ABSRACT1. An experiment was designed to compare the effects of supplementing laying hen diets with phytase and myo-inositol (inositol).2. Five diets were formulated: high balanced protein (HBP - 840 mg of Dlys/hen/day), HBP with inositol (HBP+I - 0.16%), reduced balance protein (RBP - 672 mg of Dlys/hen/day), RBP with inositol (RBP+I - 0.16%) and RBP with phytase (RBP+P - 3000 FTU/kg).3. Laying hen production, inositol concentrations, digestive tract morphology, amino acid digestibility and intestinal inositol transporters transcript abundance were evaluated. Data were analysed with a one-way ANOVA in SAS 9.4. Contrasts were used to assess the effect of protein, inositol, phytase and phytase vs. inositol. Differences were accepted when P ≤ 0.05.4. No effect on hen-day egg production or feed efficiency was found. However, feed intake and the incidence of abnormally shaped eggs were 0.77 g/h/d and 0.17% higher, respectively, in inositol treatments. Inositol decreased egg specific gravity from 1.088 to 1.0865.5. Inositol concentration in egg yolk was similar among HBP+I, RBP+I and RBP+P, and higher than for the HBP and RBP diet groups. Both gizzard and ileal digesta were enriched in inositol in all supplemented treatments, and phytase supplementation decreased the level of IP5 and IP6 in the gizzard and ileum. Generally, neither phytase or inositol affected amino acid digestibility.6. Inositol increased transcript abundance of alkaline phosphatase in the ileum, while phytase upregulated duodenal alkaline phosphatase and SMIT1, jejunal SMIT2 and reduced ileal HMIT and SMIT1 abundance.7. In conclusion, no effect of phytase or inositol was found for laying hen production performance or amino acid digestibility, but egg quality was reduced by inositol supplementation. Inositol concentration in egg yolk was similar among supplemented treatments.

There is little consensus as to the capability of poultry to utilize dietary phytate without supplemental phytase. Therefore, an experiment was conducted to examine the extent to which endogenous phytase of intestinal and cereal origin contributes to phytate degradation in birds aged 0 to 14 d posthatch. Ross 308 broilers (n = 720) were fed one of 4 experimental diets with differing dietary ingredient combinations and approximate total phytate levels of 10 g/kg, dietary phytase activity analyzed at 460 U/kg, dietary calcium (Ca) levels of 11 g/kg, and nonphytate-phosphorus (P) levels of 4 g/kg. Broiler performance, gizzard, duodenum, jejunum and ileum pH, Ca and P digestibility and solubility, amount of dietary phytate hydrolyzed in the gizzard, jejunum, and ileal digesta phytase activity were analyzed at d 4, 6, 8, 10, 12, and 14 posthatch. Intestinal endogenous phytase activity increased significantly (P < 0.001) between d 4 and 6, resulting in increased phytate hydrolysis in the gizzard (P = 0.003), jejunum (P < 0.001), and ileum (P < 0.001). Phytase activity and phytate hydrolysis continued to increase with age, with a greater phytase activity and associated increase in phytate hydrolysis and mineral utilization between d 10 and 12. Gizzard and jejunum Ca and P solubility and ileal Ca and P digestibility increased significantly (P < 0.001), and gastrointestinal pH decreased significantly (P < 0.001) between d 4 and 6. By d 14, phytase activity recovered in the ileum was approximately 45 U/kg. There were strong correlations between phytase activity measured in the ileum and phytate hydrolyzed in the gizzard (r = 0.905, P < 0.001), jejunum (r = 0.901, P = 0.023), and ileum (r = 0.938, P = 0.042). This study shows intestinal- and dietary-derived endogenous phytase activity is responsible for phytate-P hydrolysis in broilers.

