnormality caused by estrogen through inhibiting the binding of HDAC5 to the promoter of cTnI. J. Cell. RESUMO

Approaches based on the octanol-water partition coefficient are commonly used to describe sorption of neutral organic compounds in environmental systems, but they are not suitable for organic acids, which can dissociate to form anions. We here investigate the applicability of an alternative approach based on the pH-dependent distribution ratio (DOW) to describe sorption of aromatic acids to sorbents representing different degrees of carbonization. Sorption isotherms for four structurally similar acids ((2,4-dichlorophenoxy)acetic acid (2,4-D), 4-chloro-2-15 methylphenoxy)acetic acid (MCPA), 4-(2,4-dichlorophenoxy)butanoic16 acid (2,4-DB), and 5-chloro-2-(2,4-dichlorophenoxy)phenol (triclosan)) were measured for 15 sorbents: fresh and carbonized wood shavings, pig manure, sewage sludge, carbon nanotubes, and activated carbon. Dissociation greatly affected the sorption of all acids. Sorption coefficients measured in the high pH range indicated that sorption of the anions ranged over several orders of magnitude and should not be neglected. Sorption trends for all sorbates and carbonized sorbents could be very well described by a single regression equation that included DOW of the sorbate and the specific surface area of the sorbent (R(2) > 0.89).

RESUMO

BACKGROUND: Alcohol abuse during gestation may cause congenital heart diseases (CHDs). The underlying mechanisms of alcohol-induced cardiac deformities are still not clear. Recent studies suggest that histone modification may play a crucial role in this pathological process. Moreover, our previous studies reported that ethanol could induce histone3 lysine9 (H3K9) hyperacetylation and overexpression of heart development-related genes in vitro. The aim of this study was to investigate the effect of alcohol consumption during gestation on the imbalance of H3K9 acetylation and the alternation of the expression of heart development-related genes during cardiogenesis. METHODS: Pregnant mice were exposed to a single dose of alcohol (10 µl/g/d, 56% alcohol) by gavage every day in the morning from embryo day 7.5 (E7.5) to E15.5. Hematoxylin and eosin (H&E) staining was applied for observing the structure of the embryonic hearts. Western blotting and quantitative real-time polymerase chain reaction were used for detecting the level of H3K9 acetylation and gene expression. Histone acetyltransferase (HAT) and histone deacetylase (HDAC) activities were detected by colorimetric assay and fluorometric assay. RESULTS: H&E staining of cardiac tissue showed abnormalities of embryonic hearts at E17.5. The level of H3K9 acetylation reached peak at E17.5 and decreased sharply to a low level at birth and maintained at low level afterward. Alcohol exposure increased H3K9 acetylation at E11.5, E14.5, E17.5, and E18.5, respectively (p < 0.05), and enhanced the expression of Gata4 in the embryonic hearts at E14.5 and E17.5, Mef2c at E14.5, and Nkx2.5 at E14.5 and E17.5, (p < 0.05) but not for Tbx5 (p > 0.05). On embryonic day 17.5, HAT activities of embryonic hearts increased significantly, however alcohol exposure did not alter HDAC activities. CONCLUSIONS: These data indicate a time course of H3K9 acetylation change during heart development and demonstrate that alcohol exposure in utero may induce an increase of HAT activities, which results in H3K9 hyperacetylation and an increase of the expression of heart development-related genes. These findings reveal a novel epigenetic mechanism that connects the alcohol consumption during the pregnancy and the development of CHD in the fetus.

RESUMO

OBJECTIVE: We have evaluated 106 pediatric cases of infective endocarditis (IE) to elucidate clinical manifestations and pathogenic microorganism profiling of IE in China. METHODS: Clinical features, complications, pathogenic microorganisms, diagnosis and treatment of pediatric IE were reviewed in two groups of patients with IE from the different periods of time (group A, 34 cases obtained in the period from 2000 to 2011 and group B, 72 cases obtained in the period from 1964 to 1999). RESULTS: A total of 106 pediatric patients with a definite diagnosis of IE based on the modified Duke criteria were enrolled and evaluated in this study. By comparing two groups of patients from different time periods, we found that the incidence of rheumatic heart disease was significantly reduced (from 19.4 to 5.9%), whereas congenital heart disease-associated IE had a tendency to increase (from 55.6 to 79.4%). Staphylococcus aureus was detected as the most common pathogenic microorganism, and its involvement tended to increase (from 32.0 to 58.5%), whereas the percentage of infections caused by Streptococcus viridans (8.0%) had not changed. It was interesting to note that the rate of vegetations detected was increased from 50.0 to 67.6% and the incidence of right-sided IE was also increased (from 35.0 to 60.9%). The most common valves involved in recent cases were tricuspid valves (increase from 30.0 to 47.8%), while mitral valve infection was reduced (from 60.0 to 39.1%). Penicillin was still the most commonly used antibiotic for the treatment of IE; the combination of penicillin plus cephalosporin has been recommended more and more recently. CONCLUSIONS: Comparing pediatric IE patients during the past 40 years, we found that the role of rheumatic heart disease as a predisposing factor is diminishing. Pediatric IE is still predominantly caused by staphylococci. The most commonly involved valves are tricuspid valves instead of mitral valves. Gram-positive bacteria showed an increased resistance to penicillin when used alone, and the use of combination treatment with antibiotics is increasing in the area.

