Here, we screened a 10,371 library of diverse molecules using a drug-sensitive fission yeast strain to identify compounds which cause defects in chromosome segregation during mitosis. We identified a phosphorium-ylide-based compound Cutin-1 which inhibits nuclear envelope expansion and nuclear elongation during the closed mitosis of fission yeast, and showed that its target is the ß-subunit of fatty acid synthase. A point mutation in the dehydratase domain of Fas1 conferred in vivo and in vitro resistance to Cutin-1. Time-lapse photomicrography showed that the bulk of the chromosomes were only transiently separated during mitosis, and nucleoli separation was defective. Subsequently sister chromatids re-associated leading to chromosomal mis-segregation. These segregation defects were reduced when the nuclear volume was increased and were increased when the nuclear volume was reduced. We propose that there needs to be sufficient nuclear volume to allow the nuclear elongation necessary during a closed mitosis to take place for proper chromosome segregation, and that inhibition of fatty acid synthase compromises nuclear elongation and leads to defects in chromosomal segregation.

Face aftereffects are well established for static stimuli and have been used extensively as a tool for understanding the neural mechanisms underlying face recognition. It has also been argued that adaptive coding, as demonstrated by face aftereffects, plays a functional role in face recognition by calibrating our face norms to reflect current experience. If aftereffects tap high-level perceptual mechanisms that are critically involved in everyday face recognition then they should also occur for moving faces. Here we asked whether face identity aftereffects can be induced using dynamic adaptors. The face identity aftereffect occurs when adaptation to a particular identity (e.g., Dan) biases subsequent perception toward the opposite identity (e.g., antiDan). We adapted participants to video of real faces that displayed either rigid, non-rigid, or no motion and tested for aftereffects in static antifaces. Adapt and test stimuli differed in size, to minimize low-level adaptation. Aftereffects were found in all conditions, suggesting that face identity aftereffects tap high-level mechanisms important for face recognition. Aftereffects were not significantly reduced in the motion conditions relative to the static condition. Overall, our results support the view that face aftereffects reflect adaptation of high-level mechanisms important for real-world face recognition in which faces are moving.

Faces are adaptively coded relative to visual norms that are updated by experience, and this adaptive coding is linked to face recognition ability. Here we investigated whether adaptive coding of faces is disrupted in individuals (adolescents and adults) who experience face recognition difficulties following visual deprivation from congenital cataracts in infancy. We measured adaptive coding using face identity aftereffects, where smaller aftereffects indicate less adaptive updating of face-coding mechanisms by experience. We also examined whether the aftereffects increase with adaptor identity strength, consistent with norm-based coding of identity, as in typical populations, or whether they show a different pattern indicating some more fundamental disruption of face-coding mechanisms. Cataract-reversal patients showed significantly smaller face identity aftereffects than did controls (Experiments 1 and 2). However, their aftereffects increased significantly with adaptor strength, consistent with norm-based coding (Experiment 2). Thus we found reduced adaptability but no fundamental disruption of norm-based face-coding mechanisms in cataract-reversal patients. Our results suggest that early visual experience is important for the normal development of adaptive face-coding mechanisms.

INTRODUCTION: The accurate discrimination of another person's eye-gaze direction is vital as it provides a cue to the gazer's focus of attention, which in turn supports joint attention. Patients with schizophrenia have shown a "direct gaze bias" when judging gaze direction. However, current tasks do not dissociate an early perceptual bias from high-level top-down effects. We investigated early stages of gaze processing in schizophrenia by measuring perceptual sensitivity to fine deviations in gaze direction (i.e., the cone of direct gaze: CoDG) and ability to reflexively orient to locations cued by the same deviations. METHODS: Twenty-four patients and 26 controls completed a CoDG discrimination task that used realistic direct-face images with six fine degrees of deviation (i.e., 3, 6 or 9 pixels to the left and right) and direct gaze, and a gaze cueing task that assessed reflexive orienting to the same fine-grained deviations. RESULTS: Our data showed patients exhibited no impairment in gaze discrimination, nor did we observe a reduced orienting response. CONCLUSIONS: These results suggest that while patients may suffer deficits associated with interpreting another person's gaze, the earliest processes concerned with detecting averted gaze and reflexively orienting to the gazed-at location are intact.

