RESUMO

Manure storage methods can affect the concentration and prevalence of antibiotic-resistant bacteria (ARB) and antibiotic resistance genes (ARGs) in cattle manure prior to land application. The objective of this study was to compare stockpiling and composting with respect to their effectiveness in reducing ARB and ARGs in beef cattle manure in a field-scale study. Field experiments were conducted in different seasons with different bulking agents for composting. For both the winter-spring cycle and the summer-fall cycle, ARB concentrations declined below the limit of quantification rapidly in both composting piles and stockpiles; however, ARB prevalence was significantly greater in the composting piles than in the stockpiles. This was likely due to the introduction of ARB from bulking agents. There was no significant change in ARG concentrations between initial and final concentrations for either manure storage treatment during the winter-spring cycle, but a significant reduction of the ARGs erm(B), tet(O), and tet(Q) over time was observed for both the composting pile and stockpile during the summer-fall cycle. Results from this study suggest that (i) bulking agent may be an important source of ARB and ARGs for composting; (ii) during cold months, the heterogeneity of the temperature profile in composting piles could result in poor ARG reduction; and (iii) during warm months, both stockpiling and composting can be effective in reducing ARG abundance. IMPORTANCE Proper treatment of manure is essential to reduce the spread of antibiotic resistance and protect human health. Stockpiling and composting are two manure storage methods which can reduce antibiotic-resistant bacteria and resistance genes, although few field-scale studies have examined the relative efficiency of each method. This study examined the ability of both methods in both winter-spring and summer-fall cycles, while also accounting for heterogeneity within field-scale manure piles. This study determined that bulking agents used in composting could contribute antibiotic-resistant bacteria and resistance genes. Additionally, seasonal variation could hinder the efficacy of composting in colder months due to heterogeneity in temperature within the pile; however, in warmer months, either method of manure storage could be effective in reducing the spread of antibiotic resistance.

Atrazine is an herbicide that is applied in corn around the world and in sugarcane in Brazil. It is known to be hazardous for animals' health, mobile in the soil, and its analysis is considered expensive and onerous. Solute movement studies are essential to provide information about dangerous molecules movement, which can avoid contamination. While field investigations demand time and financial resources, numerical models are an alternative to describe water and solute distribution in the soil profile. Thus, the objective of this work was to use HYDRUS 2-D model for simulations of atrazine movement in containers packed with tropical soil cultivated with corn and to compare simulated and observed data through statistical parameters. The research was carried out in a greenhouse during 116 days after planting. Atrazine was analyzed in the soil solution at three different depths to validate HYDRUS-2D. Simulations were carried out using hydraulic properties fitted directly to measured retention data and parameters for corn growing and atmospheric characteristics. The mixed procedure analysis indicated that there are differences in atrazine concentration among depths and along time. In general, atrazine concentration is higher at shallow depths and right after application. However, it is possible to find atrazine in deeper soil layers, which might be a concern regarding contamination. RMSE, Willmott and Pearson coefficients indicated a favorable capacity of the model to simulate atrazine concentration on corn cultivation. HYDRUS-2D is a reliable tool to obtain trends in atrazine movement under these experiment's conditions. The uptake parameters, the crop root growth and distribution parameters depend on further specific studies to better describe the relationship between the plant and atrazine and meteorological parameters need to be updated.

Runoff from open-lot animal feeding areas contains microorganisms that may adversely affect human and animal health if not properly managed. One alternative to full manure containment systems is a vegetative treatment system (VTS) that collects runoff in a sediment basin and then applies it to a perennial vegetation (grass) treatment area that is harvested for hay. Little is known regarding the efficacy of large-scale commercial VTSs for the removal of microbial contaminants. In this study, an active, pump-based VTS designed and built for a 1200-head beef cattle feedlot operation was examined to determine the effects of repeated feedlot runoff application on fecal indicator microorganisms and pathogens over short-term (2 wk) and long-term (3 yr) operations and whether fecal bacteria were infiltrating into deeper soils within the treatment area. In a short-term study, fecal bacteria and pathogen numbers declined over time in soil. Measurements of total coliforms and Enterococcus counts taken on control soils were not effective as fecal indicators. The repeated application of manure-impacted runoff as irrigation water did not enrich the pathogens or fecal indicators in the soil, and no evidence was seen to indicate that pathogens were moving into the deeper soil at this site. These results indicate that large-scale, active VTSs reduce the potential for environmental contamination by manure-associated bacteria. Also, this study has implications to full-containment systems that apply runoff water to land application areas (cropland) and the fate of pathogens in the soils of land application sites.

