Silver eel samples, collected from the lagoons of Fogliano and Caprolace (Italy), were investigated for a broad range of contaminants (29 polychlorinated biphenyls, 9 polybrominated diphenyl ethers, 5 dichlorodiphenyltrichloroethane, 5 chlordanes, hexachlorobenzene, 3 hexachlorocyclohexane, and 5 metals). Concentrations of targeted compounds stand for a general low contamination pattern. Infestation by Anguillicola crassus and virus infections were also examined. No parasite infestation was found, while infected silver eels had a low prevalence for EVEX, and, for the first time in the Mediterranean area, for AngHV-1. Overall, a good quality status of escaping silver eels, for both lagoons, was highlighted by the use of integrative Indexes. A quality assessment of the ecological status of the two lagoons was carried out developing an expert judgment approach, in order to characterize the habitat of eel stocks. A Final Pressure Index was derived, whose values showed an overall limited global anthropogenic impact acting on both lagoons. Results stand for the suitability of an integrated approach to assess lagoon habitats and eel local stocks quality. This could be proposed as a tool to identify sites yielding high quality eel spawners in the Mediterranean region, in order to set up suitable management frameworks, providing elements to appraise and discuss the potential of coastal lagoons in the Mediterranean region towards the recovery of the eel global stock.

Karyotype and other chromosomal characteristics of the Appenine endemic cyprinid fish, Toscana stream chub Squalius lucumonis, were analysed using conventional banding and FISH with 45S and 5S rDNA probes. The diploid chromosome number (2n = 50) and karyotype characteristics including pericentromeric heterochromatic blocks and GC-rich CMA(3)-positive sites corresponding to both positive Ag-NORs and 45S rDNA loci on the short arms of a single medium-sized submetacentric chromosome pair were consistent with those found in most European leuciscine cyprinids. On other hand, 5S rDNA FISH in the Toscana stream chub and three other Italian leuciscines, S. squalus, Rutilus rubilio and Telestes muticellus, revealed a species-specific hybridization pattern, i.e. signals on four (S. lucumonis), three (S. squalus and R. rubilio) and two (T. muticellus) chromosome pairs. Whereas all the species shared the 5S rDNA loci on the largest subtelocentric chromosome pair, a "leuciscine" cytotaxonomic marker, S. lucumonis showed both classes of rDNA loci tandem aligned on the short arms of chromosome pair No. 12. The present findings suggest that the observed high variability of 5S rDNA loci provides a powerful tool for investigation of karyotype differentiation in karyologically conservative leuciscine fishes.

Italy hosts a large number of endemic freshwater fish species due to complex geological events which promoted genetic differentiation and allopatric speciation. Among them, the South European roach Sarmarutilus rubilio inhabits various freshwater environments in three different ichthyogeographic districts. We investigated the genetic diversity of S. rubilio using two different mitochondrial markers (COI and CR), aiming to define its relationship with other similar taxa from the Balkan area and, from a phylogeographic perspective, test the effects of past hydrogeological dynamics of Italian river basins on its genetic structure and demographic history. Our analysis highlighted a marked genetic divergence between S. rubilio and all other roach species and, among Italian samples, revealed the existence of three deeply divergent geographic haplogroups, named A, B and C. Haplogroup C likely corresponds to a new putative cryptic species and is located at the northern border of the South European roach range; haplogroup B is restricted to Southern Italy; and haplogroup A is widespread across the entire range and in some sites it is in co-occurrence with C or B. Their origin is probably related to the tectonic uplifting of the Apuan Alps in the north and of the Colli Albani Volcano in the south during the Pleistocene, which promoted isolation and vicariance followed by secondary contacts.

Globalization and urbanization are new challenges for the ability to protect public health. Indeed, the anthropogenic impact is changing the environment on a global scale. These changes can have direct and indirect health effects on both human and animal populations, introducing new diseases. Heat waves and floods are an example of these changes. Global Environmental Change (GEC) consequences on human health and well-being are stronger in urban areas, which are inhabited by 70% of the European population. In this context, the use of appropriate medical devices can also help mitigate the effects of climate change. Studies into lifestyle, environment quality and potential fields of application can be useful tools to identify possible types of medical device that could help to support the therapeutic needs and the prevention of health both in everyday life, and in the case of environmental alerts. A study was carried out on the potential role of medical devices (MDs) in mitigating the effects of GEC on human and animal health, by issuing two different questionnaires to specific professional clusters: the first to doctors, pharmacists, and veterinarians, the second to MD manufacturers. The data obtained from this study confirm the strong connection between GEC and the increase in the use of some MDs. Results obtained from questionnaires circulated to MD manufacturers confirmed this trend. MD manufacturers also declared that there are no longer any seasonal trends in market demand for some medical devices. This is a pilot study to consider MDs as a mitigation tool for CEGs.