Adults who missed early visual input because of congenital cataracts later have deficits in many aspects of face processing. Here we investigated whether they make normal judgments of facial attractiveness. In particular, we studied whether their perceptions are affected normally by a face's proximity to the population mean, as is true of typically developing adults, who find average faces to be more attractive than most other faces. We compared the judgments of facial attractiveness of 12 cataract-reversal patients to norms established from 36 adults with normal vision. Participants viewed pairs of adult male and adult female faces that had been transformed 50% toward and 50% away from their respective group averages, and selected which face was more attractive. Averageness influenced patients' judgments of attractiveness, but to a lesser extent than controls. The results suggest that cataract-reversal patients are able to develop a system for representing faces with a privileged position for an average face, consistent with evidence from identity aftereffects. However, early visual experience is necessary to set up the neural architecture necessary for averageness to influence perceptions of attractiveness with its normal potency.

Adaptation to facial expressions produces aftereffects that bias perception of subsequent expressions away from the adaptor. Studying the temporal dynamics of an aftereffect can help us to understand the neural processes that underlie perception, and how they change with experience. Little is known about the temporal dynamics of the expression aftereffect. We conducted two experiments to measure the timecourse of this aftereffect. In Experiment 1 we examined how the size of the aftereffect varies with changes in the duration of the adaptor and test stimuli. We found that the expression aftereffect follows the classic timecourse pattern of logarithmic build-up and exponential decay that has been demonstrated for many lower level aftereffects, as well as for facial identity and figural face aftereffects. This classic timecourse pattern suggests that the adaptive calibration mechanisms of facial expression are similar to those of lower level visual stimuli, and is consistent with a perceptual locus for the adaptation aftereffect. We also found that aftereffects could be generated by as little as 1 s of adaptation, and in some conditions lasted for as long as 3200 ms. We extended this last finding in Experiment 2, exploring the longevity of the expression aftereffect by adding a stimulus-free gap of varying duration between adaptation and test. We found that significant expression aftereffects were still present 32 s after adaptation. The persistence of the expression aftereffect suggests that they may have a considerable impact on day-to-day expression perception.

Adults are proficient in extracting identity cues from faces. This proficiency develops slowly during childhood, with performance not reaching adult levels until adolescence. Bodies are similar to faces in that they convey identity cues and rely on specialized perceptual mechanisms. However, it is currently unclear whether body recognition mirrors the slow development of face recognition during childhood. Recent evidence suggests that body recognition develops faster than face recognition. Here we measured body and face recognition in 6- and 10-year-old children and adults to determine whether these two skills show different amounts of improvement during childhood. We found no evidence that they do. Face and body recognition showed similar improvement with age, and children, like adults, were better at recognizing faces than bodies. These results suggest that the mechanisms of face and body memory mature at a similar rate or that improvement of more general cognitive and perceptual skills underlies improvement of both face and body recognition.

The face perception system partly owes its efficiency to adaptive mechanisms that constantly recalibrate face coding to our current diet of faces. Moreover, faces that are better attended produce more adaptation. Here, we investigated whether the social cues conveyed by a face can influence the amount of adaptation that face induces. We compared the magnitude of face identity aftereffects induced by adaptors with direct and averted gazes. We reasoned that faces conveying direct gaze may be more engaging and better attended and thus produce larger aftereffects than those with averted gaze. Using an adaptation duration of 5 s, we found that aftereffects for adaptors with direct and averted gazes did not differ (Experiment 1). However, when processing demands were increased by reducing adaptation duration to 1 s, we found that gaze direction did affect the magnitude of the aftereffect, but in an unexpected direction: Aftereffects were larger for adaptors with averted rather than direct gaze (Experiment 2). Eye tracking revealed that differences in looking time to the faces between the two gaze directions could not account for these findings. Subsequent ratings of the stimuli (Experiment 3) showed that adaptors with averted gaze were actually perceived as more expressive and interesting than adaptors with direct gaze. Therefore it appears that the averted-gaze faces were more engaging and better attended, leading to larger aftereffects. Overall, our results suggest that naturally occurring facial signals can modulate the adaptive impact a face exerts on our perceptual system. Specifically, the faces that we perceive as most interesting also appear to calibrate the organization of our perceptual system most strongly.

