RESUMO

Diabetic retinopathy is one of the common microvascular complications of diabetes, and it is the main cause of vision loss among working-age people. This study interpreted the roles of miR-99a-5p in DR patients and human retinal microvascular endothelial cell (hRMECs) injury induced by high glucose. The expression of miR-99a-5p was detected in patients with NDR, NPDR, and PDR. The indictive impacts of miR-99a-5p were tested by the ROC curve, and the link between miR-99a-5p and clinical information was verified by the Pearson test. HG was used to instruct cell models. The CCK-8 and transwell methods were performed to detect the proliferative and migrated cells. The targeted relationship was explained by luciferase activity. The content of miR-99a-5p was gradually lessened in NPDR and PDR patients. MiR-99a-5p might differentiate DR patients from NDR patients and PDR patients from NPDR patients. The interconnection between miR-99a-5p and clinical factors was endorsed in all DR patients. Overexpression of miR-99a-5p assuaged the abnormality of cell migration and proliferation of hRMECs triggered by HG. NOX4 was a downstream signaling component of miR-99a-5p. In conclusion, MiR-99a-5p protected hRMECs against HG damage, and the miR-99a-5p might be a novel target for diagnosis of DR.

INTRODUCTION: Acute primary angle closure (APAC) is often characterized by acute elevation of intraocular pressure (IOP) accompanied by severe ocular and systemic symptoms. Excessive collagen accumulation, which can be caused by upregulated heat shock protein 47 (HSP47) expression, can produce scarring in rat conjunctival blebs. Meanwhile, the presence of HSP47 in human aqueous humor and its levels are yet to be determined. METHODS: We examined 32 consecutive patients with APAC and 16 age-matched participants without APAC scheduled for cataract surgery who were enrolled as a control group. Aqueous humor samples were collected from all subjects at the time of surgery and compared between the subjects with and without APAC. RESULTS: The levels of HSP47 in the aqueous humor of patients with APAC (1,210.4 ± 450.2 pg/mL) were found to be significantly increased (P = 0.001) compared with those in the control group (863.4 ± 240.0 pg/mL). Notably, the levels of HSP47 negatively correlated with the age of patients with APAC (P = 0.023). CONCLUSION: HSP47 was upregulated in the aqueous humor of patients with APAC and may play a role in scarring after trabeculectomy for APAC.

Alzheimer's disease (AD) is a neurodegenerative disorder known to cause cognitive impairment among the elderly worldwide. Although physical exercise-induced adult hippocampal neurogenesis (AHN) improves cognition, understanding its underlying molecular mechanisms requires further investigation using AD mouse models. In this present work, we subjected amyloid precursor protein (APP)/PS1 mice to a 12-week aerobic treadmill exercise to investigate AHN and its potential mechanisms. We divided 3-month-old littermates wild-type and APP/PS1 transgenic male mice into four groups, and the exercise groups performed 12-week treadmill exercise. Next, we evaluated the influence of treadmill exercise on learning and memory capacity, AHN, and APP proteolytic pathway-related factors. As per our results, the treadmill exercise was able to improve the hippocampal microenvironment in APP/PS1 mice probably by regulating various neurotrophic factors and secretases resulting in APP cleavage through a non-amyloidogenic pathway, which seems to further promote new cell proliferation, survival, and differentiation, enhancing hippocampal neurogenesis. All of these effects ameliorate learning and memory capacity. This study provides a theoretical and experimental basis for understanding AHN in an AD mouse model, which is beneficial for preventing and treating AD.

BACKGROUND: This present study was to explore the association of kinesin family member 2A (KIF2A) expression with clinicopathological features and survival profiles, and the effect of KIF2A on cell proliferation and chemosensitivity in non-small cell lung cancer (NSCLC). METHODS: Tumor and paired adjacent specimens were collected from 380 patients with NSCLC underwent resection for immunohistochemistry assay of KIF2A expression. In vitro, the effect of KIF2A on cell proliferation, chemosensitivity to cisplatin/vinorelbine was detected via KIF2A plasmids transfection into NCI-H1299 NSCLC cells. RESULTS: Kinesin family member 2A expression was upregulated in tumor tissues compared with adjacent tissues, and tumor tissue KIF2A high expression was associated with higher pathological grade (P < .001), larger tumor size (P = .021), lymph node metastasis (P = .044), and increased tumor-node-metastasis stage (P = .001). As for survival profiles, disease-free survival (P < .001) and overall survival (P < .001) were worse in patients with KIF2A high expression compared with those with KIF2A low expression. Multivariate Cox's regression exhibited that KIF2A high expression was an independent predictive factor for lower DFS (P < .001) and OS (P < .001). In vitro, KIF2A promoted proliferation and decreased chemosensitivity to cisplatin but not vinorelbine in NCI-H1299 NSCLC cells. CONCLUSIONS: The correlation of KIF2A expression with tumor features, survival, and its cellular function implies its potential as a prognostic biomarker and a treatment target in NSCLC.