Total reduced sulfur (TRS) emissions from animal feeding operations are a concern with increased feeding of high-sulfur distillers by-products. Three feeding trials were conducted to evaluate feeding wet distillers grain plus solubles (WDGS) on TRS fluxes. Fresh manure was collected three times during Feeding Trial 1 from cattle fed 0, 20, 40, and 60% WDGS. Fluxes of TRS from 40 and 60% WDGS manures were 3- to 13-fold greater than the 0 and 20% WDGS manures during the first two periods. In the final period, TRS flux from 60% WDGS was 5- to 22-fold greater than other WDGS manures. During Feeding Trial 2, 0 and 40% WDGS diets on four dates were compared in feedlot-scale pens. On two dates, fluxes from mixed manure and soil near the feed bunk were 3.5-fold greater from 40% WDGS pens. After removing animals, soil TRS flux decreased 82% over 19 d but remained 50% greater in 40% WDGS pens, principally from the wetter pen edges (1.9-fold greater than the drier central mound). During two cycles of cattle production in Feeding Trial 3, TRS soil fluxes were 0.3- to 4-fold greater over six dates for pens feeding WDGS compared with dry-rolled corn diet and principally from wetter pen edges. Soil TRS flux correlated with %WDGS, total N, total P, manure pack temperature, and surface temperature. Consistent results among these three trials indicate that TRS fluxes increase by two- to fivefold when cattle were fed greater levels of WDGS, but specific manure management practices may help control TRS fluxes.

The impact of proximity to a beef cattle feedlot on Escherichia coli O157:H7 contamination of leafy greens was examined. In each of 2 years, leafy greens were planted in nine plots located 60, 120, and 180 m from a cattle feedlot (3 plots at each distance). Leafy greens (270) and feedlot manure samples (100) were collected six different times from June to September in each year. Both E. coli O157:H7 and total E. coli bacteria were recovered from leafy greens at all plot distances. E. coli O157:H7 was recovered from 3.5% of leafy green samples per plot at 60 m, which was higher (P < 0.05) than the 1.8% of positive samples per plot at 180 m, indicating a decrease in contamination as distance from the feedlot was increased. Although E. coli O157:H7 was not recovered from air samples at any distance, total E. coli was recovered from air samples at the feedlot edge and all plot distances, indicating that airborne transport of the pathogen can occur. Results suggest that risk for airborne transport of E. coli O157:H7 from cattle production is increased when cattle pen surfaces are very dry and when this situation is combined with cattle management or cattle behaviors that generate airborne dust. Current leafy green field distance guidelines of 120 m (400 feet) may not be adequate to limit the transmission of E. coli O157:H7 to produce crops planted near concentrated animal feeding operations. Additional research is needed to determine safe set-back distances between cattle feedlots and crop production that will reduce fresh produce contamination.

Agricultural runoff from areas receiving livestock manure can potentially contaminate surface water with antimicrobials and antimicrobial resistance genes (ARGs). The objective of this study was to investigate the effectiveness of narrow grass hedges (NGHs) on reducing the transport of antimicrobials and ARGs in runoff after land application of swine manure slurry. Plot-scale rainfall simulation tests were conducted on 0.75 m by 4.0 m plots designed to test three treatment factors: manure amendment (control plots receiving no manure vs. amended plots receiving manure based on 3 times N requirement), NGH (plots with a NGH vs. plots without a NGH), and rainfall events (days 1-3). Runoff generated during three 30-min simulated rainfall events was sampled and analyzed for antimicrobials and ARGs. Manure amendment was responsible for the presence of antimicrobial tylosin ( < 0.0001) and tylosin resistance gene (B) ( < 0.0001) in runoff. Narrow grass hedges proved to be effective in reducing tylosin ( < 0.0001) and (B) ( < 0.0347) in runoff. Manure amendment was responsible for the introduction of tylosin ( < 0.0482) and (B) ( = 0.0128) into the soil; however, it had no significant impact on the abundance of the 16S rRNA gene in soil. Results from this study suggest that NGHs could be a best management practice to control the transport of antimicrobials and ARGs in agricultural runoff.