Facial expression is theorized to be visually represented in a multidimensional expression space, relative to a norm. This norm-based coding is typically argued to be implemented by a two-pool opponent coding system. However, the evidence supporting the opponent coding of expression cannot rule out the presence of a third channel tuned to the center of each coded dimension. Here we used a paradigm not previously applied to facial expression to determine whether a central-channel model is necessary to explain expression coding. Participants identified expressions taken from a fear/antifear trajectory, first at baseline and then in two adaptation conditions. In one condition, participants adapted to the expression at the center of the trajectory. In the other condition, participants adapted to alternating images from the two ends of the trajectory. The range of expressions that participants perceived as lying at the center of the trajectory narrowed in both conditions, a pattern that is not predicted by the central-channel model but can be explained by the opponent-coding model. Adaptation to the center of the trajectory also increased identification of both fear and antifear, which may indicate a functional benefit for adaptive coding of facial expression.

Adult face perception mechanisms are tuned to upright faces, and this orientation selectivity is central to adult expertise with upright faces. Children are less expert than adults, and it has been argued that their face mechanisms are less orientation selective than those of adults. Here we used face aftereffects to test this hypothesis by examining whether children's aftereffects show greater transfer across changes in orientation than do adults' aftereffects. We adapted 7- to 8-year-old children and adults to figural face distortions in both upright and inverted orientations and examined the size of resulting aftereffects in both upright and inverted test faces. If children's face mechanisms are less orientation selective than those of adults, then children's aftereffects should transfer more strongly across changes in orientation. We found no evidence to support this prediction. Children's and adults' aftereffects were similarly reduced by orientation differences between adapt and test. These results indicate that children, like adults, show a high degree of orientation selectivity in face shape coding and suggest that neural tuning to face orientation may be mature by 8 years of age. Our findings are consistent with an emerging view that many of the key attributes of specialized face perception emerge much earlier in development than previously thought.

People with autism and higher levels of autistic traits often have difficulty interpreting facial emotion. Research has commonly investigated the association between autistic traits and expression labeling ability. Here, we investigated the association between two relatively understudied abilities, namely, judging whether expressions reflect genuine emotion, and using expressions to make social approach judgements, in a nonclinical sample of undergraduates at an Australian university (N = 149; data collected during 2018). Autistic traits were associated with more difficulty discriminating genuineness and less typical social approach judgements. Importantly, we also investigated whether these associations could be explained by the co-occurring personality trait alexithymia, which describes a difficulty interpreting one's own emotions. Alexithymia is hypothesized to be the source of many emotional difficulties experienced by autistic people and often accounts for expression labeling difficulties associated with autism and autistic traits. In contrast, the current results provided no evidence that alexithymia is associated with differences in genuineness discrimination and social approach judgements. Rather, differences varied as a function of individual differences in specific domains of autistic traits. More autistic-like social skills and communication predicted greater difficulty in genuineness discrimination, and more autistic-like social skills and attention to details and patterns predicted differences in approach judgements. These findings suggest that difficulties in these areas are likely to be better understood as features of the autism phenotype than of alexithymia. Finally, results highlight the importance of considering the authenticity of emotional expressions, with associations between differences in approach judgements being more pronounced for genuine emotional expressions. Research on individual differences in face recognition has provided important foundational insights: their broad range, cognitive specificity, strong heritability, and resilience to change. Elusive, however, has been the key issue of practical relevance: do these individual differences correlate with aspects of life that go beyond the recognition of faces, per se? Though often assumed, especially in social realms, such correlates remain largely theoretical, without empirical support. Here, we investigate an array of potential social correlates of face recognition. We establish social relationship quality as a reproducible correlate. This link generalises across face recognition tasks and across independent samples. In contrast, we detect no robust association with the sheer quantity of social connections, whether measured directly via number of social contacts or indirectly via extraversion-related personality indices. These findings document the existence of a key social correlate of face recognition and provide some of the first evidence to support its practical relevance. At the same time, they challenge the naive assumption that face recognition relates equally to all social outcomes. In contrast, they suggest a focused link of face recognition to the quality, not quantity, of one's social connections.

Gauging the systemic effects of non-synonymous single nucleotide polymorphisms (nsSNPs) is an important topic in the pursuit of personalized medicine. However, it is a non-trivial task to understand how a change at the protein structure level eventually affects a cell's behavior. This is because complex information at both the protein and pathway level has to be integrated. Given that the idea of integrating both protein and pathway dynamics to estimate the systemic impact of missense mutations in proteins remains predominantly unexplored, we investigate the practicality of such an approach by formulating mathematical models and comparing them with experimental data to study missense mutations. We present two case studies: (1) interpreting systemic perturbation for mutations within the cell cycle control mechanisms (G2 to mitosis transition) for yeast; (2) phenotypic classification of neuron-related human diseases associated with mutations within the mitogen-activated protein kinase (MAPK) pathway. We show that the application of simplified mathematical models is feasible for understanding the effects of small sequence changes on cellular behavior. Furthermore, we show that the systemic impact of missense mutations can be effectively quantified as a combination of protein stability change and pathway perturbation.

