RESUMO

A new fermentation method for kiwi wine was explored by developing the well-known medicinal and edible plant Cyclocarya paliurus (C. paliurus) to create more value with undersized kiwifruits. In this study, the changes in bioactive substances during the C. paliurus-kiwi winemaking process were analyzed on the basis of response surface optimization results, and the antioxidant capacity, aromatic compounds, and sensory quality of the C. paliurus-kiwi composite wine with kiwi wine and two commercial kiwi wines were compared. The results showed that DPPH radical, OH- radical, and ABTS+ scavenging rates remained at over 60.0%, 90.0%, and 70.0% in C. paliurus-kiwi wine, respectively. The total flavonoid content (TFC) and total polyphenol content (TPC) of C. paliurus-kiwi wine were significantly higher than those of the other three kiwi wines. C. paliurus-kiwi wine received the highest score and detected 43 volatile compounds. Ethyl hexanoate, which showed stronger fruity and sweet aromas, was one of the main aroma components of C. paliurus-kiwi wine and different from commercial wines. This wine has a good flavor with a natural and quality feeling of C. paliurus-kiwifruit extract, low-cost processing, and great market potential.

BACKGROUND: Citrus 'Benimadonna' (Citrus reticulata × Citrus sinensis) is a high-value perishable fruit; thus there is an urgent need for a preservation technology with high effectiveness and low safety risk from industries. Nα-Lauroyl-l-arginine ethyl ester hydrochloride (LAE) was applied to enhance preservability by compounding with natamycin, and a possible fungal inhibition mechanism based on the hypothesis of an impact on the cell membrane by surfactant was investigated. RESULTS: In vitro testing showed that the minimum inhibitory concentration of LAE against Penicillium expansum (PE), isolated as the predominant spoilage-inducing fungus, was 32 mg L-1 and it was partially synergistic with natamycin. Subsequent in vivo testing proved the inhibition capacity. During 90 days' refrigerated preservation, spoilage rate was significantly decreased by preharvest spraying versus control without extra taste loss, and LAE showed an alleviating benefit on total pectin loss. Subsequently, electron microscopic imaging and intracellular protein levels of PE exposed to LAE indicated that LAE stress led to increased permeability and decreased cell integrity. Moreover, peroxidase, superoxide dismutase and catalase revealed that LAE enhanced oxidative stress, while pectinase was antagonized. CONCLUSION: The present investigation first introduced LAE as a candidate active ingredient for citrus preservative. A theoretical basis was provided for the development of preservation technology for high-value perishable fruit. According to the authors' knowledge this study is the first report on the inhibition mechanism of LAE in terms of oxidative stress. © 2022 Society of Chemical Industry.

In order to improve the function of insoluble dietary fiber (IDF) extracted from pickle, the coupled enzymatic hydrolysis and high hydrostatic pressure treatment method (EHHP) was used to modify its structure. Compared with the unmodified IDF (U-IDF), analysis of the particle size dispersion, bulk density, surface structure monosaccharide composition, microstructure, thermodynamic properties showed that the modified IDF (EHHP-IDF) has a looser and more porous structure, reduced particle size, bulk density, crystal strength and thermal stability, and increased xylose and galactose content. Due to the special looser microstructure, EHHP-IDF has showed the notable capacity of absorption of oil, glucose, nitrite, cholesterol as well as Pb2+. Collectively, these results show that EHHP has good potential use as an ideal modification method to improve the function of IDF, and a novel functional ingredient of EHHP-IDF which could be used in future food processing was obtained in this study.

