RESUMO

The advantage of 3D printing-that is, additive manufacturing (AM) of structural materials-has been severely compromised by their disappointing fatigue properties1,2. Commonly, poor fatigue properties appear to result from the presence of microvoids induced by current printing process procedures3,4. Accordingly, the question that we pose is whether the elimination of such microvoids can provide a feasible solution for marked enhancement of the fatigue resistance of void-free AM (Net-AM) alloys. Here we successfully rebuild an approximate void-free AM microstructure in Ti-6Al-4V titanium alloy by development of a Net-AM processing technique through an understanding of the asynchronism of phase transformation and grain growth. We identify the fatigue resistance of such AM microstructures and show that they lead to a high fatigue limit of around 1 GPa, exceeding the fatigue resistance of all AM and forged titanium alloys as well as that of other metallic materials. We confirm the high fatigue resistance of Net-AM microstructures and the potential advantages of AM processing in the production of structural components with maximum fatigue strength, which is beneficial for further application of AM technologies in engineering fields.

Accumulating evidence indicates that a ligand of programmed cell death receptor-1 (PD-L1) participates in the progression and recurrence of multiple malignancies, including osteosarcoma. Nevertheless, the role of PD-L1 in chemoresistance development is not fully understood. In the current study, we aim to clarify the interaction of miR-519d-3p and PD-L1 in the development of cisplatin resistance. Immunohistochemistry, quantitative reverse-transcription polymerase reaction, and Western blot were used to evaluate PD-L1 expression. MTT and transwell migration assays were used to measure cell growth and motility, respectively. ENCORI, miRCode, and miRDB databases were recruited to predict candidate miRNAs targeting PD-L1. The binding sequences of miR-519d-3p and PD-L1 3' untranslated region were identified by dual-luciferase reporter and RNA immunoprecipitation assays. Flow cytometric analysis was conducted to measure the cycle distribution and cell apoptosis. Metastatic mouse models were generated with cisplatin-resistant sublines by intravenous injection. We found that PD-L1 expression was positively correlated to cisplatin resistance and metastasis, whereas miR-519d-3p expression was reduced in cisplatin-resistant specimens and was negatively correlated to cisplatin resistance and metastasis of osteosarcoma. We demonstrated that miR-519d-3p overexpression reversed cisplatin resistance, induced G1/S phase arrest and apoptosis. In addition, we proved that miR-519d-3p inhibited lung metastasis by establishing cisplatin-resistant MG63 metastatic xenograft models. The present findings suggest that miR-519d-3p/PD-L1 axis is a novel signaling pathway contributing to cisplatin resistance. Our study provides new clues for curing refractory osteosarcoma beyond immune checkpoint inhibitors.

OBJECTIVE: To investigate the effect of bilateral superior cervical sympathetic ganglion occlusion (SCG) on aortic dissection and its possible mechanism. METHODS: Forty-five SD rats were randomly divided into three groups with 15 in each:blank control group, sham operation group and SCG group. ß-aminopropione (666 mg·kg-1·d-1) was given by subcutaneous injection for 4 weeks to establish the aortic dissection model. Rats in SCG group were given SCG before the injection of ß-aminopropione. Blood pressure and heart rate of the rats were monitored using noninvasive tail artery blood pressure measuring instrument; sympathetic activity was monitored using drug block method; the structure of aortic wall was observed using HE staining; collagen fibers in aortic wall was observed using Sirius red staining; protein expression of Apelin was detected by immunohistochemistry; and the protein expression of matrix metalloproteinase (MMP)-2, 9 was detected by Western blotting. RESULTS: During the experiment, the body mass of the sham operation group and SCG group was smaller than that of the blank control group (all P<0.05), and the body mass of the SCG group was larger than that of the sham operation group (all P<0.05). The heart rate and sympathetic activity of the sham operation group were higher than those of the blank control group (all P<0.05), while the SCG group were lower (all P<0.05). Compared with the blank control group, the aortic wall in the sham operation group was thickening, while that in the SCG group was improved. A large number of collagen-1 in the aortic wall of the blank control group was stained brown by Sirius red, which was lighter in SCG group, and the staining in the sham operation group was the lightest. Compared with the blank control group, the expression of Apelin, MMP-2 and MMP-9 protein in the sham operation group increased (all P<0.05), while those in the SCG group decreased (all P<0.05). CONCLUSIONS: SCG can effectively reduce the incidence and mortality of aortic dissection in rats, which may be related to the inhibition of sympathetic activity and the decrease of collagen-1, Apelin, MMP-2 and MMP-9 expression.