Many aspects of faces derived from structural information appear to be neurally represented using norm-based opponent coding. Recently, however, Zhao, Seriès, Hancock, and Bednar (2011) have argued that another aspect with a strong structural component, namely face gender, is instead multichannel coded. Their conclusion was based on finding that face gender aftereffects initially increased but then decreased for adaptors with increasing levels of gender caricaturing. Critically, this interpretation rests on the untested assumption that caricaturing the differences between male and female composite faces increases perceived sexual dimorphism (masculinity/femininity) of faces. We tested this assumption in Study 1 and found that it held for male, but not female faces. A multichannel account cannot, therefore, be ruled out, although a decrease in realism of adaptors was observed that could have contributed to the decrease in aftereffects. However, their aftereffects likely reflect low-level retinotopic adaptation, which was not minimized for most of their participants. In Study 2 we minimized low-level adaptation and found that face gender aftereffects were strongly positively related to the perceived sexual dimorphism of adaptors. We found no decrease for extreme adaptors, despite testing adaptors with higher perceived sexual dimorphism levels than those used by Zhao et al. These results are consistent with opponent coding of higher-level dimensions related to the perception of face gender.

Background: Healthcare is challenged by a rapidly growing group of patients with multi-morbidity and polypharmacy. Increasing activity and specialization puts pressure on healthcare sectors. Medication errors in cross-sectoral transition of patients are often seen. The aim of the study was to explore drug-related problems (DRPs) in the transition of patients between sectors and to develop and pilot-test a cross-sectoral hospital pharmacist intervention to overcome some of these problems. Methods: DRPs in cross-sectoral transitions were explored from four perspectives; the literature, the primary and secondary healthcare sector and the patients. An intervention was developed from the findings through co-creation between pharmacists, doctors and a nurse. The intervention was piloted and evaluated from data on the included patients and the activities performed. Results: DRPs in transitions from general practice (GP) to hospital were caused by inadequate focus on updating the Shared Medication Record (SMR). For patients being discharged, DRPs were described with multiple facets; for example, missing information on medication changes, lacking patient involvement and problems with dose-dispensed medicine or electronic prescriptions. An intervention with a pharmacist in a shared employment between Hospital Pharmacy and GP was developed and piloted. The intervention included medication reconciliation and updating SMR for patients referred to hospital; and medication review, overview of medication changes and follow-up telephone calls for patients discharged from hospital. The intervention identified and solved several DRPs; in this way, medication errors were avoided. Access to health records in both sectors was important in the identification and resolution of DRPs. Conclusion: DRPs in cross-sectoral transitions are multifaceted and the experiences depend on the point of view. The cross-sectoral hospital pharmacist intervention identified and solved several DRPs and medication errors were avoided. The intervention made sense to both healthcare sectors and patients. Shared employment and unique access to health records in both sectors showed to be of importance in the identification and resolution of DRPs. Adults exhibit neural responses over the visual occipito-temporal area in response to faces that vary in how trustworthy they appear. However, it is not yet known when a mature pattern of neural sensitivity can be seen in children. Using a fast periodic visual stimulation (FPVS) paradigm, face images were presented to 8-to-9-year-old children (an age group which shows development of trust impressions; N = 31) and adult (N = 33) participants at a rate of 6 Hz (6 face images per second). Within this sequence, an 'oddball' face differing in the level of facial trustworthiness compared to the other faces, was presented at a rate of 1 Hz (once per second). Children were sensitive to variations in facial trustworthiness, showing reliable and significant neural responses at 1 Hz in the absence of instructions to respond to facial trustworthiness. Additionally, the magnitude of children's and adults' neural responses was similar, with strong Bayesian evidence that implicit neural responses to facial trustworthiness did not differ across the groups, and therefore, that visual sensitivity to differences in facial trustworthiness can show mature patterns by this age. Thus, nine or less years of social experience, perceptual and/or cognitive development may be sufficient for adult-like neural sensitivity to facial trustworthiness to emerge. We also validate the use of the FPVS methodology to examine children's implicit face-based trust processing for the first time, which is especially valuable in developmental research because this paradigm requires no explicit instructions or responses from participants.

