OBJECTIVES: The southern Brazilian shellmounds provide archaeological evidence of prolonged human activity in the coast from approximately 6000 to 1000 BP. Shellmound building populations exploited the rich coastal estuarine zones, and the human remains recovered from them are important sources of information on health and overall lifestyle of these mid-Holocene groups. Therefore, they were included in the Western Hemisphere Global History of Health project. The shellmounds contribute the highest Health Index in the Western Hemisphere, but these conclusions are based on collections that exclude postcranial remains. Here, we reconstruct the Health Index for one specific shellmound using both cranial and postcranial remains to determine whether the initial studies might misrepresent the relative health of the Brazilian shellmound builders. MATERIALS AND METHODS: The Health Index was calculated for a sample of 18 complete skeletons recovered from the shellmound Porto do Rio Vermelho 02 (Santa Catarina Island, Brazil). The Heath Index was calculated with and without postcranial markers and the results are compared with the Western Hemisphere Global History of Health data. RESULTS: The Health Index for Porto do Rio Vermelho 02 is lower than the reported average for American series in the Western Hemisphere Global History of Health Project and considerably lower than the original index reported for Brazilian shellmounds. This discrepancy is due to an increased prevalence of infectious disease and low stature. CONCLUSIONS: Although the Health Index remains a useful comparison statistic, re-evaluation of fragmentary skeletal remains demonstrates the need for caution when applying it to incomplete skeletal series.

The consumption of vertebrate tissues and eggs (hereinafter "meat") is relatively common among some primates that are highly frugivorous or eclectic omnivores, but rare or absent in those that are highly folivorous. The Neotropical howler monkeys (Alouatta spp.) belong in the latter group. Here we report the consumption of meat by free-ranging urban black and gold howler monkeys (Alouatta caraya) and discuss the potential role of the consumed meat as a source of energy, protein, or micronutrients. We studied three groups of howler monkeys (comprising four to seven individuals), living in city squares (0.6, 1.5, and 1.9 ha) in south Brazil, from July 2022 to May 2023 (65 days; 797 h of observations). All of the study groups were spontaneously supplemented daily by people with variable amounts and types of food provided. Meat was only offered in the two larger squares. The groups' diets included leaves (42-49% scan sampling feeding records), fruit (3-20%), and flowers (2-5%) from 13 to 20 plant species, and considerable amounts of supplemented food (27-50%). We recorded 33 individual events of ingestion of supplemented cooked meat, three individual events of dove egg predation, and three bird nest inspections without egg consumption. All members of the two groups in the larger squares, except an infant male, ingested meat at least once. Meat accounted for 1% of total scan feeding records of both groups with access to this supplement. We conclude that whereas the opportunistic consumption of meat probably contributed only minor amounts of energy and protein to the study subjects, it may have benefitted them with micronutrients that are scarce in plant foods.

