RESUMO

A highly sensitive optical fiber Fabry-Perot interferometer (FPI) for strain measurement with temperature compensation is proposed. Instead of using another actual reference interferometer, a virtual FPI is constructed to superpose with the sensing FPI to form the Vernier effect. The fundamental and the first-order harmonic Vernier effect are generated to increase the sensitivity by adjusting the parameter of the virtual FPI. In order to separate the strain from the environment temperature, an FBG is cascaded to distinguish the applied temperature. Experimental results demonstrate that, with the help of the fundamental Vernier effect, the sensitivity and temperature of the FPI increases from 1.05 pm/°C to 10.63 pm/°C in the temperature range of 40-120°C, and the sensitivity of strain increases from 2.635 pm/µÎµ to 33.11 pm/µÎµ in the strain range of 0-400 µÎµ. In order to access the tracking points more easily and further enhance the sensitivities, the first-order harmonic Vernier effect is generated by modifying the virtual FPI. Results show that the temperature and strain sensitivities are 21.25 pm/°C and 62.25 pm/µÎµ, respectively. In addition, with the help of the FBG, the strain can be separated from the temperature by solving the cross-sensitivity matrix.

OBJECTIVE: This pilot study aimed to observe intimal injuries related to stent retrieval in the iliac artery of a canine. BACKGROUND: In-stent restenosis remains challenging owing to permanent stent implantation. A retrievable stent may be alternative for intervention without permanent residue. METHODS: Five retrievable stents with point-to-point overlapped double-layer scaffolds were deployed into the iliac arteries and retrieved on days 14, 21, 28, 35, and 42 from five canines. RESULTS: Arterial diameter decreased by 9-10% before retrieval and 15% on day 14 after retrieval. In the 14-day-stent, the stent surface was clean without visible fibrin. In the 28-day-stent, the overlay was mainly composed of fibrin and fibroblasts. The proliferation of smooth muscle cells has not yet been observed with α-smooth muscle actin staining. In the 42-day-stent, endothelial and smooth muscle cells decreased under the struts, and the internal elastic lamina was interrupted segmentally. Neointima formation involves fibroblasts and smooth muscle cells. Neointimal thickness was negatively correlated with strut space. Stent traces on the artery wall tended to be flat at a follow-up14 days after retrieval. The primary intima was completely covered by neointima. Two stents could not be retrieved because of in-stent thrombosis or capture loss. CONCLUSIONS: The stent was covered mainly by depositional fibrin after 28 days and by typical neointima after 42 days. The stent retrieval procedure did not induce injury to vascular smooth muscle, and the intima repair was performed 14 days after stent retrieval.

BACKGROUND: Dynamic interpersonal therapy (DIT) is a brief, structured psychodynamic psychotherapy with demonstrated efficacy in treating major depressive disorder (MDD). The aim of the study was to determine whether DIT is an acceptable and efficacious treatment for MDD patients in China. METHOD: Patients were randomized to 16-week treatments with either DIT plus antidepressant medication (DIT + ADM; n = 66), general supportive therapy plus antidepressant medication (GST + ADM; n = 75) or antidepressant medication alone (ADM; n = 70). The Hamilton Depression Rating Scale (HAMD) administered by blind raters was the primary efficacy measure. Assessments were completed during the acute 16-week treatment and up to 12-month posttreatment. RESULTS: The group × time interaction was significant for the primary outcome HAMD (F = 2.900, df1 = 10, df2 = 774.72, p = 0.001) in the acute treatment phase. Pairwise comparisons showed a benefit of DIT + ADM over ADM at weeks 12 [least-squares (LS) mean difference = -3.161, p = 0.007] and 16 (LS mean difference = -3.237, p = 0.004). Because of the unexpected high attrition during the posttreatment follow-up phase, analyses of follow-up data were considered exploratory. Differences between DIT + ADM and ADM remained significant at the 1-, 6-, and 12-month follow-up (ps range from 0.001 to 0.027). DIT + ADM had no advantage over GST + ADM during the acute treatment phase. However, at the 12-month follow-up, patients who received DIT remained less depressed. CONCLUSIONS: Acute treatment with DIT or GST in combination with ADM was similarly efficacious in reducing depressive symptoms and yielded a better outcome than ADM alone. DIT may provide MDD patients with long-term benefits in symptom improvement but results must be viewed with caution.

