Mulching techniques can comprise a solution that better utilizes precipitation and irrigation water in such a manner that mitigates soil degradation and drought damage; however, there are still gaps in the literature with regard to the effect of the use of mulch materials on the development of plant-soil-microbe interactions. Waste fibers, as alternative biodegradable mulch materials, are becoming increasingly prominent. The effect of wool mulch (WM) on water use efficiency, with regard to pepper seedlings, was investigated in different soil types (sand, clay loam, peat) in a pot experiment. Two semi-field experiments were also set up to investigate the effect of WM-plant interactions on sweet pepper yields, as compared with agro textiles and straw mulches. Soil parameters (moisture, temperature, DHA, ß-glucosidase enzymes, permanganate-oxidizable carbon) were measured during the growing season. The effect of WM on yield and biomass was more significant with the less frequent irrigation and the greater water-holding capacity of soils. Microbiological activity was significantly higher in the presence of plants, and because of the water retention of WM, the metabolic products of roots and the more balanced soil temperature were caused by plants. In the sandy soil, the straw mulch had a significantly better effect on microbiological parameters and yields than the agro textiles and WM. In soils with a higher water capacity, WM is a sustainable practice for improving the biological parameters and water use efficiency of soil. The effect of WM on yields cannot solely be explained by the water retention of the mulch; indeed, the development of biological activity and plant-soil-microbe interactions in the soil are also contributing factors.

BACKGROUND: Prevention of metastatic invasion is one of the main challenges in the treatment of alveolar rhabdomyosarcoma. Still the therapeutic options are limited. Therefore, an anti-tumor screening was initiated focusing on the anti-metastatic and anti-invasion properties of selected medicinal plant extracts and phytoestrogens, already known to be effective in the prevention and treatment of different cancer entities. METHODS: Treatment effects were first evaluated by cell viability, migration, invasion, and colony forming assays on the alveolar rhabdomyosarcoma cell line RH-30 in comparison with healthy primary cells. RESULTS: Initial anti-tumor screenings of all substances analyzed in this study, identified the plant extract of Vincetoxicum arnottianum (VSM) as the most promising candidate, harboring the highest anti-metastatic potential. Those significant anti-motility properties were proven by a reduced ability for migration (60%), invasion (99%) and colony formation (61%) under 48 h exposure to 25 µg/ml VSM. The restricted motility features were due to an induction of the stabilization of the cytoskeleton - actin fibers were 2.5-fold longer and were spanning the entire cell. Decreased proliferation (PCNA, AMT, GCSH) and altered metastasis (e. g. SGPL1, CXCR4, stathmin) marker expression on transcript and protein level confirmed the significant lowered tumorigenicity under VSM treatment. Finally, significant alterations in the cell metabolism were detected for 25 metabolites, with levels of uracil, N-acetyl serine and propanoyl phosphate harboring the greatest alterations. Compared to the conventional therapy with cisplatin, VSM treated cells demonstrated a similar metabolic shutdown of the primary cell metabolism. Primary control cells were not affected by the VSM treatment. CONCLUSIONS: This study revealed the VSM root extract as a potential, new migrastatic drug candidate for the putative treatment of pediatric alveolar rhabdomyosarcoma with actin filament stabilizing properties and accompanied by a marginal effect on the vitality of primary cells.

(1) Modern, intensive agricultural practices have been attributed to the loss of insect biodiversity and abundance in agroecosystems for the last 80 years. The aim of this work is to test whether there are statistically significant differences in insect abundance between different zones and over time on the vineyard field. (2) The study was carried out in five intensive wine farms in Spain over a three-year period (2013-2015). Each field was divided into two zones, one where cover plants were planted, and another remained unchanged (without cover). (3) A clear trend to increase the average number of insect species and individuals throughout the years in all farms was observed. Moreover, the zones with cover plants showed a significant difference with respect to the zones without. (4) The use of permanent cover plants allows creating areas of refuge for the insects favouring their conservation and reducing the agriculture impact in the insect decline.

Virus neutralizing antibodies are critical correlates of protection in vaccine development and are discriminatory in the plaque reduction neutralization test when used for the diagnosis of viral infections. However, neutralization assays are time consuming, labor intensive and highly variable, thus limiting their use. Advances in automated live imaging of cells opened new possibilities for standard virus diagnostic techniques such as neutralization assays. To this end, a reporter cell line based on the translocation of the transcription factor IRF3 in response to infection is proposed. Image acquisition of signal in a microplate format allowed the setup of a rapid, semi-automated and high-throughput fluorescent neutralization test. The study is extended to the live imaging of IRF3 translocations that could potentially cut the time of analysis to few hours. The fluorescent neutralization test is suitable for high-throughput assays and expandable to other viruses of global importance such as Zika virus.

BACKGROUND: A 2 year study was conducted to evaluate the efficacy of biological control with optimally timed Trichogramma brassicae releases as an integrated pest management tool against the European corn borer (ECB), Ostrinia nubilalis (Hübner), in on-farm experiments (i.e. real field conditions) in three European regions with dissimilar geoclimatic conditions and ECB pressure and conventional management (i.e. insecticide treated and untreated). RESULTS: Biological control with Trichogramma (1) provided ECB protection comparable with conventional management, (2) in all cases maintained mycotoxin levels below the EU threshold for maize raw materials destined for food products, (3) was economically sustainable in southern France and northern Italy, but not in Slovenia where it resulted in a significant decrease in gross margin, mainly owing to the cost of Trichogramma product, and (4) enabled avoidance of detrimental environmental effects of lambda-cyhalothrin use in northern Italy. CONCLUSION: Optimally timed mass release of T. brassicae could be considered a sustainable tool for IPM programmes against ECB in southern France and northern Italy. Better involvement of regional advisory services is needed for the successful dissemination and implementation of biological control. Subsidy schemes could also motivate farmers to adopt this IPM tool and compensate for high costs of Trichogramma product.