Beef cattle manure can serve as a valuable source of nutrients for crop production. However, emissions of volatile organic compounds (VOCs) after land application may pose an odor nuisance to downwind populations. This study was conducted to evaluate the effects of land application method, diet, soil moisture content, and time since manure application on VOC emissions. Manure was collected from feedlot pens where cattle were fed diets containing 0, 10, or 30% wet distillers grains with solubles (WDGS). Land application methods included surface-applying manure (i.e., no-tillage) or incorporating manure using disk tillage. The effects of soil moisture content on VOC emissions was determined by adding water to each of the plots approximately 24 h after manure application. Isovaleric acid, butyric acid, and 4-methylphenol contributed 28.9, 18.0, and 17.7%, respectively, of the total measured odor activity values. In general, the largest emissions of volatile fatty acids and aromatics were measured during the initial collection periods on the no-tillage plots under dry soil moisture conditions. Emissions of volatile fatty acids and aromatics were reduced after water additions because these compounds were stored in the soil-water matrix rather than released into the atmosphere. In contrast, sulfide emissions generally increased with the addition of the water, especially on the plots containing manure from the 30% WDGS diet. Sulfur content of manure increases with higher percentages of WDGS feed stock. Application method, diet, soil moisture content, and time since application should be considered when estimating VOC emissions.

A lab-scale study evaluated ammonia (NH3 ) and greenhouse gases, emissions when aluminum sulfate (alum) or a microbial product were added to beef feedlot surface material (FSM). Three kilograms of FSM and 1.5 kg of water were added to stainless steel pans (50 cm × 30 cm × 6.5 cm). Treatments included a control (no amendment), 450 g alum, or 0.5 g microbial product. The pans were placed in an environmental chamber that was maintained at 23°C and 50%-60% humidity. Emissions were measured three times weekly for 3 weeks. Ammonia, N2 O, and CH4 were lower (p < 0.01) from pans with amendments compared to the control pans; emissions from the FSM that received the microbial treatment were lower than the FSM treated with alum. Methane emissions were similar for the control and alum-treated FSM but lower (p < 0.01) for the FSM that was treated with the microbial product. Nitrate-N was lower, and NH4+ -N and total sulfur concentrations were higher in FSM treated with alum compared to no treatment or the microbial treatment (p < 0.01). Results indicate that both alum and microbial amendments have benefits in reducing emissions from the feedlot surface, with the microbial product providing additional reductions in emissions compared to the alum.

Length of the menstrual cycle was positively associated with antral follicle number in women. If this pattern is consistent in cattle, a value-added benefit to using automated activity monitors to determine estrous status could be the ability to predict antral follicle count (AFC). We, therefore, hypothesized that as inter-estrous interval increased ultrasonographic AFC would be greater in crossbred beef heifers. Over 3 yr, crossbred beef heifers (n = 1,394) were fitted with automated activity monitors for 81 d. From days 42 to 46, heifers were submitted for ultrasonographic examination to determine AFC. From days 60 to 81, heifers were visually observed twice daily for 45 min for signs of behavioral estrus. Heifers that had a behavioral estrus that coincided with a sensor-based estrus and had a previous sensor-based estrus between 15 and 26 d earlier were used for the analysis (n = 850). A combination of regression analyses and correlation analyses were applied to understand the association between data collected by sensors and follicle number determined by ultrasonographic examination. Antral follicle count was analyzed using the GLM procedure of SAS with estrous cycle length (15 to 26 d) as a fixed effect. Estrus was more likely to initiate in the early morning hours and peak activity was greater (P < 0.0001) when estrus initiated between 0200 and 0800 hours then when estrus initiated at other times of the day. Antral follicle count did not differ due to length of the estrous cycle (P = 0.87). Thus, length of the estrous cycle obtained from three-axis accelerometers cannot be used to predict follicle number in crossbred beef heifers; however, machine learning approaches that combine multiple features could be used to integrate parameters of activity with other relevant environmental and management data to quantify AFC and improve reproductive management in beef cows.

Due to the use of antimicrobials in livestock production, residual antimicrobials and antimicrobial resistance genes (ARGs) could enter the environment following the land application of animal wastes and could further contaminate surface and groundwater. The objective of this study was to determine the effect of various manure land application methods on the fate and transport of antimicrobials and ARGs in soil and runoff following land application of swine manure slurry. Swine manure slurries were obtained from facilities housing pigs that were fed chlortetracyline, tylosin or bacitracin and were land applied via broadcast, incorporation, and injection methods. Three rainfall simulation tests were then performed on amended and control plots. Results show that land application methods had no statistically significant effect on the aqueous concentrations of antimicrobials in runoff. However, among the three application methods tested broadcast resulted in the highest total mass loading of antimicrobials in runoff from the three rainfall simulation tests. The aqueous concentrations of chlortetracyline and tylosin in runoff decreased in consecutive rainfall events, although the trend was only statistically significant for tylosin. For ARGs, broadcast resulted in significantly higher erm genes in runoff than did incorporation and injection methods. In soil, the effects of land application methods on the fate of antimicrobials in top soil were compound specific. No clear trend was observed in the ARG levels in soil, likely because different host cells may respond differently to the soil environments created by various land application methods.

