RESUMO

In a recent study, anthocyanins, which have a strong free radical-scavenging activity, were examined for their potential to effectively prevent cancer. However, clinical trials are limited by the purity of the anthocyanin. Multiple methods are used to extract and purify anthocyanins. Based on previous work on Solanum nigrum, which is a widely distributed plant, in this study, DM130 macroporous resin, Sephadex LH20, and a C18 column were used to separate cis-trans anthocyanin isomers. These anthocyanins constitute the majority of total S. nigrum anthocyanins. The results showed that this "DM130-LH20-C18 system" can be used to obtain a cinnamic acid-derived cis-trans anthocyanin, petunidin-3-(p-coumaroyl)-rutinoside-5-glucoside, with a purity of 98.5%, for effective quantitation. In order to determine the antioxidant ability of the petunidin-3-(p-coumaroyl)-rutinoside-5-glucoside cis-trans isomers, three ordinary methods were adopted. The maximum antioxidant ability of the cis-trans anthocyanin was dozens of times higher than that of vitamin C.

Solanum nigrum fruits have been conventionally used in beverages due to their nutritional substances such as minerals, vitamins, amino acids, proteins, sugars, polyphenols, and anthocyanins. The characterization of components and regulatory mechanism of anthocyanins in S. nigrum fruits have rarely been reported. In this study, we determined that the peel and flesh of S. nigrum fruits shared similar HPLC profiles but different contents and total antioxidant activities for anthocyanins. After an efficient purification method, mainly including extraction with pH 1.0 distilled water and then desorption with pH 1.0 95% ethanol after a DM-130 resin adsorption step to obtain more pure anthocyanin extracts, the purity of anthocyanins extracted from S. nigrum fruits reached 56.1%. Moreover, eight anthocyanins from S. nigrum fruit were identified with HPLC-MS/MS for the first time. A typical R2R3-MYB transcription factor gene, SnMYB, was also cloned for the first time by rapid amplification of cDNA ends (RACE)-PCR from S. nigrum. Moreover, the contents of anthocyanins were shown to correlate well (r = 0.93) with the expression levels of SnMYB gene during the fruit's developmental stages. Most significantly, SnMYB gene successfully produced high anthocyanin content (1.03 mg/g) when SnMYB gene was transiently expressed in tobacco leaves. Taken together, S. nigrum fruits are a promising resource for anthocyanin extraction, and SnMYB gene is an activator that positively regulates anthocyanin biosynthesis in S. nigrum.

Mo2C demonstrates excellent performance in catalysis, and it has been found to possess excellent hydrogen evolution reaction (HER) catalytic activity and highly efficient nitrogen fixation. The catalytic activity of Mo2C is greatly influenced and restricted by the preparation method. Sintering and carbon deposition, which affect the catalytic activity of Mo2C, are inevitable in the traditional vapor-solid-solid (VSS) process. In this study, we report the controllable synthesis of α-Mo2C micron flowers by adjusting the growth temperature via a vapor-liquid-solid (VLS) process. The density of the Mo2C micron flowers is closely related to the concentration of Na2MoO4 aqueous solution. The as-grown Mo2C micron flowers dispersed with Pt are validated to be an enhanced collaborative electrocatalyst for HER against Pt/VSS-Mo2C.

Due to the unique physical and chemical properties, transition metal carbides (TMCs) have many potential applications in the fields of energy conversion and catalysis. Chemical vapor deposition (CVD) is a promising method to synthesize TMCs. However, spatially inhomogeneous supply of transition metal precursor vapor in the normal CVD process generally leads to poor control of the morphology and uniformity of the products. Here, we report a vapor-liquid-solid (VLS) growth process where non-volatile Na2MoO4 is used to act as a liquid precursor for the growth of uniform ultrathin Mo2C nanosheets on Al2O3(0001). The morphology of the nanosheets can be controlled by tuning the precursor concentration, annealing time and growth temperature. The roles of Na and the liquid-solid interface in consolidating Mo atoms and promoting the epitaxial growth of Mo2C nanosheets are demonstrated. Furthermore, we show that the liquid-solid interface can cause the crystalline phase transition of Mo2C nanosheets through verification experiments.

Transition metal nitrides (TMNx) often exhibit high catalytic activity in many important reactions. Due to their low stability in a reaction environment, it remains as a crucial issue to reveal surface active structures in catalytic reactions, particularly for the cases containing both oxidative and reductive gases. Herein, MoN and Mo2N nanosheets have been constructed on Al2O3(0001) and Au foil surfaces, and in situ surface characterizations are performed on the model catalysts in ambient-pressure CO2, H2, and CO2 + H2 gases. In situ Raman spectroscopy and quasi in situ X-ray photoelectron spectroscopy (XPS) analysis indicate that MoO3 and defective MoO3-x overlayers form on both MoN and Mo2N surfaces in CO2, and the surface oxidation occurs under a milder condition on Mo2N than on MoN. Further, a hydrogenated Mo oxide (HzMoO3-y) overlayer forms in a CO2 + H2 atmosphere, as confirmed using quasi in situ XPS and time-of-flight secondary ion mass spectroscopy. The surface analysis over the model nitride catalysts suggests that O and/or H atoms may be incorporated into surface layers to form the active structure in many O and H-containing reactions.

Cotton is one of the most important fiber and oil crops in the world. Chloroplast genomes harbor their own genetic materials and are considered to be highly conserved. Transfer RNAs (tRNAs) act as "bridges" in protein synthesis by carrying amino acids. Currently, the variation and evolutionary characteristics of tRNAs in the cotton chloroplast genome are poorly understood. Here, we analyzed the structural variation and evolution of chloroplast tRNA (cp tRNA) based on eight diploid and two allotetraploid cotton species. We also investigated the nucleotide evolution of chloroplast genomes in cotton species. We found that cp tRNAs in cotton encoded 36 or 37 tRNAs, and 28 or 29 anti-codon types with lengths ranging from 60 to 93 nucleotides. Cotton chloroplast tRNA sequences possessed specific conservation and, in particular, the Ψ-loop contained the conserved U-U-C-X3-U. The cp tRNAs of Gossypium L. contained introns, and cp tRNAIle contained the anti-codon (C-A-U), which was generally the anti-codon of tRNAMet. The transition and transversion analyses showed that cp tRNAs in cotton species were iso-acceptor specific and had undergone unequal rates of evolution. The intergenic region was more variable than coding regions, and non-synonymous mutations have been fixed in cotton cp genomes. On the other hand, phylogeny analyses indicated that cp tRNAs of cotton were derived from several inferred ancestors with greater gene duplications. This study provides new insights into the structural variation and evolution of chloroplast tRNAs in cotton plants. Our findings could contribute to understanding the detailed characteristics and evolutionary variation of the tRNA family.