An experiment was conducted to evaluate the influence of a novel microbial phytase on performance, tibia ash, and the content of phytate, phytate esters, and inositol in the gizzard of young broilers. Male Cobb 500 broilers (n = 1,680) were fed 1 of 7 experimental diets: positive control (PC) formulated to meet or exceed nutrient recommendations; PC plus dicalcium phosphate (PC+DCP) formulated to provide Ca and P at 0.10% above the PC; PC plus 500 U/kg of microbial phytase (PC+500); negative control (NC) with Ca and P reduced from the PC by 0.16% and 0.15%, respectively; and the NC plus phytase at 500 (NC+500), 1,000 (NC+1,000), or 1,500 (NC+1,500) U/kg. Diets were fed in crumbled form to 20 birds/pen and 12 replicate pens/diet from d 0 to 21. On d 21, 4 birds/pen were euthanized for collection of right tibias and gizzard digesta for determination of tibia ash and gizzard phytate. In general, broilers fed the NC diet had reduced (P ≤ 0.05) feed intake and BW gain compared with broilers fed diets supplemented with phytase, but not different than the PC or PC+DCP. Phytase supplementation in the NC diet improved (P ≤ 0.05) BW gain comparable with or above that of the PC. Feed conversion ratio was improved in broilers fed the NC+1,000 or NC+1,500 compared with broilers fed all other diets. Tibia ash was reduced (P ≤ 0.05) in broilers fed the NC compared with broilers fed all other diets, and phytase supplementation improved tibia ash comparable with the PC. Phytase supplementation reduced (P ≤ 0.05) phytate (inositol hexa-phosphate) concentration in the gizzard. Inositol concentration in the gizzard was higher (P ≤ 0.05) in birds fed NC+1,000 or NC+1,500 compared with all other diets and this was correlated with growth performance (P ≤ 0.05) rather than tibia ash (P > 0.05). Improvements in feed conversion ratio associated with superdoses of phytase may be attributed to phytate destruction and the provision of inositol.

There is little consensus as to the most appropriate methodology for the measurement of gastrointestinal pH in chickens. An experiment was conducted to establish the optimum sampling method for the determination of broiler digesta pH in birds fed differing levels of dietary calcium. Ross 308 broilers (n = 60) were fed 1 of 2 experimental diets, one containing 0.8% monocalcium phosphate and 2% limestone and one containing 0.4% monocalcium phosphate and 1% limestone. Four factors were investigated to determine the most appropriate method of measuring broiler gastrointestinal digesta pH: removal from the tract, prolonged air exposure, altering the temperature of the assay, and controlling the water content of the digesta. The conditions were assessed at bird ages from 7 to 42 d posthatch. Dietary Ca content had no significant effect on in situ pH, but it contributed toward variance in ex situ pH of both gizzard and duodenum digesta. Digesta pH read higher when the digesta was removed from the tract, but the amount of time the digesta was exposed to air did not affect the reading. Digesta pH read higher when measured at room temperature than when measured at 41°C; temperature made the strongest unique contribution to explaining variance in duodenum pH, and the second strongest contribution to explaining variance in gizzard pH, after diet. When water was added to the digesta, before pH determination, the pH of the digesta read higher (P < 0.001) than when measured in situ. The method that resulted in pH readings that were most representative of bird gastrointestinal environment was insertion of a pH probe directly into the gut lumen posteuthanasia, because measurement ex situ likely encourages dissociation of carbonic acid, the major buffer in the gastrointestinal tract, which causes pH to read to be higher than when measured in situ. This study shows that the method of pH measurement needs careful consideration to ensure the validity of the result.

1. In vitro assays provide a rapid and economical tool to evaluate dietary effects, but have limitations. In this study, the effect of phytase supplementation on solubility, and presumed availability, of calcium (Ca) and phosphorus (P) in soya bean meal (SBM) and rapeseed meal (RSM) based diets were evaluated both in situ and by a two-step in vitro digestion assay that simulated the gastric and small intestine (SI) phases of digestion. 2. Comparison of the in vitro findings to in situ findings was used to evaluate the in vitro assay. Ross 308 broilers (n = 192) were fed on one of 6 SBM or RSM diets supplemented with 0, 500 or 5000 FTU/kg phytase from 0 to 28 d post hatch. The 6 diets and raw SBM and RSM were exposed to a two-step in vitro assay. Ca and P solubility and pH in the gizzard and jejunal digesta and in the gastric and SI phase of in vitro digestion were measured. 3. Both in vitro and in situ analyses detected that Ca solubility was lowest when diets were supplemented with 500 FTU/kg phytase, compared to the control diets and diets supplemented with 5000 FTU/kg phytase. Phosphorus solubility increased with increasing phytase level. Both methods also identified that mineral solubility plateaus in the gastric phase. 4. Overall relationship of the two methods was strong for both determination of gastric phase Ca and P solubility (r = 0.96 and 0.92, respectively) and also SI phase Ca and P solubility (r = 0.71 and 0.82, respectively). However, mineral solubility and pH were higher when measured in vitro than in situ, and the in situ assay identified an interaction among the effects of phase, protein source and phytase inclusion level on Ca solubility that the in vitro assay did not detect. 5. This two-step in vitro assay successfully predicted phytase efficacy, but to determine detailed response effects in the animal, in situ data is still required.

