RESUMO

Coupling characteristics between a single mode fiber (SMF) and a waveguide embedded in a glass chip via a graded index fiber (GIF) tip are investigated at a wavelength of 976 nm. The GIF tips comprise a coreless fiber section and a GIF section. A depressed cladding waveguide in a ZBLAN glass chip with a core diameter of 35 µm is coupled with GIF tips that have a range of coreless fiber and GIF lengths. An experimental coupling efficiency as high as 88% is obtained while a numerical simulation predicts 92.9% for the same GIF tip configuration. Since it is measured in the presence of Fresnel reflection, it can be further improved by anti-reflection coating. Additionally, it is demonstrated that a gap can be introduced between the chip waveguide and the GIF tip while maintaining the high coupling efficiency, thus allowing a thin planar optical component to be inserted. The results presented here will enable miniaturization and simplification of photonic chips with integrated waveguides by replacing bulk coupling lenses with integrated optical fibers.

The rapid development of optofluidic technologies in recent years has seen the need for sensing platforms with ease-of-use, simple sample manipulation, and high performance and sensitivity. Herein, an integrated optofluidic sensor consisting of a pillar array-based open microfluidic chip and caged dye-doped whispering gallery mode microspheres is demonstrated and shown to have potential for simple real-time monitoring of liquids. The open microfluidic chip allows for the wicking of a thin film of liquid across an open surface with subsequent evaporation-driven flow enabling continuous passive flow for sampling. The active dye-doped whispering gallery mode microspheres placed between pillars, avoid the use of cumbersome fibre tapers to couple light to the resonators as is required for passive microspheres. The performance of this integrated sensor is demonstrated using glucose solutions (0.05-0.3 g/mL) and the sensor response is shown to be dynamic and reversible. The sensor achieves a refractive index sensitivity of ~40 nm/RIU, with Q-factors of ~5 × 103 indicating a detection limit of ~3 × 10-3 RIU (~20 mg/mL glucose). Further enhancement of the detection limit is expected by increasing the microsphere Q-factor using high-index materials for the resonators, or alternatively, inducing lasing. The integrated sensors are expected to have significant potential for a host of downstream applications, particularly relating to point-of-care diagnostics.

RESUMO

Graded-index optical fiber probes suitable for focusing or collimating the output of an optical fiber at a wavelength of 1.3 µm have become an enabling technology in optical coherence tomography imaging applications for in vivo bioimaging. Such fiber tips however remain uncommon in other photonics applications. This paper provides the first numerical study of graded-index fiber tips covering a broad range of wavelengths spanning from the UV to short-infrared. The wavelength dependency and the influence of probe geometry on performance characteristics such as far-field divergence angle, spot size and working distance are analyzed. The paper yields easily accessible design guidelines for the fabrication of collimating or focusing fiber tips. Fiber collimators have considerable potential for use in free-space systems and could benefit a range of devices such as variable attenuators, dynamic wavelength equalisers and large 3D optical cross-connect switches, whereas focusing fiber tips have applications in high-resolution imaging.

Femtosecond laser inscribed fiber Bragg gratings in pure-silica suspended-core optical fibers have previously been demonstrated as a promising platform for high temperature sensing. However, the density of gratings that could be written on a single fiber was limited by undesired reflections associated with higher order modes in these high numerical aperture fibers. This resulted in a complex, broadband reflection spectrum with limited multiplexing capability. In this work we utilize modifications to the fine structure of the suspended core optical fibers to fine tune the relative confinement loss of the optical fiber modes, thus reducing the contribution from such higher order modes. The effects of these changes on mode propagation are modeled, giving a range of fibers with different confinement loss properties which can be tailored to the specific length scale of a desired application. We achieve single-peak reflections from individual fiber Bragg gratings, significantly improving performance for multipoint sensing and demonstrate this technique by writing 20 gratings onto a single fiber.

