BACKGROUND: High-fiber diets are supplemented with lipids to meet the required energy content, but data on the interactions between dietary fiber (DF) and lipid types on gastrointestinal fermentation in pigs are scant. OBJECTIVES: This study aimed to use a combination of in vivo and in vitro fermentation methodologies to determine the interactive effects of DF and lipid types on short-chain fatty acid (SCFA) production and absorption and organic matter (OM) fermentability in the cecum and colorectal tract of pigs. METHODS: Eight ileal- and cecal-cannulated Yorkshire barrows were fed either pectin- or cellulose-containing diets that were supplemented with either corn oil or beef tallow in 2 independent Youden squares with a 2 × 2 factorial arrangement of treatments (n = 6). Ileal and cecal digesta were collected, freeze-dried, and fermented using inoculum from fresh cecal digesta and feces, respectively, to determine individual SCFA production and absorption and fermentability of OM. RESULTS: Interactions (P < 0.001) between DF and lipid types were observed in which the addition of beef tallow decreased the quantity of cecal and colorectal acetic acid production and cecal acetic absorption, cecal butyric production, predicted cecal OM fermentability, and predicted colorectal propionic acid in pectin diets, but the effects were not observed for cellulose diets. The addition of beef tallow increased (P < 0.001) the production of cecal butyric and propionic acids during in vitro fermentation in cellulose diets and apparent total tract digestibility (ATTD) of OM in pectin diets. CONCLUSIONS: The interactions between DF and lipids on gastrointestinal fermentation largely depend on the degree of saturation of fatty acids in dietary lipids. The addition of beef tallow selectively decreased the production and absorption of individual SCFAs in pectin and cellulose diets but increased cecal butyric and propionic acid production in cellulose diets and the ATTD of OM in pectin diets.

AIMS: Soybean meal (SBM) contributes high-quality dietary protein for pigs. However, it also contains antinutritional factors such as allergenic high molecular weight proteins and non-starch polysaccharides (NSP) that limit its use. Therefore, the objective of this study was to screen and characterize a robust Bacillus sp. from camel dung for soybean meal fermentation to improve the digestibility in growing pigs. METHODS AND RESULTS: Molecular characterization revealed that isolate 9 (hereinafter referred to as "CP-9") was a Bacillus subtilis strain. It secreted cellulase (0.07 U ml-1 ), xylanase (1.91 U ml-1 ), and amylase (2.66 U ml-1 ) into the culture supernatant. Isolate CP-9 showed rapid growth on LB agar plates and grew at a wide range of pH (3.0-9.0) and temperatures (23-50°C) in LB broth. Protein profiling of SBM using SDS-PAGE showed a significant reduction of large globular proteins to small peptides after 48 h of fermentation. On a dry matter basis, neutral detergent fibre (NDF) of the fermented SBM (F-SBM) was decreased by 34.25% (from 9.72 to 7.24%) with an increase in CP content by 16.54% (from 48.74 to 56.80%). Pigs fed with a semi-purified diet formulated with F-SBM as the sole source of crude protein had higher (p < 0.05) apparent ileal digestibility (AID) of DM (80.0 vs. 71.7%), ash (55.6 vs. 36.1%), CP (84.2 vs. 78.3%), NDF (70.9 vs. 66.0%), and ADF (62.4 vs. 53.3%) compared with pigs fed with unfermented soybean meal (UF-SBM). CONCLUSIONS: A novel Bacillus subtilis strain CP-9 was isolated and characterized from camel dung for efficient fermentation of SBM. This bacterium ameliorates physico-chemical characteristics of F-SBM and improved nutrient digestibility in growing pigs. SIGNIFICANCE AND IMPACT OF THE STUDY: Our data suggest that a low-cost solid-state SBM fermentation was developed using this newly isolated bacterium. The resultant F-SBM improved the nutrient digestibility in growing pigs.

The utility of the marama bean (MB) as an alternative protein source to soybean (SB) can be limited by the high concentration of trypsin inhibitors (TI). The physical treatment of MB has the potential to ameliorate the antinutritional activities of TI and modify other chemical components. Thus, this study investigated the effects of physical treatments on the chemical components and trypsin inhibitor activity (TIA) of raw MB and SB. The bean substrates were subjected to each of the following treatment methods: (1) room temperature (20−22 °C) soaking for 24 h; (2) electric stove cooking at 100 °C for 10, 20, and 30 min; (3) steam autoclaving at a temperature of 110 °C and pressure of 7 pounds per square inch (psi), as well as a temperature of 121 °C and 7 psi for 5, 15, and 30 min; (4) pre-soaked autoclaving at 110 °C (7 psi) and 121 °C (17 psi) for 5, 15, and 30 min. Treated MB and SB had greater (p < 0.05) crude protein content than untreated samples. All the treatments (except 24 h soaking of MB) reduced (p < 0.05) the TIA and ash content. Marama and SB are similar in protein content, but their amino acids profile and TIA are quite different. Soaking for 24 h was less effective in reducing TIA in MB and SB, compared to the thermal methods, and it was detrimental to the ash and amino acids profile of the two beans. Soaking prior to autoclaving yielded beans with the lowest TI concentrations. In conclusion, thermal methods reduced the TI contents and modified the level of proximate components and amino acids profile of the beans.

