Objectives: To evaluate the clinical value of intravesical gemcitabine combined with immunotherapy in patients with non-muscle-invasive bladder carcinoma (NMIBC) after transurethral resection of bladder tumor (TURBT). Methods: Eighty patients with non-muscle-invasive urothelial carcinoma treated in Baoding No.1 Hospital from November 2016 to November 2019 were randomly divided into two groups, with 40 patients in each group. Both groups underwent TURBT. After surgery, the research group was treated with intravesical chemotherapy using gemcitabine combined with ubenimex, while the control group was given 40 mg pirarubicin by intravesical instillation. Postoperative condition was evaluated by cystoscopy every three months in both groups. The recurrence six months, one year and two years after treatment, the incidence of lower urinary tract symptoms such as dysuria, hematuria and frequent urination, general adverse drug reactions such as rashes, liver function damage and gastrointestinal reaction, as well as the changes in CD3+, CD4+, CD8+ and CD4+/CD8+ T lymphocyte subsets before and after treatment were comparatively analyzed between the two groups. Results: The recurrence rate showed no statistical significance between the two groups 6 months after treatment (p=0.17), but significant differences one year (p=0.04) and two years (p=0.03) after treatment, which were significantly lower in the research group than the control group. The incidence of adverse drug reactions was 22.5% in the research group and 7.5% in the control group, without significant difference (p=0.36). The incidence of lower urinary tract symptoms was 32.5% and 55%, respectively, in the research group and the control group. The incidence of lower urinary tract symptoms in the research group was significantly lower compared with the control group, with a statistically significant difference (p=0.04). After treatment, CD3+, CD4+ and CD4+/CD8+ levels in the research group increased significantly than those in the control group, with statistically significant differences (CD3+, p=0.01; CD4+, p=0.00; CD4+/CD8+, p=0.00). Conclusions: For NMIBC patients receiving bladder-preserving surgery, intravesical gemcitabine combined with immunotherapy can reduce the recurrence rate, relieve lower urinary tract symptoms, increase the tolerance of patients to intravesical chemotherapy and significantly improve the function of T lymphocytes, without obvious increase in adverse drug reactions. Therefore, it is safe and effective, and has certain clinical value.

Chemical genomics has been applied extensively to evaluate small molecules that modulate biological processes in Saccharomyces cerevisiae. Here, we use yeast as a surrogate system for studying compounds that are active against metazoan targets. Large-scale chemical-genetic profiling of thousands of synthetic and natural compounds from the Chinese National Compound Library identified those with high-confidence bioprocess target predictions. To discover compounds that have the potential to function like therapeutic agents with known targets, we also analyzed a reference library of approved drugs. Previously uncharacterized compounds with chemical-genetic profiles resembling existing drugs that modulate autophagy and Wnt/ß-catenin signal transduction were further examined in mammalian cells, and new modulators with specific modes of action were validated. This analysis exploits yeast as a general platform for predicting compound bioactivity in mammalian cells.

In this study, we developed a method for determining cotinine and 3-hydroxycotinine in human serum and established a methodology for an in-depth study of tobacco exposure and health. After the proteins in the human serum samples were precipitated with acetonitrile, they were separated on a ZORBAX SB-Phenyl column with a mobile phase of methanol encompassing 0.3% formic acid-water encompassing 0.15% formic acid. The measurement was performed on an API5500 triple quadrupole mass spectrometer in the multiple reaction monitoring mode. Cotinine, 3-hydroxycotinine, and cotinine-d3 isotope internal standards were held for 2.56 minutes, 1.58 minutes, and 2.56 minutes, respectively. In serum, the linear range was 0.05 to 500 ng·mL-1 for cotinine and 0.50 to 1250 ng·mL-1 for 3-hydroxycotinine. The lower limit of quantification (LLOQ) was 0.05 ng·mL-1 and 0.5 ng·mL-1 for cotinine and 3-hydroxycotinine, respectively. The intra-day and inter-day relative standard deviations were <11%, and the relative errors were within ±â€…7%. Moreover, the mean extraction recoveries of cotinine and 3-hydroxycotinine were 98.54% and 100.24%, respectively. This method is suitable for the rapid determination of cotinine and 3-hydroxycotinine in human serum because of its rapidity, sensitivity, strong specificity, and high reproducibility. The detection of cotinine levels in human serum allows for the identification of the cutoff value, providing a basis for differentiation between smoking and nonsmoking populations.

