RESUMO

Bipolaris sorokiniana is a necrotrophic fungal pathogen that causes foliar and root diseases on wheat and barley. These diseases are common in all wheat- and barley-growing regions, with more severe outbreaks occurring under warm and humid conditions. B. sorokiniana can also infect a wide range of grass species in the family Poaceae and secrete ToxA, an important necrotrophic effector also identified other wheat leaf spotting pathogens. In this study, the prevalence and virulence role of ToxA were investigated in a collection of 278 B. sorokiniana isolates collected from spring wheat and barley in the Upper Midwest of the United States or other places, including 169 from wheat leaves, 75 from wheat roots, 30 from barley leaves, and 4 from wild quack grass leaves. ToxA was present in the isolates from wheat leaves, wheat roots, and wild grass leaves but was absent from isolates collected from barley leaves. Prevalence of ToxA in wheat leaf isolates (34.3%) was much higher than that in wheat root isolates (16%). Sequencing analysis revealed the presence of two haplotypes, with the majority being BsH2. All ToxA+ isolates produced the functional effector in liquid cultures. Pathogenicity assays revealed that ToxA+ isolates caused significantly more disease on spring wheat lines harboring Tsn1 than their tsn1 mutants, suggesting that the ToxA-Tsn1 interaction plays an important role in spot blotch development. This work confirms the importance of ToxA in B. sorokiniana populations infecting wheat and, thus, the need to eliminate Tsn1 from spring wheat cultivars to reduce susceptibility to spot blotch.

OBJECTIVE: To find out the outcome in cases of vesicovaginal fistula repair. METHODS: The descriptive study was conducted at the District Headquarter Hospital, Timergara, Lower Dir, Pakistan, from November 1, 2011 to November 2013, and comprised all patients admitted in Urology unit with vesicovaginal fistula. Repair was done with either transabdominal or transvaginal approach. Repair technique involved good tissue separation, interrupted sutures, and no omental interposition. Follow-up was of three months. RESULTS: There were 30 patients available, but 2(6.6%) were excluded. Among the remaining 28(93.3%) patients dehiscence was not noted in any patient, while only 4(14.3%) patients developed mild urinary tract infection. There were no intraoperative or postoperative deaths. CONCLUSIONS: Transvaginal or Transabdominal repair of vesicovaginal fistula is successful treatment modality if good dissection and tissue separation is applied with interrupted suturing. Omental interposition is not essential for good healing.

Wheat (Triticum aestivum L.) is a prominent grain crop. The goal of the current experiment was to examine the genetic potential of advanced bread wheat genotypes for yield and stripe rust resistance. Ninety-three bread wheat genotypes including three varieties (Kohat-2017, Pakistan-2013, and Morocco) were field tested in augmented design as observational nurseries at three locations (i.e., Kohat, Nowshera, and Peshawar) during the 2018-19 crop season. Various parameters related to yield and stripe rust resistance showed significant differences among genotypes for most of the characters with few exceptions. The analysis of variance revealed significant variations for all the genotypes for all the traits at all three sites with few exceptions where nonsignificant differences were noticed among genotypes. Averaged over three locations, genotypes exhibiting maximum desirable values for yield and yield components were KT-86 (325 tillers) for tillers m-2, KT-50 (2.86 g) for grain weight spike-1, KT-49 (41.6 g) for 1000-grain weight, KT-50 (74 grains) for grains spikes-1, KT-55 (4.76 g) for spike weight, and KT-36 and KT-072 (4586 kg ha-1) for grain yield. Correlation analysis revealed that grain yield had a significant positive correlation with grain spike-1 and grain weight spike-1 at Kohat, with grains spike-1, tillers m-2, and grain weight spike-1 at Nowshera, and with plant height, spike weight, 1000-grain weight, and tillers m-2 at Peshawar. Molecular marker data and host response in the field at the adult stage revealed that Yr15 and Yr10 are both still effective in providing adequate resistance to wheat against prevalent races of stripe rust. Four lines showing desirable lower average coefficient of infection (ACI) values without carrying Yr15 and Yr10 genes show the presence of unique/new resistance gene(s) in the genetic composition of these four lines. Genotype KT-072 (4586 kg ha-1 and 1.3 ACI), KT-07 (4416 kg ha-1 and 4.3 ACI), KT-10 (4346 kg ha-1 and 1.0 ACI), and KT-62 (4338 kg ha-1 and 2.7 ACI) showed maximum values for grain yield and low desirable ACI values, and these lines could be recommended for general cultivation after procedural requirements of variety release.