This study investigated the efficacy of a single strain of Bacillus subtilis (SSB) in modulating the composition of cecal microbiota and its link to the concentration of short-chain fatty acids (SCFA) and apparent retention (AR) of components. A total of 720, 4-week-old Shaver White chicks were allotted to control (CON), 1.1E+08 (low, LSSB), 2.2E+08 (medium, MSSB), or 1.1E+09 (high, HSSB) CFU/kg of diet groups. At grower (10-week), developer (16-week), and laying (28-week) phases, excreta and cecal digesta samples were taken for AR, microbial, and SCFA analyses. Microbial analysis involved high-throughput sequencing of the V3-V4 hypervariable regions of 16S rRNA gene. Bacterial diversity decreased (P < 0.05) at the developer phase as the SSB dose increased; however, a distinct clustering pattern (P < 0.05) of bacterial community was noted. Bacteroides and Faecalibacterium were differentially enriched in the developer for SSB-fed compared to CON-fed birds. Although no differences in microbial diversity were detected in grower and layer phases, different species of Clostridium (XVIII, XIVa, IV, and XIVb)-major butyrate producers-were identified in all phases, with stronger effect sizes for SSB-fed compared to CON-fed birds. Isobutyric acid was elevated in dose response (P = 0.034) in layer phase. In addition, the relative abundances of Alistipes, Lactobacillus, and Bifidobacterium were positively correlated (P < 0.05), with AR of most components for SSB-fed birds in the pullet phase. The results suggested that supplementing chickens' diet with B. subtilis DSM 29784 may selectively enrich beneficial bacterial communities, which in turn are critical in promoting the growth and performance of hens.IMPORTANCE In egg-laying chickens, the trend in the move away from the cage to alternative housing systems and restriction in antimicrobial use requires alternative approaches to maintain health and prevent diseases. There is increased research and commercial interest toward alternative gut health solutions while improving the performance and product safety in poultry production systems. One such approach, in recognition of the importance of the gut microbial community, is the use of microbes as feed supplements (such as probiotics). Unlike meat-type chickens, studies assessing the efficacy of such microbial supplements are limited for egg-laying chickens. Thus, by conducting a comprehensive assessment of the hen microbiota in response to various levels of B. subtilis DSM 29784 during the pullet phase (grower and developer) and the layer phase, the present study demonstrates the importance of direct-fed microbes in modulating gut microbiome, which may relate to improved performance efficiency in the pullet and layer phases.

A total of eight ileal and caecal cannulated Yorkshire barrows were used to determine the interactions of dietary fibre (DF) and lipid types on apparent digestibility of DM and fatty acids (FA) and FA flows in gastrointestinal segments. Pigs were offered four diets that contained either pectin or cellulose with or without beef tallow or maize oil in two Youden square designs (n 6). Each period lasted 15 d. Faeces, ileal and caecal contents were collected to determine apparent ileal digestibility (AID), apparent caecal digestibility and apparent total tract digestibility (ATTD) of dietary components. The interactions between DF and lipid types influenced (P <0·05) the digestibility of DM and FA flows. The addition of maize oil decreased (P <0·05) AID of DM in pectin diets, and the addition of beef tallow depressed (P <0·001) ATTD of DM in cellulose diets. Dietary supplementation with beef tallow decreased (P <0·05) the AID of FA in pectin-containing diets but had no effects in cellulose-containing diets. Dietary supplementation with beef tallow increased (P <0·05) AID of SFA and PUFA and the flow of ileal oleic, vaccenic, linolenic and eicosadienoic acids and reduced the flow of faecal lauric, docosatetraenoic and docosapentaenoic acids in pectin- and cellulose-containing diets. In conclusion, the interaction between DF type and lipid saturation modulates digestibility of DM and lipids and FA flows but differs for soluble and insoluble fibre sources, SFA and unsaturated fatty acids and varies in different gastrointestinal segments.