BACKGROUND: O-negative red blood cells (ON-RBC) are a precious resource and the international blood banking community has become increasingly concerned with its inappropriate utilization. AABB recently made several recommendations to address the issue. Solutions must be multifaceted and involve donor centers, blood banks, and clinical departments. From the perspective of a hospital blood bank, it is difficult to rely solely on increased donor recruitment and ubiquitous blood typing of the entire in-patient population. We therefore focused on interventions within the blood bank to optimize inventory and policies to ensure appropriate ON-RBC utilization. STUDY DESIGN AND METHODS: Transfusion data over one year was examined for the rate of out-of-group/inappropriate ON-RBC. Furthermore, we assessed whether that rate was related to product life on the day of transfusion. We also examined our stock inventory levels and how excess inventory can contribute to inappropriate ON-RBC usage. RESULTS: The ON-RBC inventory level was decreased in order to reduce the rate of inappropriate transfusions while maintaining a safe level for optimal patient care. Compared to baseline, our intervention caused ON-RBCs to be transfused earlier in their shelf-life (9.27 vs. 11.15 days from expiration [DFE], p = 0.0012). This reduced the overall rate of inappropriate ON-RBC transfusions (67% vs. 54%, p = 0.0035), approximating 185 units of ON-RBC saved over the course of 6 months. CONCLUSIONS: A data-driven approach to optimize stock inventory levels is widely applicable; it can be adopted by numerous institutions to improve utilization and establish a benchmark for the broader blood banking community.

RESUMO

The evanescent fields along multimode fibers are usually relatively weak. To enhance the sensitivity of the resulting sensors, skew rays have been exploited for their larger number of total internal reflections and their more comprehensive spread over the fiber surface. The uniform distribution of light-matter interactions across the fiber surface facilitates high sensitivity through an increased interaction area, while mitigating the risk of laser-induced coating-material damage and photobleaching. Power-dependent measurements are less susceptible to temperature effects than interferometric techniques, and place loose requirements on the laser source. This review highlights the key developments in this area, while discussing the benefits, challenges as well as future development.

A guided-wave chip laser operating in a single longitudinal mode at 2860 nm is presented. The cavity was set in the Littman-Metcalf configuration to achieve single-frequency operation with a side-mode suppression ratio above 33 dB. The chip laser's 2 MHz linewidth on a 10 ms scale was found to be limited by mechanical fluctuations, but its Lorentzian contribution was estimated to be lower than 1 Hz using a heterodyne technique. This demonstration incorporates a high coherence source with the simplicity provided by the compactness of chip lasers.

Absorption lines of methane in the 2ν3 band centered at 1650 nm were measured with a free-running mode-locked dual-comb laser based on a single erbium-doped glass chip. The laser's spectra were broadened up to 1670 nm using amplifiers and highly nonlinear fiber. A comb was used to interrogate the complex transmission spectrum of a methane-filled gas cell with an optical point spacing of 968 MHz and an interferogram (IGM) rate of 27 kHz to yield absorption lines of the R and Q branches. A 1.28 s sequence of IGMs was measured and phase-corrected using a self-sufficient correction algorithm seeded only by the IGMs. The associated transmission spectrum was then compared to HITRAN yielding residuals limited by photodetector nonlinearity.

Ultrafast laser inscribed waveguide lasers can lead to highly efficient and compact optical devices. This Letter reports an average lasing efficiency of 65%±2.5% from a multi-waveguide 2.5 mol. % ytterbium-doped ZrF4-BaF2-LaF3-AlF3-NaF (Yb:ZBLAN) chip in an extended-cavity configuration. A maximum output power of 750 mW with a lasing efficiency of 68% is also achieved. A monolithic end-coupled configuration reached a maximum output power of 784 mW with a lasing efficiency of 70%. The lasing wavelength is tuned from 1001 to 1045 nm in a Littrow configured cavity. A beam propagation factor of the lowest-order transverse-mode output was routinely achieved with an M2 of 1.15.