OBJECTIVES: To determine the usefulness of LINC00152 and LARS2-AS1 as potential biomarkers for latent tuberculosis (LTB) and active tuberculosis (ATB), as well as their effect on Mycobacterium (Mtb) infection. METHODS: The expression levels of LINC00152 and LARS2-AS1 in the health, patients with LTB and ATB were detected by qRT-PCR. The ROC curves were constructed to show their potential as biomarkers. The intracellular survival assays for Mtb and the levels of immune-related cytokines were determined to discover the effect of LINC00152 and LARS2-AS1 on Mtb infection. The relationships of miR-485-5p with LINC00152 and LARS2-AS1 were explored. RESULTS: LINC00152 and LARS2-AS1 levels were significantly elevated in patients with ATB and LTB, and Mtb-infected macrophages. LINC00152 and LARS2-AS1 can distinguish the LTB from the health and ATB from LTB. LARS2-AS1 and LINC00152 knock-down reduced the intracellular Mtb survival and induced cellular immune response after Mtb challenge. miR-485-5p was a targeting miRNA for LINC00152 and LARS2-AS1. CONCLUSIONS: LINC00152 and LARS2-AS1 can be considered as potential biomarkers for tuberculosis disease. LINC00152 and LARS2-AS1 have anti-Mtb effects.

BACKGROUND/AIM: Triple-negative breast cancer (TNBC) is the most malignant breast cancer subtype with a short survival time and high morality. There is an urgent need for effective indicators able to predict tumor progression and provide reference for adjusting the therapeutic strategy of TNBC. lncRNA semaphorin 3B antisense RNA1-AS1 (SEMA3B-AS1) was previously identified to be correlated with the stemness and autophagy of breast cancer. SEMA3B-AS1's role in TNBC was investigated in the present study, aiming to explore a novel biomarker for the development and prognosis of TNBC. MATERIALS AND METHODS: SEMA3B-AS1 expression was detected in tissue samples from 113 TNBC patients using PCR. The clinical significance of SEMA3B-AS1 was assessed by the Chi-square test and Cox analysis. The in vitro function of SEMA3B-AS1 was investigated by CCK8 and Transwell assay. Study of molecular mechanism, correlation analysis and dual-luciferase reporter assay were employed to assess the correlation of SEMA3B-AS1 with miR-513c-5p. RESULTS: A significant down-regulation of SEMA3B-AS1 was observed in TNBC, which was related to patient TNM stage, lymph node metastasis, and Ki67 levels. SEMA3B-AS1 down-regulation indicated patients' adverse prognoses and served as an independent prognostic factor. In vitro, SEMA3B-AS1 suppressed the stemness, proliferation, and metastasis of TNBC cells. Moreover, SEMA3B-AS1 negatively regulated miR-513c-5p, which could reverse the inhibitory effects of SEMA3B-AS1 on TNBC cells. CONCLUSION: SEM3B-AS1 indicates the severity of TNBC patients and regulates tumor progression via modulating miR-513c-5p.