The current study aimed to quantify the digestible calcium (Ca) equivalency of a new phytase (HiPhorius) in broiler chickens. A total of 1,152 male Cobb 500 broiler chickens were used in an experiment in which 8 diets consisting of graded levels of Ca supplied with limestone or graded levels of the phytase were fed. The 8 dietary treatments were based on a corn-soybean meal diet containing 5.1 g/kg of Ca and 5.1 g/kg of phosphorus (P) as negative control (NC); the NC + 1.3, 2.6, or 3.9 g/kg of Ca from limestone; and the NC + 500, 1,000, 2,000, or 4,000 FYT/kg of phytase. Birds were fed the experimental diets for 3 d (from d 7 to 10) or 14 d (from d 7 to 21) to determine apparent ileal digestibility (AID) and apparent total tract retention (ATTR) of dry matter (DM), CP, Ca, and P. In the 10-day-old birds, increasing the levels of Ca decreased the AID of P and Ca (linear, P < 0.05). Increasing the levels of phytase quadratically improved (P < 0.05) the AID of Ca and P. The AID of DM and CP in the younger birds quadratically increased (P < 0.05) as the levels of Ca increased. There were linear and quadratic effects (P < 0.05) of increasing phytase level for the AID of DM and a linear effect (P < 0.05) for the AID of CP. In the 21-day-old birds, increasing Ca levels resulted in a linear decrease (P < 0.05) in the AID of CP and P, and a quadratic decrease (P < 0.05) in the ATTR of CP. Increasing phytase levels linearly and quadratically (P < 0.05) improved the AID and ATTR of CP, P, and the ATTR of Ca. The analyzed phytase activity in the diets supplemented with phytase ranged from 1,520 to 7,661 FYT/kg. The digestible Ca equivalence for dietary phytase at 1,520 to 7,661 FYT/kg ranged from 1.55 to 2.02 g/kg in the 10-day-old birds fed for 3 d and from 0.52 to 0.64 g/kg in 21-day-old birds fed for 14 d. The results showed the reduction in Ca level that could be accommodated by phytase supplementation, which is markedly different between the younger and older birds. Feeding duration influenced the impact of phytase supplementation on Ca and P digestibility, with better efficacy of phytase observed in the 10-day-old birds fed for 3 d. Also, the results showed the extra-phosphoric effects of phytase on the utilization of other essential nutrients such as protein and Ca.

The objectives of this study were to determine the ileal digestibility of P in potassium phosphate, phytase-related ileal digestible P release, bone-mineralization-based ileal digestible P equivalency of phytase, and phytase-related efficiency of ileal digestible P utilization for bone mineralization in broiler chickens at 2 dietary concentrations of highly soluble Ca (HSC). Birds were sorted by BW at d 15 posthatch and assigned to 8 cages per diet with 8 birds per cage. Twelve diets were arranged in a 2 × 6 factorial of HSC at 5 or 6 g/kg and P supply treatment at 6 levels consisting of 4 added P levels (P from KH2PO4 added at 0, 0.7, 1.4, or 2.1 g/kg of diet) or 2 added phytase levels (500 or 1,000 phytase units). On d 24 posthatch, ileal digesta were collected for ileal P digestibility (IPD) determination and the left tibia was collected from the 4 heaviest birds in each cage for bone ash determination. Weight gain, G:F, and tibia ash were higher (P < 0.05) at 5 than at 6 g of HSC/kg. Added P from KH2PO4 or added phytase linearly increased (P < 0.001) weight gain, G:F, tibia ash, and IPD. The IPD of KH2PO4 derived from multiple linear regressions of digestible on total P intake for the diets without added phytase showed a reduction (P < 0.05) from 89.5 to 84.5% with increased HSC from 5 to 6 g/kg. Polynomial regressions of digestible P intake on phytase intake indicated that 1,000 units of added phytase released 1.701 or 1.561 g of digestible P in diets containing 5 or 6 g of added HSC/kg, respectively. Polynomial regressions of tibia ash on digestible P or phytase intake in diets containing 5 or 6 g of added HSC/kg at 1,000 phytase units gave digestible P equivalency of 1.487 or 1.448 g, respectively. Thus, phytase-related efficiency of ileal digestible P utilization for bone mineralization was 87.4 and 92.8% in diets containing 5 or 6 g of added HSC/kg, respectively.

