The aims of this study were to determine the distribution and prevalence of gastroenteritis caused by human adenovirus (HAdV) in children in Yunnan province, China, in 2015-2021 and to identify preventive measures that can be taken to reduce morbidity and mortality in children.HAdV is a significant agent of diarrhea in children, but limited data are available regarding the epidemiology and genetic diversity of HAdV in children with diarrhea in Yunnan province, China. A total of 1754 fecal samples were subjected to real-time RT-PCR to detect and quantify HAdV. Positive samples were further analyzed using next-generation sequencing (NGS), and epidemiological data were analyzed as well.1754 patients with diarrhea were enrolled, of which 1041 were male and 713 were female (M:F ratio: 1.46). Seventy-two stool samples out of 1754 (4.10%) were positive for HAdV. The detection rates of all age groups varied from 2.50-4.78%. The highest incidence of HAdV was observed in children under 2 years of age, especially in children 12-24 months-old. From 2015-2021, the annual detection rate ranged from 1.62-12.26%. HAdV was detected throughout the year, but with marked seasonality. Children were most likely to be positive for HAdV in June and November. We detected HAdV in 15.53% (16/103) of samples collected in June and in 8.19% (14/171) of those collected in November. The entire viral genome was successfully sequenced for 13 of the 72 HAdV-positive samples, and 76.92% (10/13) of these were classified as genotype F41 and 23.08% (3/13) were classified as genotype C2.ConclusionsIn Yunnan province, children of all ages are susceptible to HAdV infection, but there has been marked variation in the yearly prevalence. The highest rate of HAdV detection was in June, followed by November. Priority should be given to disease prevention over the development of targeted antiviral therapies, and effective vaccines for preventing HAdV diarrhea are needed. It is also important to establish a surveillance system to collect relevant clinical and epidemiological data quickly in order to assess the potential risk of HAdV infection in children and to identify epidemic strains for the development of effective vaccines.

BACKGROUND: In China, the recommended treatment regimens for HIV-infected individuals were tenofovir in combination with lamivudine or emtricitabine as NRTIs, efavirenz or rilpivirine as NNRTIs, lopinavir/ritonavir as protease inhibitors, and raltegravir or dolutegravir as INSTIs. The development of drug resistance increases the risk of viral rebound, opportunistic infections, and ultimately treatment failure such that the early detection of resistance is ideal. This study was developed to explore primary drug resistance characteristics and genotypic distributions in newly diagnosed antiretroviral therapy (ART)-naïve HIV-1 patients in Nanjing with the goal of establishing a basis for their individualized treatment in the clinic. METHODS: Samples of serum were collected from newly diagnosed ART-naïve HIV patients from the Second Hospital of Nanjing between May 2021 and May 2022. The HIV-1 integrase (IN), protease (PR), and reverse transcriptase (RT) gene coding sequences were amplified from these samples, sequenced, and assessed for drug resistance-related mutations. RESULTS: Major integrase resistance-related mutations were detected in 4/360 amplified samples, with 5 other patient samples exhibiting accessory resistance mutations. The overall prevalence of PR and RT inhibitor-related transmitted drug resistance mutations (TDRMs) in this patient population was 16.99% (61/359). The most common mutations were non-nucleoside reverse transcriptase inhibitor-related mutations (51/359; 14.21%), followed by those associated with nucleoside reverse transcriptase inhibitors (7/359; 1.95%) and protease inhibitors (7/359; 1.95%). Dual-resistant strains were also observed in a subset of patients. CONCLUSIONS: In summary, this study is the first to have surveyed the prevalence of integrase inhibitor resistance-related mutations and other drug resistance-related mutations among newly diagnosed ART-naïve HIV-positive patients in Nanjing, China. These results highlight the need for further molecular surveillance-based monitoring of the HIV epidemic in Nanjing.