We characterize the frequency noise performance of a free-running dual-comb source based on an erbium-doped glass chip running two adjacent mode-locked waveguide lasers. This compact laser platform, contained only in a 1.2 L volume, rejects common-mode environmental noise by 20 dB thanks to the proximity of the two laser cavities. Furthermore, it displays a remarkably low mutual frequency noise floor around 10 Hz2/Hz, which is enabled by its large-mode-area waveguides and low Kerr nonlinearity. As a result, it reaches a free-running mutual coherence time of 1 s since mode-resolved dual-comb spectra are generated even on this time scale. This design greatly simplifies dual-comb interferometers by enabling mode-resolved measurements without any phase lock.

BACKGROUND: The ordering process at Stanford Health Care involved twice-daily shipments predicated upon current stock levels from the blood center to the hospital transfusion service. Manual census determination is time consuming and error prone. We aimed to enhance inventory management by developing an informatics platform to streamline the ordering process and reallocate staff productivity. STUDY DESIGN AND METHODS: The general inventory accounts for more than 50 product categories based on characteristics including component, blood type, irradiation status, and cytomegalovirus serology status. Over a 5-month calibration period, inventory levels were determined algorithmically and electronically. An in-house software program was created to determine inventory levels, optimize the electronic ordering process, and reduce labor time. A 3-month pilot period was implemented using this program. RESULTS: This system showed noninferiority while saving labor time. The average weekly transfused:stocked ratios for cryoprecipitate, plasma, and red blood cells, respectively, were 1.03, 1.21, and 1.48 before the pilot period, compared with 0.88, 1.17, and 1.40 during (p = 0.28). There were 27 (before) and 31 (during) average STAT units ordered per week (p = 0.86). The number of monthly wasted products due to expiration was 226 (before) and 196 (during) units, respectively (p = 0.28). An estimated 7 hours per week of technologist time was reallocated to other tasks. CONCLUSION: An in-house electronic ordering system can enhance information fidelity, reallocate and optimize valuable staff productivity, and further standardize ordering. This system showed noninferiority to the labor-intensive manual system while freeing up over 360 hours of staff time per year.

We present a dual-comb spectrometer based on two passively mode-locked waveguide lasers integrated in a single Er-doped ZBLAN chip. This original design yields two free-running frequency combs having a high level of mutual stability. We developed in parallel a self-correction algorithm that compensates residual relative fluctuations and yields mode-resolved spectra without the help of any reference laser or control system. Fluctuations are extracted directly from the interferograms using the concept of ambiguity function, which leads to a significant simplification of the instrument that will greatly ease its widespread adoption and commercial deployment. Comparison with a correction algorithm relying on a single-frequency laser indicates discrepancies of only 50 attoseconds on optical timings. The capacities of this instrument are finally demonstrated with the acquisition of a high-resolution molecular spectrum covering 20 nm. This new chip-based multi-laser platform is ideal for the development of high-repetition-rate, compact and fieldable comb spectrometers in the near- and mid-infrared.

We highlight the authors' recent work in the areas of current, temperature, acceleration, acoustic, humidity and ultraviolet-light sensing based on this exquisite technology, and the advantages and challenges of using optical microfibers are discussed.

We report mode-locked ~1550 nm output of transform-limited ~180 fs pulses from a large mode-area (diameter ~50 µm) guided-wave erbium fluorozirconate glass laser. The passively mode-locked oscillator generates pulses with 25 nm bandwidth at 156 MHz repetition rate and peak-power of 260 W. Scalability to higher repetition rate is demonstrated by transform-limited 410 fs pulse output at 1.3 GHz. To understand the origins of the broad spectral output, the laser cavity is simulated by using a numerical solution to the Ginzburg-Landau equation. This paper reports the widest bandwidth and shortest pulses achieved from an ultra-fast laser inscribed waveguide laser.

Microscopic defects in optical fiber coatings can be an impending catastrophe for high-power fiber laser and telecommunications systems and are difficult to detect with conventional methods. We demonstrate a highly sensitive interrogation technique that can readily identify faults such as microscopic nicks, scrapes, low-quality recoatings, and internal defects in fibers and their coatings, based on skew ray excitation and angle-resolved analysis.