The objective of this study was to determine the influence of Ca source [highly soluble calcified seaweed (HSC) or limestone], phytase supplementation, and dietary levels of Ca on bird performance and mineral digestibility (Ca and P) during a necrotic enteritis (NE) episode. Cobb 500 male broilers were weighed and randomized into 8 treatment groups (9 pens/treatment; 30 birds/pen) at day of hatch. The 21-d trial was designed as a 2 × 2 × 2 factorial, which included 2 dietary levels of Ca (0.6 and 0.9%), 2 Ca sources (limestone or HSC), and 2 levels of an Escherichia coli phytase (0 or 1,000 FTU/kg). One unit of phytase (FTU) is defined as the quantity of enzyme that releases 1 µmol of inorganic phosphorus/min from 0.00015 mol/L of sodium phytate at pH 5.5 at 37° C. Birds were placed on used litter from a previous flock that exhibited clinical signs of NE. Birds and feed were weighed on d 7, 14, and 21, and BW gain, feed intake, and feed conversion were calculated for each of these periods and cumulatively. Mortality was recorded daily and pH of the gizzard and duodenum were measured on d 7, 14, and 21. Ileal digesta (8 birds/pen) was collected on d 7, 14, and 21. Significance is reported at P < 0.05. Birds began exhibiting clinical signs of NE on d 9, and elevated NE-associated mortality persisted until the end of the trial. Significantly higher mortality was observed when broilers were fed diets with 0.9% Ca from HSC compared with birds fed diets with 0.6% Ca, regardless of Ca source. Broilers fed 0.6% Ca diets supplemented with phytase were heavier than the other treatments regardless of Ca source. Broilers fed diets formulated with HSC had significantly higher feed conversion then broilers fed diets formulated with limestone. The gizzard of broilers fed 0.9% Ca in the diet was significantly less acidic than the gizzard of broilers fed 0.6% Ca in the diet. Broilers fed 0.6% Ca in diets supplemented with phytase showed significant improvements in P and Ca digestibility. In conclusion, higher dietary Ca (0.9% vs. 0.6%) had a negative effect on mortality associated with NE and on bird performance.

An experiment was conducted to evaluate the influence of a novel microbial phytase on broiler performance from d 0 to 42 and tibia ash at d 21. Male Cobb 500 broilers (n = 2,016) were fed 1 of 7 experimental diets: positive control (PC) formulated to meet or exceed nutrient recommendations; PC plus dicalcium phosphate (PC+DCP) formulated to provide Ca and P at 0.10% above the PC; PC plus 500 U/kg of microbial phytase (PC+500); negative control (NC) with Ca and P reduced from the PC by 0.16 and 0.15%, respectively; and NC plus 500 (NC+500), 1,000 (NC+1,000), or 1,500 (NC+1,500) U/kg of microbial phytase. Diets were fed in 3 phases from d 0 to 21, d 22 to 42, and d 43 to 49 to 32 birds/pen and 9 replicate pens/diet. From d 0 to 21, broilers fed the NC diet had decreased (P ≤ 0.05) BW gain and tibia ash compared with broilers fed all other diets, except tibia ash in birds fed PC+500. Phytase supplementation at 500, 1,000, or 1,500 U/kg to the NC improved (P ≤ 0.05) BW gain and tibia ash comparable with the PC. Feed conversion ratio (FCR) was improved (P ≤ 0.05) in broilers fed NC+1,500 compared with broilers fed all other diets. From d 0 to 49, growth performance was not influenced (P > 0.05) by diet. However, FCR was improved (P ≤ 0.05) in broilers fed 1,500 U/kg of microbial phytase compared with broilers fed the PC, PC+DCP, and NC. There were no differences in performance or tibia ash between broilers fed the PC or PC+DCP, which would indicate the PC diet was sufficient in Ca and P. Therefore, the improvements in FCR in the NC+1500 may be associated with mitigation of the antinutrient effects of phytate rather than improved P utilization.

1. In a 42-d feeding trial, 264 one-d-old, as hatched, Cobb 400 broiler chickens (6 pens per group, n = 11 per pen in a 2 × 2 factorial arrangement) were fed on two concentrations of dietary calcium (Ca) (9.0 and 7.5 g/kg in starter, 7.5 and 6 g/kg in grower phases) and supplemental phytase (0 and 500 U/kg diet). 2. During d 0-21, the high Ca + phytase diet improved body weight. During d 0-42, feed intake was increased by the low Ca diet and decreased by phytase supplementation. Feed conversion ratio during d 0-21 was improved by the high Ca + phytase diet. 3. At d 42, Ca in duodenal digesta was reduced by low dietary Ca and supplemental phytase. High dietary Ca reduced P in duodenal and jejunal digesta. Phytase reduced digesta P and increased serum P concentration. 4. Relative tibia length decreased with low dietary Ca and increased with phytase. The robusticity index of tibia was improved by the low Ca diet and phytase supplementation. Phytase supplementation increased tibia ash and concentrations of Ca, magnesium (Mg), manganese (Mn), copper (Cu), zinc (Zn) and iron (Fe) in tibia. The low Ca diet increased Mg, Mn and Fe and reduced Cu and Zn in tibia. 5. It was concluded that 7.5 g Ca/kg during weeks 0-3 and 6 g Ca/kg during weeks 3-6 sustained broiler performance and bone ash, while phytase supplementation facilitated tibia mineralisation, particularly during the grower phase.