We surveyed macronutrients and dissolved iron (DFe) concentrations and speciation in a transect over the shelf of the South Brazil Bight (SBB) at Santa Marta Grande Cape (SE Brazil) during a coastal downwelling episode. Driven by dominant NE winds, coastal downwelling is a common feature during the austral summer and force after water convergence, with contribution of internal wave breaking at the shelf edge, upwelling of macronutrients into the nutrient-depleted waters of the southbound Brazil Current at ~100 km from the coastline. As a result, we found a plume of high turbidity that reached the euphotic layer, a deepening of the silicate, nitrate, and phosphate isolines over the shelf and a bulging of the nitrate and phosphate isolines over the shelf edge and the slope. Our first measurements of DFe concentration and speciation in the area revealed that against prior findings in other coastal areas, macronutrients, DFe, and iron ligand cycles were disentangled. Higher DFe concentrations were often found at the surface indicating aerial deposition. Secondary DFe maxima over the sediment-water interface and in the upwelled plume indicated DFe fluxes from the sediment and from resuspended instable colloids. Iron ligand concentrations were higher than DFe concentrations in most stations with a clear land-to-ocean gradient. Subtraction of HS iron ligands revealed that except in upwelled water, the bulk of surface ligands was the result of local biological processes. The analysis of the concentrations of Fe-HS complexes showed that the contribution of HS to DFe was dominant in upwelled waters, significant in waters close to the coast, but nearly negligible in the rest of the studied area. We hypothesize that the injection of iron-humic complexes into the euphotic layer during summer upwelling episodes is the key to understanding the persistent high chlorophyll meanders found over the shelf edge of the SBB coast.
RESUMO
In early 2017, an outbreak caused by an unknown and supposedly viral agent in the Marilena region of southern Brazil was investigated. Since the etiological agent causing the outbreak was not identified from human samples, mosquitoes from this region were collected. Three out of 121 mosquito pools collected from the region tested positive for alphavirus in molecular tests. Next generation sequencing results revealed the presence of a novel alphavirus, tentatively named here as Caainguá virus (CAAV). DNA barcoding analyses indicated that different species of Culex are hosts for CAAV. This new virus was basal to the New World encephalitic alphaviruses in a comprehensive and robust phylogenetic approach using complete genomes. Viral particles were observed in the cytosol and inside of intracellular compartments of cells in mosquito-derived cell cultures. Despite being noninfectious in vertebrate derived cell cultures, primary culturing of CAAV in human mononuclear cells suggests monocytes and lymphocytes as CAAV targets. However, the epidemiological link of CAAV on the human outbreak should be further explored.

Five new species-group taxa of Clitellata of the genus Glossoscolex from Rio Grande do Sul State, Brazil are described in this paper: Glossoscolex (Glossoscolex) riograndensis n. sp. has three subspecies, G. (G.) riograndensis riograndensis, G. (G.) riograndensis pollulus n. ssp., G. (G.) riograndensis nativus n. ssp. The two other new species are named G. (G.) pastivus n. sp., and G. (G.) pampas n. sp. All new species are part of the truncatus species group within the subgenus, characterized by the presence of male pores in segment xvii. A single unnamed specimen, also described here, is deemed to belong to a sixth new species-group taxon of the truncatus group. A tabular character comparison of the new taxa and known and morphologically similar species is given. Information on the habitat, such as vegetation cover and soil characteristics, is given as well. G. (G.) r. riograndensis and G. (G.) r. pollulus had an unusual storage of sperm in the nephridia. Histological sections were performed to verify it.

With the advent of Next-Generation Sequencing technology, sequencing of short tandem repeats (STRs) allows for a more detailed analysis when compared to size-based fragment methods (capillary electrophoresis-CE). The implementation of high-throughput sequencing can help uncover deeper genetic diversities of different populations. Subjects from the South region of Brazil present a particular and more homogeneous ancestry background when compared to other regions of the country. Both autosomal and Y- STRs have been analyzed in these individuals; however, all analyses published to date encompass data from CE-based fragment analysis. In this study, a genetic analysis of 59 individuals from Southern Brazil was performed on STR sequences. Forensically relevant STRs were PCR-enriched using a prototype of the PowerSeq™ AUTO/Y system (Promega Corp.). Next-generation sequencing was performed on an Illumina MiSeq instrument. The raw data (FASTQ files) were processed using a custom designed sequence processing tool, Altius. Isoalleles, which are sequence-based allelic variants that do not differ in length, were observed in nine autosomal and in six Y- STRs from the core global forensic marker set. The number of distinctive alleles based on sequence was higher when compared to those based on length, 37.3% higher in autosomal STRs and 13.8% higher in Y-STRs. The most polymorphic autosomal locus was D12S391, which presented 38 different sequence-based alleles. Among the loci in the Y chromosome, DYS389II presented the highest number of isoalleles. In comparison to CE analysis, Observed and Expected Heterozygosity, Polymorphic Information Content (PIC) and Genetic Diversity also presented higher values when the alleles were analyzed based on their sequence. For autosomal loci, Polymorphic Information Content (PIC) was 2.6% higher for sequence-based data. Diversity was 9.3% and 6.5% higher for autosomal and Y markers, respectively. In the analysis of the repeat structures for the STR loci, a new allele variant was found for allele 18 in the vWA locus. The STR flanking regions were also further investigated and sixteen variations were observed at nine autosomal STR loci and one Y-STR locus. The results obtained in this study demonstrate the importance of genetic analysis based on sequencing and highlight the diversity of the South Brazilian population when characterized by STR sequencing.