RESUMO

In order to enhance traditional building materials, High-performance concrete (HPC) is being modified by adding carbon and basalt fibers with volume contents of 0.75-1.25% and 0.15-0.35%, respectively. The original mechanical properties are maintained while developing the material's intelligent self-sensing and self-heating functions, which are tested for pressure sensitivity and bending sensitivity, and with electrothermal tests. The results demonstrate that carbon fiber can significantly reduce the matrix resistivity of high-performance concrete, reaching the percolation threshold at a content of 1%. The inclusion of basalt fibers in the material results in a decrease in resistivity. However, the addition of mixed fibers leads to improved mechanical-electrical sensitivity under compression and bending, with a positive hybrid effect. The optimal contents for carbon fiber and basalt are 0.75% and 0.3%, respectively. In electrothermal tests, the specimen can reach a temperature of 104.5 °C with a heating rate of 25.86 °C/h, indicating the potential for self-monitoring and the electric melting of ice and snow. These findings provide support for the intelligent improvement of building structures in the new era.

RESUMO

Alcohol exposure during pregnancy may cause congenital heart disease (CHD). In our previous studies, we found that alcohol selectively increased acetylation of histone H3 at lysine 9 (H3K9) and enhanced the expression of heart development-related genes in cardiac progenitor cells. The objective of this study is to investigate the protective effects of histone acetyltransferases (HATs) inhibitor, curcumin, on histone hyperacetylation and the over-expression of heart development genes induced by alcohol. Western blot analysis was employed to detect the acetylation levels of histone H3K9 and real-time PCR was applied to measure the expressions of heart development-related transcription factors, GATA4, Mef2c and Tbx5 (GMT). Our results showed that alcohol increased the acetylation of H3K9 by 2.76-fold (P<0.05) and significantly enhanced the expression of GATA4 and Mef2c (P<0.05). When cells were treated with alcohol plus 25 µM curcumin, the hyperacetylation of H3K9 and over-expression of GATA4 and Mef2c by alcohol was reversed. These data indicate that curcumin can correct the over-expression of cardiac genes by reversing the alcohol induced hyperacetylation of histone H3 at lysine 9 in cardiac progenitor cells, suggesting that curcumin is protective against alcohol-induced cardiac gene over-expression that may result in heart malformations.

RESUMO

Background and Aim: Heart failure in children differs substantially from the adult population. Clinical characteristics of pediatric diastolic heart failure has rarely been reported. In this study, we aimed to summary the causes, clinical features, lab tests, and treatment effect of pediatric diastolic heart failure. Methods: This study was a single center, retrospective study conducted in Children's Hospital of Chongqing Medical University. Children who were diagnosed with diastolic heart failure (DHF) without systolic heart failure (SHF) between 2006 and 2014 were included. Meanwhile, SHF (without DHF) cases were also collected from 2013 to 2014. Results: A total of 421 DHF and 42 SHF cases were included. The average age of pediatric DHF was 1.89 ± 3.29 years old, significant younger than that of SHF (4.65 ± 4.90). The top three cardiovascular causes of DHF were complex congenital heart malformations (53.4%), simple congenital heart defect (15.7%), and cardiomyopathy (7.4%). Alternatively, number of cardiomyopathy cases (57.1%) ranked first in SHF group. Simple congenital heart diseases (CHDs) rarely caused SHF. The most common symptom and sign were tachypnea and hepatomegaly in pediatric HF. Symptoms like cyanosis, feeding difficulty, be fidgety, pale, fatigue, and edema were valuable in differential diagnosis of DHF and SHF in children. B-type natriuretic peptide (BNP) increase was found in 36.9% of DHF children, and 60% in SHF patients. Sensitivity of BNP greater than 100 pg/ml in diagnosis of DHF was 0.37, and specificity of it was 0.86. Diastolic function indicators, such as E/A (early wave/late wave) ratio, IVRT (isovolumic relaxation time) were significant recovered after treatment in DHF patients. Less therapeutic benefits were achieved in children with cardiomyopathy induced DHF, in compared with non-cardiomyopathy patients. Conclusion: Pediatric DHF and SHF were largely different in primary causes, clinical symptoms and signs and short-term prognosis. There was a limit diagnostic value of BNP with 100 pg/ml as cut-off value in pediatric DHF. Larger, multicenter studies of pediatric DHF are required in the future.