Phytate is not only an unavailable source of phosphorus (P) for broilers but it also acts as an anti-nutrient, reducing protein and mineral absorption, increasing endogenous losses and reducing broiler performance. The objective of this study was to investigate the anti-nutritional effects of phytate by including high levels of phytase in diets not severely limited in available P. A total of 768 male Arbor Acres broilers were distributed in six treatments of eight replicate pens of 16 birds each consisting of a positive control diet (PC), positive control with 500 FTU/kg phytase, negative control (NC) diet with lower available P and calcium (Ca) levels and the same NC diet with 500, 1,000 or 1,500 FTU/kg phytase. Body weight gain (BWG), feed intake (FI), feed conversion ratio (FCR) and mortality were determined at 21 and 35 d of age while foot ash was determined in four birds per pen at 21 d of age. FI, FCR and foot ash where not affected by the lower mineral diets at 21 d of age nor by the enzyme inclusion but broilers fed lower Ca and available P diets had lower BWG. At 35 d of age no difference was observed between broilers fed the positive or NC diets but broilers fed 500, 1,000 and 1,500 FTU/kg on top of the NC diet had better FCR than broilers fed the positive control diet. When compared to birds fed a diet adequate in P, birds fed the same diet included with 500, 1,000 and 1,500 FTU/kg of phytase in marginally deficient available P and Ca diets had an improvement of performance. These results support the concept that hydrolysing phytate and reducing the anti-nutritional effects of phytate improves bird performance on marginally deficient diets that were not covering the P requirement of birds.

Muramidase is an enzyme that hydrolyzes peptidoglycans of bacterial cell walls and improves performance of broilers in a dose-dependent manner. An experiment was conducted to evaluate muramidase supplementation, at a high or step-down dose, in turkeys from hatch to market. Male, B.U.T. 6 turkey poults were placed in 24 floor pens at 32 birds per pen. Poults were fed 1 of 3 diets from d 1 to 126 of age. There were 8 replicate pens per treatment. The treatments were a control (CTL) diet, the CTL plus muramidase at 45,000 LSU(F)/kg from phase 1 to 6 (BAL45), and the CTL plus muramidase at 45,000 LSU(F)/kg from phase 1 to 3 and decreased to 25,000 LSU(F)/kg from phase 4 to 6 (BAL45-25). Data were analyzed using SAS. The model included treatment and block and means were separated by Fisher LSD test. Birds fed BAL45 were heavier (P < 0.05) and had a greater (P < 0.05) average daily gain compared with birds fed the CTL from hatch to d 126 of age. Birds fed BAL45-25 had a final BW and average daily gain intermediate to or equivalent to birds fed BAL45 at the same phases. Feed conversion ratio was improved (P < 0.05) in birds fed BAL45 compared with birds fed the CTL and intermediate in birds fed BAL45-25. Breast meat yield was greater (P < 0.05) in turkeys fed muramidase, regardless of dose, compared with birds fed the CTL. There was no effect of treatment on muramic acid content in the jejunum digesta or litter scores. The frequency of pododermatitis score 1 was greater (P < 0.05) and score 2 was lower (P < 0.05) in birds fed muramidase, regardless of dose, compared with birds fed the CTL diet. In conclusion, muramidase supplementation improved performance, breast meat yield, feed efficiency and some markers of welfare, proportional to the dose in the diets.

A series of in vitro trials was conducted to evaluate the influence of limestone, dicalcium phosphate, phytase, and the digestion phase on Ca and P solubility. Experimental samples were arranged as a 2 × 2 × 2 × 2 factorial and contained corn (experiment 1) or soybean meal (experiment 2) plus limestone, dicalcium phosphate, phytase, and all combinations. Calcium and available P in the samples were maintained at 1.0% with limestone and 0.45% with dicalcium phosphate, respectively. Phytase was added to the samples at 1,000 FTU/kg. Samples were exposed to a 2-step in vitro digestion assay to simulate the gastric and the small intestinal (SI) phases of digestion. In experiment 1, dicalcium phosphate improved (P ≤ 0.05) P solubility in the gastric phase, which did not change in the SI phase, except when phytase was supplemented, which reduced (P ≤ 0.05) P solubility in the SI phase. The small amount of Ca present in corn is highly soluble, but limestone, dicalcium phosphate, or phytase reduced (P ≤ 0.05) Ca solubility in the gastric phase. Solubility was further reduced (P ≤ 0.05) in the SI phase in the presence of limestone. In experiment 2, P was more soluble (P ≤ 0.05) in the presence of limestone, dicalcium phosphate, or phytase in the gastric phase and compared with P solubility in the SI phase. Calcium solubility was reduced (P ≤ 0.05) in the SI phase compared with the gastric phase, except when dicalcium phosphate or limestone was supplemented. In conclusion, P and Ca solubility were influenced by the change in pH between the gastric and SI phases, differences in diet composition, and the Ca:P ratio. Limestone, dicalcium phosphate, and phytase increased P solubility in the gastric phase and reduced P solubility in the SI phase. Phytase had more of an effect on P and Ca solubility in soybean meal than in corn, and this is possibly a result of the high amount of phytate in soybean meal.

