RESUMO

Thousands of tourists visit certain Antarctic sites each year, generating a wide variety of environmental impacts. Scientific knowledge of human activities and their impacts can help in the effective design of management measures and impact mitigation. We present a case study from Barrientos Island in which a management measure was originally put in place with the goal of minimizing environmental impacts but resulted in new undesired impacts. Two alternative footpaths used by tourist groups were compared. Both affected extensive moss carpets that cover the middle part of the island and that are very vulnerable to trampling. The first path has been used by tourists and scientists since over a decade and is a marked route that is clearly visible. The second one was created more recently. Several physical and biological indicators were measured in order to assess the environmental conditions for both paths. Some physical variables related to human impact were lower for the first path (e.g. soil penetration resistance and secondary treads), while other biochemical and microbiological variables were higher for the second path (e.g. ß-glucosidase and phosphatase activities, soil respiration). Moss communities located along the new path were also more diverse and sensitive to trampling. Soil biota (Collembola) was also more abundant and richer. These data indicate that the decision to adopt the second path did not lead to the reduction of environmental impacts as this path runs over a more vulnerable area with more outstanding biological features (e.g. microbiota activity, flora and soil fauna diversity). In addition, the adoption of a new route effectively doubles the human footprint on the island. We propose using only the original path that is less vulnerable to the impacts of trampling. Finally from this process, we identify several key issues that may be taken into account when carrying out impact assessment and environmental management decision-making in the Antarctic area.

Background: Prehospital emergency medical services (EMS) are the main gateway for trauma patients. Recent advances in point-of-care testing and the development of early warning scores have allowed EMS to improve patient classification. We aimed to identify patients presenting with major trauma involving life-saving interventions (LSI) using the modified Sequential Organ Failure Assessment (mSOFA) score in the prehospital scenario, and to compare these results with those of other trauma scores. Methods: This was a prospective, ambulance-based, multicenter, training-validation study in trauma patients who were treated in a prehospital setting and subsequently transported to a hospital. The study involved six Advanced Life Support units, 38 Basic Life Support units, and four hospitals. The primary outcome was LSI performed at the scene or en route and intensive care unit (ICU) admission and all-cause two-day in-hospital mortality. We collected epidemiological variables, creatinine, lactate, base excess, international normalized ratio, and vital signs. Discriminative power (area under the receiver operating characteristic curve [AUC]), calibration (observed vs predicted outcome agreement), and decision-curve analysis (DCA, clinical utility) were used to assess the reliability of the mSOFA in comparison to other scores. Results: Between January 1, 2020-April 30, 2022, a total of 763 patients were selected. The mSOFA score's AUC was 0.927 (95% confidence interval [CI] 0.898-0.957) for LSI, 0.845 (95% CI 0.808-0.882) for ICU admission, and 0.979 (95% CI 0.966-0.991) for two-day mortality. Conclusion: The mSOFA score outperformed the other scores, allowing a quick identification of high-risk patients. The routine implementation in EMS of mSOFA could provide critical support in the decision-making process in time-dependent trauma injuries.

Cyanobacteria are key organisms in the Antarctic ecosystem, but the primary succession of its communities in recently deglaciated soils remains poorly understood. In this study, we surveyed the primary succession of cyanobacterial communities with an in-depth Next Generation Sequencing approach in three Antarctic recently glacier forefields. Despite the similar physicochemical characteristics of the soils, we did not find a common pattern in the distribution of the cyanobacterial communities at the finest level of taxonomic resolution. However, the metabarcoding analysis revealed a common community of 14 cyanobacterial identical sequences in all the studied soils, whose lineages were not restricted to polar or alpine biotopes. These ASVs comprised a relative abundance within the cyanobacterial community of 51.5%-81.7% among the three locations and were also found in two cyanobacterial mats from the Antarctic Peninsula. Our results suggest that (micro)biotic interactions act as a key driver of the community composition and dynamics of Cyanobacteria during the early stages of succession in recently deglaciated soils of Antarctica. A few common genera might play a key role in the ecosystem, due to its ubiquitous presence not only in these soils but also in microbial mats, conforming probably the most widely disperse and dominant single genotypes in Antarctic soils.

The biological activity of marine vertebrates represents an input of nutrients for Antarctic terrestrial biota, with relevant consequences for the entire ecosystem. Even though microbial mats assemble most of the biological diversity of the non-marine Antarctica, the effects of the local macrofauna on these microecosystems remain understudied. Using 16S rRNA gene sequencing, 13C and 15N stable isotopes, and by characterizing the P and N-derived nutrient levels, we evaluated the effects of penguins and other marine vertebrates on four microbial mats located along the Antarctic Peninsula. Our results show that P concentrations, C/N and N/P ratios, and δ15N values of "penguin-impacted" microbial mats were significantly higher than values obtained for "macrofauna-free" sample. Nutrients derived from penguin colonies and other marine vertebrates altered the trophic interactions of communities within microbial mats, as well as the relative abundance and trophic position of meiofaunal groups. Twenty-nine bacterial families from eight different phyla significantly changed with the presence of penguins, with inorganic nitrogen (NH4 + and NO3 -) and δ15N appearing as key factors in driving bacterial community composition. An apparent change in richness, diversity, and dominance of prokaryotes was also related to penguin-derived nutrients, affecting N utilization strategies of microbial mats and relating oligotrophic systems to communities with a higher metabolic versatility. The interdisciplinary approach of this study makes these results advance our understanding of interactions and composition of communities inhabiting microbial mats from Antarctica, revealing how they are deeply associated with marine animals.