The study examined the effects of successive feeding of sources of n-3 PUFA to broiler breeders (BB) and their progeny in broiler chickens challenged with Eimeria. The BB were fed: 1) control (CON), corn-soybean meal diet, 2) CON + 1 % microalgae (DMA), as a source of DHA and 3) CON + 2.50% co-extruded full fat flaxseed (FFF), as a source of ALA. Eggs were hatched at 34, 44, and 54 wk of age. Posthatch treatments (BB-progeny) were: CON-CON, DMA-CON, FFF-CON, DMA-DMA and FFF-FFF with diets formulated for starter (d 1-10) and grower/finisher (d 11-42) phases. All chicks were orally challenged with Eimeria (E. acervulina and E. maxima) on d 10. Relative to CON, DMA and FFF increased concentration of n-3 PUFA by ≥ 2-fold in hatching eggs and progeny diets. There were no (P > 0.05) interactions between treatment and BB age on d 0 to 10 growth. In general, BB age affected (P < 0.05) growth performance throughout the study. In the starter phase, successive exposure to DHA and ALA improved FCR over CON-CON (P < 0.01). The interaction between treatment and BB age in grower/finisher was such that DHA exposure to younger BB resulted in poor growth performance (P < 0.05) relative to exposure to older BB. In contrast, exposure to ALA had similar (P > 0.05) growth performance irrespective of BB age. Moreover, successive exposure to ALA resulted in higher BWG, breast weight and lower FCR compared to successive exposure to DHA (P < 0.05). There were no (P > 0.05) interactions between treatment and BB age on the intestinal lesion scores, lymphoid organ weights and concentration of plasma immunoglobulin A (IgA). Successive exposure to DHA resulted in higher (P = 0.006) jejunal lesion scores than CON-CON birds. The results showed that successive exposure of DHA and ALA improved FCR relative to non-exposed birds in the starter phase. However, responses in the grower/finisher phase depended on n-3 PUFA type, with birds on successive ALA exposure supporting better growth and breast yield than birds on successive DHA exposure.

The study examined the effects of feeding broiler breeder pullets hydrolyzed whole yeast (HY) from hatch to 22 wk of age (WOA). A total of 524-day-old Ross 708 pullets were placed in floor pens (∼24 birds/pen) for the starter (0-4 WOA) and grower (5-18 WOA) phases, then transferred to the egg production facility and redistributed to ∼20 birds/pen for the prelay phase (19-22 WOA). Two diets were allocated to pens (0-18 WOA; n = 11) and (19-22 WOA; n-12). The diets were a control corn and soybean meal diet formulated to meet specifications and control plus 0.05% HY (HY). Birds had ad libitum access to feed in the first week and daily feed allocation based on pen BW from 2 WOA. Birds had free access to water throughout the trial. Body weight (BW) and uniformity (BW CV) were monitored. Boosters for infectious bronchitis and New Castle disease vaccines were administered at 18 WOA, and samples of pullets bled for antibody titer 5-day later. One pullet/pen was randomly selected, weighed, bled for plasma biochemistry, and necropsied for organ weights, ceca digesta for short-chain fatty acids (SCFA), and leg bones morphometry. In the starter and grower phases, birds fed HY were lighter and gained less (P < 0.05) than control birds. However, there were no diet effects (P > 0.05) on growth, the BW prelay phase, or BW uniformity throughout the trial. There were no (P > 0.05) diet effects on breast, gastrointestinal, liver and bursa weights, serum antibody titers, plasma biochemistry, SCFA and bone attributes. However, pullets fed HY had heavier (P = 0.047) spleen and tended to have lower (P = 0.080) plasma concentrations of aspartate aminotransferase (AST) relative to control pullets. In conclusion, the parameters assessed showed no negative consequences of feeding HY to broiler breeder pullets. However, effects on the spleen and plasma AST may indicate modest modulation of immunity and metabolism. The impact of the provision of HY during broiler breeder pullet phase on reproductive performance and chick quality should be investigated.

Comparative efficacy of hydroxychloride (HC) and organic (OR) sources of Zn, Cu and Mn on performance of broiler breeders (BB) between 42 and 63 weeks of age (WOA) was investigated. A total of 408 ♀ Ross 708 and 48 ♂ Yield Plus cockerels were placed in pens (17 ♀ and 2 ♂) housed in 2 rooms (12 pens/room) and allocated to one of 2 diets in a completely randomized block design (n=12). The diets had similar nutrient specifications but differed in Zn, Cu, and Mn sources: 1) HO, a blend of 80% HC and 20% OR sources, and 2) OR, 100% OR sources. Birds were fed and managed according to breeder guidelines. The egg count was recorded daily and categorized as normal or abnormal. Egg yolk color, albumen height, Haugh unit, eggshell thickness, and eggshell breaking strength were assessed every 4 wk. Individual hen body weight (BW) was recorded at 5-wk intervals to determine BW uniformity. At 52 and 63 WOA, the eggs and excreta samples were collected. At the end of the trial, 4 hens per pen were bled for plasma concentration of trace minerals and organs (liver, gizzard, spleen, kidney, and thymus) weight. There were no interactions between source and age on any parameters (P > 0.05). There were no main effects of source on egg production, eggshell quality, BW, and organs weight (P > 0.05). Hens fed HO diets had darker yolk compared to those fed OR diets (P = 0.014). The concentration of Zn in the eggs of OR BB was higher (P = 0.022) than for HO birds. However, there were no dietary effects on the concentration of trace minerals in the egg, plasma, and excreta (P > 0.05). The results indicated that a mixture of HC and O as sources for Zn, Cu, and Mn was as effective as OR sources in supporting egg production, egg quality, and trace mineral utilization in broiler breeders.