Spoofing is becoming a serious threat to various Global Navigation Satellite System (GNSS) applications, especially for those that require high reliability and security such as power grid synchronization and applications related to first responders and aviation safety. Most current works on anti-spoofing focus on spoofing detection from the individual receiver side, which identifies spoofing when it is under an attack. This paper proposes a novel spoofing network monitoring (SNM) mechanism aiming to reveal the presence of spoofing within an area. Consisting of several receivers and one central processing component, it keeps detecting spoofing even when the network is not attacked. The mechanism is based on the different time difference of arrival (TDOA) properties between spoofing and authentic signals. Normally, TDOAs of spoofing signals from a common spoofer are identical while those of authentic signals from diverse directions are dispersed. The TDOA is measured as the differential pseudorange to carrier frequency ratio (DPF). In a spoofing case, the DPFs include those of both authentic and spoofing signals, among which the DPFs of authentic are dispersed while those of spoofing are almost overlapped. An algorithm is proposed to search for the DPFs that are within a pre-defined small range, and an alarm will be raised if several DPFs are found within such range. The proposed SNM methodology is validated by simulations and a partial field trial. Results show 99.99% detection and 0.01% false alarm probabilities are achieved. The SNM has the potential to be adopted in various applications such as (1) alerting dedicated users when spoofing is occurring, which could significantly shorten the receiver side spoofing cost; (2) in combination with GNSS performance monitoring systems, such as the Continuous Operating Reference System (CORS) and GNSS Availability, Accuracy, Reliability anD Integrity Assessment for Timing and Navigation (GAARDIAN) System, to provide more reliable monitoring services.

Based on the application of finite element analysis in spine biomechanics,the research progress of finite element method applied in lumbar spine mechanics is reviewed and the prospect is forecasted.The related works,including lumbar ontology modeling,clinical application research,and occupational injury and protection,are summarized.The main research areas of finite element method are as follows:new accurate modeling process,the optimized simulation method,diversified clinical effect evaluation,and the clinical application of artificial lumbar disc.According to the recent research progress,the application prospects of finite element method,such as automation and individuation of modeling process,evaluation and analysis of new operation methods and simulation of mechanical damage and dynamic response,are discussed.The purpose of this paper is to provide the theoretical reference and practical guidance for the clinical lumbar problems by reviewing the application of finite element method in the field of the lumbar spine biomechanics.

RESUMO

Serotonin type 3 (5-HT3) receptor partial agonists have been targeted as potential new drugs for the symptomatic relief of irritable bowel syndrome (IBS). Multiple diazepinone-based compounds have been discovered, which exhibit nanomolar binding affinity for the h5-HT3A receptor and display a range of intrinsic activities (IA=7-87% of 5-HT Emax) in HEK cells heterologously expressing the h5-HT3A receptor. Favorable physicochemical properties and in vitro ADME profile coupled with oral activity in the murine von Bezold-Jarisch reflex model demonstrates the series has promise for producing low to moderate IA partial agonists suitable for an IBS indication.

RESUMO

The sodium glucose co-transporter 2 (SGLT2) has received considerable attention in recent years as a target for the treatment of type 2 diabetes mellitus. This report describes the design, synthesis and structure-activity relationship (SAR) of C-glycosides with benzyltriazolopyridinone and phenylhydantoin as the aglycone moieties as novel SGLT2 inhibitors. Compounds 5p and 33b demonstrated high potency in inhibiting SGLT2 and high selectivity against SGLT1. The in vitro ADMET properties of these compounds will also be discussed.

