OBJECTIVE: This study aimed to translate and culturally adapt the standardized outcomes in nephrology-hemodialysis fatigue (SONG-HD fatigue) scale and to assess the psychometric properties of the Chinese version of the SONG-HD fatigue (C-SONG-HD fatigue) scale. METHODS: Forward and back translations were used to translate the SONG-HD fatigue scale into Chinese. We used the C-SONG-HD fatigue scale to survey Chinese patients undergoing hemodialysis (HD) in China. We examined the distribution of responses and floor and ceiling effects. Cronbach's alpha and McDonald's omega coefficient, intraclass coefficients, and Spearman correlations were used to assess internal consistency reliability, test-retest reliability, and convergent validity, respectively. Responsiveness was also evaluated. RESULTS: In total, 489 participants across southeast China, northwest China, and central China completed the study. The C-SONG-HD fatigue scale had good internal consistency (Cronbach's alpha coefficient 0.861, omega coefficient 0.916), test-retest reliability (intraclass correlation coefficient 0.695), and convergent validity (Spearman correlation 0.691). The analysis of all first-time HD patients did not show notable responsiveness, and only patients with temporary vascular access had good responsiveness with an effect size (ES) of 0.54, a standardized response mean (SRM) of 0.85, and a standard error of measurement (SEM) of 0.77. CONCLUSION: The Chinese version of the SONG-HD fatigue scale showed satisfactory reliability and validity in patients undergoing hemodialysis (HD) in China. It could be used as a tool to measure the fatigue of Chinese HD patients.

OBJECTIVE: To investigate the expression of endothelium tight junction protein Claudin-5 and intercellular adhesion molecule CD99 in solid-pseudopapillary neoplasms (SPN) and neuroendocrine tumors of pancreas (P-NET), and their significance in the differential diagnoses. METHODS: Immunohistochemical staining of Claudin-5 and CD99 was performed in 37 cases SPN and 21 cases of P-NET. RESULTS: Membranous Claudin-5 expression was observed in all cases of SPN but was absent in all cases of P-NET. The difference was significant (P < 0.01). In SPN, 91.9% (34/37) of the cases displayed paranuclear dot-like immunoreactivity for CD99; in contrast, 61.9% (13/21) of the cases of P-NET displayed membranous staining (P < 0.01). There was a positive association between the expression of Claudin-5 and CD99 in SPN (r = 0.421,P = 0.001). CONCLUSIONS: Although the macroscopic and microscopic features of SPN are quite characteristic, they may not allow confident differentiation from P-NET in all cases, especially when these characteristics are not classical. If necessary, immunostaining for Claudin-5 and CD99 can help to differentiate between these entities.

OBJECTIVE: In contrast to the newly-planted plants, through measuring and analyzing the chlorophyll content, photosynthetic characteristics, root activity and enzyme activity of Rehmannia glutinosa in growth stages, the differentiation manifestation of R. glutinosa physiological activity mediated by continuous cropping was studied. METHODS: SPAD-502 chlorophyll meter was used to measure chlorophyll content and LI-6400 portable photosynthetic apparatus to determine plant photosynthetic characteristics. Plant root vigor and enzyme system were measured following reference literature. RESULTS: The problems of Rehmannia caused by continuous cropping had happened since the early stage of its growth period, and lasted throughout the whole growth period. Under the condition of continuous cropping, the chlorophyll content, photosynthetic characteristics and root activity remained at a lower level compared with the newly-planted plants, among which, the chlorophyll content and the root activity (100 days after planting) had significant differences. CONCLUSION: The insufficient photosynthesis source and the reducing of the storage capacity (root tuber) under the condition of continuous cropping might be the main reasons for these problems of R. glutinosa.