RESUMO

Histone acetyltransferases (HATs), p300 and cAMP response element binding protein (CREB)-binding protein (CBP) are two structurally related transcriptional co-activators that activate expression of many eukaryotic genes involved in cellular growth and signaling, muscle differentiation and embryogenesis. However, whether these proteins play important and different roles in mouse cardiogenesis is not clear. Here, we investigate the protein distributions and mRNA expression of the two HATs in embryonic and adult mouse heart during normal heart development by using immunohistochemical and RT-PCR techniques. The data from immunohistochemical experiments revealed that p300 was extensively present in nearly every region of the hearts from embryonic stages to the adulthood. However, no CBP expression was detected in embryonic hearts at day E7.5. CBP expression appeared at the later stages, and the distribution of CBP was less than that of p300. In the developmental hearts after E10.5, both for p300 and CBP, the mRNA expression levels reached a peak on day E10.5, and then were gradually decreased afterwards. These results reveal that both p300 and CBP are related to embryonic heart development. The dynamic expression patterns of these two enzymes during mouse heart development indicate that they may play an important role on heart development. However, there is a difference in spatiotemporal expression patterns between these two enzymes during heart development. The expression of p300 is earlier and more predominate, suggesting that p300 may play a more important role in embryonic heart development especially during cardiac precursor cell induction and interventricular septum formation.

RESUMO

While coronary artery abnormality (CAA) has been established as the most serious complication of Kawasaki disease (KD), there have been no detailed systematic reviews of the risk factors associated with this condition. We searched six databases and performed a systematic review and meta-analysis. Study-specific odds ratios (ORs) for each factor were pooled using a random effects model. We identified four risk factors for CAA children with KD: gender (OR, 1.75; 95% confidence interval [CI], 1.59-1.92), intravenous immunoglobulin (IVIG) resistance (OR, 3.43; 95% CI, 2.07-5.67), IVIG treatment beyond 10 days of onset of symptoms (OR, 3.65; 95% CI, 2.23-5.97), and increased C-reactive protein levels (OR, 1.02; 95% CI, 1.01-1.02). More number of the five typical symptoms of KD was identified as a protective factor against CAA (OR, 0.47; 95% CI, 0.33-0.66). Pediatric patients with IVIG resistant were more likely to develop CAA within 1 month of the onset of KD than the general population, even in patients with other risk factors for CAA. Thus, there is a potential risk of CAA misdiagnosis if echocardiography is not carried out frequently. In summary, we report four risk factors for CAA and a protective factor against CAA in children with KD. We recommend that pediatricians consider these factors much more closely. With accurate prediction and early preventive treatment in high-risk patients, we can expect a reduction in CAA rates. Further research is now required to investigate the associations between CAA and other factors including the interval between KD onset and IVIG administration, platelet count, and the duration of fever. We also need to confirm whether the frequency of echocardiography within a month of KD onset should be increased in IVIG-resistant patients.

RESUMO

The silicone rubber passive sampling technique is extensively applied to monitor the aqueous freely dissolved concentration of hydrophobic organic compounds (HOCs). The silicone rubber-water partition coefficient (Ksrw) is an important parameter to accurately measure the concentrations of chemicals using passive sampling devices. In this study, two theoretical linear solvation energy relationship (TLSER) models and a quantitative structure-property relationship (QSPR) model were developed for predicting the Ksrw of HOCs. The 119 model compounds studied here included 31 personal care products, such as musks, UV-filters, and organophosphate flame retardants, as well as "conventional" pollutants, such as polycyclic aromatic hydrocarbons and polychlorinated biphenyls. The statistical parameters indicated that the final QSPR model with seven descriptors for all 119 chemicals had a satisfactory goodness-of-fit (Radj2 = 0.898), robustness (QLOO2 = 0.881) and predictive ability (Qext-F1,2,32 = 0.897-0.926). In comparison, the results of one TLSER model with four descriptors for 113 chemicals (Radj2 = 0.826, QLOO2 = 0.790, Qext-F1,2,32 = 0.805-0.837) and another TLSER model with one descriptor for 5 chemicals (Radj2 = 0.747, QLOO2 = 0.647) were also acceptable. The applicability domains of the obtained models covered chemicals containing hydroxyl, imino groups, carbonyl groups, ether bonds, halogen atoms, sulfur atoms, phosphorus atoms, nitro groups, and cyano groups. In addition, the structural features governing the partition behavior of chemicals between silicone rubber and water were explored through interpretation of appropriate mechanisms.