Extracting rare earth elements (REEs) from wastewater is crucial for saving the environment, sustainable use of natural resources and economic growth. Reported here is a simple, low cost and one-step synthesis of Fe nanoparticles (FeNPs) based on two plant extracts having the ability to recover REEs. The synthesis of FeNPs using Excoecaria cochinchinensis leaves extract (Ec-FeNPs) exhibited high selectivity for heavy rare earth due to unique biomolecules, achieving separation coefficients (Kd) of 3.16 × 103-4.04 × 106 mL/g and recovery efficiencies ranging from 71.7 to 100 %. Conversely, the synthesis of FeNPs using Pinus massoniana lamb extract (PML-FeNPs) revealed poorer REE recovery efficiencies of 7.2-86.7 %. To understand the differences between Ec-FeNPs and PML-FeNPs in terms of selectivity and efficiency, LC-QTOF-MS served to analyze the biomolecules differences of two plant extracts. In addition, various types of characterization were carried out to identify the different functional groups encapsulated on the surface of FeNPs. These results reveal the source of the difference in the selectivity of Ec-FeNPs and PML-FeNPs for REEs. Furthermore, during DFT calculations, it was found that biomolecules with varying affinities for the surface of FeNPs interact with each other, leading to the formation of structures that exhibit high reactivity towards REEs. Finally, incorporating Spearman correlation analysis demonstrates that the selective removal efficiency of REEs was closely linked to surface complexation, ion exchange, and electrostatic adsorption. Consequently, this work strongly highlights the potential for the practical application of novel adsorbents in this field.

Globally extensive research into how silver nanoparticles (AgNPs) affect enzyme activity in soils with differing properties has been limited by cost-prohibitive sampling. In this study, customized machine learning (ML) was used to extract data patterns from complex research, with a hit rate of Random Forest > Multiple Imputation by Chained Equations > Decision Tree > K-Nearest Neighbors. Results showed that soil properties played a pivotal role in determining AgNPs' effect on soil enzymes, with the order being pH > organic matter (OM) > soil texture ≈ cation exchange capacity (CEC). Notably, soil enzyme activity was more sensitive to AgNPs in acidic soil (pH < 5.5), while elevated OM content (>1.9 %) attenuated AgNPs toxicity. Compared to soil acidification, reducing soil OM content is more detrimental in exacerbating AgNPs' toxicity and it emerged that clay particles were deemed effective in curbing their toxicity. Meanwhile sand particles played a very different role, and a sandy soil sample at > 40 % of the water holding capacity (WHC), amplified the toxicity of AgNPs. Perturbation mapping of how soil texture alters enzyme activity under AgNPs exposure was generated, where soils with sand (45-65 %), silt (< 22 %), and clay (35-55 %) exhibited even higher probability of positive effects of AgNPs. The average calculation results indicate the sandy clay loam (75.6 %), clay (74.8 %), silt clay (65.8 %), and sandy clay (55.9 %) texture soil demonstrate less AgNPs inhibition effect. The results herein advance the prediction of the effect of AgNPs on soil enzymes globally and determine the soil types that are more sensitive to AgNPs worldwide.

While nitrogen (N) deposition and over-fertilization enrich N in soil, it is unclear how it impacts soil organic carbon (SOC) transformation at the aggregate scale. Herein, a 90-day study reveals the transformation mechanisms of SOC in soil aggregates under nitrate and ammonium enrichment conditions. Results showed that nitrate treatment (NT) and ammonium treatment (AT) significantly increased SOC content by 15.6 % and 18.9 %, respectively. In addition, NT increased SOC accrual in large macro-aggregates (LMA), while AT increased SOC accrual in small macro-aggregates (SMA) and micro-aggregates (MA). Further analysis of pyrolysis products showed that N enrichment drove the transformation of labile soil organic matter (SOM) composition into recalcitrant SOM, with polysaccharides declining from 19-30 % to 2-13 %, while lipids rose from 18-27 % to 33-45 %. LMA and SMA contained more aromatic compounds than MA. This is linked to the inhibition of the expression of C degradation function genes, while almost all genes encoding SOC degradation are down-regulated under N enrichment. In the meantime, NT increased the abundance of genes encoding the degradation of N-containing compounds in LMA. Moreover, NO3- enrichment exerted a higher inhibitory effect on labile SOC degradation while NH4+ enrichment substantially inhibited recalcitrant SOC. Finally, Random Forest analysis confirmed that N enrichment elevated the importance of N-containing compounds' metabolism, which diminished when the size of soil aggregates decreased. In contrast, the importance of genes encoding saccharides and cellulose metabolism increased in smaller aggregates. This study highlights that both N type and aggregate size were determining factors in shaping SOC transformation in the N enrichment process.