The genetic and morphometric population structures of the semiterrestrial crab Armases angustipes from along the Brazilian coast were examined. The influence of the Central South Equatorial Current on larval dispersal of A. angustipes also was evaluated. Six populations were sampled from estuarine areas in São Luis do Maranhão, Maranhão; Natal, Rio Grande do Norte; Maceió, Alagoas; Ilhéus, Bahia; Aracruz, Espírito Santo; and Guaratuba, Paraná. Patterns of genetic differentiation were assessed using DNA sequence data corresponding to parts of the mitochondrial cytochrome c oxidase subunit 1. Geometric morphometric techniques were used to evaluate morphological variation in shape and size of the carapace and right cheliped propodus. Our results revealed low genetic variability and lack of phylogeographic structure; geometric morphometrics showed statistically significant morphological differentiation and geographic structuring. Our data indicate the absence of possible barriers to gene flow for this mobile species, and no clear correlation of morphological or genetic variation with ocean currents and/or geographic distance. Our results also suggest that historical geological and climatological events and/or possible bottleneck effects influenced the current low genetic variability among the populations of A. angustipes.

Abstract INTRODUCTION This study registers Ascogregarina spp. infection in field populations of Aedes aegypti and Aedes albopictus in a subtropical region of Brazil. METHODS Mosquito larvae collected in tires placed in four municipalities of Santa Catarina were identified morphologically and assessed for Ascogregarina sp. infection using morphological and molecular methods. RESULTS Both mosquito species harbored Ascogregarina taiwanensis, whose genomic DNA was confirmed in both the Aedes species by PCR. DNA sequences were deposited in GenBank. Conclusion: Both Ae. albopictus e Ae. aegypti harbor Ascogregarina sp.

BACKGROUND Hepatitis B virus (HBV) infection is a major public health problem in Brazil. Several risk factors are involved in HBV infection and their identification by a rational and essential approach is required to prevent the transmission of this infection in Brazil. OBJECTIVES To evaluate risk factors associated with HBV infection in South Brazil. METHODS A total of 260 patients with HBV and 260 controls from Caxias do Sul (state of Rio Grande do Sul, Brazil) participated in this study. All participants were given a standard questionnaire to yield the sociodemographic information and to identify HBV risk factors. HBV infection was detected by HBsAg test in all participants. FINDINGS HBV infection in these cases was strongly associated with history of a family member HBV-infected, mainly mother [odds ratio (OR) = 4.86; 95% confidence intervals (CI): 1.69-13.91], father (OR = 5.28; 95% CI: 1.58-17.71), and/or siblings (OR = 22.16; 95% CI: 9.39-52.25); sharing personal objects (OR = 1.40; 95% CI: 1.37-2.38); and having history of blood transfusion (OR = 2.05; 95% CI: 1.10-2.84). CONCLUSIONS HBV infection was strongly associated with having a family member infected with hepatitis B, sharing personal objects, and having history of blood transfusion.