Systemic lupus erythematosus (SLE) is an autoimmune disease associated with an imbalance of T helper 17 (Th17) to regulatory T cells (Tregs). However, the underlying mechanism remains unclear. Increasing evidence suggests that circular RNAs play a crucial role in SLE. Although circETS1 was discovered 30 years ago, detailed exploration of its functions remains limited. In this study, we measured the expression levels of circETS1 in peripheral blood mononuclear cells (PBMCs) and CD4+ T cells of patients with SLE by quantitative polymerase chain reaction. The impact of circETS1 expression on the Th17/Treg balance and underlying mechanism were evaluated using double-luciferase reporter, gain-/loss-of-function, and rescue assays. Receiver operating characteristic (ROC) curve analysis was conducted to assess the diagnostic value of circETS1. Both circETS1 and FOXP3 expression were downregulated in the PBMCs and CD4+ T cells of patients with SLE (n = 28) compared with those in the cells of healthy controls (n = 20). Mechanistically, we found that circETS1 can bind directly to the microRNA miR-1205, acting as a sponge to upregulate the transcription of FOXP3, thereby maintaining the Th17/Treg balance. Notably, ROC analysis showed that the expression level of circETS1 in PBMCs had an area under the curve of 0.873 (95% confidence interval: 0.771-0.976; p < .001) for diagnosing SLE, with a sensitivity of 80.00% and a specificity of 89.29%. Finally, we found negative correlations between the level of circETS1 in PBMCs and disease severity (according to the Systemic Lupus Erythematosus Disease Activity Index) in patients with SLE (r = -0.7712, 95% CI: -0.8910 to -0.5509; p < .001). The imbalance in Th17/Treg cells in SLE may be attributed, in part, to the circETS1/miR-1205/FOXP3 pathway. CircETS1 has potential to serve as a valuable biomarker for the diagnosis and evaluation of SLE.

Water-in-salt (WIS) electrolytes composed of 21 m LiTFSI have recently emerged as a safe and environmentally friendly alternative to conventional organic electrolytes in Li-ion batteries. Several studies have emphasized the relation between the high conductivity of WIS electrolytes and their nanoscale structure. Combining force measurements with a surface forces apparatus and atomic force microscopy, this study describes the nanoheterogeneity of LiTFSI solutions as a function of concentration and distance from a negatively charged (mica) surface. We report various nanostructures coexisting in the WIS electrolyte, whose size increases with concentration and is influenced by the proximity of the mica surface. Two key concentration thresholds are identified, beyond which a transition of behavior is observed. The careful scrutinization on the concentration-dependent nanostructures lays groundwork for designing novel electrolytes in future energy storage devices.

1D tubular micro-nano structural materials have been attracting extensive attention in the microwave absorption (MA) field for their anisotropy feature, outstanding impedance matching, and electromagnetic energy loss capability. Herein, unique double-shelled Sn@Mo2 C/C tubes with porous Sn inner layer and 2D Mo2 C/C outer layer are successfully designed and synthesized via a dual-template method. The composites possess favorable MA performance with an effective absorption bandwidth of 6.76 GHz and a maximum reflection loss value of -52.1 dB. Specifically, the rational and appropriate construction of Sn@Mo2 C/C tubes promotes the multi-path electron transfer in the composites to optimize the dielectric constant and consequently to enhance the capacity of electromagnetic wave energy dissipation. Three mechanisms dominate the MA process: i) the conductive loss resulted from the rapid electron transmission due to the novel 1D hollow coaxial multi-shelled structure, especially the metallic Sn inner layer; ii) the polarization loss caused by abundant heterogeneous interfaces of Sn-Mo2 C/C and Mo2 CC from the precise double-shelled structure; iii) the capacitor-like loss by the potential difference between Mo2 C/C nanosheets. This work hereby sheds light on the design of the 1D hierarchical structure and lays out a profound insight into the MA mechanism.