An experiment was conducted to determine the influence of 2 levels of dietary Ca from limestone and 3 levels of phytase on broiler performance, bone ash, gastrointestinal pH, and apparent ileal digestibility (AID) of Ca, P, and amino acids. Cobb 500 broilers (n = 576) were allowed access to one of 6 corn-soy diets from 0 to 16 d. Experimental diets contained 1.03% or 0.64% Ca from limestone and 0.61% total P. Each diet was supplemented with 0, 500, or 5,000 FTU/kg of phytase to create a 2 × 3 factorial experiment. Broiler feed intake (FI) and BW gain were not affected by dietary Ca or phytase. Feed conversion ratio was improved (P < 0.05) as dietary phytase increased (1.36, 1.34, and 1.31, respectively). Tibia ash percent was reduced (P < 0.05) from 41.4 to 40.0% as dietary Ca decreased but increased with phytase addition (P < 0.05). Gizzard and ileal pH were reduced (P < 0.05) in broilers fed 0.64% Ca compared with broilers fed 1.03% Ca. Phytase at 5,000 FTU/kg increased (P < 0.05) pH in the gizzard, duodenum, jejunum, and ileum. Apparent ileal P digestibility was increased (P < 0.05) in broilers fed 0.64% Ca compared with broilers fed 1.03% Ca (0.68 vs. 0.73, respectively). Apparent ileal Ca digestibility was increased (P < 0.05) in broilers fed 1.03% Ca compared with broilers fed 0.64% Ca (0.67 vs. 0.53, respectively). Phytase improved AID of CP in broilers fed 1.0% Ca but did not have an effect on AID of CP in broilers fed 0.64% Ca, which resulted in a Ca × phytase interaction (P < 0.05). In conclusion, high dietary Ca increased pH in gizzard and ileum and interfered with the AID of P and CP. The interactions between Ca and phytase in the gastrointestinal tract are complex, and feeding phytase at doses above industry recommendations may allow for reduced-Ca diets while maintaining broiler performance, bone ash, and improving amino acid digestibility.

An experiment was conducted to evaluate the influence of a highly soluble Ca (HSC) source on performance, bone ash and mineralization, and apparent ileal digestibility of Ca, P, N, and energy in Ross 708 broiler chickens. Dietary Ca was supplied by the HSC source and monocalcium phosphate to provide 4 levels of Ca (0.45, 0.60, 0.75, or 0.90%). Available P (aP) was maintained at 0.32% in all the diets. Each diet was supplemented with 0, 500, or 2,500 U/kg of phytase as a 4 × 3 factorial. An additional diet was formulated using limestone to contain 0.90% Ca and 0.45% aP as a positive control (PC). Diets were fed to 7 replicate cages of 8 birds per cage from d 0 to 21 posthatch. Feed intake (FI) was linearly (P ≤ 0.05) reduced, and BW gain was quadratically (P ≤ 0.05) reduced as Ca from HSC increased from 0.45 to 0.90%. Phytase supplementation linearly (P ≤ 0.05) increased FI and BW gain and improved feed conversion ratio. Tibia ash and P weights were lower in birds fed 0.90% Ca from HSC compared with broilers fed 0.90% Ca from limestone. Phytase supplementation increased tibia ash and P weights in broilers fed 0.60, 0.75, or 0.90% HSC (Ca level × phytase P ≤ 0.05). Phytase supplementation linearly (P ≤ 0.05) increased tibia Ca weight. Reducing HSC from 0.90 to 0.45% or increasing phytase supplementation linearly (P ≤ 0.05) improved ileal P digestibility. Ileal Ca digestibility increased linearly only in broilers fed 0.45% Ca from HSC as phytase supplementation increased (Ca level × phytase P ≤ 0.05). Nitrogen or energy digestibility were quadratically (P ≤ 0.05) influenced as HSC or phytase increased. In conclusion, feeding HSC with phytase allowed for reductions in dietary Ca while maintaining broiler performance and bone ash. In addition, P digestibility and P in the bones were increased in broilers fed reduced dietary Ca in the presence of phytase.