Liangshan Prefecture is one of the three major forest areas in Sichuan Province and one of the three major disaster areas of forest fire. We measured the physicochemical properties and combustion performances of different organs (leaves and branches) of 15 main economic tree species in Liangshan, and analyzed the bioecology characteristics, silviculture characteristics and value characteristics of different tree species. We investigated the fire resistance of different tree species to screen out fire-resistant species suitable for economic forest development in Liangshan Prefecture, and improve the biological fire prevention ability. The seven physicochemical properties and combustion performances indices of 15 tree species showed significant differences. Except for crude ash and lignin, the weights of moisture content, caloric value, ignition point, crude fat, and crude fibre of leaves were higher than those of branches. Crude fibre index of leaves (9.6%) and the crude ash index of branches (9.9%) were the highest weight indices of the two organs, respectively. Based on the fire resistance, we divided all the species into three classes, i.e., class Ⅰ (excellent fire-resistance trees) Juglans regia and Morus alba; class Ⅱ (better fire-resistant trees) Sapium sebiferum, Mangifera indica, Phyllanthus emblica, Eriobotrya japonica, Ligustrum lucidum, Castanea mollissima, and Punica granatum; class Ⅲ (poor fire-resistant trees) Pinus armandii, Illicium simonsii, Morella rubra, Sapindus mukorossi, Olea europaea and Camellia oleifera. J. regia and M. alba had fireproof solid performance and could be used as the preferred species for fireproof economic forest in Liangshan region. It was suggested that to use class Ⅰ to Ⅱ fire-resistant tree species built the main fireproof isolated forest belt, and pay attention to fire prevention after planting class Ⅲ tree species in a large area.

Additive manufacture (AM) has been widely and rapidly applied in fabrication of 3D porous scaffolds for tissue engineering applications. For synthetic polymers of high melting temperature, the melting-extruding technique is the most applied AM method for such fabrication of polymer porous scaffolds. This results in a big challenge to directly process the scaffolds using the polymers and thermosensitive substances simultaneously because of deactivation under high temperature. In this article, the selective laser sintering (SLS) method was proposed to make a poly(l-lactic acid) (PLLA) porous scaffold containing dexamethasone (Dex) simultaneously. Dex was encapsulated in two groups of PLLA-bioactive glass (BG) composite microspheres with an average diameter of 115-120 µm and loading amounts of 0.68 ± 0.09 and 0.84 ± 0.10 µg/mg, respectively. The drug-loading composite microspheres were then fabricated into scaffolds under a laser fluence of 0.83-2.08 J/mm2. The average pore size and compressive modulus for the porous scaffold were 450-500 µm and 18-25 MPa, respectively. Drug release experiments showed that Dex was released from the scaffold in a controlled manner until about a month. The eluting time of HPLC tests before or after SLS processing both presented at 4 min indicated no chemical structure changes for the drug. Ex vivo cell experiments also testified the comparable effect of released Dex with commercial products, showing that the bioactivities were not affected after SLS. Implantation of the composite scaffolds in rat cranium defects demonstrated that new bone and blood vessel formation was faster in the Dex-releasing scaffolds than in the groups without drug loading.

BACKGROUND: The lack of a robust small animal model for hepatitis C virus (HCV) has hindered the development of novel drugs, including internal ribosome entry site (IRES) inhibitors. Phage phiC31 integrase has emerged as a potent tool for achieving long-term gene expression in vivo. This study utilized phiC31 integrase to develop a stable, reproducible and easily accessible HCV IRES mouse model. METHODS: phiC31 integrase plasmid and the reporter vector, HCV-IRES-luciferase expression cassette (containing an attB site), was codelivered to murine livers using high pressure tail vein injection. HCV IRES-dependent translation reflected by luciferase expression was accurately monitored in vivo by bioluminescence imaging. Genomic integration of the transgene was confirmed by partial hepatectomy and nested PCR. An HCV IRES-targeted short hairpin RNA (shRNA) expression plasmid, sh184, was hydrodynamically transfected into mouse liver to study its inhibition efficacy in vivo. RESULTS: phiC31 integrase mediated intramolecular recombination between wild-type attB and attP sites in mice. The expression of luciferase was stable after 30 days post-transfection and remained so for 300 days only in the livers of mice that were coinjected with the integrase-encoding plasmid. Luciferase levels reduced dramatically after hydrodynamic transfection of sh184. CONCLUSIONS: These results indicate that this mouse model provides a powerful tool for accurate and long-term evaluation of potential anti-IRES compounds in vivo.