Ice-free areas represent less than 1% of the Antarctic surface. However, climate change models predict a significant increase in temperatures in the coming decades, triggering a relevant reduction of the ice-covered surface. Microorganisms, adapted to the extreme and fluctuating conditions, are the dominant biota. In this article we analyze the diversity and composition of soil bacterial communities in 52 soil samples on three scales: (i) fine scale, where we compare the differences in the microbial community between top-stratum soils (0-2 cm) and deeper-stratum soils (5-10 cm) at the same sampling point; (ii) medium scale, in which we compare the composition of the microbial community of top-stratum soils from different sampling points within the same sampling location; and (iii) coarse scale, where we compare communities between comparable ecosystems located hundreds of kilometers apart along the Antarctic Peninsula. The results suggest that in ice-free soils exposed for longer periods of time (millennia) microbial communities are significantly different along the soil profiles. However, in recently (decades) deglaciated soils the communities are not different along the soil profile. Furthermore, the microbial communities found in soils at the different sampling locations show a high degree of heterogeneity, with a relevant proportion of unique amplicon sequence variants (ASV) that appeared mainly in low abundance, and only at a single sampling location. The Core90 community, defined as the ASVs shared by 90% of the soils from the 4 sampling locations, was composed of 26 ASVs, representing a small percentage of the total sequences. Nevertheless, the taxonomic composition of the Core80 (ASVs shared by 80% of sampling points per location) of the different sampling locations, was very similar, as they were mostly defined by 20 common taxa, representing up to 75.7% of the sequences of the Core80 communities, suggesting a greater homogeneity of soil bacterial taxa among distant locations.

The atmosphere plays an important role in the dispersal of microorganisms, as well as in the connectivity of most of the planet's ecosystems. In recent decades, interest in microbial diversity and dispersion in the atmosphere has increased due to its importance in various fields. However, there are few studies on the abundance of airborne microorganisms and the factors, such as meteorology, that affect their distribution. Likewise, the physical-mathematical models attempting to reproduce their possible origins also require integrating some biological features. We collected airborne microorganisms under different meteorological conditions at a sampling station over a 12-day period to expand the knowledge about abundance of airborne microorganisms, their relationship with atmospheric conditions and their possible origins with a biological perspective. Total abundance and size distribution of microorganisms were measured in all samples using epifluorescence techniques. Their possible origins were estimated using refined mathematical simulation models of the air masses back-trajectories considering dry deposition. Our results showed microbial abundance values similar to those found in temperate regions over land surface. In our contribution we report a clear relationship between the abundance and, considered as a whole, local meteorological conditions. Despite most of the captured particles were small spherical microorganisms (diameter < 20 µm), large filamentous microorganisms, surprisingly up to 400 µm, were also found. We demonstrate the possibility that these large microorganisms can have their origin at long distances, showing thus probability of remarkable long dispersal, without ruling out a nearby origin, when their equivalent spherical diameter (ESD) and drying capacity are considered.

Microbial mats are complex communities that represent a large biomass fraction in non-marine Antarctic ecosystems. They confer structure to soils and constitute, by themselves, intricate microecosystems, where a great variety of microorganisms and microfauna contributes to the ecosystem functions. Although in recent years Antarctic microbial mats have been thoroughly investigated, trophic relationships within the communities remain unresolved. We therefore conducted a study of the trophic relationships of a microbial mat from Byers Peninsula, Antarctica, using DNA analysis and stable isotopes as trophic tracers. Our results suggested, based on a Bayesian mixing model, that at least four trophic levels are present within this microecosystem: primary producers (cyanobacteria and diatoms), primary consumers (rotifers and tardigrades), secondary consumers (nematodes) and decomposers (fungi). Nematodes would play a key role as top consumers of the community, connecting the two carbon inputs described into the system, as omnivores at the secondary trophic level. In addition, carbon pathways from primary trophic level to consumers take place quickly during the first 24 h after its incorporation in the primary producers, dispersing across all the trophic levels and reaching secondary consumers in less than 11 days. This suggests that, given the changing physical conditions and presumably short periods of activity, there is a fine temporal coupling among the organisms in the community, minimizing the redundancy in function performance among trophic levels.

CD69 is rapidly upregulated on T cells upon activation. In this work we show that this is also the case for CD69 expression on dendritic cells (DC). Thus, the expression kinetics of CD69 on both cell types is reminiscent of the one of costimulatory molecules. Using mouse models of transgenic T cells, we aimed at evaluating the effect of monoclonal antibody (MAb)-based targeting and gene deficiency of CD69 expressed by either DC or T cells on the extent of antigen (Ag)-specific T cell priming, which could be the result of a putative role in costimulation as well as on DC maturation and Ag-processing and presentation. CD69 targeting or deficiency of DC did not affect their expression of costimulatory molecules nor their capacity to induce Ag-specific T cell proliferation in in vitro assays. Also, CD69 targeting or deficiency of transgenic T cells did not affect the minimal proliferative dose for different peptide agonists in vitro. In in vivo models of transgenic T cell transfer and local Ag injection, CD69 deficiency of transferred T cells did not affect the extent of the proliferative response in Ag-draining lymph nodes (LN). In agreement with these results, CD69 MAb targeting or gene deficiency of Vaccinia-virus (VACV) infected mice did not affect the endogenous formation of virus-specific CD8(+) T cell populations at the peak of the primary immune response. Altogether our results argue against a possible role in costimulation or an effect on Ag processing and presentation for CD69.