Age-related cataract (ARC) is one of the most common chronic diseases. Circular RNA (circ)_HIPK3 is reported to be involved in the advancement of ARC, but its molecular mechanism has not been clarified. Our study provides a new perspective on the clinical treatment of ARC. Our data showed that the expression levels of circ_HIPK3 and histone deacetylase 4 (HDAC4) were downregulated, while microRNA (miR)-495-3p level was increased in ARC tissues and H2O2-induced SRA01/04 cells. Functional experiments showed that circ_HIPK3 and HDAC4 overexpression could inhibit H2O2-induced lens epithelial cell apoptosis and fibrosis. In terms of mechanism, we found that circ_HIPK3 could sponge miR-495-3p, miR-495-3p could target HDAC4. Besides, we confirmed that circ_HIPK3 sponged miR-495-3p to positively regulate HDAC4. Additionally, miR-495-3p overexpression or HDAC4 knockdown reversed the inhibition effect of circ_HIPK3 on H2O2-induced lens epithelial cell injury. In conclusion, our data showed that circ_HIPK3 suppressed H2O2-induced lens epithelial cell injury by regulating miR-495-3p/HDAC4 axis.

Either effortful swallowing exercise or repetitive transcranial magnetic stimulation (rTMS) is considered as an effective method to treat dysphagia after stroke. Thus, synchronizing these two interventions may improve the efficiency of treatment. This trial intended to explore the effects of rTMS and effortful swallowing exercise on the recovery of swallowing function in patients after stroke. A total of 56 patients with post-stroke dysphagia who were able to actively cooperate with the training were analyzed in this study. We experimented with different intervention effects of rTMS synchronization with effortful swallowing training (group 1), rTMS (group 2), and traditional swallowing training alone (group 3). Every patient completed conventional swallowing training 5 days a week for 2 weeks. Patients in group 1 and group 2 underwent 10 consecutive sessions of 5 Hz rTMS over the affected mylohyoid cortical region. Fiberoptic endoscopic dysphagia severity scale (FEDSS), penetration/aspiration scale (PAS), standardized swallowing assessment (SSA), and functional oral intake scale (FOIS) were assessed and compared across the groups. No significant difference in FEDSS, PAS, SSA, or FOIS scores was found at baseline among the three groups. The mean change values of the FEDSS score, PAS score, SSA score, and FOIS score between baseline and post-intervention of the three groups (H = 16.05, P < 0.001; H = 21.70, P < 0.001; F (2, 53) = 9.68, P < 0.001; H = 18.26, P < 0.001; respectively) were statistically significant. In addition, the mean change values of FEDSS, PAS, SSA, and FOIS scores in participants in group 1 (all P < 0.001) and group 2 (P = 0.046; P = 0.045; P = 0.028; P = 0.032; respectively) were significantly higher than in group 3. Similarly, the mean change values of FEDSS, PAS, SSA, and FOIS scores were significantly higher in participants in group 1 than in group 2 (P = 0.046; P = 0.038; P = 0.042; P = 0.044; respectively). The results revealed that the conjunction of rTMS and effortful swallowing training was an effective method to facilitate the recovery of swallowing function in stroke patients. The present clinical trial provided a new treatment method for the functional restoration of swallowing in stroke patients, which may further facilitate the recovery of swallowing function in stroke patients with swallowing disorders.

A post chemical etching process to a tapered seven-core fiber (TSCF) is proposed and experimentally demonstrated to effectively adjust the mode profiles of high-order supermodes, aimed to improve the figure of merit (FOM). The experimental results show that the FOM of an etched TSCF is as high as 1431.36 1/RIU, a 7.32-times enhancement compared with that of TSCF without etching, provided the TSCF has the same taper waist diameter of 19.20 µm. The proposed method opens a new, to the best of our knowledge, method for optimizing optical fiber sensor performance.

Sclerotinia sclerotiorum is a well-known phytopathogenic fungus with a wide host range. Identifying novel mycoviruses in phytopathogenic fungi is necessary to develop novel strategies for plant health protection and contribute to understanding the origin of viruses. Six new mycoviruses with positive single-stranded RNA genomes co-infecting the hypovirulent strain SCH733 of S. sclerotiorum were identified using a metatranscriptomic approach, and their complete genome sequences were molecularly determined. These mycoviruses belong to the following five families: Narnaviridae, Mitoviridae, Deltaflexviridae, Botourmiaviridae, and Ambiguiviridae. Three of these mycoviruses belong to existing International Committee on Taxonomy of Viruses (ICTV)-recognized species. Two of these newly identified mycoviruses have unique genomic features that are significantly different from those of all known mycoviruses. Phylogenetic analysis revealed that these six mycoviruses included close as well as distant relatives of known mycoviruses, thereby providing new insight into virus evolution and classification. Mycovirus horizontal transmission and elimination experiments revealed that Sclerotinia sclerotiorum narnavirus 5 is associated with hypovirulence of S. sclerotiorum, although we have not shown that it is independently responsible for the hypovirulence phenotype. This study broadens the diversity of known mycoviruses infecting S. sclerotiorum and provides a clue toward limiting hypovirulence in S. sclerotiorum.