INTRODUCTION: Human immunodeficiency virus type 1 (HIV-1) has spread worldwide, with several subtypes and circulating recombinant forms. Brazil has an incidence of 20.5 HIV-1/acquired immunodeficiency syndrome (AIDS) patients per 100,000 inhabitants; however, the Southernmost State of Rio Grande do Sul (RS) has more than twice the number of HIV-1-infected people (41.3/100,000 inhabitants) and a different pattern of subtype frequencies, as previously reported in studies conducted in the capital (Porto Alegre) and its metropolitan region. This study examined HIV-1/AIDS epidemiological and molecular aspects in the countryside of Rio Grande do Sul. METHODS: Socio-demographic, clinical and risk behavioral characteristics were obtained from HIV-1-positive adult patients using a structured questionnaire. HIV-1 subtypes were determined by nested-polymerase chain reaction (PCR) and sequencing of the pol and env genes. RESULTS: The study sample included 149 (55% women) patients with a mean age of 41.8 ± 11.9 years. Most (73.8%) patients had a low education level and reported heterosexual practices as the most (91.9%) probable transmission route. HIV-1 subtypes were detected in 26 patients: 18 (69.2%) infected with subtype C, six (23.1%) infected with subtype B and two (7.7%) infected with BC recombinant forms. CONCLUSIONS: These data highlight the increasing number of HIV-1 subtype C infections in the countryside of South Brazil. .

Ethnobotanical data can be an important tool in the search for new drugs. The Brazilian Health Surveillance Agency accepts the registration of herbal medicines based on ethnopharmacological and ethnobotanical studies. With the purpose of increasing the knowledge of potentially useful plants for the treatment of painful conditions, we analyzed the ethnobotanical studies carried out in Rio Grande do Sul state (RS-Southern Brazil); we had access to nineteen studies.To our knowledge, this is the first compilation of ethnobotanical studies that focus on pain relief carried out in RS. The species native to RS cited in at least nine (about 50%) of these studies were selected. The search retrieved 28 native species cited as used to alleviate painful conditions, which are distributed in eighteen botanical families, being Asteraceae the most mentioned. The species more frequently cited for pain relief were Achyrocline satureioides, Baccharis articulata, Baccharis crispa, Lepidium didymum, Eugenia uniflora and Maytenus ilicifolia. The only species not reported in any pre-clinical study associated with pain relief was B. articulata. Among the six species cited, no studies on clinical efficacy were found. In conclusion, the folk use of native plants with therapeutic purposes is widespread in RS State (Brazil), being pain relief an important property.
RESUMO
The method used by YAGYU et al. for the subtype-specific polymerase chain reaction (PCR) amplification of the gp41 transmembrane region of the human immunodeficiency virus type-1 (HIV-1) env gene, was tested. HIV-1 proviral DNA from 100 infected individuals in Itajaí, South Brazil was used to analyze this method. Seventy individuals were determined according to this method as having PCR products at the expected size for subtypes B, C, D and F. Of these individuals, 26 (37.1%) were observed as having the expected amplification for subtype C, and 42 (60%) were observed as having the expected products for subtypes B and D. Of the subtype B and D amplicons, 16 (22.9%) were classified as subtype D, and 26 (37.1%) were classified as subtype B. Two individuals (2.9%) had amplicons that were observed after subtype F-specific amplification was performed. Sequencing and comparing the patient sequences to reference sequences confirmed the classification of sequences of subtypes C and B. However, sequences that were falsely determined as being D and F in the PCR assay were determined as being subtypes C and B, respectively, by sequence analysis. For those individuals from whom no amplified products were obtained, a low viral load that was indicated in their patient history may explain the difficulty in subtyping by PCR methods. This issue was demonstrated by the results of ANOVA when testing the effect of viral load on the success of PCR amplification. The alignment of the obtained sequences with HIV-1 reference sequences demonstrated that there is high intra-subtype diversity. This indicates that the subtype-specific primer binding sites were not conserved or representative of the subtypes that are observed in the Brazilian populations, and that they did not allow the correct classification of HIV-1 subtypes. Therefore, the proposed method by YAGYU et al. is not applicable for the classification of Brazilian HIV-1 subtypes.
A metodologia para amplificação subtipo-específica por PCR da região transmembrana do gene env (gp41) do HIV-1, descrita por Yagyu e colaboradores, foi testada a partir de DNA proviral de 100 pacientes infectados pelo HIV-1 de Itajaí, Sul do Brasil. Setenta indivíduos apresentaram produtos amplificados e correspondentes aos subtipos B, C, D e F de acordo com a metodologia escolhida. Destes indivíduos, 26 (37,1%) apresentaram a amplificação esperada para o subtipo C de acordo com a metodologia; 42 (60%) apresentaram os produtos esperados para os subtipos B e D, sendo que na etapa seguinte de diferenciação destes subtipos, 16 (22,9%) corresponderam ao subtipo D e 26 (37,1%) ao subtipo B. Dois indivíduos (2,9%) mostraram produtos amplificados após a amplificação específica para o subtipo F. O sequenciamento e a comparação com sequências referências confirmou a subtipagem de HIV-1 C e B obtida pela metodologia. No entanto, indivíduos subtipados erroneamente como HIV-1 D e F pela metodologia, foram classificados pela comparação com sequências referências como subtipos C e B, respectivamente. Em relação aos indivíduos que não mostraram produtos amplificados, a baixa carga viral observada no histórico destes pacientes seria em parte responsável pela dificuldade na subtipagem pela metodologia de PCR, como demonstrado pelo resultado significativo no ANOVA ao testar o efeito da carga viral no sucesso da amplificação. O alinhamento das sequências obtidas com sequências referências de HIV-1 correspondentes à região da gp41 demonstrou que há uma alta diversidade intra-subtipo e que as regiões a partir das quais foram desenhados os oligonucleotídeos iniciadores HIV-1 subtipo-específicos não são conservadas nem suficientemente representativas dos subtipos observados nas populações brasileiras para permitir sua correta identificação. Portanto, esta metodologia não é aplicável para populações virais brasileiras.