Objective: To evaluate and compare the clinical features, imaging, overlapping antibodies, and prognosis of pediatric and adult patients with anti-GFAP antibodies. Methods: This study included 59 patients with anti-GFAP antibodies (28 females and 31 males) who were admitted between December 2019 and September 2022. Results: Out of 59 patients, 18 were children (under 18 years old), and 31 were adults. The overall cohort's median age at onset was 32 years old, 7 for children, and 42 for adults. There were 23 (41.1%) patients with prodromic infection, 1 (1.7%) patient with a tumor, 29 (53.7%) patients with other non-neurological autoimmune diseases, and 17 (22.8%) patients with hyponatremia. Fourteen (23.7%) patients had multiple neural autoantibodies, with the AQP4 antibody being the most common. Encephalitis (30.5%) was the most common phenotypic syndrome. Common clinical symptoms included fever (59.3%), headache (47.5%), nausea and vomiting (35.6%), limb weakness (35.6%), and disturbance of consciousness (33.9%). Brain MRI lesions were primarily located in the cortex/subcortex (37.3%), brainstem (27.1%), thalamus (23.7%), and basal ganglia (22.0%). Spinal cord MRI lesions often involved the cervical and thoracic spinal cord. There was no statistically significant difference in the MRI lesion site between children and adults. Out of 58 patients, 47 (81.0%) had a monophasic course, and 4 died. The last follow-up showed that 41/58 (80.7%) patients had an improved functional outcome (mRS <3), and children were more likely than adults to have no residual disability symptoms (p = 0.001). Conclusion: There was no statistically significant difference in clinical symptoms and imaging findings between children and adult patients with anti-GFAP antibodies; Patients with anti-GFAP antibodies may present with normal MRI findings or delayed MRI abnormalities, and patients with overlapping antibodies were common. Most patients had monophasic courses, and those with overlapping antibodies were more likely to relapse. Children were more likely than adults to have no disability. Finally, we hypothesize that the presence of anti-GFAP antibodies is a non-specific witness of inflammation.

The utilization of atomically confined plasmonic fields has revolutionized the imaging technique. According to the fundamental position-momentum uncertainty principle, such a narrow spatial distribution certainly leads to a broad momentum distribution in the fields, which has however been overlooked. Here we propose a novel exploitation for the momentum distribution by adaptively satisfying the conservation law of momentum in inelastic Raman scatterings in periodic systems, providing a unique optical means of directly measuring the whole phonon dispersions. The proposed technique is particularly useful for measuring phonon dispersions of low-dimensional hydrogen-rich materials, which are completely inaccessible via other techniques. The numerical results for a single all-trans polyacetylene chain demonstrate that all phonon dispersion branches can be conclusively measured from their Raman images for the first time. Our findings highlight a unique advantage of the emerging momentum-based nanophotonics and open the door for exploiting highly confined plasmonic fields in another dimension.

Objective: The aims of this study were to evaluate the treatment adherence and the main factors associated with adherence for Chinese hypertensive patients between urban and rural areas. Participants and Methods: A total of 529 hypertensive patients with 328 from urban areas and 201 from rural areas hospitalized in a tertiary hospital in Xi'an, China during the period from May 1 to December 31, 2021 were invited to participate in the cross-sectional study. The adherence to treatment was assessed by "modified Chinese Hill-Bone compliance to high blood pressure therapy scale". Binary logistic regression analysis was used to analyze independent risk factor for treatment adherence of hypertensive patients. General linear model was used to analyze the association of the independent risk factors to treatment adherence in each subgroup of the scale. Results: The treatment adherence of Chinese hypertensive patients in rural areas was 23.88% and it was significantly lower than that in urban areas (36.59%). Age and gender were the factors that independently associated with treatment adherence of hypertensive patients regardless of urban or rural status. The duration of antihypertensive drugs used was identified to be the factor that independently associated with treatment adherence only in urban patients. The subgroups including reduced sodium intake and medication taking had significant effects on treatment adherence in certain factors. Conclusion: The treatment adherence of Chinese hypertensive patients was low, especially in rural areas. More emphasis should be placed on patients in rural areas. Furthermore, more attention and effective strategies should be designed to address factors affecting treatment adherence in both urban and rural areas. Health programs should focus on education including the importance of taking continuous antihypertensive drugs and low sodium diet.

Nuclear receptor subfamily 4 group A1 (NR4A1) is an orphan nuclear receptor, which is expressed in the majority of cells. NR4A1 expression in peripheral blood mononuclear cells is low during the preclinical stage of multiple sclerosis. Knockout of the Nr4a1 gene in mice can aggravate the symptoms of experimental autoimmune encephalomyelitis (EAE), which is an animal model of multiple sclerosis. In this study, we intragastrically administered the NR4A1 agonist cytosporone B (Csn-B) to mice after inducing EAE. After treatment with Csn-B, the clinical symptoms in the EAE mice were substantially attenuated compared with that in PBS-treated control mice. The percentages of CD4+ T cells and F4/80+ cells in the central nervous system were decreased. In addition, interferon-γ and interleukin-17 production by proinflammatory Th1/Th17 cells in the central nervous system and interferon-γ levels in splenocytes were decreased after Csn-B treatment. These findings suggest that the NR4A1 agonist Csn-B can alleviate nerve injury after EAE induction, and, therefore, may be useful as a potential treatment for multiple sclerosis.