Fifty gilts (initial body weight [BW] 190.7 ±â€…4.2 kg) were recruited on day 85 of gestation and were used until day 19 of lactation to assess the dose-response of inactivated yeast via hydrolyzation (HY) inclusion on offspring growth and immunoglobulin (Ig) transfer prior to weaning. Gilts were assigned to one of the 5 experimental diets: a control with no HY (HY0) or inclusion of 0.25% (HY0.25), 0.5% (HY0.5), 1.0% (HY1.0), or 1.2% (HY1.2) HY. Gilts were weighed on days 85 and 110 of gestation and days 1 and 19 (weaning) after farrowing. Offspring were weighed on days 1 and 19 of age. On lactation day 1 (approximately 24 h after farrowing), colostrum, gilt plasma, and plasma from 2 median BW piglets were collected and on day 19, plasma from each gilt and 2 median BW piglets per litter were collected for determination of Ig concentrations. Contrast statements were used to assess the linear, quadratic, cubic, and quartic effects of HY inclusion. The inclusion of HY had minimal effects on gilt BW or litter characteristics at birth (total number born and born alive, piglet birth weight). Lactation average daily feed intake of the gilts tended to increase then decrease with increasing HY inclusion (quadratic; P = 0.085). Piglet preweaning average daily gain (linear, quadratic, and quartic; P < 0.05) and BW at weaning (quadratic and quartic; P < 0.05) increased then decreased with increasing HY inclusion. On lactation day 1, colostrum and gilt plasma Ig concentrations were not affected by dietary treatment (P > 0.10) but piglet IgA and IgM decreased then increased with HY inclusion level (cubic; P < 0.05). On lactation day 19, piglet plasma IgG tended to increase with HY inclusion (linear; P = 0.099). In summary, increasing HY inclusion in late gestating and lactating gilt diets improved immune transfer in the first 24 h after birth and piglet preweaning growth rates and BW at weaning. Therefore, maternal feeding of HY could be used as a strategy to improve offspring immunocompetence and BW at weaning, with possible carryover benefits for the postweaning phase.


Abrupt weaning exposes piglets to various stressors that result in a period after weaning with little or no weight gain or feed intake and increased incidence of morbidity and mortality. Inactivated yeast via hydrolyzation (HY) is a functional feed additive that can improve the immune response in pigs. The low and variable feed intakes immediately after weaning render feed additives less useful in nursery pig diets, therefore, enhancing immunocompetence prior to weaning could be a strategy to improve offspring outcomes. This study tested 4 levels of HY (0.25%, 0.5%, 1.0%, and 1.2%) and control (0%) fed to gestating and lactating gilts from day 85 of gestation until day 19 of lactation when piglets were weaned. Plasma immunoglobulin (Ig) concentrations and preweaning offspring growth rates were assessed. It was found that piglet preweaning average daily gain and body weight at weaning were improved with increasing inclusion of HY in the maternal diet, which corresponded to increased plasma IgA and IgM concentrations for the offspring after birth. Greater body weight at weaning and greater plasma IgA and IgM concentration have the potential to attenuate the postweaning growth lag in addition to improving immunocompetence around weaning.

Diets fed to commercial chicken breeders are high in n-6 fatty acids (n-6 FAs) and low in n-3 fatty acids (n-3 FAs). N-3 FAs are essential for embryonic brain development. In precocial birds, like chickens, brain development and brain n-3 FA accrual occur primarily before hatching. In two experiments, broiler and layer breeders were fed diets with or without flaxseed as the source of n-3 FAs from plant-based alpha-linolenic acid. Day-old broiler (n = 80) and layer (n = 96) offspring were dissected to calculate the percentage brain-to-body weight. Brain FA analyses from total lipid extracts were determined in the broiler (n = 24) and layer (n = 24) offspring brains, and the percentage FA composition and concentration (µg FAs per g brain) were calculated for each n-3 and n-6 FA. The brain size was only increased in broiler offspring from mothers fed flaxseed (χ2 = 9.22, p = 0.002). In layer offspring only, the maternal flaxseed diet increased the brain concentration and percentage of n-3 FAs and decreased n-6 FAs (p < 0.05). We showed that feeding flaxseed to mothers increased the brain size in broiler offspring and altered brain FA composition in layer offspring. These results may have implications for poultry and other captive bird species fed diets low in n-3 FAs.