Soil biotic and abiotic factors strongly influence nitrogen (N) availability and increases in nitrification rates associated with the application of manure. In this study, we examine the effects of edaphic properties and a dairy (Bos taurus) slurry amendment on N availability, nitrification rates and nitrifier communities. Soils of variable texture and clay mineralogy were collected from six USDA-ARS research sites and incubated for 28 d with and without dairy slurry applied at a rate of ~300 kg N ha(-1). Periodically, subsamples were removed for analyses of 2 M KCl extractable N and nitrification potential, as well as gene copy numbers of ammonia-oxidizing bacteria (AOB) and archaea (AOA). Spearman coefficients for nitrification potentials and AOB copy number were positively correlated with total soil C, total soil N, cation exchange capacity, and clay mineralogy in treatments with and without slurry application. Our data show that the quantity and type of clay minerals present in a soil affect nitrifier populations, nitrification rates, and the release of inorganic N. Nitrogen mineralization, nitrification potentials, and edaphic properties were positively correlated with AOB gene copy numbers. On average, AOA gene copy numbers were an order of magnitude lower than those of AOB across the six soils and did not increase with slurry application. Our research suggests that the two nitrifier communities overlap but have different optimum environmental conditions for growth and activity that are partly determined by the interaction of manure-derived ammonium with soil properties.

Manure from cattle fed distillers' grain or corn diets was applied to fields, and the fields were subjected to rainfall simulation tests. Manure was added at three rates on till and no-till plots. Correlations between microbial transport and runoff characteristics were identified. Results indicate that diet affects phage but not bacterial transport from manure-amended fields.

Subsurface measures have been adapted to identify manure accumulation on feedlot surfaces. Understanding where manure accumulates can be useful to develop management practices that mitigate air emissions from manure, such as odor or greenhouse gases. Objectives were to determine if electromagnetic induction could be used to predict differences in volatile fatty acids (VFA) and other volatiles produced in vitro from feedlot surface material following a simulated rain event. Twenty soil samples per pen were collected from eight pens with cattle fed two different diets using a predictive sampling approach. These samples were incubated at room temperature for 3 d to determine fermentation products formed. Fermentation products were categorized into acetate, straight-, branched-chained, and total VFAs. These data were used to develop calibration prediction models on the basis of properties measured by electromagnetic induction (EMI). Diet had no significant effect on mean volatile solids (VS) concentration of accumulated manure. However, manure from cattle fed a corn (Zea mays L.)-based diet had significantly ( P ≤ 0.1) greater mean straight-chained and total VFA generation than pens where wet distillers grain with solubles (WDGS) were fed. Alternately, pens with cattle fed a WDGS-based diet had significantly (P ≤ 0.05) greater branched-chained VFAs than pens with cattle fed a corn-based diet. Many branched-chain VFAs have a lower odor threshold than straight-chained VFAs; therefore, emissions from WDGS-based diet manure would probably have a lower odor threshold. We concluded that diets can affect the types and quantities of VFAs produced following a rain event. Understanding odorant accumulation patterns and the ability to predict generation can be used to develop precision management practices to mitigate odor emissions.

Reducing Escherichia coli O157:H7 in cattle and their manure is critical for reducing the risk for human foodborne and waterborne illness. The objective of this study was to evaluate the effects of soil and pond ash surfaces for feedlot pens on the prevalence, levels, and/or persistence of naturally occurring E. coli O157:H7 and total E. coli in cattle (feces and hides) and manure. Cattle (128 beef heifers) were sorted among 16 pens: 8 surfaced with soil and 8 surfaced with pond ash. The prevalence of E. coli O157:H7 in feces decreased (P < 0.0001) during the study from 57.0% on day 0 to 3.9% on day 84 but did not differ (P > or = 0.05) between cattle on soil and on pond ash pens at any sampling period. The prevalence of the pathogen on hides and in feedlot surface material (FSM) also decreased (P < 0.0001), with no effect of soil or pond ash surface (P > or = 0.05). Similarly, levels of E. coli in FSM did not differ (P > or = 0.05) at any sampling period, and there were no clear trends for survival differences of E. coli O157:H7 or E. coli in FSM between pond ash and soil surfaces, although E. coli populations survived at 5.0 log CFU/g of FSM on the pen surfaces 6 weeks after the cattle were removed. These results indicate that housing cattle on pens surfaced with pond ash versus pens surfaced with soil does not affect E. coli O157:H7 in cattle or their manure.