Magnoliae Officinalis Cortex( MOC),the stem bark of Magnolia officinalis( MO) and M. officinalis var. biloba( MOB),is a main ingredient in more than 200 types of Chinese formulae commonly used in clinics. MO and MOB are widely distributed in China,from Sichuan of the west to Zhejiang province of the east and from Shannxi province in the north to Guangxi province in the south. This review summarizes new findings on geo-heralism of MOC concerning textual research,plants taxonomy,genetic study,chemical study,and pharmacological activity,resulting in the following views. ①The original plants of MOC are suggested to be divided into three geographic clans according to the form of leave and the result of genetic research; ②Concentrations of magnolol,honokiol,magnoloside A,magnoloside B,magnoflorine,and ß-eudesmol in samples collected from different geographic areas are varied;③Samples of MOC produced in Hubei and Sichuan were traditionally regarded as Dao-di herbs,which were called Chuanpo,and the pure haplotype of MOC produced in Hubei may become a genetic index.

The methods of literature researching on species and medical parts of Chinese herbs in traditional Chinese medicine(TCM)classical prescription were discussed. The first thing is to get literature by searching, sorting and selecting literature. Then textural research based on the selected literature was performed in several ways of studying descriptions of botanic features of herbs, studying pictures of herbs, studying archaeological founding, field survey, distribution of producing area, etc. A systemic analysis on both horizontal level and longitudinal level was conducted to get the result of the species and medical parts of Chinese herbs in TCM classical prescription.

Through comprehensive study on the descriptions of dragon's blood in traditional medical books, we pointed out that before Tang Dynasty, Butea monosperma was used as dragon's blood; during Tang and Song Dynasty, Liquidambar formosana. was used as dragon's blood; in Commentaries on the illustrations of Song Dynasty, the author made a mistake by combined the descriptions of Butea monosperma with the descriptions of the prepared medicine of Dracaena cochinchinensis in dragon's blood, and thus the oversea species D. cochinchinensis became the mainstream of dragon's blood source in homeland. Until the foundation of the Republic of China, Daemonorops draco replaced D. cochinchinensis to the main source of dragon's blood.

The achievement of Qimin Yaoshu in the area of herbalogical textual research was illustrated in this paper. Qimin Yaoshu quoted and keeped many contents from ancient agriculture books related in herbalogical literature. It also recorded the information of lots of tropical and subtropical plants. The author bringed forwarded many experiences indistinguishing plants and clarified some long last confused or wrong opinions. The studying philosophy and methods of the authors gave us much enlightenment in our herbalogical textual research. It should be taken into account when the herbalogical textual research was carried out.

Wuyi is one of the Chinese medicine recorded in Shennong's Herbal Classic and many other herbal books during the long history period. Up to now, Wuyi is still an important medicine used for insecticidal and removing food retention. Recent researches indicated that Wuyi also had the function of anti-malaria and treating herpetic simplex keratitis. Therefore, Wuyi had notable value on clinic and development of new medicine. The herbalogical textual research on Wuyi was conducted through investigation of the literature of materia medica during the long historical period from Han Dynasty. It was demonstrated by this work that the original plants of Wuyi include Ulmus macrocarpa Hance and Hemiptelea davidii (Hance) Planch, in which, U. macrocarpa was the quality specie. U. pumila was the false specie which cause confusing. The medicine Wuyi was the processed product made from the nutlet of the plants through fermentation with other additive agents.

It also quoted many contents from lots of ancient agriculture hooks related in herbalogical literature. Many agriculture researchers were encouraged to have done much work in area of herhalogical textual research It also included the materia medica hook Jiuhuang Bencao and Yecaipu therein and promoted their delivery.

All previous literatures about Chinese herbal medicines show distinctive traditional Chinese medicine (TCM) flavors. Compendium of Materia Medica is an influential book in TCM history. The TCM flavor theory and flavor standardization principle in this book has important significance for modern TCM flavor standardization. Compendium of Materia Medica pays attention to the flavor theory, explain the relations between the flavor of medicine and its therapeutic effects by means of Neo-Confucianism of the Song and Ming Dynasties. However,the book has not reflected and further developed the systemic theory, which originated in the Jin and Yuan dynasty. In Compendium of Materia Medica , flavor are standardized just by tasting medicines, instead of deducing flavors. Therefore, medicine tasting should be adopted as the major method to standardize the flavor of medicine.