Hypoviruses are positive-sense single-stranded RNA mycovirus that infect filamentous fungi. However, hypoviruses have not been reported in Bipolaris oryzae, an important phytopathogenic fungus in water bamboo and rice. Here, we report the characterization of a novel hypovirus, tentatively named "Bipolaris oryzae hypovirus 1" (BoHV1), isolated from strain ES35 of B. oryzae infecting water bamboo. The complete genome of BoHV1 consists of 13,596 nucleotides and a poly(A) tail at the 3' end. BoHV1 has single open reading frame (ORF) and encodes a putative polyprotein (4,218 amino acids) containing four potential conserved domains for a papain-like protease, a protein of unknown function (DUF3525), RNA-dependent RNA polymerase (RdRp), and helicase. Phylogenetic analysis of the polyprotein, RdRp, and helicase domains suggested that BoHV1 belongs to the genus Hypovirus within the family Hypoviridae. This is the first report of the presence of a hypovirus in the phytopathogenic fungus B. oryzae.

The development of paper-based SERS substrates that can allow multi-component detection in real-word scenarios is of great value for applications in molecule detection under complex conditions. Here, a multifunctional SERS-based paper sensing substrate has been developed through the uniform patterning of high-density arrays of GO-isolated Ag nanoparticles on the hydrophilic porous cellulose paper strip (GO@AgNP@paper). Wet-chemical synthesis was used to provide the cover of SERS hot spots on any part of the paper, not just limited surface deposition. In virtue of the inherent ability of paper to deliver analytes by the capillary force, the detection ability of the GO@AgNP@paper substrate was greatly promoted, allowing as low as 10-19M R6G detection from microliter-volume (50 µL) samples. For the components with different polarity, the paper substrate can be used as an all-in-one machine to achieve the integration of separation and high-sensitive detection for ultralow mixture components, which improves the practical application value of SERS-based paper devices.

We investigate a new class of nondiffracting optical beams carrying fractional orbital angular momentum (FOAM Bessel beams), which can be constructed by modifying the phase step of the spiral phase plate in a setup originally for the generation of traditional Bessel beams. The FOAM Bessel beams have entirely stable vortex structures, and form an infinite number of orthogonal and complete subsets for monochromatic scalar light in free space. With the advantages of structural stability and orthogonality-completeness, the FOAM Bessel beams are expected to be useful for study and application in various fields of modern optics.

We have proposed a synthetic approach to produce self-supported and bendable surface-enhanced Raman scattering (SERS)-based 3D chemical sensors with high adsorptivity. Such 3D substrates consist of foam-like graphene macrostructures obtained by template-directed chemical vapour deposition on nickel foams (interconnected 3D scaffold of nickel) and uniform and high-density Ag nanoparticles wrapping around the foam graphene, via seed-mediated in situ growth process. Such 3D AgNPs/G@Ni foam substrates show high-quality SERS performance in terms of Raman signal reproducibility and sensitivity for the analyte, resulting from the high density and homogeneity of "hot spots" on AgNPs/G@Ni foam, multiple cascaded amplication (localized surface plasmon mode and optical standing waves or optical refraction) of incident laser to the 3D foam structures and powerful support from nickel scaffold. Moreover, in virtue of the high adsorptivity and sensitivity of AgNPs/G@Ni foam, the low-concentration crystal violet molecules can be easily traced in the curvilinear fish surface, by simply swabbing the surface to achieve molecules concentration effect in the practical applicability. This work shows promising potential in developing the applications of SERS in the foodstuffs processing and security field.

The physical phenomenon, surface-enhanced Raman scattering (SERS), is mainly based on the local electromagnetic fields enhancement located at the nano-gaps between metal nanostructures attributed to localized surface plasmon resonance. Therefore, nano-gaps are very important for obtaining high-density hot spots and optimal and uniform SERS signals. However, it remains a challenge to form the three-dimensional ultra-narrow nano-gaps. Here, a gyrus-inspired Gyrus-SERS substrate was fabricated with the nanostructure of Ag gyrus/graphene/Au film using an extremely simple method. The lateral and vertical hot spots respectively were obtained from the dense nano-gaps (~3 nm) between gyrus and the coupling of Ag gyrus and Au film in bilayer graphene nano-gaps (0.68 nm), which were demonstrated in experiment and theory. The proposed Gyrus-SERS platform performs an excellent SERS activity (EF~5 × 109), high sensitivity (the minimum detected concentration of R6G and CV respectively is 10-13 and 10-12 M), and outstanding reproducibility (RSD~7.11%). For practical application, the in situ detection of Malachite green (MG) residue on prawn skin was executed using the prepared flexible Gyrus-SERS substrate, which shows the wide potential in food safety field.