Based on the early transcriptome and digital differentially expressed profiling library construction in consecutive monoculture (two-year culturing) Rehmannia glutinosa, we screened and chose the twelve differentially expressed protein genes which might be related with calcium signal system. The spatiotemporal expression of these genes was measured by the real-time quantitative PCR, and the relative expression values of the genes related with calcium signal system in different development stages and tissues of normal growth (one-year culturing) and succession cropping of R. glutinosa (two-year culturing) was elaborated in detail. In addition, disposed succession cropping of R. glutinosa was treated with different levels of calcium signal blocking agents in order to verify the mode of action of calcium signal system on consecutive monoculture problem in R. glutinosa. Among the twelve genes, two calcium channels away from the cytoplasm were down-regulated expressed, while the ten calcium channels toward the cytoplasm were up-regulated expressed. The results implied that succession cropping caused calcium ions flowing from endoplasmic reticulum to cytoplasm. While the key genes in calcium signal respond components such as CBL, CBP, CIBP, PLC, etc. were down-regulated expressed significantly in succession cropping of R. glutinosa which were disposed with calcium signal blocking agents, the extent of the damage was relieved, and approached the normal growth (one-year culturing) level. This result strongly showed that calcium signal system participated in the perceiving, conducting and magnifying processes of succession cropping obstacles of R. glutinosa.

OBJECTIVE: Based on previous study, authors used the suppression subtractive hybridization (SSH) technique to construct the forward and reverse subtractive cDNA libraries of consecutive monoulture Rehmannia glutinosa. Five genes related with consecutive monoculture problem of R. glutinosa were chosen from the each of two subtractive libraries. And their spatiotemporal expression was measured in order to explore the functions in consecutive monoculture problem of R. glutinosa. METHOD: Using the real-time quantitative PCR, we tested the relative expression values of the genes in different development stages and tissues of normal growth (one-year culturing) and consecutive monoculture (two-year culturing) R. glutinosa. RESULT: The five genes (calcium-dependent protein kinase, s-adenosyl-methionine synthetase, Aminocyclopropane-1-carboxylate oxidase, methyltransferase, calpain), which were chosen from the forward library had high expression in consecutive monoculture R. glutinosa, especially in root, and were hardly expression in normal growth R. glutinosa. On the contrary, the other five genes (RNA-dependent RNA polymerase, RNA replicase, DNA-directed RNA polymerase IIa, cyclin D, RNA binding protein) chosen from the reverse library had high expression in one-year R. glutinosa, but were down regulated or shut down in consecutive monoculture R. glutinosa. CONCLUSION: The key genes, which regulate inessential metabolism parthway (such as cyclin D, DNA-directed RNA polymerase IIa), were restrained or shut down in consecutive monoculture R. glutinosa. Calcium and ethylene signaling might played key roles in the formation of consecutive monoculture problem, resulting in disturbing normal metabolic process and syndrome of disease in R. glutinosa appeared in turn.

BACKGROUND: Transjugular intrahepatic portosystemic shunt (TIPS) is widely applied to decrease portal hypertension. Because of the lack of strong evidence, it is controversial whether anticoagulation should be performed after TIPS. This meta-analysis aimed to assess the safety and efficacy of anticoagulation for patients with portal hypertension following TIPS. METHODS: Studies making comparisons between combination treatment and TIPS alone were searched in China National Knowledge Infrastructure, Cochrane Library, PubMed, the Wan Fang electronic databases, and EMBASE, delivered between the earliest accessible date and September 4, 2021. The RevMan version 5.3 was applied to conduct all statistical analyses. I2 index statistic was used to assess heterogeneity. RESULTS: Five eligible studies were selected, and total 707 patients were enrolled. According to the meta-analysis, compared to TIPS alone, TIPS + anticoagulation led to much lower incidence of portal vein thrombosis (PVT; odds ratio [OR] = 0.39, 95% confidence interval [CI] 0.18-0.84, P = .02) as well as low heterogeneity (P = 0.36, I2 = 3%). Other index like the stent dysfunction rate (OR = 1.27, 95% CI 0.71-2.77, P = .42), bleeding rate (OR = 1.27, 95% CI 0.71-2.77, P = .42), and incidence of hepatic encephalopathy (OR = 0.87, 95% CI 0.56-1.36, P = .55) showed no statistical significance. CONCLUSIONS: In certain patients with portal hypertension, anticoagulation following TIPS may not be required. However, for patients who do not have a PVT before TIPS, post-TIPS anticoagulation can decrease the incidence of PVT. Nonetheless, further research is still required.