Dynein is a large macromolecular motor complex that moves cargo along microtubules. A motor-independent role for the light chain of dynein, Dyn2p, in peroxisome biology in Saccharomyces cerevisiae was suggested from its interaction with Pex14p, a component of the peroxisomal matrix protein import docking complex. Here we show that cells of the yeast Yarrowia lipolytica deleted for the gene encoding the homologue of Dyn2p are impaired in peroxisome function and biogenesis. These cells exhibit compromised growth on medium containing oleic acid as the carbon source, the metabolism of which requires functional peroxisomes. Their peroxisomes have abnormal morphology, atypical matrix protein localization, and an absence of proteolytic processing of the matrix enzyme thiolase, which normally occurs upon its import into the peroxisome. We also show physical and genetic interactions between Dyn2p and members of the docking complex, particularly Pex17p. Together, our results demonstrate a role for Dyn2p in the assembly of functional peroxisomes and provide evidence that Dyn2p acts in cooperation with the peroxisomal matrix protein import docking complex to effect optimal matrix protein import.

Prions are proteins that can adopt different infectious conformations known as "strains" or "variants," each with a distinct, epigenetically inheritable phenotype. Mechanisms by which prion variants are determined remain unclear. Here we use the Saccharomyces cerevisiae prion Rnq1p/[PIN(+)] as a model to investigate the effects of chaperone proteins upon prion variant determination. We show that deletion of specific chaperone genes alters [PIN(+)] variant phenotypes, including [PSI(+)] induction efficiency, Rnq1p aggregate morphology/size and variant dominance. Mating assays demonstrate that gene deletion-induced phenotypic changes are stably inherited in a non-Mendelian manner even after restoration of the deleted gene, confirming that they are due to a bona fide change in the [PIN(+)] variant. Together, our results demonstrate a role for chaperones in regulating the prion variant complement of a cell.

RESUMO

Rapid detection of pathogens and analytes at the point of care offers an opportunity for prompt patient management and public health control. This paper reports an open microfluidic platform coupled with active whispering gallery mode (WGM) microsphere resonators for the rapid detection of influenza viruses. The WGM microsphere resonators, precoated with influenza A polyclonal antibodies, are mechanically trapped in the open micropillar array, where the evaporation-driven flow continuously transports a small volume (∼µL) of sample to the resonators without auxiliaries. Selective chemical modification of the pillar array changes surface wettability and flow pattern, which enhances the detection sensitivity of the WGM resonator-based virus sensor. The optofluidic sensing platform is able to specifically detect influenza A viruses within 15 min using a few microliters of sample and displays a linear response to different virus concentrations.

We report the first Yb:ZBLAN and Yb:IOG10 waveguide lasers fabricated by the fs-laser direct-writing technique. Pulses from a Titanium-Sapphire laser oscillator with 5.1 MHz repetition rate were utilized to generate negative refractive index modifications in both glasses. Multiple modifications were aligned in a depressed cladding geometry to create a waveguide. For Yb:ZBLAN we demonstrate high laser slope efficiency of 84% with a maximum output power of 170 mW. By using Yb:IOG10 a laser performance of 25% slope efficiency and 72 mW output power was achieved and we measured a remarkably high refractive index change exceeding Δn = 2.3 × 10(-2).

We report a novel fiber-optic sensing architecture for the detection of paramagnetic gases. By interacting a modulated magnetic field with guided light within a microstructured optical fiber, it is possible to exploit Faraday Rotation Spectroscopy (FRS) within unprecedentedly small sample volumes. This approach, which utilizes magnetic circular birefringence and magnetic circular dichroism effects, is applied to a photonic bandgap fiber to detect molecular oxygen and operates at a wavelength of 762.309 nm. The optical fiber sensor has a 4.2 nL detection volume and 14.8 cm long sensing region. The observed FRS spectra are compared with a theoretical model that provides a first understanding of guided-mode FRS signals. This FRS guided-wave sensor offers the prospect of new compact sensing schemes.