Background and objective: Drug-related problems (DRPs) are often seen when a patient is transitioning from one healthcare sector to another, for example, when a patient moves from the hospital to a General Practice (GP) setting. This transition creates an opportunity for information on medication changes and follow-up plans to be lost. A cross-sectoral hospital pharmacist intervention was developed and pilot-tested in a large GP clinic. The intervention included medication history, medication reconciliation, medication review, follow-up telephone calls, identification of possible DRPs and communication with the GP. It is unknown whether the intervention is transferable to other GP clinics. The aim of the study was to explore similarities and differences between GP clinics in descriptive data and intervention acceptability. Methods: A convergent mixed methods study design was used. The intervention was tested in four GP clinics with differing characteristics. Quantitative data on the GP clinics, patients and pharmacist activities were collected. Qualitative data on the acceptability were collected through focus group interviews with general practitioners, nurses and pharmacists. The Theoretical Framework of Acceptability was used. Results: Overall, the intervention was found acceptable and relevant by all. There were differences between the GP clinics in terms of size, daily physician work form and their use of pharmacists for ad hoc tasks. There were similarities in patient characteristics across GP clinics. Therefore, the intervention was found equally relevant for all of the clinics. Shared employment with unique access to health records in both sectors was important in the identification and resolution of DRPs. Economy was a barrier for further implementation. Conclusions: The intervention was found acceptable and relevant by all; therefore, it was considered transferable to other GP clinics. Hospital pharmacists were perceived to be relevant healthcare professionals to be utilized in GP, in hospitals and in the cross-sectoral transition of patients.


Acceptability of a pharmacist activity for patients transitioning between hospital and general practice Why was the study done? Introduction: Caring for patients with multimorbidity in general practice is increasing in amount and complexity. To integrate care for patients with multimorbidity and to support general practitioners (GPs), the Clinic for Multimorbidity (CM) was established in 2012 at Silkeborg Regional Hospital, Denmark. This case study aims to describe the CM and the patients seen in it. Results: CM is an outpatient clinic that offers a comprehensive one-day assessment of the patient's complete health status and medication. GPs can refer patients with complex multimorbidity (≥2 chronic conditions). It involves collaboration across medical specialties and healthcare professions. The assessment is completed with a multidisciplinary conference and recommendation.In all, 141 patients were referred to the CM between May 2012 and November 2017. The median age was 70 years, 80% had more than five diagnoses, and in median patients had a usage of 11 drugs (IQI, 7-15). Physical and mental health was reported low (SF-12 score: 26 and 42). In median four specialties were involved and 4 examinations (IQI, 3-5) conducted. Conclusion: The CM offers innovative care by bridging and exceeding conventional boundaries of disciplines, professions, organizations, and primary and specialized care. The patients represented a very complex group, requiring many examinations and involvement of several specialists.

Historically, it has been argued that face individuation develops very slowly, not reaching adult levels until adolescence, with experience being the driving force behind this protracted improvement. Here, we challenge this view based on extensive review of behavioural and neural findings. Results demonstrate qualitative presence of all key phenomena related to face individuation (encoding of novel faces, holistic processing effects, face-space effects, face-selective responses in neuroimaging) at the earliest ages tested, typically 3-5 years of age and in many cases even infancy. Results further argue for quantitative maturity by early childhood, based on an increasing number of behavioural studies that have avoided the common methodological problem of restriction of range, as well as event-related potential (ERP), but not functional magnetic resonance imaging (fMRI) studies. We raise a new possibility that could account for the discrepant fMRI findings-namely, the use of adult-sized head coils on child-sized heads. We review genetic and innate contributions to face individuation (twin studies, neonates, visually deprived monkeys, critical periods, perceptual narrowing). We conclude that the role of experience in the development of the mechanisms of face identification has been overestimated. The emerging picture is that the mechanisms supporting face individuation are mature early, consistent with the social needs of children for reliable person identification in everyday life, and are also driven to an important extent by our evolutionary history.

Face recognition difficulties are common in autism and could be a consequence of perceptual atypicalities that disrupt the ability to integrate current and prior information. We tested this theory by measuring the strength of serial dependence for faces (i.e. how likely is it that current perception of a face is biased towards a previously seen face) across the broader autism phenotype. Though serial dependence was not weaker in individuals with more autistic traits, more autistic traits were associated with greater integration of less similar faces. These results suggest that serial dependence is less specialised, and may not operate optimally, in individuals with more autistic traits and could therefore be a contributing factor to autism-linked face recognition difficulties.