The in-situ leaching of rare earth minerals results in ecological differences between sunlit and shady slopes, which may be related to differences in the distribution REEs in the associated soil matrices. Studies of REEs mine tailings in Southern China indicated higher total concentrations of REEs on sunlit slopes compared to shady ones. Specifically, the exchangeable REEs fraction (F1-REEs) was higher on the shady slopes, whereas the Fe/Mn oxides bound REEs fraction (F3-REEs) was higher on the sunlit slopes. In addition, light REE (LREE) concentrations were lower at lower elevations. With the exception of the Ce fraction which remained stable, this indicated a change in all REEs distributions, moving from F1-REEs towards the residual fraction. Hierarchical cluster and principal component analysis revealed a strong correlation between F3-REEs, organic matter bound REEs (F4-REEs), and LREEs, and a positive association of F3-REEs with sunlight exposure. Partial Least Squares Path Modeling analysis suggested that OM promoted the conversion of LREEs to F3 and F4-REEs in soil driven by sunlight exposure. Additionally, as the Feo/Fed ratio decreased, more LREEs were converted to F3. This study suggests that sunlight and elevation both play a critical role in the geochemical dynamics of REEs in in-situ tailings, advocating for environmental evaluations to be undertaken in order to accurately understand the ecological impacts of rare earth mining.

In this study, machine learning models predicted the impact of silver nanoparticles (AgNPs) on soil enzymes. Artificial neural network (ANN) optimized with genetic algorithm (GA) (MAE = 0.1174) was more suitable for simulating overall trends, while the gradient boosting machine (GBM) and random forest (RF) were ideal for small-scale analysis. According to partial dependency profile (PDP) analysis, polyvinylpyrrolidone coated AgNPs (PVP-AgNPs) had the most inhibitory effect (average of 49.5%) on soil enzyme activity among the three types of AgNPs at the same dose (0.02-50 mg/kg). The ANN model predicted that enzyme activity first declined and then rose when AgNPs increased in size. Based on predictions from the ANN and RF models, when exposed to uncoated AgNPs, soil enzyme activities continued to decrease before 30 d, but gradually rose from 30 to 90 d, and fell slightly after 90 d. The ANN model indicated the importance order of four factors: dose > type > size > exposure time. The RF model suggested the enzyme was more sensitive when experiments were conducted at doses, sizes, and exposure times of 0.01-1 mg/kg, 50-100 nm, and 30-90 d, respectively. This study presents new insights on the regularity of soil enzyme responses to AgNPs.

Neuroblastoma (NB) is a common childhood cancer. Circular RNA RAN binding protein 17 (circRANBP17) has been identified to participate in diverse tumor progression. This study aims to explore the function and mechanism of circRANBP17 in NB. The levels of circRANBP17, miR-27b-3p and KDM1A in NB tissues and cells were measured by qRT-PCR. Mouse model assay was performed to investigate the effect of circRANBP17 knockdown on tumor formation in vivo. The levels of circRANBP17 and KDM1A were significantly up-regulated, and the level of miR-27b-3p was strikingly down-regulated in NB tissues and cells (SK-N-SH and SK-N-AS). Functional studies indicated that miR-27b-3p inhibitor mitigated the inhibitory effects on cell proliferation, migration, invasion and the promoting effect on cell apoptosis in SK-N-SH and SK-N-AS cells induced by circRANBP17 knockdown. Also, miR-27b-3p regulated NB cell malignancy by targeting KDM1A. Further studies revealed that miR-27b-3p inhibitor reversed the low expression of KDM1A induced by circRANBP17 knockdown. In support, circRANBP17 knockdown led to inhibition of tumor formation in vivo. In conclusion, circRANBP17 modulated KDM1A to promote cell proliferation, migration, invasion and restrain cell apoptosis in NB by sponging miR-27b-3p, and the new regulatory network may provide a theoretical basis for the further study of NB.