BACKGROUND: Maintaining standards of living donor liver transplantation (LDLT) can be a challenge during the corona virus disease 2019 (COVID-19) pandemic. Center-specific protocols have been developed and transplant societies propose limiting elective LDLT. We have looked at outcomes of LDLT during the pandemic in an exclusively LDLT center. METHODS: Patients were grouped into pre-COVID (January 2019-February 2020) (n = 162) and COVID (March 2020-January 2021) (n = 53) cohorts. We looked at patient characteristics, 30-day morbidity, and mortality. Outcomes were also assessed in donors and recipients who underwent surgery after recovery from COVID-19. RESULTS: The average number of transplants reduced from 11.5/month to 4.8/month. Fewer patients with MELD > 20 underwent LDLT in the COVID cohort (41.3% versus 24.5%, P = 0.03). Out of nine patients with a positive pretransplant COVID-19 PCR, there were 2 (22.3%) deaths on the waiting list. Seven patients underwent LT after recovery from COVID-19 with one 30-day mortality due to biliary sepsis. Three donors with positive COVID-19 PCR underwent uneventful donation after testing negative for COVID-19. No significant difference in 30-day survival was observed in the pre-COVID and COVID cohorts (93.2% versus 90.6%) (P = 0.3). Out of two recipients who developed COVID-19 pneumonia within 30 days after LT, there was one mortality. The 1-year survival for the entire cohort with a MELD cutoff of 20 was 90% and 84% (P = 0.2). CONCLUSION: Despite comparable outcomes, fewer sick patients might undergo LDLT during the pandemic. Individuals recovered from COVID-19 might be safely considered for donation or transplantation.

BACKGROUND: Diverticula of small intestine are rare. Jejunal diverticula can be single or multiple. Diverticula in the jejunum tend to be large and multiple. Clinically they may be asymptomatic or may give rise to symptoms like pain, flatulence and borborygmi, may produce malabsorption syndrome or may present in emergency with different acute pathologies like perforation, haemorrhage, obstruction, enterolith formation and inflammation. The Objective was to see the pattern of complications in jejunal diverticula presenting as a surgical emergency. METHODS: This descriptive study was conducted at Surgical Units of the 3 tertiary care Hospitals of Peshawar, for 7 years from January 1, 2002 to December 31, 2008. Study included all patients presenting to and admitted in Surgical Unit, Hayatabad Medical Complex, Peshawar with complicated jejunal diverticula during the above mentioned period. Name, age, sex, other relevant data, history and examination findings and results of investigation were recorded. Uncomplicated jejuna diverticula were excluded from study. The operative findings and the type of complication were recorded. RESULTS: Ten patients were admitted during 7 years of study. Out of all patients 9 were male and 1 was female. Eight out of 10 patients presented with perforation of diverticula while 1 patient had severe inflammation of diverticulum causing pain, ileus and acute abdomen. One patient had acute pain due to adhesion formation. CONCLUSION: It is seen that complicated jejunal diverticulae are quite rare and the most common complication is perforation. Inflammation and adhesion are other complications with which jejunal diverticula presented during this study.

Germplasm collections are rich sources of genetic variation to improve crops for many valuable traits. Nested association mapping (NAM) populations can overcome the limitations of genome-wide association studies (GWAS) in germplasm collections by reducing the effect of population structure. We exploited the genetic diversity of the USDA-ARS wheat (Triticum aestivum L.) core collection by developing the Spring Wheat Multiparent Introgression Population (SWMIP). To develop this population, twenty-five core parents were crossed and backcrossed to the Minnesota spring wheat cultivar RB07. The NAM population and 26 founder parents were genotyped using genotyping-by-sequencing and phenotyped for heading date, height, test weight, and grain protein content. After quality control, 20,312 markers with physical map positions were generated for 2,038 recombinant inbred lines (RILs). The number of RILs in each family varied between 58 and 96. Three GWAS models were utilized for quantitative trait loci (QTL) detection and accounted for known family stratification, genetic kinship, and both covariates. GWAS was performed on the whole population and also by bootstrap sampling of an equal number of RILs from each family. Greater power of QTL detection was achieved by treating families equally through bootstrapping. In total 16, 15, 12, and 13 marker-trait associations (MTAs) were identified for heading date, height, test weight, and grain protein content, respectively. Some of these MTAs were coincident with major genes known to control the traits, but others were novel and contributed by the wheat core parents. The SWMIP will be a valuable source of genetic variation for spring wheat breeding.
