RESUMO

China has the highest incidence of hepatitis B virus (HBV) infection worldwide. HBV genotypes have variable impacts on disease pathogenesis and drug tolerance. We have developed a technically simple and accurate method for HBV genotyping that will be applicable to pre-treatment diagnosis and individualized treatment. Multiple sequence alignments of HBV genomes from GenBank were used to design primers and probes for genotyping of HBV A through H. The hybridization was carried out on nitrocellulose (NC) membranes with probes fixed in an array format, which was followed by hybrid amplification by an extension step with DNA polymerase to reinforce the double-stranded DNA hybrids on the NC membrane and subsequent visualization using an avidin-biotin system. Genotyping results were confirmed by DNA sequencing and bioinformatics analysis using the National Center for Biotechnology Information genotyping database, and compared with results from the line probe assay. The data show that multiple sequence alignment defined a 630 bp region in the HBV PreS and S regions that was suitable for genotyping. All genotyping significant single nucleotides in the region were defined. Two-hundred-and-ninety-one HBV-positive serum samples from Northwest Chinese patients were genotyped, and the genotyping rate from the new modified hybridization-extension method was 100% compared with direct sequencing. Compared with line probe assay, the newly developed method is superior, featuring reduced reaction time, lower risk of contamination, and increased accuracy for detecting single nucleotide mutation. In conclusion, a novel hybridization-extension method for HBV genotyping was established, which represents a new tool for accurate and rapid SNP detection that will benefit clinical testing.

Biomass-based aerogel fibers have attracted increasing attention due to their renewable nature. However, their disadvantages, such as low mechanical strength, poor long-range order, and easy combustion, are still significant challenges. Herein, a directed freezing-assisted forced stretching strategy is developed to fabricate sheath-core structured Ca-alginate/polyvinyl alcohol (Ca-A/PVA) aerogel fibers with Long-range-ordered pores. The Ca-A/PVA aerogel fibers (3:2 m/m) exhibit the best comprehensive mechanical properties in terms of low thermal conductivity of 0.0524 W·m-1·K-1, a density of 0.1614 g·cm-3, a porosity of 89.9 %, a tensile strength of 8.72 MPa, a tensile modulus of 249.7 MPa, a toughness of 1.98 MJ∙m-3, and a self-extinguishing time from the fire of <1.2 s. The Ca-A/PVA fabrics showed a maximum absolute temperature difference of 11.4 °C at -20 °C and 14.0 °C at 60 °C compared to the plate temperature. The presented strategy is generalizable to other alginate-based aerogel fibers (e.g., alginate/guar gum).

We investigate the effect of vibrational resonance in small-world neuronal networks with hybrid chemical and electrical synapses. It is shown that, irrespective of the probability of chemical synapses, an optimal amplitude of high-frequency component of the signal can optimize the dynamical response of neuron populations to the low-frequency component, which encodes the information. This effect of vibrational resonance of neuronal systems depends extensively on the network structure and parameters, which determine the ability of neuronal networks to enhance the outreach of localized subthreshold low-frequency signal. In particular, chemical synaptic coupling is more efficient than the electrical coupling for the transmission of local input signal due to its selective coupling. Moreover, there exists an optimal small-world topology characterized by an optimal value of rewiring probability, warranting the largest peak value of the system response. Considering that two-frequency signals are ubiquity in brain dynamics, we expect the presented results could have important implications for signal processing in neuronal systems.

To investigate the different expression of epidermal growth factor receptor 1 (EGFR) and human epidermal growth factor receptor 2 (HER2) in gastric cancer based on tumor locations and its impact on patients survival.Gastric cancer is heterogeneous disease, recent years have established a molecular classification and described distribution of molecular subtypes in stomach. However, the difference of EGFR and HER-2 expression among tumor location is still unknown.Between January 2010 and August 2014, 2477 consecutive patients with gastric cancer were treated in our surgery department. The tumor locations were classified into 4 groups: cardia, fundus, corpus, and antrum. Based on tumor locations, the clinicopathologic characteristics, EGFR and HER-2 expression, and follow-up data were analyzed by univariant analysis and Kaplan-Meier analysis retrospectively.There were difference of gender, age, Borrmann type, pathological type, differentiation, T-stage, tumor size, gastrectomy method, and complications among the locations. The positive rate of EGFR expression in fundus was 18.18%, which was lower than cardia (46.21%), corpus (43.62%), and antrum (48.83%) (P < .001). The 5-year survival rate in EGFR positive patients was 50.8%, which was significantly lower than EGFR negative patients (64.0%, P = .021). The positive rate of HER-2 expression in cardia was 48.15%, which was significantly higher than fundus (37.5%), corpus (35.45%), and antrum (38.54%) (P = .009), but HER-2 expression did not correlate with 5-year survive (P = .548).Our results suggest that there exist difference of EGFR and HER-2 expression based on tumor locations, and the distribution of EGFR impact on patients survival. Emphasizing the role of EGFR and HER-2 in the context of location contribute to make appropriate treatment strategy and improve prognosis of gastric cancer.

We investigate the detectability of weak electric field in a noisy neural network based on Izhikevich neuron model systematically. The neural network is composed of excitatory and inhibitory neurons with similar ratio as that in the mammalian neocortex, and the axonal conduction delays between neurons are also considered. It is found that the noise intensity can modulate the detectability of weak electric field. Stochastic resonance (SR) phenomenon induced by white noise is observed when the weak electric field is added to the network. It is interesting that SR almost disappeared when the connections between neurons are cancelled, suggesting the amplification effects of the neural coupling on the synchronization of neuronal spiking. Furthermore, the network parameters, such as the connection probability, the synaptic coupling strength, the scale of neuron population and the neuron heterogeneity, can also affect the detectability of the weak electric field. Finally, the model sensitivity is studied in detail, and results show that the neural network model has an optimal region for the detectability of weak electric field signal.