A chip-based spectral-domain optical coherence tomography (SD-OCT) system consists of a broadband source, interferometer, and spectrometer. The optical power divider flatness in the interferometer's wavelength is crucial to higher signal-to-noise ratios. A Mach-Zehnder directional coupler (MZDC) structure could be utilized to smoothly maximize the splitting ratio of 50:50 on a silicon platform, with a sub-micrometer of decoupler optical path difference insensitive to the process variation up to 20 nanometers. However, the optical signal reflected from the reference and sample will go back to the same interferometer MZDC. The so-called bidirectional coupler MZDC will not illustrate a flat optical power response in the operating wavelength range but could still demonstrate at least 20 dB signal-to-noise ratio improvement in OCT after the echelle grating spectrum compensation is applied. For maintaining the axial resolution and sensitivity, the echelle grating is also insensitive to process shifts such as MZDC and could be further utilized to compensate a 3 dB bidirectional MZDC structure for a broad and flat 100 nm wavelength response in the interferometer-based on-chip SD-OCT.

OBJECTIVE: To investigate the features of the multifocal electroretinogram (mfERG) first-order kernel P(1) wave in anisometropic amblyopic eyes. METHODS: mfERG of fifteen anisometropic amblyopic eyes and their normal control eyes were recorded with ROLAND RETIscan visual evoked response system, the mean amplitude density and eclipse period of the P(1) wave were analyzed and compared between the amblyopic and normal eyes with t-test. RESULTS: The P(1) wave amplitude density of mfERG first-order kernel in amblyopic eyes (164.7 ± 73.1) nV×deg(-2) was significantly attenuated in the central region of the visual field as compared with that of the control eyes (227.0 ± 61.3) nV×deg(-2) (t = 2.554, P = 0.016). The eclipse period of the amblyopic eyes (30.3 ± 4.3) ms was shorter than that of the control eyes (34.4 ± 3.2) ms (t = 2.92, P = 0.007). CONCLUSIONS: The significant change of the multifocal electroretinogram (mfERG) first-order kernel P(1) wave may reflect the abnormality of the retinal on-bipolar cells function and the visual information transmission, whether the P(1) wave could be used as an objective index of amblyopic eyes is a promising research topic.

Limited data are available on seasonal associations of polycyclic aromatic hydrocarbons (PAHs) exposure with oxidative DNA damage. We conducted a pilot study with 20 postgraduates, and measured urinary levels of mono-hydroxyl PAHs (OH-PAHs) and 8-hydroxy-2'-deoxyguanosine (8-OHdG) for 7 consecutive days in the four seasons. We assessed the relationships of urinary OH-PAHs with urinary 8-OHdG in the whole year as well as cold- and warm-seasons. Summed OH-PAHs (∑OH-PAHs) were higher in cold season than in warm season. Each ln-unit (ln-transformed unit) increase in ∑OH-PAHs in the whole year corresponded to a 34%, 16% or 23% increase in urinary 8-OHdG levels at lag0, lag1 or lag2 day as well as a 26% increase in urinary 8-OHdG levels at lag0-2 days (cumulative effects). Each ln-unit increase in ∑OH-PAHs corresponded to a 36%, 26% or 46% increase in urinary 8-OHdG levels in cold season at lag0 day, lag1 day or lag2 day as well as a 36% increase in urinary 8-OHdG in warm season at lag0 day. Distributed non-linear cumulative lag models (DLNMs) indicated that each ln-unit increase in ∑OH-PAHs within the range of 5.7-8.1 nmol/mmol Cr had a stronger effect (coefficient ß: 1.11-2.97 nmol/mmol Cr) on urinary 8-OHdG rather than non-cumulative DLNMs (coefficient ß: 1.08-1.43 nmol/mmol Cr) as well as the non-linear dose-response relationships of ∑OH-PAHs with urinary 8-OHdG. PAHs exposure exhibited the lagged and cumulative effects on urinary 8-OHdG levels.