Honeysuckle flower is a traditional herbal medicine in China Through systemically sorting and studying literature of Chinese medicine, this article pointed out that leech used by the traditional Chinese medicine in ancient time has the features of twist vine, slight purple stem with clothing hair; opposite growing leaves, ovule shape with clothing hair on both side; two flowers growing from one pedicel, labiate corolla with 3.2 cm longth, flower grows from white color to yellow color, each branch axil grows only one pedicel, the involucre is ovoid shape, and the flower season is from mid-March to mid-May. Among all species of caprifoliaceae, only Lonicera japonica Thunb. meets these botanic features. Therefore, L. japonica Thunb. should be used as the orthodox species of herbal honeysuckle flower.

PURPOSE: This study aimed to explore the mechanism by which systemic lupus erythematosus (SLE) activity is promoted through Treg inhibition from the perspective of ceRNA. METHODS: qRT-PCR was used to detect the expressions of circETS1, miR-1205, and FoxP3 in clinical SLE patient samples. Overexpression of circETS1and miR-1205, along with knockdown of miR-1205 and FoxP3 were conducted in CD4+ T cells, while the proliferation of helper T cell 17 (Th17) and regulatory T cell (Treg) was detected. Arescue assay was performed to verify the molecular mechanism of circETS1/miR-1205/Foxp3 mRNA axis in regulating CD4+ T cell differentiation. In the in vivo experiment, the expression of miR-1205 in SLE mice was intervened, and renal function, inflammatory factors, and serum complement were measured. Additionally, Treg/Th17 cell ratio was detected by flow cytometry. RESULTS: In SLE patients, Treg cells were found to decrease, while Th17 cells increased. Transfection with circETS1 overexpression led to CD4+ T cells differentiating into Treg cells, causing an imbalance in the Th17/Treg ratio. Transfection of miR-1205 mimic and si-FoxP3 could reverse the effect of circETS1 overexpression. Moreover, inhibiting the expression of miR-1205 showed therapeutic effects on SLE mice. CONCLUSION: circETS1 inhibits Treg via the miR-1205/FoxP3 axis, thereby promoting SLE activity, which may become a new target for SLE treatment.

Background: The rapid detection of Mycobacterium tuberculosis (MTB) is essential for controlling tuberculosis. Methods We designed a portable thermocycler-based real-time fluorescence loop-mediated isothermal amplification assay (cyp141-RealAmp) using six oligonucleotide primers derived from cyp141 to detect MTB. A combined number of 213 sputum samples (169 obtained from clinically diagnosed cases of pulmonary TB and 44 from a control group without tuberculosis) underwent Acid-fast bacillus (AFB) smear, culture, Xpert MTB/RIF assays, and cyp141-RealAmp assay. Results: By targeting MTB cyp141, this technique could detect as low as 10 copies/reaction within 30 min, and it was successfully rejected by other mycobacteria and other bacterial species tested. Of the 169 patients, there was no statistical difference between the detection rate of cyp141-RealAmp (92.90%, 95% CI: 89.03-96.07) and that of Xpert MTB/RIF (94.67%, 95% CI: 91.28-98.06) (P > 0.05), but both were statistically higher than that of culture (65.68%, 95% CI: 58.52-72.84) (P< 0.05) and AFB (57.40%, 95% CI: 49.94-64.86) (P< 0.05). Both cyp141-RealAmp and Xpert MTB/RIF had a specificity of 100%. Furthermore, a high concordance between cyp141-RealAmp and Xpert MTB/RIF was found (Kappa = 0.89). Conclusion: The cyp141-RealAmp assay was shown to be effective, responsive, and accurate in this study. This method offers a prospective strategy for the speedy and precise detection of MTB.