Transient quantum hyperdiffusion, namely, faster-than-ballistic wave packet spreading for a certain time scale, is found to be a typical feature in tight-binding lattices if a sublattice with on-site potential is embedded in a uniform lattice without on-site potential. The strength of the sublattice on-site potential, which can be periodic, disordered, or quasiperiodic, must be below certain threshold values for quantum hyperdiffusion to occur. This is explained by an energy band mismatch between the sublattice and the rest uniform lattice and by the structure of the underlying eigenstates. Cases with a quasiperiodic sublattice can yield remarkable hyperdiffusion exponents that are beyond three. A phenomenological explanation of hyperdiffusion exponents is also discussed.

During the process of peripheral nerve regeneration, a single neuron can regenerate and maintain more than one collateral in a regenerative distal stump. Furthermore, some of the new shoots can mature gradually through remyelination and grow into the remote target organ to play a physiological function. Our study found that when neonatal nerve fibers are subjected to a second injury, the regenerative distal stump can regenerate and maintain more than one collateral in the second regenerative distal stump. The neonatal nerve contributed to the functional recovery of the nerve, but the restoration of nerve function was not complete.

Dramatic imaging viewpoint variation is the critical challenge toward action recognition for depth video. To address this, one feasible way is to enhance view-tolerance of visual feature, while still maintaining strong discriminative capacity. Multi-view dynamic image (MVDI) is the most recently proposed 3-D action representation manner that is able to compactly encode human motion information and 3-D visual clue well. However, it is still view-sensitive. To leverage its performance, a discriminative MVDI fusion method is proposed by us via multi-instance learning (MIL). Specifically, the dynamic images (DIs) from different observation viewpoints are regarded as the instances for 3-D action characterization. After being encoded using Fisher vector (FV), they are then aggregated by sum-pooling to yield the representative 3-D action signature. Our insight is that viewpoint aggregation helps to enhance view-tolerance. And, FV can map the raw DI feature to the higher dimensional feature space to promote the discriminative power. Meanwhile, a discriminative viewpoint instance discovery method is also proposed to discard the viewpoint instances unfavorable for action characterization. The wide-range experiments on five data sets demonstrate that our proposition can significantly enhance the performance of cross-view 3-D action recognition. And, it is also applicable to cross-view 3-D object recognition. The source code is available at https://github.com/3huo/ActionView.

OBJECTIVE: Low serum adiponectin level can predict hypertension development, and adiponectin gene (ADIPOQ) polymorphisms have been reported to be linked with hypertension risk. Whereas, the interaction between ADIPOQ polymorphisms and environmental factors on the susceptibility of hypertension remained unclear. The purpose of this study was to explore the relationship of ADIPOQ polymorphisms with hypertension risk and their interaction with lipid levels in coal miners. METHODS: A matched case-control study with 296 case-control pairs was performed in a large coal mining group located in North China. The participants were questioned by trained interviewers, and their ADIPOQ genotype and lipid levels were determined. Logistic regression, stratified analysis, and crossover analysis were applied to evaluate the effects of rs2241766, rs1501299, and rs266729 genotypes and gene-lipid interaction on hypertension risk. RESULTS: In this matched case-control study, the genotypes of rs2241766 TG+GG, rs1501299 GT+TT, and rs266729 CG+GG were marginally related to hypertension risk. Individuals with high total cholesterol (TC), low-density lipoprotein cholesterol (LDL-C), and high-density lipoprotein cholesterol (HDL-C) level were susceptible to hypertension (TC: odds ratio [OR] = 1.807, 95% confidence intervals [95%CI] = 1.266-2.581; LDL-C: OR = 1.981, 95%CI = 1.400-2.803; HDL-C: OR = 1.559, 95%CI = 1.093-2.223). Antagonistic interactions were detected between rs2241766 and TC, rs1501299 and TC, rs2241766 and LDL-C, and rs1501299 and HDL-C (rs2241766 and TC: OR = 0.393, 95%CI = 0.191-0.806; rs1501299 and TC: OR = 0.445, 95%CI = 0.216-0.918; rs2241766 and LDL-C: OR = 0.440, 95%CI = 0.221-0.877; rs1501299 and HDL-C: OR = 0.479, 95%CI = 0.237-0.967). Stratified analysis showed that hypertension risk was high for the subjects with rs2241766 TG+GG or rs1501299 GG under the low lipid level but low for those under the high lipid level. In the case group, the TC and LDL-C levels for rs2241766 TG+GG were lower than those for rs2241766 GG, and the TC and HDL-C levels for rs1501299 GT+TT were higher than those for rs1501299 GG. CONCLUSIONS: Although the effects of ADIPOQ polymorphisms alone were not remarkable, an antagonistic interaction was observed between ADIPOQ polymorphisms and lipid levels.