Na região sul do Rio Grande do Sul a infestação dos bovinos por Rhipicephalus (Boophilus) microplus ocorre, principalmente, entre os meses de outubro e abril, devido às condições climáticas. Além do conhecimento do ciclo biológico desse parasito, também é fundamental conhecer a epidemiologia, para estabelecer estratégias de controle. No Rio Grande do Sul, e também no Brasil, existem poucos estudos epidemiológicos a respeito da resistência aos acaricidas. Além disso, a grande área geográfica e a deficiência estrutural quanto ao uso e acesso a bancos de dados dificultam a obtenção de dados confiáveis. O presente estudo teve como objetivo realizar um inquérito abordando a percepção dos produtores da região sul do Rio Grande do Sul, quanto à identificação de populações de R. (B.) microplus difíceis de controlar com acaricidas e os fatores de risco para a seleção de populações de carrapatos resistentes. Para execução do trabalho foram coletados dados sobre o controle do carrapato de bovinos de corte, em 85 propriedades de sete municípios, localizados na região sul do Estado. Os resultados revelaram a existência de associação positiva entre a dificuldade de controlar o carrapato com os acaricidas e o grau de instrução do proprietário (até o ensino fundamental com OR=3,67 e p=0,01) e o número de aplicação de carrapaticida por ano (superior a 4 com OR=4,05 e p=0,006). Esses resultados indicam também que propriedades com mais de 100 bovinos de corte em criação extensiva, na região sul do rio Grande do Sul apresentam características que podem contribuir para uma maior vida útil dos carrapaticidas do que as verificadas em outras regiões do País.
In the southern region of Rio Grande do Sul, cattle become infested with Rhipicephalus (B.) microplus mainly between October and April due to the climatic conditions. In addition to knowing its life cycle, knowledge of parasite's epidemiology is essential to establish management strategies. Epidemiological studies on resistance to acaricides in Rio Grande do Sul as well as in the rest of Brazil are scarce. Moreover, the large geographical area and the structural deficiency with respect to the use and access to databases make reliable data difficult to obtain. The present study surveyed the perception by cattle breeders in the southern region of Rio Grande do Sul in regards of identification of R. (B.) microplus populations that are difficult to manage using acaricides, as well as the risk factors for the selection of resistant tick populations. Tick management data on beef cattle in 85 properties of seven municipalities were collected. The results revealed that the difficulty in tick management correlated positively with levels of education of the farm owners (up to elementary school, OR = 3.67 and p = 0.01) and with the yearly number of acaricide applications (over four, with OR = 4.05 and p= 0.006). These results also suggest that properties with more than 100 beef cattle under extensive farming conditions in the southern region of Rio Grande do Sul show characteristics that may contribute to longer acaricide lifetimes as compared to other regions in the Country.