Amur virus was recently identified as the causative agent of hemorrhagic fever with renal syndrome. Here we report the complete genome sequence of an Amur virus isolated from Apodemus peninsulae in Northeastern China. The sequence information provided here is critical for the molecular epidemiology and evolution of Amur virus in China.

OBJECTIVE: To normalize bacteriostasis and relieving itching external therapeutic function of Kochiae Fructus. METHODS: Itching guinea pig model caused by histamine, itching mice model, eczema guinea pig model caused by OVA, and inhibitory effect on pathogens in vitro were used to observe the itching threshold, symptoms and other related physiological index, as well as the inhibitory effect on the normal skin fungi by water extraction of Kochiae Fructus to evaluate the external therapeutic function of Kochiae Fructus. RESULTS: The itching threshold of guinea pig itching model treated by water extraction of Kochiae Fructus at high, medium and low three dosage level, could be significantly increased when compared with negative control group (P < 0.05 or P < 0.01); Red speckle of OVA guinea pig model treated by water extraction of Kochiae Fructus at high, medium and low three dosage level, could be significantly decreased when compared with negative control group (P < 0.05 or P < 0.01); The number of itching and total time of itching within 30 minutes of mice model caused by R-glycose anhydride treated by water extraction of Kochiae Fructus at high, medium and low three dosage level, could be significantly decreased when compared with negative control group (P < 0.05 or P < 0.01); Several common skin fungi could be significantly inhibited by the water extraction of Kochiae Fructus. MIC of the water extraction of Kochiae Fructus on Trichophyton mentagrophytes, Trichophyton rubrum, Microsporum canis, Trichophyton violaceum, and Trichophyton schoenleini were 3.12%, 0.78%, 0.78%, 0.78%, 0.78%, respectively. CONCLUSION: Kochiae fructus has the effect of bacteriostasis and relieving itching.

The title complex, [Ni(N(3))(2)(C(10)H(8)N(2))(CH(3)OH)(2)], lies on a twofold roation axis which runs through the Ni(II) ion and the mid-point of the bipyridine ligand. The Ni(II) ion is coordinated in a distorted octa-hedral environment by two azide ligands in a trans configuration. The methanol ligands are in a cis configuration and their hy-droxy groups form intra-molecular O-H⋯(N,N) hydrogen bonds with the azide ligands.

OBJECTIVE: To study the chemical constituents from Radix Boehmeriae. METHODS: Compounds were isolated by various column chromatographies with silica gel, their structures were elucidated by spectral analysis and chemical evidence. RESULTS: Eight compounds were isolated and their structures were identified as emodin (1), emodin-8-O-beta-glucopyranoside (2), physcion (3), polydatin (4), catechin (5), epicatechin (6), potassium nitrate (7), sitosterol (8). CONCLUSION: Among these compounds, 3 -7 are isolated from Radix Boehmeriae for the first time.

To gain more insights into epidemiologic characteristics and genotype of hantavirus in Apodemus agrarius in Changbai Area. Complete hantavirus S segment sequences were amplified by RT-PCR and sequenced. The phylogenetic trees were constructed for analysis of genetic characters of hantavirus. A total of 58 Apodemus agrarius were trapped in the epidemic areas, and complete hantavirus S segment sequences were obtained from 4 lung samples of these rodents (6. 90%0). Phylogenetic analysis of the four S segment sequences indicated that all viruses isolated from Apodemu sagrarius were closely related to genotype 6 of Hantaan virus (95. 8%-96. 3%, nucleotide identity; 98. 6%-99. 5%, amino acid identity), all of them had a specific S387 different from other genotypes of Hantaan virus.

RESUMO

Human cytomegalovirus is an important pathogen which widely infects human. In this study, an eukaryotic expression vector containing human cytomegalovirus gB680 and pp65m genes was injected into BALB/c mice to explore the immune response. Firstly, the recombinant plasmid pVAX1/gB680+pp65m and plasmid pVAX1 were transfected into COS-7 cells respectively and the transiently expressed product was detected by RT-PCR and Western blot. The maxiprepared plasmid pVAX1/gB680 + pp65m and pVAX1 by alkaline lysis with SDS and purified by Sepharose 4FF column chromatography were then used to immunize BALB/c mice and the humoral and cellular immune responses were determined. Recombinant plasmid pVAX1/gB680+pp65m could be expressed in COS-7 cells, and could induce antibodies and cellular immune responses in BALB/c mice.
