RESUMO

This paper summarizes recent insights into causal biological mechanisms underlying the carcinogenicity of asbestos. It addresses their implications for the shapes of exposure-response curves and considers recent epidemiologic trends in malignant mesotheliomas (MMs) and lung fiber burden studies. Since the commercial amphiboles crocidolite and amosite pose the highest risk of MMs and contain high levels of iron, endogenous and exogenous pathways of iron injury and repair are discussed. Some practical implications of recent developments are that: (1) Asbestos-cancer exposure-response relationships should be expected to have non-zero background rates; (2) Evidence from inflammation biology and other sources suggests that there are exposure concentration thresholds below which exposures do not increase inflammasome-mediated inflammation or resulting inflammation-mediated cancer risks above background risk rates; and (3) The size of the suggested exposure concentration threshold depends on both the detailed time patterns of exposure on a time scale of hours to days and also on the composition of asbestos fibers in terms of their physiochemical properties. These conclusions are supported by complementary strands of evidence including biomathematical modeling, cell biology and biochemistry of asbestos-cell interactions in vitro and in vivo, lung fiber burden analyses and epidemiology showing trends in human exposures and MM rates.

PURPOSE: The German Aerospace Center (DLR) investigated in the NORAH sleep study the association between a distinct change in nocturnal aircraft noise exposure due to the introduction of a night curfew (11:00 p.m.-5:00 a.m.) at Frankfurt Airport and short-term annoyance reactions of residents in the surrounding community. Exposure-response curves were calculated by random effects logistic regression to evaluate the aircraft noise-related parameters (1) number of overflights and (2) energy equivalent noise level LASeq for the prediction of short-term annoyance. Data of the NORAH sleep study were compared with the STRAIN sleep study which was conducted by DLR near Cologne-Bonn Airport in 2001/2002 (N = 64), representing a steady-state/low-rate change. METHODS: The NORAH sleep study was based on questionnaire surveys with 187 residents living in the vicinity of Frankfurt Airport. Noise-induced short-term annoyance and related non-acoustical variables were assessed. Nocturnal aircraft noise exposure was measured inside the residents' home. RESULTS: A statistically significant rise in the portion of annoyed residents with increasing number of overflights was found. Similarly, the portion of annoyed subjects increased with rising LASeq. Importance of the frequency of fly-overs for the prediction of annoyance reactions was emphasized. The annoyance probability was significantly higher in the NORAH than in the STRAIN sleep study. CONCLUSIONS: Results confirm the importance of both acoustical parameters for the prediction of short-term annoyance due to nocturnal aircraft noise. Quantitative annoyance models that were derived at steady-state/low-rate change airports cannot be directly applied to airports that underwent a distinct change in operational and noise exposure patterns.

Over the last decade, numerous companies have marketed aqueous ozone sprayers for insect and disease management, but little to no data has been published on their efficacy. Thus, we evaluated the potential of both gaseous and aqueous ozone as a potential preharvest insecticide against the adult life stage of the invasive fruit pest, spotted-wing drosophila, Drosophila suzukii (Matsumura). Gaseous ozone was applied at two dosages, 14,600 and 30,100 ppmv, for varying durations and the respective concentration-time (CT) exposure responses were modeled for sex-specific mortalities recorded at 0, 24, 48, and 72 h following treatment. We found that gaseous ozone primarily caused mortality immediately following exposure, with slight increases 72 h following ozone treatments. The female and male lethal concentration-time (LCT) 50 estimates were significantly different at 0, 24, 48, and 72 h after 30,100 ppmv treatments, where males observed an increased mortality response. However, the LCT 99 estimates confidence intervals (95%) of adult female and male D. suzukii were similar at 0, 24, 48, and 72 h after 14,600 or 30,100 ppmv ozone treatments. In contrast, ozone dissolved in distilled water at 18.52 ppm (mg/L) did not provide any mortality after total immersion of subjects for 30 s. While gaseous ozone may have some utility as a fumigant for D. suzukii in closed vessels where concentrations could be maintained, we did not identify any insecticidal potential for ozone dissolved in aqueous solution when simulating a preharvest treatment under optimal laboratory conditions.