Dye pollution in the aquatic environment can harm ecosystems and human health. Here, we developed a new green adsorbent by applying an improved drying process. Diatomite was embedded in a network structure formed between chitosan and polyvinyl alcohol without using any crosslinking agent to prepare chitosan-polyvinyl alcohol-diatomite hydrogel beads through alkali solidification. The beads were tested for removing a cationic dye (methylene blue (MB)) from water. The structure of the adsorbent beads was analysed using scanning electron microscopy, energy-dispersive spectroscopy, X-ray diffraction, X-ray photoelectron spectroscopy, and Fourier-transform infrared spectroscopy. The adsorption capacity was investigated, and the results indicated excellent MB adsorption properties. The adsorbents had a rough surface and high swelling capacity of 66.9 g/g. The maximum MB adsorption capacity was 414.70 mg/g, and the adsorption followed the Freundlich isothermal and quasi-second-order kinetic models. The adsorption was an endothermic spontaneous process governed by both intra-particle and external diffusion processes. The proposed adsorption mechanisms involved hydrogen bonding and electrostatic interactions. These adsorbent beads have considerable application potentials owing to their high adsorption capacity, green composition, and non-polluting nature.

Paragonimiasis is a common zoonotic parasitic disease. The retinoic acid-inducible gene I (RIG-I) signaling is very important for the host to recognize invading pathogens (especially viruses and bacteria). However, the role of RIG-I signaling in the early stages of P. proliferus infection remains unclear. Therefore, in this study, Sprague-Dawley (SD) rat models with lung damage caused by P. proliferus were established. Experimental methods including Enzyme-linked Immuno Sorbent Assay (ELISA), real-time fluorescent quantitative polymerase chain reaction (PCR), western blotting, and hematoxylin and eosin (HE) staining were used to explore the mechanisms of lung injury caused by P. proliferus. As a result, the expression of the mRNA and proteins of RIG-I signal-related key target molecules, including RIG-I, tumor necrosis factor (TNF) receptor associated factor 6 (TRAF6), interferon regulatory Factor 7 (IRF7), IPS-1, and downstream C-X-C chemokine ligand 10 (CXCL10), were significantly up-regulated immediately after infection, peaked at 3 or 7 days, and showed a downward trend on after 14 days. The levels of pro-inflammatory cytokines interleukin-1 (IL-1), interferon (IFN)-α, -ß, and -γ, which represent type 1 immune response, gradually increased and reached a peak by 14 days, which was consistent with the changes in the degree of inflammatory damage observed under HE staining of lung tissues. In conclusion, RIG-I signaling is activated in the early stage (before 14 days) of P. proliferus infection, it is inferred that the lung injury of the host may be related to the activation of RIG-I like signaling to induce type I immune response.

RESUMO

Chinese herbal medicine are the main means to prevent and treat diseases in traditional Chinese medicine, thus the quality of traditional Chinese medicinal herbs decides the clinical efficacy. Though the producing areas of genuine medical herbs had been recorded in the Qing and Han Dynasties, genuine medical herbs had not been finally established and thrived until the Ming and Qing Dynasties. Their development in more than 1 000 years was affected by many factors. Thepractice examination and theoretical direction of TCM were the theoretical basis for the generation of genuine medical herbs, while the differences in the ecological environment and herbal germplasm indicated their essential causes. The social development in the Ming and Qing Dynasties was also an intangible impetus driving force that could not be neglected.

Geo-authentic herbs lay stress on their producing areas. The producing areas of most geo-authentic herbs have never changed since the ancient times. However, many other geo-authentic herbs have experienced significant changes in the long history. There are two main causes for the change in producing areas of herbs-change of natural environment and development of human society, which are restricted by each other and play a great role throughout the development process of geo-authentic herbs.

RESUMO

The science of geo-authentic herbs is a characteristic discipline of traditional Chinese medicine established during thousands of years of clinical practices. It has a long history under the guidance of profound theories of traditional Chinese medicine. The words of "geo-authentic product" were derived from an administrative division unit in the ancient times, which layed stress on the good quality of products in particular regions. In ancient records of traditional Chinese medicine, the words of "geo-authentic product" were first found in Concise Herbal Foundation Compilation of the Ming dynasty, and the words of "geo-authentic herbs" were first discovered in Peony Pavilion of the late Ming dynasty. After all, clinical effect is the fundamental evaluation standard of geo-authentic herbs.