The biodiversity of marine heterotrophic protists is poorly known in the South Atlantic coastal zone (~32 ºS) especially regarding the nanoflagellates. The presence of loricate choanoflagellates was reported for the first time in the Patos Lagoon estuary and the adjacent coastal zone. Seventeen species of eleven genera of loricate choanoflagellates were observed between October 1998 and May 2000 in fixed water samples (lugol's solution + glutaraldehyde) in a JEM 100-SX transmission electron microscope. Most species were recorded in euhaline and mixopolyhaline waters during the spring and summer, none in autumn and a few (four) in winter. The absence of choanoflagellates at the more sheltered inshore stations is due freshwater influence, and at the beach station, probably due the strong wave action. The probably cosmopolitan species Pleurasiga minima, Cosmoeca norvegica, C. ventricosa and Parvicorbicula circularis were present in spring or summer in the estuary channel and coastal area while Stephanoeca diplocostata which apparently prefers lower temperature, was recorded in winter. Calotheca alata and Campyloacantha spinifera are mainly temperate species and were present in spring. The new species Diplotheca tricyclica was recorded at the estuary channel in the summer 1999, in high salinity water.
A biodiversidade de protistas heterotróficos é pouco conhecida na zona costeira do Atlântico Sul (~32 ºS), especialmente dos nanoflagelados. No presente estudo, foi registrada pela primeira vez a presença de coanoflagelados loricados no estuário da Lagoa dos Patos e na zona costeira adjacente. Dezessete espécies de onze gêneros de coanoflagelados loricados foram observadas em amostras de água fixadas (solução de lugol + glutaraldeido) coletadas entre outubro de 1998 e maio de 2000, e analisadas em microscópio de transmissão JEM 100-SX. A maior parte das espécies foi registrada em águas euhalinas e mixopolihalinas durante a primavera e o verão, nenhuma no outono e poucas (quatro) no inverno. A ausência de coanoflagelados nas estações mais protegidas deve-se a influência da água doce, e na estação da praia deve-se, provavelmente, a forte ação das ondas. As espécies, provavelmente cosmopolitas, Pleurasiga minima, Cosmoeca norvegica, C. ventricosa and Parvicorbicula circularis ocorreram na primavera ou no verão no canal principal do estuário e na área costeira, enquanto Stephanoeca diplocostata, que aparentemente prefere baixas temperaturas, foi registrada no inverno. Calotheca alata e Campyloacantha spinifera são principalmente de ambientes temperados e estiveram presentes na primavera. Uma espécie nova, Diplotheca tricyclica, foi registrada na área do canal principal do estuário, no verão de 1999, em águas de alta salinidade.

Screening sorológico e avaliação de fatores de risco para a transmissão de Toxoplasma gondii foi conduzido em 2.126 gestantes no sul do Brasil. Entre as gestantes avaliadas, 74,5% (n=1.583) mostraram anticorpos específicos contra Toxoplasma gondii. O contato com o solo foi o maior fator associado à infecção.