RESUMO

Bioretention is one of the most popular technical practices for urban runoff pollution control. However, the efficiency of nutrient removal from urban stormwater runoff by bioretention systems varies significantly. To improve the nutrient removal performance, innovative up-flow and mixed-flow bioretention systems were proposed in this study, and a laboratory study was conducted to investigate the runoff retention and nutrient removal performance. During the leaching experiment using tap water as the inflow, turbidity, chemical oxygen demand (COD), total nitrogen (TN), and total phosphorus (TP) leaching phenomenon was obvious. COD and TN leaching controls were obviously improved when the up-flow and mixed-flow bioretention systems were adopted comparing with the conventional bioretention. During the semi-synthetic runoff experiments, after the leaching experiments' performance (accumulated 2.78 times of empty bed volume), there were no significant differences in COD mass removal efficiencies of conventional and up-flow bioretention processes (p > 0.05); however, the COD mass removal efficiencies of the mixed-flow bioretention processes increased by 10% when compared with conventional bioretention. The TN mass removal efficiencies of the up-flow and mixed-flow bioretention increased obviously from 17% ± 13% (conventional) to 41% ± 23% (up-flow) and 31% ± 16% (mixed-flow). However, there were no significant differences in TP mass removal or runoff reduction among the three bioretention columns (p > 0.05). Both up-flow and mixed-flow bioretention can effectively improve TN mass removal, and the mixed-flow bioretention did not show a better TN removal performance than the up-flow bioretention because these two bioretention had almost the same volume of the saturated zone. Overall, the results indicate the mixed-flow bioretention proposed in this study can effectively improve TN mass removal and slightly improve COD mass removal relative to conventional methods via increases in hydraulic retention time and in-flow paths, respectively.

RESUMO

GATA4 is a particularly important cardiogenic transcription factor and serves as a potent driver of cardiogenesis. Recent progress in the field has made it clear that histone acetylation can influence gene expression through changing the structure of chromatin. Our previous research had revealed that hypo-acetylation could repress gata4 expression in cardiocytes, however the underlying mechanism by which this occurred was still unclear. To reveal the mechanism of histone acetylation involved in the regulation of gata4 transcription, we concentrated on P300, one of the important histone acetyltransferase associated with cardiogenesis. We found that P300 participated in gata4 expression through regulating histone acetylation in embryonic mouse hearts. RNAi-mediated downregulation of P300 modulated the global acetylation of H3 and the acetylation of H3K4, H3K9, and H3K27 in gata4 and Tbx5 promoters. Interestingly, there was an obvious inhibition of gata4 transcription, whereas Tbx5 was not influenced. Furthermore, SGC-CBP30, the selective inhibitor of the bromodomain in CBP/P300, downregulated gata4 transcription by repressing the acetylation of H3K4, H3K9, and H3K27 in the gata4 promoters. Taken together, our results identified that acetylation of H3K4, H3K9, and H3K27 mediated by P300 plays an important role in regulation of gata4 expression in cardiogenesis.

RESUMO

Patients with familial hypercholesterolemia usually present with high levels of serum low-density lipoprotein, xanthomas and early coronary artery disease. A 13 years old female patient was admitted to Children's Hospital of Chongqing Medical University presenting symptoms of heart failure. Laboratory tests showed that her cholesterol and low-density lipid levels were extremely high. Electrocardiogram test revealed that she had sinus tachycardia, QT lengthening and ST-T change. Multiple cardiac function abnormalities were diagnosed by echocardiogram. Multiple coronary artery stenosis was determined by computed tomography angiography. After the combination of lipid lowering, anti-thrombosis, and cardiac remodeling therapies, the patient's symptoms were significantly improved and the patient was discharged.