RESUMO

Serotonin type 3 (5-HT(3)) receptor partial agonists are being targeted as potential new drugs for the treatment of irritable bowel syndrome (IBS). Two new chemical series bearing indazole and indole cores have exhibited nanomolar binding affinity for the h5-HT(3)A receptor. A range of partial agonist activities in HEK cells heterologously expressing the h5-HT(3)A receptor were measured for the indazole series. Excellent 5-HT(3) receptor selectivity, favorable in vitro metabolic stability and CYP inhibition properties, and good oral in vivo potency in the murine von Bezold-Jarisch reflex model is exemplified thereby indicating the series to have potential utility as improved IBS agents.

OBJECTIVE: To analyze the time distributions of traffic accidents happening in different age groups in Beijing from 2004 to 2008, and to provide information on the prevention and rescue. METHODS: The traffic injury cases recorded by Beijing Emergency Medical Center from 2004 to 2008 were analyzed, and the data were separated by age: the youth (0-17 years old), the adult (18-64 years old) and the senior adult (above 65 years old). The constituent ratios of hours and months were calculated to describe the states and time distributions of all the cases. RESULTS: The high peaks of accidents for the youth group occurred in the periods 7:00 to 8:00 (6.77%, 39) and 16:00 to 18:00 (10.38%, 60;10.73%, 62), and for the senior adult group appeared in the period 9:00 to 11:00 (11.19%, 151; 11.04%, 149). The adult group showed a stable trend during the period 8:00 to 24:00. The amount of traffic injury happening in these three groups had the same status monthly, which turned out that most traffic accidents happened in October for all the groups (12.11%, 70; 10.38%, 1 257; 12.30%, 166), and February had the smallest number (4.15%, 24; 5.28%, 640; 5.26%, 71). CONCLUSION: The time distributions of traffic injury within these three groups do not show the same situation daily but do monthly. The emergency treatment team and traffic control personnel should pay attention to this and have special protocol for different cases to increase the efficiency of the prehospital rescue.

OBJECTIVE: To analyze the characteristics and laws of road traffic accidents in Beijing, and provide some accident-prevention measures. METHODS: The data from Beijing Emergency Medical Center were managed by computer,then the descriptive statistical analyses were made. RESULTS: (1) A total of 2 984 traffic accidents with relatively complete information were recorded in our research,in which 42 people were led to death and 2 942 injured. Most of the casualties were male. The ages of the injured segments were mainly between 18 to 59 years.(2) The number of the injured in accidents between the motor vehicles was the largest, which was 1 883; The deaths caused by accidents between the motor vehicles and pedestrians were the most, with the number of 26;Major vicious accidents were mainly caused by accidents between the motor vehicles and the number was 11.(3) In one day accidents mainly happened at 8:00-10:00 ,and 14:00-18:00.(4) The most common injuries were the head and face injuries, with the number of 921, followed by multi-site combined injuries with the number of 761, and lower limb injuries, with the number of 541.(5) Human factors were the main causes of accidents, followed by vehicle factors. CONCLUSION: The illegal driving and walking through the pedestrian lane were the main causes of car accidents. Strengthening traffic safety education of the public, especially of the floating population, rigorous training of drivers for traffic rules and regulations, enhancing the public awareness of road safety, and improving the road traffic management and control measures were the main measures to prevent and control traffic injuries.