The extraordinary polymorphism of major histocompatibility complex (MHC) genes is considered a paradigm of pathogen-mediated balancing selection, although empirical evidence is still scarce. Furthermore, the relative contribution of balancing selection to shape MHC population structure and diversity, compared to that of neutral forces, as well as its interaction with other evolutionary processes such as hybridization, remains largely unclear. To investigate these issues, we analyzed adaptive (MHC-DAB gene) and neutral (11 microsatellite loci) variation in 156 brown trout (Salmo trutta complex) from six wild populations in central Italy exposed to introgression from domestic hatchery lineages (assessed with the LDH gene). MHC diversity and structuring correlated with those at microsatellites, indicating the substantial role of neutral forces. However, individuals carrying locally rare MHC alleles/supertypes were in better body condition (a proxy of individual fitness/parasite load) regardless of the zygosity status and degree of sequence dissimilarity of MHC, hence supporting balancing selection under rare allele advantage, but not heterozygote advantage or divergent allele advantage. The association between specific MHC supertypes and body condition confirmed in part this finding. Across populations, MHC allelic richness increased with increasing admixture between native and domestic lineages, indicating introgression as a source of MHC variation. Furthermore, introgression across populations appeared more pronounced for MHC than microsatellites, possibly because initially rare MHC variants are expected to introgress more readily under rare allele advantage. Providing evidence for the complex interplay among neutral evolutionary forces, balancing selection, and human-mediated introgression in shaping the pattern of MHC (functional) variation, our findings contribute to a deeper understanding of the evolution of MHC genes in wild populations exposed to anthropogenic disturbance.

Sensitivity analysis applied to Artificial Neural Networks (ANNs) as well as to other types of empirical ecological models allows assessing the importance of environmental predictive variables in affecting species distribution or other target variables. However, approaches that only consider values of the environmental variables that are likely to be observed in real-world conditions, given the underlying ecological relationships with other variables, have not yet been proposed. Here, a constrained sensitivity analysis procedure is presented, which evaluates the importance of the environmental variables considering only their plausible changes, thereby exploring only ecological meaningful scenarios. To demonstrate the procedure, we applied it to an ANN model predicting fish species richness, as identifying relationships between environmental variables and fish species occurrence in river ecosystems is a recurring topic in freshwater ecology. Results showed that several environmental variables played a less relevant role in driving the model output when that sensitivity analysis allowed them to vary only within an ecologically meaningful range of values, i.e. avoiding values that the model would never handle in its practical applications. By comparing percent changes in MSE between constrained and unconstrained sensitivity analysis, the relative importance of environmental variables was found to be different, with habitat descriptors and urbanization factors that played a more relevant role according to the constrained procedure. The ecologically constrained procedure can be applied to any sensitivity analysis method for ANNs, but obviously it can also be applied to other types of empirical ecological models.

Species distribution is the result of complex interactions that involve environmental parameters as well as biotic factors. However, methodological approaches that consider the use of biotic variables during the prediction process are still largely lacking. Here, a cascaded Artificial Neural Networks (ANN) approach is proposed in order to increase the accuracy of fish species occurrence estimates and a case study for Leucos aula in NE Italy is presented as a demonstration case. Potentially useful biotic information (i.e. occurrence of other species) was selected by means of tetrachoric correlation analysis and on the basis of the improvements it allowed to obtain relative to models based on environmental variables only. The prediction accuracy of the L. aula model based on environmental variables only was improved by the addition of occurrence data for A. arborella and S. erythrophthalmus. While biotic information was needed to train the ANNs, the final cascaded ANN model was able to predict L. aula better than a conventional ANN using environmental variables only as inputs. Results highlighted that biotic information provided by occurrence estimates for non-target species whose distribution can be more easily and accurately modeled may play a very useful role, providing additional predictive variables to target species distribution models.

The use of chlorinated disinfectants during drinking-water production has been shown to generate halogenated compounds as a result of interactions of humic acids with chlorine. Such chlorinated by-products have been shown to induce genotoxic effects and consumption of chlorinated drinking-water has been correlated with increased risk for cancer induction in human populations. The aim of this work was to test the potential genotoxic effects on circulating erythrocytes of the fish Cyprinus carpio exposed in vivo to well-waters disinfected with sodium hypochlorite (NaClO), chlorine dioxide (ClO2) or peracetic acid (CH3COO2H, PAA), in the absence or presence of standard humic acids (HA). The effects were measured by use of the micronucleus (MN) and the single-cell gel electrophoresis (Comet) assays at different sampling times after a 3-day exposure period. The exposure to chlorine disinfectants without the addition of HA produced a clear toxic effect. Significant cytogenetic damage (i.e. MN induction) was detected in fish populations exposed to both NaClO and ClO2 with humic acids. In the Comet assay, a significant decrease of DNA migration was observed in erythrocytes of specimens after exposure to NaClO-disinfected water without HA. No effects were observed in any other experimental condition.

RESUMO

A short overview of fish-based assessment methods for aquatic systems is presented. Multimetric indices, as, e.g., the index of biotic integrity (IBI), firstly developed in USA and later adapted for European river basins and other countries, are shortly described. Non-multimetric indices are also discussed, e.g. the ichthyological index (II) and the index of ecological status of fish communities (ISECI), both proposed for monitoring Italian rivers. Moreover, statistical and predicting methods based on machine learning techniques are described. Finally, a new approach for developing standardised fish-based methods useful to assess ecological status of Italian rivers is proposed. Although rather complex, the use of bony fish in biomonitoring is promising and requires a multidisciplinary approach to be adopted.