Lung cancer is the most commonly occurring cancer attributed to the leading cause of cancer-related deaths globally. Non-small cell lung cancer (NSCLC) comprises 85% to 90% of lung cancers. The survival rate of patients with advanced stage NSCLC is in months. Moreover, the underlying molecular mechanisms still remain to be understood.We used 2 sets of microarray data in combination with various bioinformatic approaches to identify the differentially expressed genes (DEGs) in NSCLC patients.We identified a total of 419 DEGs using the Limma package. Gene set enrichment analysis demonstrated that "Citrate cycle (TCA cycle)," "RNA degradation," and "Pyrimidine metabolism" pathways were significantly enriched in the NSCLC samples. Gene Ontology annotations of the 419 DEGs primarily comprised "glycosaminoglycan binding," "cargo receptor activity," and "organic acid binding." Kyoto Encyclopedia of Genes and Genomes analysis revealed that DEGs were enriched in pathways related to "Malaria," "Cell cycle," and "IL-17 signaling pathway." Protein protein interaction network analysis showed that the hub genes constituted of CDK1, CDC20, BUB1, BUB1B, TOP2A, CCNA2, KIF20A, CCNB1, KIF2C, and NUSAP1.Taken together, the identified hub genes and pathways will help understand NSCLC tumorigenesis and develop prognostic markers and therapeutic targets against NSCLC.

BACKGROUND: The C242T polymorphisms of P22(phox) and plasma vitamin E have been associated with the risk of coronary heart disease (CHD) in several studies, but the results have been inconsistent. In this study, we sought to examine potential interactions between P22(phox) genotypes, plasma vitamin E concentrations, and smoking in relation to CHD risk. METHODS: We determined C242T genotype frequency in the P22(phox) gene and plasma levels of vitamin E in 565 Chinese patients with CHD and 609 control subjects. Logistic regression was used to control for potential confounders in multivariate analyses. RESULTS: Compared with subjects with a CC genotype, subjects with a CT or TT genotype had a lower risk of CHD (multivariate-adjusted odds ratio [OR] 0.45, 95% CI 0.28-0.74, P = .001). Plasma concentrations of vitamin E were lower in case patients than in control subjects (multivariate-adjusted OR 0.65, 95% CI 0.50-0.86, P = .025). Compared with nonsmokers with a CC genotype, nonsmokers with a CT or TT genotype had a decreased risk of CHD (OR 0.25, 95% CI 0.12-0.53), but smokers with a CT or TT genotype had an increased risk (OR 2.04, 95% CI 0.74-5.61, P for interaction = .039). Smokers with a lower vitamin E concentration had a >3.0-fold greater risk of CHD than did nonsmokers with a higher vitamin E concentration (OR 3.52, 95% CI 2.36-5.24, P for interaction = .041). CONCLUSIONS: These data provide evidence that P22(phox) genotypes are significantly associated with CHD risk in a Chinese population and suggest potential interactions among smoking, P22(phox) genotypes, and vitamin E in relation to CHD risk.