Overcoming proteins adsorption on the inner surface of capillary has attracted increasing attention recently. By using the unique photochemistry reaction of diazoresin (DR), a new covalent capillary coating was prepared on the fused-silica capillary through layer-by-layer self-assembly of DR with polyglycerol (PG) dendrimer. The separation performance of covalently DR/PG-dendrimer coated capillary noticeably exceeded the bare capillary and the noncovalently linked DR/PG-dendrimer capillary. A baseline separation of lysozyme, myoglobin, bovine serum albumin, and ribonuclease A was achieved using CE within 20 min. Besides, the covalently linked DR/PG-dendrimer coating has the remarkable stability and reproducibility. Especially, compared with the traditional method which use highly toxic and moisture-sensitive silane coupling agent, this method seems to be a simple and environmental friendly way to prepare the covalently coated capillaries for CE.

UNLABELLED: Mycoviruses have been detected in all major groups of filamentous fungi, and their study represents an important branch of virology. Here, we characterized a novel double-stranded RNA (dsRNA) mycovirus, Sclerotinia sclerotiorum megabirnavirus 1 (SsMBV1), in an apparently hypovirulent strain (SX466) of Sclerotinia sclerotiorum. Two similarly sized dsRNA segments (L1- and L2-dsRNA), the genome of SsMBV1, are packaged in rigid spherical particles purified from strain SX466. The full-length cDNA sequence of L1-dsRNA/SsMBV1 comprises two large open reading frames (ORF1 and ORF2), which encode a putative coat protein and an RNA-dependent RNA polymerase (RdRp), respectively. Phylogenetic analysis of the RdRp domain clearly indicates that SsMBV1 is related to Rosellinia necatrix megabirnavirus 1 (RnMBV1). L2-dsRNA/SsMBV1 comprises two nonoverlapping ORFs (ORFA and ORFB) encoding two hypothetical proteins with unknown functions. The 5'-terminal regions of L1- and L2-dsRNA/SsMBV1 share strictly conserved sequences and form stable stem-loop structures. Although L2-dsRNA/SsMBV1 is dispensable for replication, genome packaging, and pathogenicity of SsMBV1, it enhances transcript accumulation of L1-dsRNA/SsMBV1 and stability of virus-like particles (VLPs). Interestingly, a conserved papain-like protease domain similar to a multifunctional protein (p29) of Cryphonectria hypovirus 1 was detected in the ORFA-encoded protein of L2-dsRNA/SsMBV1. Phylogenetic analysis based on the protease domain suggests that horizontal gene transfer may have occurred from a single-stranded RNA (ssRNA) virus (hypovirus) to a dsRNA virus, SsMBV1. Our results reveal that SsMBV1 has a slight impact on the fundamental biological characteristics of its host regardless of the presence or absence of L2-dsRNA/SsMBV1. IMPORTANCE: Mycoviruses are widespread in all major fungal groups, and they possess diverse genomes of mostly ssRNA and dsRNA and, recently, circular ssDNA. Here, we have characterized a novel dsRNA virus (Sclerotinia sclerotiorum megabirnavirus 1 [SsMBV1]) that was isolated from an apparently hypovirulent strain, SX466, of Sclerotinia sclerotiorum. Although SsMBV1 is phylogenetically related to RnMBV1, SsMBV1 is markedly distinct from other reported megabirnaviruses with two features of VLPs and conserved domains. Our results convincingly showed that SsMBV1 is viable in the absence of L2-dsRNA/SsMBV1 (a potential large satellite-like RNA or genuine genomic virus component). More interestingly, we detected a conserved papain-like protease domain that commonly exists in ssRNA viruses, including members of the families Potyviridae and Hypoviridae. Phylogenetic analysis based on the protease domain suggests that horizontal gene transfer might have occurred from an ssRNA virus to a dsRNA virus, which may provide new insights into the evolutionary history of dsRNA and ssRNA viruses.