The aim of this study was to evaluate the use of gonadal alterations in the thinlip grey mullet (Liza ramada) as a biological indicator in assessing aquatic ecosystems health, with particular emphasis to river ecosystems exposed to sewage discharges. For this purpose, the reproductive status and the presence of gonadal alterations were studied in 206 mullets collected from two sites on the low course of the Tiber River, downstream of a large urban sewage treatment plant and in the estuarine area, and from an uncontaminated pond considered as reference site. Intersex and irregularly shaped gonads were observed in 20.8% of the mullets from the most polluted site, and intersex gonads in 10.3% of those from the estuarine area. No alterations were detected in the fish from the reference site, which also showed distinct stages of gonadal development. Conversely, unclear stages of testicular and ovary development were observed in the fish from the two polluted river sites. The results of this study suggest that L. ramada may represent a sentinel species in environmental risk assessment and support the use of gonadal alterations of this species as a bioindicator for extensive monitoring of pollution in lower stretches of rivers and estuarine areas.

Leuciscine fishes represent an important component of freshwater ichthyofauna endemic to northern Mediterranean areas. This lineage shows high intra-specific morphological variability and exhibits high levels of hybridization, two characteristics that contribute to systematic uncertainties, misclassification of taxa and, potentially, the mismanagement of biodiversity. This study focused on brook chub, Squalius lucumonis, an endemic taxon of Central Italy. The taxonomic status of this species has long been questioned, and a hybrid origin from sympatric leusciscines (S. squalus x Rutilus rubilio, or S. squalus x Telestes muticellus) has been hypothesised. A phenotypic (evaluating shape and meristic counts) and genetic (using mitochondrial and nuclear markers) investigation of these four taxa was conducted to test species delimitation in sympatric areas and to evaluate the taxonomic status of S. lucumonis. One hundred and forty-five individuals of all four taxa were collected within streams of the lowest portion of the Tiber River basin and analysed; this region encompasses a large portion of the S. lucumonis distribution. The different morphological and genetic approaches were individually examined, compared, and then combined in a quantitative model to both investigate the limits of each approach and to identify cases of misclassification. The results obtained confirm the cladogenetic non-hybrid origin of S. lucumonis, highlight the need for immediate conservation actions and emphasise the value of an integrated approach in the study of leuciscines evolution.

Although anthropogenic degradation of riverine systems stimulated a multi-taxon bioassessment of their ecological integrity in EU countries, specific responses of different taxonomic groups to human pressure are poorly investigated in Mediterranean rivers. Here, we assess if richness and composition of macroinvertebrate and fish assemblages show concordant variation along a gradient of anthropogenic pressure in 31 reaches across 13 wadeable streams in central Italy. Fish and invertebrate taxonomic richness was not correlated across sites. However, Mantel test showed that the two groups were significantly, albeit weakly, correlated even after statistically controlling for the effect of environmental variables and site proximity. Variance partitioning with partial Canonical Correspondence Analysis showed that the assemblages of the two groups were influenced by different set of environmental drivers: invertebrates were influenced by water organic content, channel and substratum features, while fish were related to stream temperature (mirroring elevation) and local land-use. Variance partitioning revealed the importance of biotic interactions between the two groups as a possible mechanisms determining concordance. Although significant, the congruence between the groups was weak, indicating that they should not be used as surrogate of each other for environmental assessments in these Mediterranean catchments. Indeed, both richness and patterns in nestedness (i.e. where depauperate locations host only a subset of taxa found in richer locations) appeared influenced by different environmental drivers suggesting that the observed concordance did not result from a co-loss of taxa along similar environmental gradients. As fish and macroinvertebrates appeared sensitive to different environmental factors, we argue that monitoring programmes should consider a multi-assemblage assessment, as also required by the Water Framework Directive.

Trout farming, that represents the most important sector for aquaculture inland production in Italy, can cause negative effects on aquatic ecosystems. Recently, in the framework of Water Frame Directive 2000/60/EC and national law DL 152/2006, concerning the sustainable uses of water resources, multi-criteria approaches have been suggested to evaluate the impact of fish farming on aquatic ecosystems. In this study trout farms of central Italy were selected to investigate the effects of their effluents, on receiving water bodies using a multi-criteria approach based on physico-chemical parameters, microbiological and macrobenthonic indicators, detected in sampling stations located upstream/downstream the trout farm. Moreover, antibiotic susceptibility against antibiotics allowed and/or forbidden by current law (D.lgs 193/56/06) was tested on E. coli strains. The results indicate variations of chemical parameters and biological indicators from upstream to downstream sites in some of the investigated farms. Antibiotic resistance of E. coli strains suggested a large use of tetracycline and a possible past use of chloramphenicol. This study represents a first contribute to the knowledge of fish farm impacts on aquatic systems in Central Italy.