The breast cancer stem cells (BCSC) play important roles in breast cancer occurrence, recurrence and metastasis. However, the role of estrogen signaling, a signaling pathway important in development and progression of breast cancer, in regulation of BCSC has not been well established. Previously, we identified and cloned a variant of estrogen receptor α, ER-α36, with a molecular weight of 36 kDa. ER-α36 lacks both transactivation domains AF-1 and AF-2 of the 66 kDa full-length ER-α (ER-α66) and mediates rapid estrogen signaling to promote proliferation of breast cancer cells. In this study, we aim to investigate the function and the underlying mechanism of ER-α36-mediated rapid estrogen signaling in growth regulation of the ER-positive breast cancer stem/progenitor cells. ER-positive breast cancer cells MCF7 and T47D as well as the variants with different levels of ER-α36 expression were used. The effects of estrogen on BCSC's abilities of growth, self-renewal, differentiation and tumor-seeding were examined using tumorsphere formation, flow cytometry, indirect immunofluorence staining and in vivo xenograft assays. The underlying mechanisms were also studied with Western-blot analysis. We found that 17-ß-estradiol (E2ß) treatment increased the population of ER-positive breast cancer stem/progenitor cells while failed to do so in the cells with knocked-down levels of ER-α36 expression. Cells with forced expression of recombinant ER-α36, however, responded strongly to E2ß treatment by increasing growth in vitro and tumor-seeding efficiency in vivo. The rapid estrogen signaling via the AKT/GSK3ß pathway is involved in estrogen-stimulated growth of ER-positive breast cancer stem/progenitor cells. We concluded that ER-α36-mediated rapid estrogen signaling plays an important role in regulation and maintenance of ER-positive breast cancer stem/progenitor cells.

BACKGROUND: Di-(2-ethylhexyl) phthalate (DEHP) is a common plasticizer used in industrial and diverse consumer products. Animal studies indicate DEHP caused developmental, reproductive, and hepatic toxicities. However, human studies of the potential effects of DEHP are limited. METHODS: The exposed site with a history of over 20 years of waste plastic recycling was located in Hunan Province, China. The reference site without known DEHP pollution source was about 50 km far away from the exposed site. In this study, 181 workers working in plastic waste recycling and 160 gender-age matched farmers were recruited. DEHP concentrations in water and cultivated soil samples, serum thyroid-stimulating hormone, malondialdehyde (MDA), superoxide dismutase (SOD), urinary 8-hydroxy-2'-deoxyguanosine (8-OHdG), and micronuclei frequency in human capillary blood lymphocytes were analyzed. RESULTS: Mean levels of DEHP were greater in environment at the recycling site than at reference site (industry wastewater for the exposed: 42.43 µg/l; well water: 14.20 vs. 0.79 µg/l, pond water: 135.68 vs. 0.37 µg/l, cultivated soil: 13.07 vs. 0.81 mg/kg, p < 0.05 for all). The workers had higher median levels of MDA (3.80 vs. 3.14 nmol/ml) and urinary 8-OHdG (340.37 vs. 268.18 µmol/mol creatinine) and decreased SOD activities (112.15 vs. 123.82 U/ml) than the reference group (p < 0.01 for all). Multivariate analysis revealed that the history of working in waste plastic recycling was an independent risk factor for the increased urinary 8-OHdG levels in the male workers (p < 0.01). CONCLUSIONS: The occupational DEHP exposure might contribute to oxidative deoxyribonucleic acid damage in the male workers.

BACKGROUND/PURPOSE: The efficacy of cosmetics on human skin measured under normal mild laboratory environment might be discounted by exterior environment factors such as wind, UV exposure, etc. Few studies have focused on the 'genuine' efficacy of cosmetics on human skin during exposure to external rigorous environment. The aim of this study was to develop a model for the evaluation of the efficacy of cosmetics on human skin under simulated rigorous environment. METHODS: We measured skin water content and transepidermal water loss (TEWL) before and after products application under simulated windy and sub-erythema UV exposure treatment in a constant temperature and humidity laboratory. RESULTS: The results showed that the products had higher water content and lower TEWL at 2, 4 and 8 h of products application, and ameliorated the skin moisturization situation after a 3-week continuous use of the products. In addition, the products might protect the skin to maintain the normal water content and TEWL under the simulated windy or sub-erythema UV exposure treatment in our laboratory when compared with untreated area. CONCLUSION: Our data indicate that this model may provide a more accurate evaluation for the genuine moisturizing effect of cosmetics under external natural climate.