The contribution of the Green's function to the scattered magnetic field is not well expressed in the traditionally used magnetic field integral equation (MFIE) when the source and field points lie on the same plane. This decreases the accuracy of MFIE for objects with flat surfaces. To solve this problem, the normal magnetic field integral equation resulting from the use of the normal boundary condition is considered. It is then combined with the traditionally used MFIE into a new MFIE formulation, named the combined magnetic field integral equation (CMFIE). Through the use of an appropriate combination parameter, the omitted contribution of the Green's function to the scattered magnetic fields in the traditionally used MFIE can be recovered in this new CMFIE. Numerical results validate the analysis and show that the proposed MFIE formulation is much more accurate than the traditional one for small objects with flat surfaces.

The performance of a diode end-pumped passively Q-switched dual-wavelength Nd:GYSGG laser operating at 1057.28 and 1060.65 nm with graphene oxide as the saturable absorber was demonstrated. The maximum dual-wavelength average output power of 521 mW was achieved under the absorbed pump power of 5.4 W, corresponding to the optical-to-optical conversion and slope efficiency of 9.8% and 21%, respectively. The minimum pulse width was 115 ns with a pulse repetition rate of 338 kHz.

RESUMO

Passively Q-switched mode-locking performance of Nd:GdTaO4 crystal using molybdenum disulfide (MoS2) as a saturable absorber at 1066 nm was demonstrated for the first time. The MoS2 saturable absorber was prepared simply by transferring the MoS2 suspension onto a quartz substrate. By inserting the MoS2 saturable absorber into the Nd:GdTaO4 laser, stable Q-switched modelocked operation can be achieved. At the pump power of 4 W, the maximum average output power of 0.156 W was obtained with the optical conversion efficiency of 3.9%.

RESUMO

A facile and controllable interface-directed coassembly (IDCA) approach is developed for the first time to synthesize uniform discrete mesoporous silica particles with a large pore size (ca. 8 nm) by using 3-dimensional macroporous carbon (3DOMC) as the nanoreactor for the confined coassembly of template molecules and silica source. By controlling the amount of the precursor solution and using Pluronic templates with different compositions, we can synthesize mesoporous silica particles with diverse morphologies (spheres, hollow spheres, and hemispheres) and different mesostructure (e.g., 2-D hexagonal and 3D face centered cubic symmetry), high surface area of about 790 m(2)/g, and large pore volume (0.98 cm(3)/g). The particle size can be tunable from submicrometer to micrometer regimes by changing the macropore diameter of 3DOMC. Importantly, this synthesis concept can be extended to fabricate multifunctional mesoporous composite spheres with a magnetic core and a mesoporous silica shell, large saturated magnetization (23.5 emu/g), and high surface area (280 m(2)/g). With the use of the magnetic mesoporous silica spheres as a magnetically recyclable absorbent, a fast and efficient removal of microcystin from water is achieved, and they can be recycled for 10 times without a significant decrease of removal efficiency for microcystin.

BRCA1 and BRCA2 genes are crucial for double-strand break repair by homologous recombination, and mutations in these genes are responsible for most familial breast carcinomas. Cells with inactivating mutations of the BRCA1 or BRCA2 tumor suppressor genes are sensitive to poly (ADP-ribose) polymerase-1 (PARP1) inhibitors. Already in 2010, it has been predicted, that BRCA1 hypermethylation might be sensitive to PARP1 inhibitor. However, till today, a statistically significant proof has been missing, and the effectiveness of PARP1 inhibitors for breast cancer caused by BRCA1 promoter hypermethylation remained elusive. Pyrosequencing has been proposed as an optimal method to investigate the methylation status of the BRCA1 genes. Here, we show for the first time that BRCA1 CpG island hypermethylation is sensitive to PARP1 inhibitors. In clinical settings, this might improve treatment response and provide a more personalized therapy for breast cancer patients. Furthermore, the determination of methylation status of BRCA1 and other genes of the BRCA/homologous recombination (HR) pathway may be an important predictive classifier of response to PARP inhibitor therapy.

The higher heating value of biochar is an important parameter for the utilization of biomass energy. In this work, extreme gradient boosting regression and artificial neural network were used to predict it based on the characteristics of biomass and pyrolysis conditions. Besides, empirical correlations were developed for comparison. Results showed that the extreme gradient boosting regression models showed better performance (R2 = 0.83-0.94). The shapley additive explanations and partial dependence plot indicated that lignin content and higher heating value of raw material were highly positively correlated with higher heating value of biochar, and found the better conditions such as pyrolysis temperature (>550 °C), lignin content (>40 wt%) for high-higher heating value biochar preparation. What's more, a program that predicted higher heating value of biochar was developed through PySimpleGUI library. It offered a new optimization idea for the directional preparation process of biochar.