Male broilers (n = 576) were obtained and allocated to 96 cages with 6 birds per cage. From hatch to d 24, 16 randomly allocated cages were fed a nutrient adequate reference diet. The reference diet was formulated to contain 0.96 and 0.87% total Ca from hatch to d 10 and d 11 to 24, respectively. The remaining 80 cages were fed a nutrient adequate diet, formulated to contain 0.50% standardized ileal digestible (SID) Ca from hatch to d 10. On d 11, five diets containing 0.56, 0.46, 0.36, 0.26, or 0.16% SID Ca were randomly assigned to 16 cages per diet. Means were separated using polynomial contrasts and Dunnett's Multiple Comparison tests. From hatch to d 10, birds fed diets formulated to contain 0.50% SID Ca gained more (P < 0.05) compared with birds fed the reference diet. From d 11 to 24, there was no effect of diet on feed intake or BW gain. Birds fed 0.46 or 0.16% SID Ca were less (P < 0.05) efficient compared with birds fed the reference diet. Birds fed 0.26 or 0.36% SID Ca were more efficient compared with birds fed all other levels of SID Ca (quadratic, P < 0.05). Tibia ash percent was greatest in birds fed 0.56 or 0.46% SID Ca, decreased (quadratic, P < 0.05) as dietary SID Ca decreased and was lower (P < 0.001) in birds fed the 0.26 or 0.16% SID Ca diets compared with birds fed the reference diet. Apparent ileal digestibility (AID) of Ca or retention of P was greater (P < 0.05) in birds fed the diets formulated using SID Ca compared with birds fed the reference diet. Apparent P retention was greatest in birds fed 0.36% SID Ca (quadratic, P < 0.05). Regression equations developed using P retention, AID of Ca, and percent tibia ash estimated the SID Ca requirement of 11 to 24-d-old broilers was 0.380, 0.488, and 0.515%, respectively. This corresponds to a SID Ca to available P ratio of 0.86 to 1.17.

An experiment was conducted to compare the use of a total and digestible Ca formulation system in broilers from hatch to d 37 post-hatch. Ross 308 (n = 288) male broilers were obtained and allocated to one of 2 dietary treatments in floor pens. There were 18 birds per pen and 8 replicate pens per diet. One diet was formulated using ingredient and nutrient recommendations for total Ca and the second diet was formulated using ingredient and nutrient recommendations for standardized ileal digestible (SID) Ca. All diets were formulated to be nutrient adequate using a 2-phase feeding program and including 0.19% available P (avP) and 0.209% total Ca or 0.073% digestible Ca from 1,000 FYT/kg of phytase. On d 17 and 37, tibias and ileal contents were obtained. From hatch to d 37, birds fed diets formulated using digestible Ca gained (P < 0.05) more and were more efficient (P < 0.05) compared with birds fed diets formulated using total Ca. There was no impact of formulation system on tibia ash or minerals. Litter pH (P < 0.05) was greater and litter dry matter (P < 0.05) was lower in birds fed the diets formulated using digestible Ca compared with those fed diets formulated using total Ca. Apparent ileal digestibility (AID) of Ca (P < 0.05), AID of P (P < 0.05) and digestible P intake (P < 0.05) were lower in birds fed diets formulated using digestible Ca compared with those formulated using total Ca at d 17 or 37. However, apparent ileal digested Ca and digestible Ca intake were not different between the experimental diets on d 17 or 37. In conclusion, formulating diets using digestible Ca improved weight gain and feed conversion ratio, but reduced the AID of Ca and P compared with birds fed diets formulated using total Ca. These findings might be reflective of the higher total Ca concentration in the diets formulated using digestible Ca compared with those formulated using total Ca.

An experiment was conducted to determine the standardized ileal digestible (SID) Ca requirement of Arbor Acres Plus male broilers from d 25 to 42 post-hatch. Broilers were obtained at hatch, placed in floor pens, and fed a nutrient adequate diet until d 24 post-hatch. On d 25, twelve hundred birds were weighed and allocated to one of 4 treatments. There were 25 birds per pen and 12 pens per diet. The diets were formulated to contain 0.46, 0.35, 0.24, or 0.13% SID Ca. Available P (avP) was 0.39% in all diets, including 0.16% avP expected from 2,500 FYT/kg of phytase. The SID Ca requirement was estimated using nonlinear models, including quadratic, straight broken-line, and quadratic broken-line. There was no effect of SID Ca on feed intake, body weight gain, feed conversion ratio, or livability. Tibia ash percent was greatest in birds fed 0.35% SID Ca and lowest in birds fed 0.13% SID Ca (quadratic, P = 0.063). Apparent ileal digestibility (AID) of Ca was highest in birds fed the diets containing 0.13% SID Ca and decreased (quadratic, P = 0.014) as dietary SID Ca increased to 0.46%. Apparent digested Ca was highest in birds fed 0.35% SID Ca and lowest in birds fed 0.13% SID Ca (quadratic, P = 0.005). Decreasing the concentration of SID Ca in the diet from 0.46 to 0.13% (P < 0.0001) increased the AID of P and apparent digested P. Litter N or P were lowest in birds fed 0.35% SID Ca and increased (quadratic, P ≤ 0.05) as dietary SID Ca decreased to 0.13%. Non-linear equations, developed using tibia ash percent, digested Ca, or litter P, estimate the SID Ca requirement of Arbor Acres Plus broilers from d 25 to 42 was 0.37, 0.35, or 0.35%, respectively. This corresponds to an SID Ca to available P ratio of 0.95 to 0.90.