Determining the survival of zoonotic pathogens in livestock manure and runoff is critical for understanding the environmental and public health risks associated with these wastes. The occurrence and persistence of the bacterial pathogens Escherichia coli O157:H7 and Campylobacter spp. in a passive beef cattle feedlot runoff control-vegetative treatment system were examined over a 26-mo period. Incidence of the protozoans Cryptosporidium spp. and Giardia spp. was also assessed. The control system utilizes a shallow basin to collect liquid runoff and accumulate eroded solids from the pen surfaces; when an adequate liquid volume is attained, the liquid is discharged from the basin onto a 4.5-ha vegetative treatment area (VTA) of bromegrass which is harvested as hay. Basin discharge transported E. coli O157, Campylobacter spp., and generic E. coli into the VTA soil, but without additional discharge from the basin, the pathogen prevalences decreased over time. Similarly, the VTA soil concentrations of generic E. coli initially decreased rapidly, but lower residual populations persisted. Isolation of Cryptosporidium oocysts and Giardia cysts from VTA samples was infrequent, indicating differences in sedimentation and/or transport in comparison to bacteria. Isolation of generic E. coli from freshly cut hay from VTA regions that received basin discharge (12 of 30 vs. 1 of 30 control samples) provided evidence for the risk of contamination; however, neither E. coli O157 or Campylobacter spp. were recovered from the hay following baling. This work indicates that the runoff control system is effective for reducing environmental risk by containing and removing pathogens from feedlot runoff.

Odors from livestock operations are a complex mixture of volatile carbon, sulfur, and nitrogen compounds. Currently, detailed volatiles analysis is both time consuming and requires specialized equipment and methods. This work describes a new method that utilizes a dynamic flux chamber, solid-phase microextraction (SPME), and gas chromatography-mass spectroscopy (GC-MS) to describe and compare the odorous compounds emitted from cattle and swine feces. Evaluation of method parameters produced a protocol for comparing relative emissions based on fixed sample temperature (20 degrees C) and exposed surface area (approximately 523 cm(2)), air flow rates (1 L min(-1) or 16 cm s(-1)), SPME exposure time (5 min), and chamber cleaning procedures (70% ethanol rinse and drying for 30 min at 105 degrees C) to minimize cross-contamination between samples. A variety of volatile organic compounds (VOCs) including alcohols, volatile fatty acids, aromatic ring compounds, ketones, esters, and sulfides were routinely detected and the relative emissions from fresh and incubated (37 degrees C overnight) swine and cattle feces were compared as a measure of potential to produce odorants during manure storage. Differences in the types and relative quantities of volatiles emitted were detected when animal species (cattle or swine), diet, fecal incubation, or sample storage conditions (20, 4, or -20 degrees C) were varied.

Cattle feedlot dust is an annoyance and may be a route for nutrient transport, odor emission, and pathogen dispersion, but important environmental factors that contribute to dust emissions are poorly characterized. A general protocol was devised to test feedlot samples for their ability to produce dust under a variety of environmental conditions. A blender was modified to produce dust from a variety of dried feedlot surface and soil samples and collect airborne particles on glass fiber filters by vacuum collection. A general blending protocol optimized for sample volume (150-175 cm3), blending time (5 min of pre-blending), and dust collection time (15 s) provided consistent dust measurements for all samples tested. The procedure performed well on samples that varied in organic matter content, but was restricted to samples containing less than 200 to 700 g H2O kg(-1) dry matter (DM). When applied to field samples, the technique demonstrated considerable spatial variability between feedlot pen sites. Mechanistically, dust potential was related to moisture and organic matter content. An alternative protocol also demonstrated differences within pen sites in maximum dust potential and dust airborne residence time. The two protocols were not intended, nor are they suitable, for predicting actual particulate matter emissions from agricultural sources. Rather, the protocols rapidly and inexpensively compared the potential for dust emission from samples of differing composition under a variety of environmental conditions.

The objective of the research was to determine the optimum application rates of soybean peroxidase (SBP) plus peroxide (SBPP) for reducing odorous VOC emissions from swine manure. Industrial-grade SBP was applied in combination with liquid hydrogen peroxide (H(2)O(2)) or powdered calcium peroxide (CaO(2)) to standard phenolic solutions and swine manure, and emissions were measured in a wind tunnel. The primary odorant in the untreated manure was 4-methylphenol, which accounted for 68-81% of the odor activity value. At the optimum application rate of SBPP (50 g L(-1)), 4-methylphenol emissions were reduced from the swine manure by 62% (H(2)O(2)) and 98% (CaO(2)) after 24h (P<0.0001). The CaO(2) had a longer residence time, remaining effective for 48 h with 92% reduction in emission rates (P<0.0001), while H(2)O(2) was similar to the control at 48 h (P=0.28).
