RESUMO

The homeostasis of histone methylation is maintained by histone methyltransferases and demethylases, which are important for the regulation of gene expression. Here, we report a histone demethylase from rice (Oryza sativa), Jumonji C domain-containing protein (JMJ710), which belongs to the JMJD6 group and plays an important role in the response to drought stress. Overexpression of JMJ710 causes a drought-sensitive phenotype, while RNAi and clustered regularly interspaced short palindromic repeats (CRISPR)-knockout mutant lines show drought tolerance. In vitro and in vivo assays showed that JMJ710 is a histone demethylase. It targets to MYB TRANSCRIPTION FACTOR 48 (MYB48-1) chromatin, demethylates H3K36me2, and negatively regulates the expression of MYB48-1, a positive regulator of drought tolerance. Under drought stress, JMJ710 is downregulated and the expression of MYB48-1 increases, and the subsequent activation of its downstream drought-responsive genes leads to drought tolerance. This research reports a negative regulator of drought stress-responsive genes, JMJ710, that ensures that the drought tolerance mechanism is not mis-activated under normal conditions but allows quick activation upon drought stress.

Bone infection and implant secondary removal remains a clinical challenge. We used ultrasonic micro-arc oxidation (UMAO) and conversion of phytic acid copper plating to prepare a pure magnesium polyhydric biofilm; we evaluated the surface microstructures, phase, element composition, and wettability of the film in vitro. The antibacterial activity of films with different Cu contents was confirmed by coating method, imaging examination, and microbiological cultures in vitro. The biocompatibility of biofilms was confirmed by cell proliferation, vitality, and morphology assays in vitro and histological evaluation in vivo. MC3T3-E1 cells were co-cultured with different films to assess cell viability on the films. The results showed that the mass fraction of Cu increased with increasing time of copper plating, and the surface of the Cu group was more dense and uniform. Additionally, copper coating significantly inhibited the growth of E. coli and Staphylococcus aurous. We also found that the adhesion, proliferation, and differentiation of the cells on the surface of copper plating were enhanced. Copper implantation of animals in vivo showed fine ability to promote bone growth. Antibacterial activity and biocompatibility of pure magnesium UMAO-phytic acid-Cu3min implant film are excellent, so the film has potential application value in the treatment of bone implantation.

An aspherical lens is fabricated from an ultraviolet (UV) curable polymer and is characterized by measuring its focal spot. Electrostatic force is employed to manipulate the shape of the liquid polymer lens. Experiment results show that a liquid lens in a strong electrostatic field can be distorted from initial spherical shape to parabolic to even near cone shape. With in situ measurement of the surface profile and focal spot, an aspherical liquid lens with good performance can be cured to a solid aspherical lens by UV light. A probe scanning microscope is employed to accurately measure the focal spot of the aspherical lens, and the modulation transfer function (MTF) of the aspherical lens is calculated to characterize it. A focal spot of 1.825 microm diameter, an MTF of 800 line pairs/mm cutoff spatial frequency, and a Strehl ratio of 0.742 are achieved. These demonstrate the feasibility of fabricating an aspherical lens with small aberrations by using this method.