An experiment was conducted to evaluate the effects of dietary enzymes on performance, tibia ash, and intestinal goblet cells of broilers administered a live coccidia oocyst vaccine (Coccivac B, Schering Plough, Kenilworth, NJ). Cobb 500 straight-run broilers were obtained and one-half of the chicks were sprayed with the live coccidia oocyst vaccine. Chicks were weighed and placed in battery brooders with respect to nonvaccinated or vaccinated group according to dietary treatment. The 8 dietary treatments were a positive control (0.90% Ca and 0.45% available P), a negative control (NC; 0.80% Ca and 0.35% available P), NC + phytase (PHY), NC + protease (PRO), NC + xylanase (XYL), NC + PHY+ PRO, NC + PHY + XYL, and NC + PHY + PRO + XYL. A diet × vaccination interaction (P > 0.05) was not observed for feed intake or BW gain. Feed conversion ratio was improved (P ≤ 0.05) in birds fed NC + PHY + XYL compared with NC. Vaccination reduced (P ≤ 0.05) feed intake and BW gain from d 0 to 18. Tibia ash was reduced (P ≤ 0.05) in the NC and PRO or XYL diets. Vaccination increased goblet cell numbers in the duodenum of birds fed XYL, whereas no differences were found in goblet cell numbers between nonvaccinated and vaccinated birds in other dietary treatments, which resulted in a diet × vaccination interaction (P ≤ 0.05). Protease decreased and NC + PHY+ PRO increased goblet cells in the jejunum at d 7, which resulted in a diet × vaccination interaction (P ≤ 0.05). At d 18, NC + PHY + XYL was the only diet in which vaccination decreased goblet cells in the jejunum, resulting in a diet × vaccination interaction (P ≤ 0.05). The data indicate that NC + PHY + XYL improved the feed conversion ratio in broilers fed corn-soybean meal diets. The vaccination × dietary enzyme interaction altered the number of goblet cells in the small intestine. Dietary enzyme supplementation did not alleviate reductions in growth performance associated with the use of a live coccidia oocyst vaccine.

An experiment was conducted to evaluate effects of inositol and gradient phytase supplementation on growth performance and meat yield of broilers from 1 to 41 d of age. A total of 1,920 Yield Plus × Ross 708 male chicks were placed in 64 floor pens (30 birds per pen). Each pen received one of the 8 dietary treatments (8 replicate pens) from 1 to 15, 16 to 29, and 30 to 40 d of age. Treatment 1 was formulated to contain 0.165 and 0.150% lower calcium and phosphorus, respectively, than treatment 7 (positive control). Phytase was added to treatment 1 at concentration of 500, 1,500, 4,500, 13,500, and 40,500 phytase units (FTU)/kg to establish treatments 2 to 6, respectively. Treatment 8 was formulated by adding inositol to treatment 7 based on the expected inositol liberation in treatment 6. Feed and birds were weighed at 1, 15, 29, and 40 d of age to determine BW gain, feed intake, and feed conversion. Twelve birds per pen were processed at 41 d of age to determine carcass characteristics. From 1 to 40 d of age, log-quadratic effects of phytase (treatments 1-6) were observed for BW gain (P = 0.002) and feed conversion in broilers (P = 0.018), whereas feed intake increased log-linearly (P = 0.045). The addition of 40,500 FTU/kg of phytase increased cumulative BW gain (P = 0.001) and decreased cumulative feed conversion (P = 0.005) by 4.7 and 2.6%, respectively, compared with birds subjected to treatment 8. Log-quadratic effects of phytase additions were observed for carcass (P < 0.001) and breast meat weights (P = 0.004). Growth performance and carcass characteristics of broilers subjected to treatment 7 were similar (P > 0.05) to those of birds subjected to treatment 8. These data demonstrate that the extraphosphoric effects of phytase may be associated with increased feed intake of broilers. Inositol supplementation did not provide additional benefits to broilers in this study.