To improve the efficiency of expression of reporter transgenes delivered by hydrodynamic injection, we generated expression cassettes carrying different liver-specific regulatory elements using the firefly luciferase gene as a reporter. From our studies the human alpha-antitrypsin promoter together with the apolipoprotein E/C-I and albumin enhancer was the combination of choice for prolonged transgene expression, but reporter gene expression in vivo lasted for no more than 7 weeks. Subsequently, phage phiC31 integrase was introduced as a potential tool to improve further the efficiency of expression of the delivered transgene. Long-term transgene expression in vivo was achieved by specific integration of the target gene into mouse livers. This study demonstrates the use of a combination of phage phiC31 integrase and liver-specific regulatory elements for generation of transgenic mice.

RESUMO

A non-invasive orthotopic hepatocellular carcinoma (HCC) model was created with human HCC cells (HepG-Luc) constitutively expressing luciferase (Luc) in nude mice. Development of tumor growth and response to anti-tumor therapy combined with 5-fluorouracil and cisplatin was monitored by whole-body bioluminescent imaging (BLI). Luciferase activity in the tumor, determined by BLI, correlated with the tumor volume and weight. The anti-tumor therapy proved effective by BLI monitoring. In conclusion, BLI by luciferase provides a non-invasive method of monitoring tumor activities that can prove useful for therapeutic intervention studies.

OBJECTIVE: To explore the vegetation landscapes in marshland snail habitats using satellite image. METHODS: The false-color composition image from band 2, 3 and 4 of LANDSAT ETM + images was classified in the ERDAS IMAGINE 8.5 to analyze the vegetation types in the marshland of Jiangning county, using the unsupervised classification. The efficiency of classification was evaluated by the transformed divergence. The overlaid layers of the classified vegetation image and the vector layer of snail habitats distribution were used to analyze the relationship between the snail distribution and the landscape types. RESULTS: The land-cover of marshland in LANDSAT ETM234 image in Jiangning county could be classified into 10 types, including water, bare soil, sandy and other landscapes while the transformed divergence analysis showed that there were misclassified pixes between some types especially for the non-continuous types. The study indicated that through adding the NDVI image in the process of classification efficiency of classification and eliminate misclassification in the non-continuous type could be improved. Analysis on the overlaid layer of the vector of snail distribution and the classified image proved that the vegetation covers in marshland snail habitats in Jiangning mainly belonged to type 3, 5 and 6, that responded to the beach with sparse vegetation, exuberant weed and bulrush respectively. The density of snails in the bulrush was higher than that in other 2 landscapes. CONCLUSION: The vegetation type in the marshland snail habitats could be distinguished from the satellite image, which was helpful for the surveillance of snail habitat in marshland and for the prevention of schistosomiasis.

OBJECTIVE: To analyze the relationship between the normalized difference vegetation index (NDVI) and the snail distribution in marshland of Jiangning county in Jiangsu province, and to explore the utility of Terra-MODIS image map in the small scale snail habitats surveillance. METHODS: NDVI were extracted from MODIS image by vector chart of the snail distribution using ArcView 8.1 and ERDAS 8.5 software. The relationship between NDVI and the snail distribution were Investigated using Bivariate correlations and stepwise linear regression. RESULTS: The snail density on marshland was positively correlated with the mean NDVI in the first ten-day of May and the maximum NDVI (N(20max)) in the last ten-day of May. Incidence of pixel with the live snail on marshland was positively correlated with the mean NDVI (N(2mean)) in the first ten-day of May. An equation Y(1) = 0.009 47 x N(20max) (R(2) = 0.73), Y(2) = 0.018 6 x N(2mean) (R(2) = 0.906) was established. CONCLUSION: This study showed that the Terra-MODIS satellite images reflecting the status of the vegetation on marshland in Jiangning county could be applied to the study to supervise the snail habitat. The results suggested that MODIS images could be used to survey the small scale snail habitats on marshland.