OBJECTIVE: To identify potential tumor antigen by immunoscreening the urinary bladder cancer cDNA library with monoclonal antibodies. METHODS: Monoclonal antibodies were prepared. A cDNA expression library was constructed from bladder cancer cell line BLZ211. Immunogenic proteins were identified by immunoscreening the cDNA library with ten monoclonal antibodies. RESULTS: The cDNA library of BLZ211 cells was established using lambdaZAP as vector. The titer of unamplified cDNA library was 1.39x10(6) pfu/ml with a recombinant rate of 97.72%, and titer of amplified one was 8.4x10(9) pfu/ml. After immunoscreening, ten positive clones representing ten different antigens were identified, which include two proteins with unknown function; coactosin-like 1, eukaryotic translation elongation factor, HNRPA1, histidine triad nucleotide binding protein, KRT7, LCN2, TSTA3, zinc finger protein, C11orf48 and HSPC148. CONCLUSION: The cDNA library was of high quality and can be used in further study. By immunoscreening the bladder cancer cDNA library with ten monoclonal antibodies, we identified ten immunogenic proteins that otherwise would not have been identified as potential diagnostic marker and vaccinogens of bladder cancer using the gene discovery effort.

BACKGROUND: For companies, employee creativity is vital to gaining competitive business advantages. Research regarding creativity has focused on contextual factors such as feedback, but results of studies on the relationship between feedback and creativity are inconsistent; further, only a handful of studies have been carried out from the perspective of coworkers. In this study, we aimed to analyze the association between the coworker feedback environment and creativity, to test the mediating role of feedback monitoring in this relationship and to test the moderating role of self-motivation among employees in China. METHODS: A coworker feedback environment survey, a coworker feedback monitoring questionnaire, a self-motivation scale and a creativity scale were used. The staff submitted 264 questionnaires, of which 235 (74.6%) were completed by the participants. Among the respondents in the sample, 132 (56.2%) were men, and 103 (43.8%) were women. The mean age is 30 and age range from 24-49. Mplus 7.11 software was used to perform descriptive analysis, Spearman's correlation analysis, mediating analysis and moderation analysis. RESULTS: Coworker feedback environment was positively associated with creativity (ß=0.60, p<0.01) after controlling for the demographic variable. Coworker feedback monitoring partially mediated the relationship between coworker feedback environment and creativity (χ2/df=2.5, RMSEA=0.05; CFI=0.93; GFI=0.93). When self-assessment motivation is lower, the positive relationship between coworker feedback monitoring and creativity becomes stronger (ß=0.19); the same relationship becomes weaker (ß=0.01) when self-assessment motivation is higher. CONCLUSION: It was suggested that a coworker feedback environment could improve employee creativity by promoting coworker feedback monitoring; creativity could be improved especially among employees who have lower self-assessment motivation.

The focus of the current work attempts to propose a purely data-based model for generating residuals for non-Gaussian process monitoring purposes, the idea of residual generation is borrowed from the field of model-based fault detection and applied in statistical monitoring, the generated residual instead of the measured variables is thus modeled and monitored. The proposed approach first employs the modified independent component analysis (MICA) algorithm to extract independent components (ICs) from a given dataset. Secondly, through assuming but only one variable is missing at one time, the known data regression (KDR) method dealing with missing data problem is then used for estimating the corresponding ICs. The inconsistency between the actual and estimated ICs is called residual and may present much lower level of non-Gaussianity, in contrast to the actual ICs. Thirdly, a principal component analysis based statistical monitoring model can be utilized for online fault detection based on the generated residual. Finally, the superiority and efficiency of the MICA-KDR approach over its counterparts are validated by implementing comparisons on two industrial processes, the proposed MICA-KDR method is demonstrated to be a comparative alternative in monitoring non-Gaussian processes.
